1. Chemistry in the Museum: Elucidation of 1920s Medical Kits

Abstract

This project report describes the process of a team of undergraduate researchers (Chemistry and Nursing majors), who analyzed 20th-century medical kits housed at The Columbus Museum (Columbus, GA, USA). Curators and museum personnel were unfamiliar with the contents and needed assistance in identifying the various chemical contents. Items were identified by the Taylor Lab, which was followed by fully elucidating the chemical information in a chemical report and student-curated exhibit. The intent of this project was to help the museum be aware of how to properly curate and store the medical collections for an extended period. Laboratory analyses were executed to determine the composition of the aged items in the collections. The historical context of these kits and their contents provided knowledge of medicine to the community of Columbus, Georgia, in addition to explaining the use of medically related items in the 20th century.

2. Supporting Teachers' Effective Classroom and Behavior Management: What Do Teachers Tell Us?

Abstract

Elementary teachers (N = 160) from two districts responded to survey questions about inservice professional development and administrator feedback and support for implementing research-based, effective classroom and behavior management (C/BM) strategies. Results indicated slightly more than half (55%) of teachers reported their participation in formal, inservice professional development activities improved their ability to implement research-based C/BM strategies. A higher percentage (77%) of teachers reported they receive adequate feedback from their current school supervisor (e.g., principal, curriculum/instructional coach) on their use of C/BM. Teachers' reported specific ways for school and district administrators to better prepare and support them for challenges related to C/BM. Implications are provided for improving inservice professional development and administrator feedback to promote teachers' use of effective classroom and behavior management practices.

Abstract

Background Implementation of the cardiovascular disease (CVD) prevention guidelines in the UK has been repeatedly evaluated under the auspices of the British Cardiovascular Society in three Action on Secondary and Primary Prevention by Intervention to Reduce Events (ASPIRE) surveys in 1994–1995, 2008–2010 and 2017–2019. The primary care arm of ASPIRE-2-PREVENT (A-3-P) was conducted to evaluate lifestyle and medical risk factor management in people at high risk of atherosclerotic CVD in everyday clinical practice.Methods A-3-P was a cross-sectional survey in 27 general practices and health centres across 5 English National Health Service regions. Patients with no history of atherosclerotic CVD started on blood pressure and/or lipid and/or glucose lowering treatments were identified retrospectively and interviewed at least 6 months after the initiation of medication.Results 557 patients attended the interview and examination (45.8% women; mean age 61.7±10.8 years). The risk factor control was poor: 9.3% of patients were smokers, 38.1% obese (body mass index≥30 kg/m2) and 53.5% centrally obese (waist circumference≥88 cm for women, ≥102 cm for men). Abstract

Introduction The COVID-19 pandemic has had an unprecedented ef- fect on hospital systems. Policy changes lead to de- creased hospital visits as well as surgical case volume. The literature on pediatric surgical case volume during the pandemic is sparse. Throughout the country, hospitals sought various policies to preserve personal protective equipment and other hospital resources, and to minimize avoidable peri- and postoperative sequelae due to COVID-19 infection. Our hospital first placed a hold on all elective surgeries. Later, all elective cases required a preoperative negative COVID test prior to proceed- ing. We sought to review the sequelae of our hospital’s policy in response to COVID-19. We identified trends in surgical case volume and cancellations due to a positive COVID-19 test. We also reviewed postoperative out- comes of cases with a positive test. Material and Methods This study was approved by the institutional IRB. Data was retrospectively collected on all surgical cases at our children’s hospital between March 2019 and March 2021. We marked the start of the COVID-19 pandemic as March 2020, when elective cases were suspended. A required preoperative negative COVID-19 test was im- plemented in May 2020. We identified pre-operative COVID-19 test results,the posted urgency of each case and 30-day outcomes from medical records. Results From March 2019 to March 2021, we identified 25,496 completed surgeries and 3,503 cancellations. 12,024 ca- ses proceeded during the first year of the pandemic, which appeared lower, compared to pre-pandemic case numbers. Of those, 2,785 (23%) cases were considered urgent or emergent. The average number of completed monthly cases fell from a pre-pandemic number of 1,123 to a pandemic number of 925. When comparing to a pre- pandemic month, average monthly case volume declined by 19%, with the largest decline noted to be 66%. There was a monthly average of 189 total cancellations between March 2020 and March 2021. 34 (18%) of those were for a positive preoperative COVID test. A total of 139 sur- geries commenced despite concomitant COVID-19 in- fection. 25 (18 %) had identifiable respiratory symptomsdocumented preoperatively. 13 (9 %) were deemed to have a respiratory complication afterward. Of those, three patients (2%) had a prolonged, and one (1%) had an unexpected reintubation. The remaining nine (6%) pa- tients had a prolonged oxygen requirement. Conclusion The COVID pandemic left operating rooms struggling to determine how to safely provide care to patients. This study demonstrated how the policies of one hospital af- fected the operating room case volume and how conco- mitant COVID infection affected outcomes in those that proceeded with surgery.

5. Improving nitrogen cycling in a land surface model (CLM5) to quantify soil N2O, NO, and NH3 emissions from enhanced rock weathering with croplands

Abstract

Surficial enhanced rock weathering (ERW) is a land-based carbon dioxide removal (CDR) strategy that involves applying crushed silicate rock (e.g., basalt) to agricultural soils. However, unintended biogeochemical interactions with the nitrogen cycle may arise through ERW increasing soil pH as basalt grains undergo dissolution that may reinforce, counteract, or even offset the climate benefits from carbon sequestration. Increases in soil pH could drive changes in the soil emissions of key non-CO2 greenhouse gases, e.g., nitrous oxide (N2O), and trace gases, e.g., nitric oxide (NO) and ammonia (NH3), that affect air quality and crop and human health. We present the development and implementation of a new improved nitrogen cycling scheme for the Community Land Model v5 (CLM5), the land component of the Community Earth System Model, allowing evaluation of ERW effects on soil gas emissions. We base the new parameterizations on datasets derived from soil pH responses of N2O, NO, and NH3 in ERW field trial and mesocosm experiments with crushed basalt. These new capabilities involve the direct implementation of routines within the CLM5 N cycle framework, along with asynchronous coupling of soil pH changes estimated through an ERW model. We successfully validated simulated “control” (i.e., no ERW) seasonal cycles of soil N2O, NO, and NH3 emissions against a wide range of global emission inventories. We benchmark simulated mitigation of soil N2O fluxes in response to ERW against a subset of data from ERW field trials in the US Corn Belt. Using the new scheme, we provide a specific example of the effect of large-scale ERW deployment with croplands on soil nitrogen fluxes across five key regions with high potential for CDR with ERW (North America, Brazil, Europe, India, and China). Across these regions, ERW implementation led to marked reductions in N2O and NO (both 18 %), with moderate increases in NH3 (2 %). While further developments are still required in our implementations when additional ERW data become available, our improved N cycle scheme within CLM5 has utility for investigating the potential of ERW point-source and regional effects of soil N2O, NO, and NH3 fluxes in response to current and future climates. This framework also provides the basis for assessing the implications of ERW for air quality given the role of NO in tropospheric ozone formation, as well as both NO and NH3 in inorganic aerosol formation.

Abstract

Glacier surges are prevalent in the Karakoram and occasionally threaten local residents by inundating land and initiating mass movement events. The Kyagar Glacier is well known for its surge history, and in particular its frequent blocking of the downstream valley, leading to a series of high-magnitude glacial lake outburst floods (GLOFs). Although the surge dynamics of the Kyagar Glacier have been broadly described in the literature, there remains an extensive archive of remote sensing observations that have great potential for revealing specific surge characteristics and their relationship with historic lake outburst floods. In this study, we propose a new perspective on quantifying the surging process using successive digital elevation models (DEMs), which could be applied to other sites where glacier surges are known to occur. Advanced Spaceborne Thermal Emission and Reflection Radiometer DEMs, High Mountain Asia 8-meter DEMs, and the Shuttle Radar Topography Mission DEM were used to characterize surface elevation changes throughout the period from 2000 to 2021. We also used Landsat time series imagery to quantify glacier surface velocities and associated lake changes over the course of two surge events between 1989 and 2021. Using these datasets, we reconstruct the surging process of the Kyagar Glacier in unprecedented detail and find a clear signal of surface uplift over the lower glacier tongue, along with uniformly increasing velocities, associated with the period of surge initiation. Seasonal variations in surface flow are still evident throughout the surge phase, indicating the presence of water at the glacier bed. Surge activity of the Kyagar Glacier is strongly related to the development and drainage of the terminal ice-dammed lake, which itself is controlled by the drainage system beneath the glacier terminus.

7. Evaluation of low-cost Raspberry Pi sensors for structure-from-motion reconstructions of glacier calving fronts

Abstract

Glacier calving fronts are highly dynamic environments that are becoming ubiquitous as glaciers recede and, in many cases, develop proglacial lakes. Monitoring of calving fronts is necessary to fully quantify the glacier ablation budget and to warn nearby communities of the threat of hazards, such as glacial lake outburst floods (GLOFs), tsunami waves, and iceberg collapses. Time-lapse camera arrays, with structure-from-motion photogrammetry, can produce regular 3D models of glaciers to monitor changes in the ice but are seldom incorporated into monitoring systems owing to the high cost of equipment. In this proof-of-concept study at Fjallsjökull, Iceland, we present and test a low-cost, highly adaptable camera system based on Raspberry Pi computers and compare the resulting point cloud data to a reference cloud generated using an unoccupied aerial vehicle (UAV). The mean absolute difference between the Raspberry Pi and UAV point clouds is found to be 0.301 m with a standard deviation of 0.738 m. We find that high-resolution point clouds can be robustly generated from cameras positioned up to 1.5 km from the glacier (mean absolute difference 0.341 m, standard deviation 0.742 m). Combined, these experiments suggest that for monitoring calving events in glaciers, Raspberry Pi cameras are an affordable, flexible, and practical option for future scientific research. Owing to the connectivity capabilities of Raspberry Pi computers, this opens the possibility for real-time structure-from-motion reconstructions of glacier calving fronts for deployment as an early warning system to calving-triggered GLOFs.

Abstract

Abstract There are two major challenges to improving interannual to decadal forecasts: (a) consistently initializing the coupled system so that variability is not dominated by initial imbalances, and (b) having a large sample of different initial conditions on which to test forecast skill. The second challenge requires consideration of time periods not only outside the recent period of intensive ocean observation, but also before the instrumental era, which increases the importance of the first challenge. Forecasts prior to the 1850s isolate internally generated sources of variability by removing the majority of anthropogenic forcing, and the sparse observational record during this time period motivates the use of paleoclimate proxy data. We address these issues by using a linear inverse model (LIM) approach and a recent proxy‐based reconstruction over the last millennium at annual resolution. The reconstruction is used to train, initialize, and validate LIM forecasts. The LIM trained on paleo‐data assimilated using a LIM trained on global climate model (GCM) simulation data outperforms a LIM trained on raw GCM data at forecast leads longer than 2 years for in‐sample experiments, and beyond 4‐year leads in most out‐of‐sample experiments validated on instrumental data. The most skillful normal mode of the paleo‐data LIM for the instrumental experiment represents a persistent pattern with a longer decay time than for the GCM‐LIM's modes, which accounts for the outperformance at longer leads. The paleo‐data LIM is consequently more sensitive to ocean initialization, which is reflected in forecasts during the instrumental era where ocean reanalyses exhibit large uncertainty.

11. Increased carbon capture by a silicate-treated forested watershed affected by acid deposition

Abstract

Meeting internationally agreed-upon climate targets requires carbon dioxide removal (CDR) strategies coupled with an urgent phase-down of fossil fuel emissions. However, the efficacy and wider impacts of CDR are poorly understood. Enhanced rock weathering (ERW) is a land-based CDR strategy requiring large-scale field trials. Here we show that a low 3.44 t ha−1 wollastonite treatment in an 11.8 ha acid-rain-impacted forested watershed in New Hampshire, USA, led to cumulative carbon capture by carbonic acid weathering of 0.025–0.13 t CO2 ha−1 over 15 years. Despite a 0.8–2.4 t CO2 ha−1 logistical carbon penalty from mining, grinding, transportation, and spreading, by 2015 weathering together with increased forest productivity led to net CDR of 8.5–11.5 t CO2 ha−1. Our results demonstrate that ERW may be an effective, scalable CDR strategy for acid-impacted forests but at large scales requires sustainable sources of silicate rock dust.

Abstract

Rationale & Objective: Older adults with end-stage kidney disease have increased morbidity, fatigue, and decreased physical function, which can inhibit self-care and social engagement. We pilot tested a home-based program to improve physical and social functioning of low socioeconomic status older adults treated with hemodialysis (HD). Study Design: Qualitative study and randomized waitlist control intervention. Setting & Participants: Older adult HD patients in Baltimore, MD. Interventions: We identified functional needs and home environmental barriers to social engagement through focus groups; mapped findings onto aspects of an established program, which includes home visits with an occupational therapist, nurse, and handyman to provide ≤$1,300 worth of repairs, modifications, and devices; and piloted the program (Seniors Optimizing Community Integration to Advance Better Living with ESRD [SOCIABLE]) among 12 older adult HD patients. We delivered the services over 5 months in a staggered fashion. Outcomes: Feasibility and acceptability of the intervention and change in disability scores. Results: Focus group themes included fatigue, lack of social support, and desire to live independently. SOCIABLE pilot participants were recruited from 2 dialysis units and all were African American (50% men); mean age was 69 years. At baseline, the mean disability score for activities of daily living (ADLs) was 4.4 and for instrumental ADLs (IADLs) was 6.3 (both out of a possible 16). Among the 9 participants alive at follow-up, there was 100% intervention completion and outcomes assessment. All treated participants improved a mean score of 2.3 for ADL and 2.6 for IADL disability, and social support and social network scores improved by 4.8 and 4.6, respectively. Limitations: Small sample size; all participants were African American. Conclusions: A home-based intervention addressing physical and social functioning of low socioeconomic status older adults on HD therapy was feasible and acceptable. Abstract

Background: Previous research showed that weight-reducing diets increase appetite sensations and/or circulating ghrelin concentrations for up to 36 months, with transient or enduring perturbations in circulating concentrations of the satiety hormone peptide YY.Objective: This study assessed whether a diet that is higher in protein and low in glycemic index (GI) may attenuate these changes.Methods: 136 adults with pre-diabetes and a body mass index of ≥25 kg/m2 underwent a 2-month weight-reducing total meal replacement diet. Participants who lost ≥8% body weight were randomized to one of two 34-month weight-maintenance diets: a higher-protein and moderate-carbohydrate (CHO) diet with low GI, or a moderate-protein and higher-CHO diet with moderate GI. Both arms involved recommendations to increase physical activity. Fasting plasma concentrations of total ghrelin and total peptide YY, and appetite sensations, were measured at 0 months (pre-weight loss), at 2 months (immediately post-weight loss), and at 6, 12, 24, and 36 months.Results: There was a decrease in plasma peptide YY concentrations and an increase in ghrelin after the 2-month weight-reducing diet, and these values approached pre-weight-loss values by 6 and 24 months, respectively (P = 0.32 and P = 0.08, respectively, vs. 0 months). However, there were no differences between the two weight-maintenance diets. Subjective appetite sensations were not affected by the weight-reducing diet nor the weight-maintenance diets. While participants regained an average of ~50% of the weight they had lost by 36 months, the changes in ghrelin and peptide YY during the weight-reducing phase did not correlate with weight regain.Conclusion: A higher-protein, low-GI diet for weight maintenance does not attenuate changes in ghrelin or peptide YY compared with a moderate-protein, moderate-GI diet.Clinical Trial Registry:ClinicalTrials.gov registry ID NCT01777893 (PREVIEW) and ID NCT02030249 (Sub-study).

We have previously shown that a severely energy-restricted diet leads to greater loss of weight, fat, lean mass and bone mineral density (BMD) at 12 months in postmenopausal women with obesity than a moderately energy-restricted diet. We now aim to evaluate whether these effects are sustained longer term (ie, at 36 months). 101 postmenopausal women were randomized to either 12 months of moderate (25 to 35%) energy restriction with a food-based diet (moderate intervention), or 4 months of severe (65 to 75%) energy restriction with a total meal replacement diet followed by moderate energy restriction for 8 months (severe intervention). Body weight and composition were measured at 0, 24 and 36 months. Participants in the severe intervention lost ~1.5 to 1.7 times as much weight, waist circumference, whole-body fat mass and visceral adipose tissue compared to those in the moderate intervention, and were 2.6 times more likely (42% versus 16%) to have lost 10% or more of their initial body weight at 36 months (P < 0.01 for all). However, those in the severe versus moderate intervention lost ~1.4 times as much whole-body lean mass (P < 0.01), albeit this was proportional to total weight lost and there was no greater loss of handgrip strength, and they also lost ~2 times as much total hip BMD between 0 and 36 months (P < 0.05), with this bone loss occurring in the first 12 months. Thus, severe energy restriction is more effective than moderate energy restriction for reducing weight and adiposity in postmenopausal women in the long term (3 years), but attention to BMD loss in the first year is required. Abstract

Donor infection status should be considered when accepting an organ for transplant. Here we present a case of Chagas disease developing after a lung transplant where the donor was known to be Trypanosoma cruzi antibody positive. The recipient developed acute Trypanosoma cruzi infection with reactivation after treatment. Chagas disease-positive donors are likely to be encountered in the United States; donor targeted screening is needed to guide decisions regarding organ transplant and posttransplant monitoring.

Abstract

Trees dominate terrestrial biotic weathering of silicate minerals by converting solar energy into chemical energy that fuels roots and their ubiquitous nutrient-mobilising fungal symbionts. These biological activities regulate atmospheric CO2 concentrations ([CO2]a) over geologic timescales by driving calcium and magnesium fluvial ion export and marine carbonate formation. However, the important stabilising feedbacks between [CO2]a and biotic weathering anticipated by geochemical carbon cycle models remain untested. We report experimental evidence for a negative feedback across a declining Cenozoic [CO2]a range from 1500 to 200 ppm, whereby low [CO2]a curtails mineral surface alteration via trenching and etch pitting by arbuscular mycorrhizal (AM) and ectomycorrhizal (EM) fungal partners of tree roots. Optical profile imaging using vertical scanning interferometry reveals changes in nanoscale surface topography consistent with a dual mode of attack involving delamination and trenching by AM and EM fungal hyphae on phyllosilicate mineral flakes. This is consistent with field observations of micropores in feldspar, hornblende and basalt, purportedly caused by EM fungi, but with little confirmatory evidence. Integrating these findings into a process-based biotic weathering model revealed that low [CO2]a effectively acts as a "carbon starvation" brake, causing a three-fold drop in tree-driven fungal weathering fluxes of calcium and magnesium from silicate rock grains as [CO2]a falls from 1500 to 200 ppm. The feedback is regulated through the action of low [CO2]a on host tree productivity and provides empirical evidence for the role of [CO2]a starvation in diminishing the contribution of trees and mycorrhizal fungi to rates of biological weathering. More broadly, diminished tree-driven weathering under declining [CO2]a may provide an important contributory mechanism stabilising Earth's [CO2]a minimum over the past 24 million years.

Abstract

With obesity being a leading cause of preventable death, it is vital to understand how best to identify individuals with greater risk of metabolic disease, especially those with high visceral adipose tissue (VAT). This study aimed to determine whether three commonly used waist circumference (WC) measurement sites could provide accurate estimations of VAT, as determined by magnetic resonance imaging (MRI), which is a gold standard for measuring VAT, in postmenopausal women with obesity. VAT volume was measured by MRI of the total abdomen in 97 women aged 57.7 ± 0.4 years (mean ± SEM), mean body mass index 34.5 ± 0.2 kg/m2. WC was measured at the midpoint between the lowest rib and the iliac crest (WCmid), the narrowest point of the torso (WCnarrow), and at the level of the umbilicus (WCumbilicus). WC differed significantly according to measurement site, with WCnarrow (102.1 ± 0.7 cm) < WCmid (108.3 ± 0.7 cm) < WCumbilicus (115.7 ± 0.8 cm) (p < 0.001). WCmid, WCnarrow and WCumbilicus were all significantly correlated with VAT, as measured by MRI (r = 0.581, 0.563 and 0.390, respectively; p < 0.001 for all), but the relationships between WCmid or WCnarrow and VAT determined by MRI were stronger than for WCumbilicus. Measurement of either WCmid or WCnarrow provides valid estimates of VAT in postmenopausal women with obesity, with WCnarrow being favoured in light of its greater ease and speed of measurement in this population.

Abstract

The Schwarzschild-Couder Telescope (SCT) is a medium-sized telescope technology proposed for the Cherenkov Telescope Array. It uses a novel dual-mirror optical design that removes comatic aberrations across its entire field of view. The SCT camera employs high-resolution silicon photomultiplier (SiPM) sensors with a pixel size of 4 arcminutes. A prototype SCT (pSCT) has been constructed at the Fred Lawrence Whipple Observatory in Arizona, USA. An observing campaign in 2020, with a partial camera of 1600 pixels (2.7 degrees by 2.7 degrees field of view) resulted in detection of the Crab Nebula at 8.6 sigma statistical significance. Work on the pSCT camera and optical system is ongoing to improve performance and prepare for an upcoming camera upgrade. The pSCT camera upgrade will replace the current camera modules with improved SiPMs and readout electronics and will expand the camera to its full design field of view of 8 degrees in diameter (11,328 pixels). The fully upgraded pSCT will enable next-generation very-high-energy gamma-ray astrophysics through excellent background rejection and angular resolution. Abstract

The Cherenkov Telescope Array (CTA) is the next-generation ground-based observatory for very-high-energy gamma rays. One candidate design for CTA's medium-sized telescopes consists of the Schwarzschild-Couder Telescope (SCT), featuring innovative dual-mirror optics. The SCT project has built and is currently operating a 9.7-m prototype SCT (pSCT) at the Fred Lawrence Whipple Observatory (FLWO); such optical design enables the use of a compact camera with state-of-the art silicon photomultiplier detectors. A partially-equipped camera has recently successfully detected the Crab Nebula with a statistical significance of 8.6 standard deviations. A funded upgrade of the pSCT focal plane sensors and electronics is currently ongoing, which will bring the total number of channels from 1600 to 11328 and the telescope field of view from about 2.7° to 8° . In this work, we will describe the technical and scientific performance of the pSCT.

Abstract

The Schwarzschild-Couder Telescope (SCT) is a telescope concept proposed for the Cherenkov Telescope Array. It employs a dual-mirror optical design to remove comatic aberrations over an $8^{\circ}$ field of view, and a high-density silicon photomultiplier camera (with a pixel resolution of 4 arcmin) to record Cherenkov emission from cosmic ray and gamma-ray initiated particle cascades in the atmosphere. The prototype SCT (pSCT), comprising a 9.7 m diameter primary mirror and a partially instrumented camera with 1536 pixels, has been constructed at the Fred Lawrence Whipple Observatory. The telescope was inaugurated in January 2019, with commissioning continuing throughout 2019. Abstract

A prototype Schwarzschild-Couder Telescope (pSCT) has been constructed at the Fred Lawrence Whipple Observatory as a candidate for the medium-sized telescopes of the Cherenkov Telescope Array Observatory (CTAO). CTAO is currently entering early construction phase of the project and once completed it will vastly improve very high energy gamma-ray detection component in multi-wavelength and multi-messenger observations due to significantly improved sensitivity, angular resolution and field of view comparing to the current generation of the ground-based gamma-ray observatories H.E.S.S., MAGIC and VERITAS. The pSCT uses a dual aspheric mirror design with a $9.7$ m primary mirror and $5.4$ m secondary mirror, both of which are segmented. The Schwarzschild-Couder (SC) optical system (OS) selected for the prototype telescope achieves wide field of view of $8$ degrees and simultaneously reduces the focal plane plate scale allowing an unprecedented compact ($0.78$m diameter) implementation of the high-resolution camera ($6$mm/ $0.067$deg per imaging pixel with $11,328$ pixels) based on the silicon photo-multipliers (SiPMs). The OS of the telescope is designed to eliminate spherical and comatic aberrations and minimize astigmatism to radically improve off-axis imaging and consequently angular resolution across all the field of view with respect to the conventional single-mirror telescopes. Fast and high imaging resolution OS of the pSCT comes with the challenging submillimeter-precision custom alignment system, which was successfully demonstrated with an on-axis point spread function (PSF) of $2.9$ arcmin prior to the first-light detection of the Crab Nebula in 2020. Ongoing and future commissioning activities are reported.

Abstract

The novel 9.7m Schwarzschild-Couder Telescope (SCT), utilizing aspheric dual-mirror optical system, has been constructed as a prototype medium size x-ray telescope for the Cherenkov Telescope Array (CTA) observatory. The prototype SCT (pSCT) is designed to achieve simultaneously the wide (>= 8°) field of view and the superior imaging resolution (0.067 per pixel) to significantly improve scientific capabilities of the observatory in conducting the sky surveys, the follow-up observations of multi-messenger transients with poorly known initial localization and the morphology studies of x-ray sources with angular extent. In this submission, we describe the hardware and software implementations of the telescope optical system as well as the methods specifically developed to align its complex optical system, in which both primary and secondary mirrors are segmented. The pSCT has detected Crab Nebula in June 2020 during ongoing commissioning, which was delayed due to worldwide pandemic and is not yet completed. Verification of pSCT performance is continuing and further improvement of optical alignment is anticipated.

Abstract

For the first time in the history of ground-based $\gamma$-ray astronomy, the on-axis performance of the dual mirror, aspheric, aplanatic Schwarzschild-Couder optical system has been demonstrated in a $9.7$-m aperture imaging atmospheric Cherenkov telescope. The novel design of the prototype Schwarzschild-Couder Telescope (pSCT) is motivated by the need of the next-generation Cherenkov Telescope Array (CTA) observatory to have the ability to perform wide ($\geq 8^{\circ}$) field-of-view observations simultaneously with superior imaging of atmospheric cascades (resolution of $0.067^{\circ}$ per pixel or better). The pSCT design, if implemented in the CTA installation, has the potential to improve significantly both the $\gamma$-ray angular resolution and the off-axis sensitivity of the observatory, reaching nearly the theoretical limit of the technique and thereby making a major impact on the CTA observatory sky survey programs, follow-up observations of multi-messenger transients with poorly known initial localization, as well as on the spatially resolved spectroscopic studies of extended $\gamma$-ray sources. Abstract

The first prototype of the Schwarzschild Couder Medium Size Telescope (pSCT) proposed for the CTA observatory has been installed in 2018 at the Fred Lawrence Whipple Observatory. The pSCT camera is composed of 25 modules with 64 channels each, covering only a small portion of the full focal plane of the telescope. The Italian Institute of Nuclear Physics (INFN) has developed and characterized in collaboration with Fondazione Bruno Kessler (FBK) a new generation of Silicon Photomultipliers (SiPMs) sensitive to the Near Ultraviolet wavelengths, based on the High Density technology (NUV-HD devices). The latest generation of 6×6 mm2 SiPMs (called NUV-HD3) have been used to equip a subsection of 9 out of 25 modules of the pSCT camera. An upgrade of this camera is foreseen between 2019 and 2020 using the same sensors, aiming to equip the full focal plane with 177 modules, for a total of more than 11000 pixels. We will present a full characterization of the performance of these devices, highlighting why they are suitable for Cherenkov light detection. An overview on the overall behavior of the installed sensors will be also given, providing information on the uniformity of the sensors and of the performance of the camera.

Abstract

The Cherenkov Telescope Array (CTA) is the major next-generation observatory for ground-based very-high-energy gamma-ray astronomy. It will improve the sensitivity of current ground-based instruments by a factor of five to twenty, depending on the energy, greatly improving both their angular and energy resolutions over four decades in energy (from 20 GeV to 300 TeV). This achievement will be possible by using tens of imaging Cherenkov telescopes of three successive sizes. They will be arranged into two arrays, one per hemisphere, located on the La Palma island (Spain) and in Paranal (Chile). Abstract

Near UltraViolet High Density (NUV-HD) SiPMs produced by Fondazione Bruno Kessler in collaboration with INFN have been tested and characterized in INFN laboratories. The third generation of these devices (HD3) has proven to be suitable to equip the focal plane of the prototype Schwarzschild–Couder Medium Size Telescope (pSCT) proposed for the Cherenkov Telescope Array Observatory. Photosensors have been assembled in 4 16-pixel optical units coupled with TARGET–7 ASIC front-end electronics for amplification and digitization of the signal. At present, 9 modules have been successfully integrated on the pSCT camera and are currently taking data. In this contribution we report on the performances of the HD3 technology as single sensor and as assembled optical units, showing their performance and homogeneity in terms of gain and dark count rate.

In recent decades, there has been considerable growth in neurologic subspecialties. In response to this trend, the American Academy of Neurology (AAN), American Neurological Association (ANA), Association of University Professors of Neurology (AUPN), Child Neurology Society (CNS), and Professors of Child Neurology (PCN) helped establish the United Council for Neurologic Subspecialties (UCNS) in order to provide accreditation of training (fellowship) programs in neurologic subspecialties and certification of graduates of these programs (and, during a limited “practice track” period, practitioners qualified by their substantive experience). This article reviews the reasons why UCNS was established, how it operates, and what it has accomplished. In the first issue of Neurology ® in 1951, an article by Pearce Bailey, MD, entitled “The past, present, and future of neurology in the United States,” noted that by the end of World War II, neurology had not yet attained “unchallenged professional autonomy as a medical specialty.”1 Bailey pointed to the establishment of the AAN in 1948 as reason for hope that there would be “a new renaissance of neurology in the near future” and he concluded that “[t]he time has come for neurology as a specialty to assume responsibility for the total medical treatment of neurologic patients.” In the decades since Bailey's article, AAN has undertaken many initiatives to help neurologists achieve this goal. In 1998, AAN President Francis I. Kittredge, Jr, MD, FAAN, appointed a Commission on Subspecialty Certification that was chaired by Stephen M. Sergay, MB BCh, FAAN. This was a time of major change in neurology. Neuroscience was advancing at an explosive rate. New technologies and treatments were developed that promised dramatic advances in neurologic care. Abstract

Can manufacturers produce hundreds of millions of cans annually, increasingly, food cans are being opened by lifting a tab on the can end that initiates a fracture, which then propagates around a circumferential score. The damage and fracture mechanisms that lead to crack initiation and propagation in the opening process, however, are not fully understood, therefore optimisation of easy open end scores is largely based on trial and error. This paper presents an experimental analysis that concentrates on the combined shear and bending forces as applied to the particular industrial method concerning full aperture easy open ends. The influence of a gradually increasing gap measured between the score and shear force location on traditional groove geometries and depths are examined for two different packaging steels. Earlier studies have shown that the complete opening cycle depends on fracture modes I, II & III as well as their combination. Experimental results for Modes I, II & III will be presented, however attention will focus on the behaviour of the initial fracture point, whereby prior investigations have shown it to be influenced primarily by mode II shearing. After initial specimen manufacture, where the score is formed by pressing a punch into a thin steel sheet the predeformed scored specimens are loaded in shear to simulate the local stress field found during the initial opening phase. Experiments have been completed using a novel Mode II experimental technique that has been designed for use in the majority of commercially available tensile test machines. Experimental results indicate that opening forces can change radically with different gap sizes and that there is considerable potential for the industrialised process of can end manufacture to be optimised through the efficient management and control of the can ends dimensional parameters.

Abstract

Introduction: Effective management of major depressive disorder (MDD) continues to be a challenging task for psychiatrists and primary care physicians. This trial evaluated the efficacy and safety of adjunctive aripiprazole versus antidepressant monotherapy in patients with MDD and independently replicated the positive findings of two similar trials.Methods: Patients (N=1, 147) with MDD experiencing a major depressive episode and a history of inadequate response to antidepressant monotherapy were enrolled (week 0); 827 received single-blind adjunctive placebo plus open-label antidepressant (escitalopram, fluoxetine, paroxetine controlled release, sertraline, or venlafaxine extended release) for 8 weeks to confirm inadequate response to antidepressants; 349 patients with inadequate response were randomized (1:1) to double-blind, adjunctive placebo (n=172) or adjunctive aripiprazole (n=177; 2–20 mg/day). Primary outcome was the mean change in Montgomery-Åsberg Depression Rating Scale (MADRS) Total score from baseline (week 8) to endpoint (week 14).Results: Clinically significant improvements in depressive symptoms as assessed by decreases in the MADRS Total score were greater with adjunctive aripiprazole (−10.1) than placebo (−6.4; PPConclusion: For some patients with MDD who do not obtain adequate symptom relief with antidepressant monotherapy, adjunctive therapies can significantly improve depressive symptoms. As reported, adjunctive aripiprazole was associated with a two-fold higher remission rate than adjunctive placebo. This, and previous studies, have shown that discontinuations due to adverse events were low and completion rates were high, and has indicated that both antidepressant and aripiprazole in combination were relatively well-tolerated and safe. This is the third consecutive clinical trial, in the absence of a failed trial, to demonstrate that aripiprazole augmentation to antidepressants is an efficacious and well-tolerated treatment for patients with MDD who do not respond adequately to standard antidepressant monotherapy (ClinicalTrials.gov study NCT00105196).

33. The role of glycosylation in flavonol-induced pollen germination

Abstract

Flavonols are small (C15) plant-specific molecules that are required for petunia and maize pollen to germinate. They exist in two chemical forms: the aglycone or glycosyl conjugates. Flavonol-deficient pollen is biochemically complemented by flavonol aglycones but not by the glycosylated forms that accumulate in wild type (WT) pollen. Coincident with the biochemical induction of germination, the added flavonol aglycone is rapidly converted to a galactoside and then to a glucosyl galactoside (diglycoside) that is identical to the compound present in WT pollen. A flavonol 3-O-galactosyltransferase (F3GalTase) activity has been identified that controls the formation of glycosylated flavonols in pollen. Importantly, this enzyme also catalyzes the reverse reaction, i.e. the production of the flavonol aglycone from the galactoside and UDP (Fig. 1). F3GalTase/RevGalTase therefore has the potential to control the level of the bioactive flavonol species and as a result, pollen germination.

Abstract

Nimbus and the University of Pittsburgh (UOP) have continued the development of a totally implanted axial flow blood pump under the National Institutes of Health (NIH) Innovative Ventricular Assist System (IVAS) program. This 62 cc device has an overall length of 84 mm and an outer diameter of 34.5 mm. The inner diameter of the blood pump is 12 mm. It is being designed to be a totally implanted permanent device. A key achievement during the past year was the completion of the Model 2 pump design. Ten of these pumps have been fabricated and are being used to conduct in vitro and in vivo experiments to evaluate the performance of different materials and hydraulic components. Efforts for optimizing the closed loop speed control have continued using mathematical modeling, computer simulations, and in vitro and in vivo testing. New hydraulic blade designs have been tested using computational fluid dynamics (CFD) and flow visualization. A second generation motor was designed with improved efficiency. To support the new motor, a new motor controller fabricated as a surface mount PC board has been completed. The program is now operating under a formal QA system.

Abstract

This paper summarizes the authors' in vivo experience in evaluating a miniature centrifugal blood pump designed for pediatric/neonatal ventricular support. Left ventricular bypass was accomplished in two adult sheep and five juvenile lambs (5.5-80.0 kg) via either central (left ventricle to carotid artery) or peripheral (jugular vein to carotid artery) cannulation. Animals were weaned from mechanical ventilation and continuously monitored. Hemodynamic parameters remained within a normal range over the duration of the bypass. Two of five lambs were electively killed at 8, and 76 hours; the remaining three lambs died from respiratory complications at 33, 44, and 156 hours. There were no mechanical complications, and blood seal integrity was confirmed beyond 6 days. The pump speed was maintained at 3,000-4,500 rpm with pump flow rates between 0.4-1.5 L/min. Average plasma free hemoglobin was below 20 mg/dl in the five lamb experiments. Renal, hepatic, and hematologic indices also remained within physiologic ranges. Histopathologic analyses of major organs revealed renal cortical infarctions in two of five lambs. Examination of the pump surfaces after explant indicated small areas of thrombus in the housing adjacent to the outflow ports in two experiments. These encouraging results support further testing and refinement of this miniature centrifugal pump.

Abstract

This paper summarizes the authors' in vivo experience to date with an implantable axial flow blood pump designed for long-term ventricular support. This small, valveless pump with blood-lubricated bearings has been implanted in six calves (83 +/- 6 kg) as a left ventricular assist device (LVAS). The left ventricle and descending thoracic aorta were cannulated by left thoracotomy, and the pump was placed in a subcutaneous pocket below the costal margin. Animals remained hemodynamically stable throughout the course of support during partial left ventricular bypass. Five animals were killed after 15, 27, 52, 57, and 181 days. The longest survivor (181 days) demonstrated normal pump function at the time death. Pump speed was maintained at 10,100 +/- 100 rpm, with an average pump flow rate of 4.9 +/- 0.5 L/min under resting physiologic conditions. Average plasma free hemoglobin was 17.4 +/- 7.5 mg/dl. Renal, hepatic, and hematologic indices remained within physiologic range in all of these animals, except during the immediate postoperative period. Histopathologic analyses of major organs after death revealed small renal cortical infarcts in five of six animals; the remaining organs were normal. These animal studies support the feasibility of this small implanted axial flow pump for long-term ventricular assistance.

Abstract

Two peptides, which have no significant homology with known protein structures, were obtained by microsequencing of a mu-opioid binding protein purified to homogeneity from bovine striatal membranes. Polyclonal antibodies generated against portions of these peptides immunoprecipitated up to 65% of radiolabeled purified opioid binding protein. Sequential immunoprecipitations, using antibodies directed against portions of the two different peptides, confirmed that the peptides are derived from the same protein. Immunoblots of the protein with antipeptide antibodies revealed a protein band corresponding to the molecular weight of denatured reduced mu-opioid binding protein. The immunoresponse was blocked by the appropriate peptide and was not observed with irrelevant antisera. The antipeptide antibodies were used for immunoblots of sodium dodecyl sulfate extracts of tissues from bovine brain regions and of the mu receptor-containing cell line SK-N-SH. Affinity-purified antipeptide antibody detected an immunoreactive protein of molecular weight 65,000 in brain regions containing high levels of mu-opioid receptors (striatum, thalamus, hippocampus, and frontal cortex) and in the cell line SK-N-SH. Pons, which contains low levels of receptors, produced a a barely detectable signal, whereas white matter, HeLa cells, and C6 glioma cells, devoid of opioid binding activity, produced no detectable signal. The correlation between immunoreactivity and the presence of mu-opioid binding in brain regions and cell lines and the correspondence of the molecular weight of the immunoreactive protein to that of mu-opioid receptors provide strong evidence that the peptide antisera recognize mu receptors.

Abstract

In this work, we used two approaches to localize the 90-kDa heat shock protein (hsp90)-binding site within the hormone-binding domain of the glucocorticoid receptor. In the first approach, derivatives of the glucocorticoid receptor deleted for increasing portions of the COOH terminus were translated in rabbit reticulocyte lysate, and the [35S]methionine-labeled translation products were immunoadsorbed with the 8D3 monoclonal antibody against hsp90. The data suggest that a segment from amino acids 604 to 659 (mouse) of the receptor is required for hsp90 binding. We have recently shown that the internal deletion mutant of the mouse receptor (delta 574-632) binds hsp90, although the complex is somewhat unstable (Housley, P. R., Sanchez, E. R., Danielsen, M., Ringold, G. M., and Pratt, W. B. (1990) J. Biol. Chem. 265, 12778-12781). The two observations indicate that amino acids 574-659 are involved in forming a stable receptor-hsp90 complex and that region 632-659 is especially important. To test this hypothesis directly, we synthesized three peptides corresponding to segments in region 624-665 and three peptides spanning the highly conserved sequence at amino acids 582-617, and we then tested the ability of the peptides to compete for the association of hsp90 with the L cell glucocorticoid receptor. In this assay, the immunopurified hsp90-free mouse receptor is incubated with rabbit reticulocyte lysate, which directs the association of rabbit hsp90 with the mouse receptor, simultaneously converting the receptor to the steroid binding state. All three peptides spanning region 624-665 and a peptide corresponding to segment 587-606 inhibited both hsp90 association with the receptor and reconstitution of steroid binding capacity. The data from all of the approaches support a two-site model for the hsp90-binding site in which the critical contact site occurs in region 632-659, which contains a short proline-containing hydrophobic segment and adjacent dipole-plus-cysteine motif that are conserved among all of the hsp90-binding receptors in the superfamily. A second hsp90 contact site is predicted in region 574-632, which contains the only highly conserved amino acid sequence in the receptor superfamily outside of the DNA-binding domain.

Abstract

Context Extracts of Hypericum perforatum (St John's wort) are widely used for the treatment of depression of varying severity. Their efficacy in major depressive disorder, however, has not been conclusively demonstrated. Objective To test the efficacy and safety of a well-characterized H perforatum extract (LI-160) in major depressive disorder. Design and setting Double-blind, randomized, placebo-controlled trial conducted in 12 academic and community psychiatric research clinics in the United States. Participants Adult outpatients (n = 340) recruited between December 1998 and June 2000 with major depression and a baseline total score on the Hamilton Depression Scale (HAM-D) of at least 20. Interventions Patients were randomly assigned to receive H perforatum, placebo, or sertraline (as an active comparator) for 8 weeks. Based on clinical response, the daily dose of H perforatum could range from 900 to 1500 mg and that of sertraline from 50 to 100 mg. Responders at week 8 could continue blinded treatment for another 18 weeks. Main outcome measures Change in the HAM-D total score from baseline to 8 weeks; rates of full response, determined by the HAM-D and Clinical Global Impressions (CGI) scores. Results On the 2 primary outcome measures, neither sertraline nor H perforatum was significantly different from placebo. The random regression parameter estimate for mean (SE) change in HAM-D total score from baseline to week 8 (with a greater decline indicating more improvement) was -9.20 (0.67) (95% confidence interval [CI], -10.51 to -7.89) for placebo vs -8.68 (0.68) (95% CI, -10.01 to -7.35) for H perforatum (P =.59) and -10.53 (0.72) (95% CI, -11.94 to -9.12) for sertraline (P =.18). Full response occurred in 31.9% of the placebo-treated patients vs 23.9% of the H perforatum-treated patients (P =.21) and 24.8% of sertraline-treated patients (P =.26). Sertraline was better than placebo on the CGI improvement scale (P =.02), which was a secondary measure in this study. Adverse-effect profiles for H perforatum and sertraline differed relative to placebo. Conclusion This study fails to support the efficacy of H perforatum in moderately severe major depression. The result may be due to low assay sensitivity of the trial, but the complete absence of trends suggestive of efficacy for H perforatum is noteworthy.

We reviewed the charts of 429 patients with primary brain tumors treated at Memorial Sloan-Kettering Cancer Center to determine the frequency, clinical characteristics, and provocative factors involved in the development of the shoulder-hand syndrome. Five percent (20/429) of patients developed this syndrome, which was strongly associated with phenobarbital use (p less than 0.0001) and often produced more short-term disability than the brain tumor itself.

Abstract

The Schwarzschild-Couder Telescope (SCT) is a candidate technology for a medium-sized telescope within the Cherenkov Telescope Array, the next generation ground based observatory for very high energy gamma ray astronomy. The SCT uses a novel two-mirror design and is expected to yield improvements in field of view and image resolution compared to traditional Cherenkov telescopes based on single-mirror-dish optics. To match the improved optical resolution, challenging requirements of high channel count and density at low power consumption must be overcome by the camera. The prototype camera, currently commissioned and tested on the prototype SCT, has been developed based on millimeter scale SiPM pixels and a custom high density digitizer ASIC, TARGET, to provide 1600 pixels spanning a 2.7 degree field of view while being able to sample nanosecond photon pulses. It is mechanically designed to allow for an upgrade to 11,328 pixels covering a field of view of 8 degrees and demonstrating the full potential of the technology. Abstract

The Cherenkov Telescope Array (CTA), with more than 100 telescopes, will be the largest ever ground-based gamma-ray observatory and is expected to greatly improve on both gamma-ray detection sensitivity and energy coverage compared to current-generation detectors. The 9.7-m Schwarzschild-Couder telescope (SCT) is one of the two candidates for the medium size telescope (MST) design for CTA. The novel aplanatic dual-mirror SCT design offers a wide field-of-view with a compact plate scale, allowing for a large number of camera pixels that improves the angular resolution and reduce the night sky background noise per pixel compared to the traditional single-mirror Davies-Cotton (DC) design of ground-based gamma-ray telescopes. The production, installation, and the alignment of the segmented aspherical mirrors are the main challenges for the realization of the SCT optical system.
