The present work aims to establish of Visual Interpretation Criterias of the land-use and land-cover (LULC) classes of the Brazilian biomes. The process relies on the efforts of experts from each biome, Ph.D. and Master's students, and undergraduate students in research. Due to the particularities, the criterias were elaborated individually for each biome. The classes correspond to MapBiomas collection 04 legend. In each LULC class, the user has the following information: class definition, patterns (e.g., color, texture, roughness), and historical Landsat images (RGB 564) from the dry and rainy periods, as well as high-resolution images and field photos of the class. These visual interpretation criterias was used to generate data of samples for MapBiomas mapping validation. With the help of Visual Interpretation Criterias, experienced and inexperienced interpreters were able to produce high-quality sample data without visual inspection. This initiative, a pioneer in Brazil, is a tool to support future interpretations of Brazilian biomes. The results can be found on Lapig website.

Unlike the central nervous system, peripheral nerves can regenerate after injury. However, depending on the size of the lesion, the endogenous regenerative potential is not enough to replace the lost nerve tissue. Many strategies have been used to generate biomaterials capable of restoring nerve functions. Here, we set out to investigate whether adsorbing the extracellular matrix protein, laminin (LM), to poly-ℇ-caprolactone (PCL) filaments would enhance functional nerve regeneration. Initial in vitro studies showed that explants of dorsal root ganglia (DRGs) of P1 neonate mice exhibited stronger neuritogenesis on a substrate of LM that had been previously polymerized (polylaminin [polyLM]) than on ordinary LM. On the other hand, when silicone tubes filled with PCL filaments were used to bridge a 10-mm sciatic nerve gap in rats, only filaments coated with LM improved tissue replacement beyond that obtained with empty tubes. Motor function recovery correlated with tissue replacement as only LM-coated filaments consistently improved motor skills. Finally, analysis of the lateral gastrocnemius muscle revealed that the LM group presented twice the amount of α-bungarotixin–labeled motor plates. In conclusion, although polyLM was more effective in stimulating growth of sensory fibers out of DRGs in vitro, LM adsorbed to PCL filaments exhibited the best regenerative properties in inducing functional motor recovery after peripheral injury in vivo. Este trabalho, realizado no âmbito da disciplina de Seminário de Projecto da Licenciatura em Engenharia das Indústrias Agro-Alimentares, consistiu na elaboração de um projecto para a instalação de uma Adega e de um Centro de Aproveitamento de Subprodutos da Vinificação. Foram analisadas várias questões relacionadas com a instalação dos edifícios, características das estruturas, aquisição e instalação dos equipamentos, circuitos de produção, balanços de materiais, entre outros, culminando com uma breve análise económica. Dos resultados obtidos foi possível concluir que ambas as unidades têm viabilidade económica.

The Primeiras and Segundas Archipelago Reserve is a recently established marine protected area, the largest in Africa, located in the waters of Northern Mozambique. This protected area is of significant local economic importance and global ecological relevance, containing the southernmost coral reefs in Eastern Africa. However, information related to the marine ecosystem, notably benthic habitat is very scarce. Twelve atolls were mapped in the region using object-based image classification of very-high resolution satellite imagery (IKONOS, Quickbird, and WorldView-2). Geographically referenced data on benthic cover and depth were gathered in the course of three fieldwork expeditions covering a total of four atolls and two shallow reef structures in the Segundas Archipelago. The resulting map allows the estimation of three distinct types of coral cover (field, patches, spurs and grooves); the differentiation of sand, rubble and rock substrate; and the detection of seagrass and brown macroalgae, identifying up to 24 benthic habitats. Average overall accuracy was above 50%. The high variability of the optical properties on the reef systems, in large due to the connectivity with the mainland via plumes, while interesting from an ecological perspective increases the challenges for remote sensing of bottom cover. New information indicates the presence of deep benthic cover extending from the atolls, suggesting the need for further research on Coastal Eastern African corals, namely on their resilience and connectivity, and supporting current knowledge of the existence of an almost continuous coral reef from Kenya to Mozambique. Coral and fish biodiversity data have been analysed together with the satellite-derived maps. Results support the local perception that ecosystems are in decline and uncover new information about biodiversity’s spatial patterns. Our work provides a detailed depiction of marine habitats that may aid the management of the protected area, namely in the definition of fishing zones and coral cover monitoring.

About one-third of major depressive disorder (MDD) patients demonstrate unresponsiveness to classic antidepressants, and even the clinical efficacy of fast-acting drugs such as ketamine varies significantly among patients with treatment-resistant depression. Nevertheless, the lack of suitable animal models that mimic a possible ketamine-resistant phenotype challenges the understanding of resistance to drug treatment. In this study, we showed that PI3Kγ knock-out (KO) mice do not respond to classical doses of ketamine and classical antidepressants. PI3Kγ KO mice were unresponsive to both the rapid and sustained antidepressant-like effects of a single dose of ketamine in the forced swimming test. Additionally, they were unresponsive to the antidepressant-like effects induced by the tricyclic antidepressant imipramine and the selective serotonin reuptake inhibitor fluoxetine. However, acute pharmacological inhibition of PI3Kγ did not block the antidepressant-like effect of ketamine, showing that a chronic deficiency of the PI3Kγ-mediated pathway is necessary for the effects of classic doses of ketamine and antidepressants. Therefore, we propose that PI3Kγ participates in the antidepressant activity and is likely implicated in the neurobiology and phenotype observed in patients with MDD who demonstrate treatment resistance.

Introduction: PTCL accounts for 10-15% of all NHL. As previously demonstrated, Latin America has its own epidemiological distribution, with a high frequency of ATLL and ENKT, likely influenced by distinct genetic profiles and viral epidemiology. 3-year OS is about 40%. Treatment advances have also been limited, except for BV-CHP in some countries. The IPI, which includes extranodal (EN) site as a variable, has been validated for PTCL. However, the specific impact of EN involvement on nodal PTCL (such as PTCL-NOS, AIT, ALCL ALK+/ALK-) and its biological implications remain unclear. Simplification could improve the reproducibility and applicability of these models, especially in low-income countries. Objective: To evaluate number of EN sites in nodal PTCL lymphomas as a risk factors or surrogate for outcomes, as OS and PFS in Latin America cohort. Methodology: Patients (pts) aged ≥18 years with newly diagnosed nodal PTCL-NOS, AITL and ALCL ALK+/ALK-) from GELL (n = 339, 2000-2023, retrospective), TCBP (n = 427, 2015-2022, ambispective). Treatment outcome was determined by OS and PFS. REDcap Platform (by Vanderbilt) was used to collect and store data, whereas statistical analysis the IBM-SPSS v.24. This trial is registered at Clinical trials (NCT03207789). Results: 766 pts [427pts - TCBP and 339 - GELL] diagnosed with nodal PTCL were grouped according to the number of EN: no EN involvement (No EN - 383); one EN involvement (EN1 -168); and 32 (EN2 - 215). Considering all, 61% male; median age 56 y/o; 74% were staged III/IV; 69% IPI 32; 60% was PTCL-NOS, 19% ALCL ALK- and 12% AITL. 61% had B symptoms and 55% elevated LDH. CHOEP was used in 47% and 34% CHOP, and 47% achieved CR after first line; 16% used transplant as consolidation. Resumo: A Paracoccidioidomicose é uma micose sistémica endémica nas áreas rurais da América Latina, uma fonte importante de imigrantes e destino de emigração e turismo europeu, a maioria dos casos ocorrendo no Brasil, Argentina, Venezuela e Colômbia. Os autores descrevem o caso clínico de um paciente com 43 anos, anteriormente emigrado na Venezuela e residente em Portugal há 8 anos, que se apresenta com lesão cutânea isolada. Embora sem queixas relevantes do foro respiratório, apresentava extensas lesões do parênquima pulmonar caracterizadas por tomografia computorizada de alta resolução (TCAR). Foi realizada biópsia da lesão cutânea e exame micológico da expetoração que revelaram infeção por Paracoccidioides brasiliensis (PB). Abstract: Paracoccidioidomycosis is a systemic mycosis which is endemic in rural areas of Latin America, an important European source of immigrants and a growing European touristic destination as well, with most cases occurring in Brazil, Argentina, Venezuela and Colombia. The authors report a case of a 43 year old man who previously worked in Venezuela and is living in Portugal for 8 years, presenting with a single cutaneous lesion. Despite the absence of valuable respiratory complaints, severe lung damage was found with high-resolution computed tomography (HRCT). Biopsy of the cutaneous lesion and mycologic sputum examination were performed revealing Paracoccidioides brasiliensis infection. Annelid biodiversity studies in the Red Sea are limited and integrative taxonomy is needed to accurately improve reference libraries in the region. As part of the bioblitz effort in Saudi Arabia to assess the invertebrate biodiversity in the northern Red Sea and Gulf of Aqaba, Perinereis specimens from intertidal marine and lagoon-like rocky environments were selected for an independent assessment, given the known taxonomic ambiguities in this genus. This study used an integrative approach, combining molecular with morphological and geographic data. Our results demonstrate that specimens found mainly in the Gulf of Aqaba are not only morphologically different from other five similar Perinereis Group I species reported in the region, but phylogenetic analysis using available COI sequences from GenBank revealed different molecular operational taxonomic units, suggesting an undescribed species, P. kaustiana sp. nov. The new species is genetically close and shares a similar paragnath pattern to the Indo-Pacific distributed P. helleri, in particular in Area III and Areas VII–VIII. Therefore, we suggest it may belong to the same species complex. However, P. kaustiana sp. nov. differs from the latter mainly in the shorter length of the postero-dorsal tentacular cirri, median parapodia with much longer dorsal Tentacular cirri, posteriormost parapodia with much wider and greatly expanded dorsal ligules. Additionally, two new records are reported for the Saudi Neom area belonging to P. damietta and P. suezensis, previously described only for the Egyptian coast (Suez Canal) and are distributed sympatrically with the new species, but apparently not sympatric with each other.

Abstract The increasing need for sustainable alternatives to synthetic insecticides has driven the analysis of extracts from Solanum habrochaites, a wild tomato, through fractionated column chromatography. Potential bioactive compounds for pest management, a clean and promising biotechnological solution, have been reported from this plant. The objective is to provide detailed gas chromatography data, including peaks, structural formulas, and retention indices for the extracts of S. habrochaites aerial parts. Column chromatographic analysis was conducted with five fractions (F1, F2, F5, F3, and F4) of S. habrochaites extracts. Long-chain hydrocarbons such as hexadecanoic acid and docosano were identified in the F1 fraction; fatty acid esters, including hexadecanoate and octadecenoate ethyls in the F2 and methyl ketones, with tridecan-2-one as the major component in the F5, while no identifiable compounds were disclosed in the F3 and F4 fractions. The column chromatography provided valuable insights into compounds in the F1, F2, and F5 fractions of S. habrochaites extracts, highlighting fatty acid esters, long-chain hydrocarbons, and methyl ketones. The bioactive compounds, from extracts of this plant, including the first record of the docosanoate, hexadecanoate and octadecanoate ethyls in S. habrochaites and Solanaceae, reinforces their promising biological application in different areas of science.

Abstract Background Physiology is widely recognized as a difficult course, which can potentially increase students’ withdrawal and failures rates. Several factors are likely contributing to the difficulties in learning physiology, including inherent features of the discipline as well as aspects related to instructions and/or students’ perception. With regards to the later, it is currently unknown how students of exercise physiology think and explain physiology in terms of its cause or consequence (i.e., teleological or mechanistic thinking). Therefore, the aims of the present study were to determine 1) whether undergraduate students’ perception of cardiorespiratory physiology during exercise follows a predominant teleological or mechanistic thinking, and 2) whether prior enrollment in physiology courses can influence the predominance of teleological vs. mechanistic thinking. Methods The test instrument was an online questionnaire about exercise physiology consisting of nine incomplete sentences about exercise physiology where students had to choose between a teleological or a mechanistic complement. The questionnaire was administered to undergraduate students in the following areas: 1) Movement Sciences (n = 152), 2) Health-related (n = 81) and, 3) Health-unrelated programs (n = 64). Students in Movement Sciences and Health-related programs were also analyzed separately in the following categories: 1) students who previously undertook physiology courses, and 2) students who did not take physiology courses. Results Overall, all groups presented a percentage of teleological thinking above 58%, which is considerably high. Abstract Objective This study aimed at investigating a set of peripheral cytokines in elderly female patients with MDD, comparing them to controls, and assessing the potential influence of clinical comorbidities on inflammatory markers. Methods Twenty-five elderly female patients diagnosed with MDD and 19 age-matched female controls were enrolled on this study. Plasma levels of interleukin (IL)-4, IL-6, IL-10, interferon (IFN)-γ and tumor necrosis factor (TNF)-α were evaluated with commercially-available assays. Results Elderly female patients with MDD exhibited higher plasma IL-6 and IL-4 levels when compared to controls. In a logistic regression model taking cytokine levels, comorbidities, and age into account, only type 2 diabetes mellitus (DM2) remained associated with MDD. Conclusion Diabetes influences the association between MDD and higher levels of cytokines in elderly female patients. Future studies should take this evidence into account in order to mitigate confounding factors.

Introduction. Since the advent and development of transcatheter aortic valve replacement (TAVR) in the contemporary era, balloon aortic valvuloplasty (BAV) has seen renewed interest. We aimed to compare 30-day and 1-year all-cause mortality between patients submitted to BAV as a bridging therapy before definite TAVR and patients submitted directly to TAVR. Methods. This was an observational, retrospective study of patients who underwent TAVR between 2009 and 2022 in a tertiary center. Patients with severe aortic stenosis (SAS) who underwent TAVR without prior BAV (woBAV group) and patients who were performed TAVR with prior BAV (wBAV group) as a bridging therapy were included. Primary endpoint was all-cause mortality at 30 days and 1 year after TAVR between wBAV and woBAV groups. Results. 800 patients were included, of which 767 were in woBAV group and 33 were in wBAV group. 30-day all-cause mortality rate was 21% in wBAV group compared to 4.4% in woBAV (unadjusted hazard ratio [HR], 5.19; 95% confidence interval [CI], 2.3–11.7, p

Anhedonia is clinically defined as difficulty or inability to feel pleasure or to be motivated to perform activities that were previously pleasurable. Anhedonia is a core feature of depressive disorders but can be present in other conditions such as substance use and anxiety disorders. Herein we report the case of a 34-year-old female who developed marked anhedonia after left cortico-amygdalohippocampectomy. Despite optimal seizure control, the person struggled with anhedonia and other depressive symptoms. After ruling out medico-neurologic complications, she was prescribed with a selective serotonin reuptake inhibitor and cognitive-behavioral therapy. Anhedonia can be a challenging neuropsychiatric presentation that requires ruling out the effects of antiseizure medications, neurosurgery, and other drugs before prescribing antidepressants.

TNF-α is essential for induction and maintenance of inflammatory responses and its dysregulation is associated with susceptibility to various pathogens that infect the central nervous system. Activation of both microglia and astrocytes leads to TNF-α production, which in turn triggers further activation of these cells. Astrocytes have been implicated in the pathophysiology of a wide range of neurodegenerative diseases with either harmful or protective roles, as these cells are capable of secreting several inflammatory factors and also promote synapse elimination and remodeling. These responses are possible because they sense their surroundings via several receptors, including the metabotropic glutamate receptor 5 (mGluR5). Under neuroinflammatory conditions, mGluR5 activation in astrocytes can be neuroprotective or have the opposite effect. In the current study, we investigated the role of mGluR5 in hiPSC-derived astrocytes subjected to pro-inflammatory stimulation by recombinant TNF-α (rTNF-α). Our results show that mGluR5 blockade by CTEP decreases the secreted levels of pro-inflammatory cytokines (IL-6 and IL-8) following short rTNF-α stimulation, although this effect subsides with time. Additionally, CTEP enhances synaptoneurosome phagocytosis by astrocytes in both non-stimulated and rTNF-α-stimulated conditions, indicating that mGluR5 blockade alone is enough to drive synaptic material engulfment. Finally, mGluR5 antagonism as well as rTNF-α stimulation augment the expression of the reactivity marker SERPINA3 and reduces the expression of synaptogenic molecules. Altogether, these data suggest a complex role for mGluR5 in human astrocytes, since its blockade may have beneficial and detrimental effects under inflammatory conditions.

Abstract Background Apathy is among the most common behavioral symptoms in dementia and is consistently associated with negative outcomes in Alzheimer’s disease (AD). Despite its prevalence and clinical relevance, available pharmacological and non-pharmacological strategies to treat apathy in AD have been marked, respectively, by potentially severe side effects and/or limited efficacy. Transcranial direct current stimulation (tDCS) is a relatively novel non-pharmacological method of neuromodulation with promising results. Compared to previous tDCS formats, recent technological advances have increased the portability of tDCS, which creates the potential for caregiver-administered, home use. Our study aims to evaluate the feasibility, safety, and efficacy of home-based tDCS for the treatment of apathy in AD. Methods/design This is an experimenter- and participant-blinded, randomized, sham-controlled, parallel-group (1:1 for two groups) pilot clinical trial, involving 40 subjects with AD. After a brief training, caregivers will administer tDCS for participants at home under remote televideo supervision by research staff to ensure the use of proper technique. Participants will be assessed at baseline, during treatment (week 2, week 4, and week 6), and 6 weeks post-treatment. Dependent measures will cover cognitive performance, apathy, and other behavioral symptoms. Data about side effects and acceptability will also be collected. Discussion Our study will address apathy, an overlooked clinical problem in AD. Our findings will advance the field of non-pharmacological strategies for neuropsychiatric symptoms, presenting a great potential for clinical translation. Abstract Objectives BDNF has been implicated in the pathophysiology of systemic lupus erythematosus (SLE), especially its neuropsychiatric symptoms. The purpose of this study was to investigate the profile of blood BDNF levels in patients with SLE. Methods We searched PubMed, EMBASE, and the Cochrane Library for papers that compared BDNF levels in SLE patients and healthy controls (HCs). The Newcastle–Ottawa scale was used to assess the quality of the included publications, and statistical analyses were carried out using R 4.0.4. Results The final analysis included eight studies totaling 323 healthy controls and 658 SLE patients. Meta-analysis did not show statistically significant differences in blood BDNF concentrations in SLE patients compared to HCs (SMD 0.08, 95% CI [ − 1.15; 1.32], P value = 0.89). After removing outliers, there was no significant change in the results: SMD -0.3868 (95% CI [ − 1.17; 0.39], P value = 0.33. Univariate meta-regression analysis revealed that sample size, number of males, NOS score, and mean age of the SLE participants accounted for the heterogeneity of the studies (R2 were 26.89%, 16.53%, 18.8%, and 49.96%, respectively). Conclusion In conclusion, our meta-analysis found no significant association between blood BDNF levels and SLE. The potential role and relevance of BDNF in SLE need to be further examined in higher quality studies.

Biomedical engineering has been constantly looking for the development of new materials that have bioactive surfaces and that are responsible for new bone formation, induce osteoblast differentiation and present antibacterial properties. Here, we present innovative and applied research to evaluate the influence of niobium pentoxide thin films (Nb2O5) deposited on the surface of a Ti-6Al-4V alloy via the reactive sputtering technique on antibacterial activity, on the cellular proliferation of MC3T3 cells and on biomineralization capacity. Results demonstrated the reactive sputtering technique improved the cell viability, the osteogenic performance of cells involved in the osseointegration process, the ability to delay bacterial proliferation within the first six hours of contact with S. aureus as well as the capacity to promote rapid amorphous apatite formation in vitro. These findings confirm the great potential of the Ti-6Al-4V alloy functionalized with Nb2O5 for future applications in implantable devices.

Background: Venous Thromboembolism (VTE) is a common cause of death and complications in patients with cancer. Choice of anticoagulant is challenging because of high risk of bleeding and recurrent VTE. Some tumor oncogenes, such as KRAS, a GTPase signaling protein that regulates proliferation, differentiation and cell survival can modulate the expression of genes at the core of hemostasis control. Clinically, KRAS mutation status is a validated predictive biomarker that is currently used to determine those mCRC (metastatic colorectal cancer) patients likely not to benefit from anti-EGFR therapy. A retrospective cohort study with 172 patients with mCRC has suggested that KRAS mutation (codons 12/13) could increase the risk of VTE. Aim: To analyze the risk VTE of patients with mCRC based on KRAS status. Methods: Records of 111 patients with metastatic or unresectable locally advanced colorectal cancer, attended at Clinica AMO (Salvador, Brazil) between January 2015 and January 2020, tested for mutational status of KRAS were reviewed. Seven mutations within codons 12 e 13 of KRAS detected by PCR (Idylla technique) or were considered for this study. Non-metastatic cases, those with insufficient clinical data, VTE provoked by catheter use and superficial vein thrombosis were excluded. This study was approved by an institutional Ethics Committee. Odds Ratios and 95% Confidence Intervals were obtained from the regression coefficients. A value of p < 0.05 was statistically significant. Results and discussion: One-hundred eleven patients were selected; the median age was 64 (22‒79) and 44.1% were male. Sixty-three patients (56.7%) had a KRAS activating mutation and 48 (43.2%) were found to be KRAS wild-type (Fig. 1), the incidence being comparable to the literature (approximately 30%–50%) (6). Twenty-three (20,7%) patients suffered at least 1 episode of VTE, 5 (4,5%) Pulmonary Embolism (PE), 15 (13.5%) low-extremity Deep-Vein Thrombosis (DVT) and 3 (2.7%) presented both PE and DVT. All six patients with thrombosis in catheter had PE or DVT and there was no need to be excluded. KRAS mutated status was not associated with an elevated risk of thrombosis (15.8% vs. 27%, OR = 0.50, 95% CI 0.20‒1.28) as well for the outcome of PE, DVT and both PE and DVT (Table 1). Although Ades, et al. (2015) found mutated KRAS as risk factor for VTE and remained significant despite adjustment for Khorana score and bevacizumab treatment, data are insufficient and conflicting. A Cohort with 194 patients, 41 patients (21.1%) who experienced VTE, ECOG ≥2 was the only independent predictor. Moreover, an abstract of 184 patients with mCRC shown limited evidence that mCRC patients harboring KRAS mutations may be at greater risk for VTE. In Table 2, we described VTE patient's characteristics according to oncogenic status. More than a half were metastatic and all patients had performance status ECOG 0‒1 at diagnosis. Personal characteristics and treatment may interfere in VTE outcome and may be considered in further investigations to analyze if KRAS has impact on patient care and associated morbidity. Conclusion: In our study, biomarker KRAS have not been associated with an increased risk of VTE in advanced stage CRC patients and there is no evidence to select patients to whom VTE thromboprophylaxis should be offered.

Background: and purpose: Fatigue is among the most common persistent symptoms following post-acute sequelae of Sars-COV-2 infection (PASC). The current study investigated the potential therapeutic effects of High-Definition transcranial Direct Current Stimulation (HD-tDCS) associated with rehabilitation program for the management of PASC-related fatigue. Methods: Seventy patients with PASC-related fatigue were randomized to receive 3 mA or sham HD-tDCS targeting the left primary motor cortex (M1) for 30 min paired with a rehabilitation program. Each patient underwent 10 sessions (2 sessions/week) over five weeks. Fatigue was measured as the primary outcome before and after the intervention using the Modified Fatigue Impact Scale (MFIS). Pain level, anxiety severity and quality of life were secondary outcomes assessed, respectively, through the McGill Questionnaire, Hamilton Anxiety Rating Scale (HAM-A) and WHOQOL. Results: Active HD-tDCS resulted in significantly greater reduction in fatigue compared to sham HD-tDCS (mean group MFIS reduction of 22.11 points vs 10.34 points). Objectives The main goal of this study was to characterise the frequency and phenotype of B, T follicular helper (Tfh) and T follicular regulatory (Tfr) cells in peripheral blood and the cytokine environment present in circulation in children with extended oligoarticular juvenile idiopathic arthritis (extended oligo JIA) and polyarticular JIA (poly JIA) when compared with healthy controls, children with persistent oligoarticular JIA (persistent oligo JIA) and adult JIA patients.Methods Blood samples were collected from 105 JIA patients (children and adults) and 50 age-matched healthy individuals. The frequency and phenotype of B, Tfh and Tfr cells were evaluated by flow cytometry. Serum levels of APRIL, BAFF, IL-1β, IL-2, IL-4, IL-6, IL-10, IL-17A, IL-21, IL-22, IFN-γ, PD-1, PD-L1, sCD40L, CXCL13 and TNF were measured by multiplex bead-based immunoassay and/or ELISA in all groups included.Results The frequency of B, Tfh and Tfr cells was similar between JIA patients and controls. Children with extended oligo JIA and poly JIA, but not persistent oligo JIA, had significantly lower frequencies of plasmablasts, regulatory T cells and higher levels of Th17-like Tfh cells in circulation when compared with controls. Furthermore, APRIL, BAFF, IL-6 and IL-17A serum levels were significantly higher in paediatric extended oligo JIA and poly JIA patients when compared with controls. These immunological alterations were not found in adult JIA patients in comparison to controls.Conclusions Our results suggest a potential role and/or activation profile of B and Th17-like Tfh cells in the pathogenesis of extended oligo JIA and poly JIA, but not persistent oligo JIA.

Abstract Management programs and efficient techniques are necessary to recover degraded ecosystems. The sewage sludge is rich in nitrogen (N) and with the potential to fertilize Sapindus saponaria L. (Sapinales: Sapindaceae), used in the recovery of degraded areas; this can affect the insect fauna. The study's objective was to evaluate, for 24 months, the abundance of chewing insects, dipterans, pollinators, and predators on S. saponaria plants fertilized with or without dehydrated sewage sludge in a degraded area. The experimental design was completely randomized (with the same characteristics) with two treatments (with or without dehydrated sewage sludge) and 24 replicates, each with one plant. The abundance of Anastrepha sp. (Tephritidae), Cerotoma sp. (Chrysomelidae), Curculionidae, Musca domestica L. (Muscidae), Mantis religiosa L. (Mantodea: Mantidae), Oxyopidae, Salticidae, Tettigoniidae (Orthoptera), and Teudis sp. (Anyphaenidae) was higher on fertilized plants. The abundances of Teudis sp. and Tmarus sp. (Thomisidae) and M. religiosa and Teudis sp. were positively correlated with chewing insects and Diptera, respectively. The population increase of insects and spiders on S. saponaria plants fertilized with dehydrated sewage sludge (bigger crowns) has shown to be suitable for recovering degraded areas with a higher number of niches and better food quality, improving the ecological indices of the area.

Propellant mass gauging under micro-gravity conditions is a challenging task due to the unpredictable position and shape of the fuel body inside the tank. Micro-gravity conditions are common for orbiting satellites and rockets that operate on limited fuel supplies. Capacitance sensors have been investigated for this task in recent years; however, the effect of various positions and shapes of the fuel body is not analyzed in detail. In this paper, we investigate this with various fill types, such as annular, core-annular, and stratified fills at different positions. We compare the performance among several curve-fitting-based approaches and a machine-learning-based approach, the latter of which offers superior performance in estimating the fuel content.

Electrical Capacitance Volume Tomography (ECVT) is a low-cost and high-speed sensing modality with great potential for industrial multiphase flow monitoring. In this work, we examine the use of the slope of the capacitance vector residual curve, as directly provided by the measurement data, to formulate a robust stopping criterion for the iterative image reconstruction in ECVT. The methodology is illustrated based on experimental data from a gas-solid fluidized bed. We show that the proposed stopping criterion can improve the performance of both the image reconstruction and the flow velocity profiling in ECVT applications. For concreteness, we focus on the popular Landweber iterative reconstruction algorithm although other reconstruction algorithms exhibiting semi-convergent behavior might benefit from the present analysis as well.

Background Long non-coding RNAs (lncRNAs) have been reported to be involved in the pathogenesis of neurodegenerative diseases. It has also been hypothesized that plasma exosomal lncRNAs may be used as Alzheimer’s disease (AD) biomarkers. In this systematic review, we compiled all studies on the subject to evaluate the accuracy of lncRNAs in identifying AD cases through meta-analysis. Methods A PRISMA-compliant systematic search was conducted in PubMed/MEDLINE, EMBASE, and Web of Science databases for English publications till September 2022. We included all observational studies published which investigated the sensitivity and specificity of various lncRNAs in plasma samples of AD diagnosis. Our search strategy included lncRNA and all the related spelling and abbreviation variations combined with the keyword Alzheimer’s disease. Methodological quality was assessed using the Strengthening the Reporting of Observational Studies in Epidemiology (STROBE) guidelines and the Quality Assessment of Diagnostic Accuracy Studies (QUADAS-II) tool. The meta-analysis was carried out using the area under the Receiver Operator Characteristic (ROC) curves (AUC) and sensitivity and specificity values to assess the accuracy of the identified lncRNAs in AD diagnosis. To account for the predicted heterogeneity of the study, a random-effects model was used. All the statistical analyses and visualizations were conducted using Stata 17.0 software. Results A total of seven studies (AD patients = 553, healthy controls = 513) were included in the meta-analysis. Three lncRNAs were upregulated (RNA BACE-AS1, RNA NEAT1, RNA GAS5), and one lncRNA (MALAT1) was downregulated in plasma samples of AD patients. RNA 51A and RNA BC200 were reported to have variable expression patterns. A lncRNA (RNA 17A) was not significantly different between AD and control groups. The pooled sensitivity, specificity, and AUC values of lncRNAs in identifying AD were (0.74; 95% CI [0.63, 0.82], I2 = 79.2%), (0.88; 95% CI [0.75, 0.94], I2 = 88.9%), and 0.86; 95% CI [0.82, 0.88], respectively. In addition, the pooled diagnostic odds ratio (DOR) of the five individual lncRNAs in AD diagnosis was 20. Conclusion lncRNAs had high accuracy in identifying AD and must be seen as a promising diagnostic biomarker of the disease.

Abstract Sapindus saponaria L. (Sapindaceae) is a pioneer species used in programs to recover degraded areas. The trees also assist in the pest control of some insects due to the composition of saponins on their leaves. In addition, these chemical components are important to pharmaceutical product production. The objective was to evaluate the impact of spatial distribution, indices and ecological relationship of arthropods on S. saponaria leaves to preserve the balance of biodiversity. Aggregated distribution of arthropods was observed; the numbers of phytophagous arthropods were higher on the adaxial leaf face than on the abaxial part. Only Aleyrodidae (Hemiptera) had a higher presence on the abaxial leaf face of S. saponaria saplings. Abundance, diversity, and species richness of natural enemies correlated positively with phytophagous and pollinators insects. On the other hand, the number of Lyriomyza sp. mines correlated negatively with Pseudomyrmex termitarius (Smith) (Hymenoptera: Formicidae). All this information can assist and guide integrated pest management programs.

IntroductionTo understand mechanisms and identify potential targets for intervention in the current crisis of opioid use disorder (OUD), postmortem brains represent an under-utilized resource. To refine previously reported gene signatures of neurobiological alterations in OUD from the dorsolateral prefrontal cortex (Brodmann Area 9, BA9), we explored the role of microRNAs (miRNA) as powerful epigenetic regulators of gene function.MethodsBuilding on the growing appreciation that miRNAs can cross the blood-brain barrier, we carried out miRNA profiling in same-subject postmortem samples from BA9 and blood tissues.ResultsmiRNA–mRNA network analysis showed that even though miRNAs identified in BA9 and blood were fairly distinct, their target genes and corresponding enriched pathways overlapped strongly. Among the dominant enriched biological processes were tissue development and morphogenesis, and MAPK signaling pathways. These findings point to robust, redundant, and systemic opioid-induced miRNA dysregulation with a potential functional impact on transcriptomic changes. Further, using correlation network analysis, we identified cell-type specific miRNA targets, specifically in astrocytes, neurons, and endothelial cells, associated with OUD transcriptomic dysregulation. Finally, leveraging a collection of control brain transcriptomes from the Genotype-Tissue Expression (GTEx) project, we identified a correlation of OUD miRNA targets with TGF beta, hypoxia, angiogenesis, coagulation, immune system, and inflammatory pathways.DiscussionThese findings support previous reports of neurovascular and immune system alterations as a consequence of opioid abuse and shed new light on miRNA network regulators of cellular response to opioid drugs.

Introduction The combination of a CDK4/6 inhibitor with an aromatase inhibitor (AI) has recently become the gold standard for AI-sensitive first line treatment of oestrogen receptor-positive (ER+) HER2-negative (HER2−) advanced breast cancer. However, most patients receiving this combination will ultimately progress and require further therapies.Several studies have demonstrated that the onset of a ESR1 gene mutation lead to AIs resistance in the advanced setting. ESR1 mutations can be detected in circulating tumour DNA (ctDNA) using a digital PCR assay. Our study aims to prove the clinical efficacy of periodic monitoring for emerging or rise of ESR1 mutations in ctDNA to trigger an early change from AI plus palbociclib to fulvestrant plus palbociclib treatment while assessing global safety.Methods PADA-1 is a randomised, open-label, multicentric, phase III trial conducted in patients receiving AI and palbociclib as first line therapy for metastatic ER +HER2- breast cancer. 1000 patients will be included and treated with palbociclib in combination with an AI. Patients will be screened for circulating blood ESR1 mutation detection at regular intervals. Patients for whom a rising circulating ESR1 mutation is detected without tumour progression (up to N=200) will be randomised (1:1) between (1) Arm A: no modification of therapy; and (2) Arm B: palbociclib in combination with fulvestrant, a selective ER down-regulator. At tumour progression, an optional crossover will be offered to patients randomised in arm A. The coprimary endpoints are (1) Grade ≥3 haematological toxicities and their associations with baseline characteristics and (2) progression-free survival in randomised patients.Ethics and dissemination The study has been approved by the French medicines agency (ANSM) and by an ethics committee (ref 01/17_1 CPP Ouest-IV Nantes) in January 2017. The trial results will be published in academic conference presentations and international peer-reviewed journals.Trial registration numbers EudraCT: 2016-004360-18; NCT03079011.
