1. Las poblaciones ibéricas de pino albar ante el cambio climático: con la muerte en los talones

Abstract

El pino albar (Pinus sylvestris) es uno de los árboles más ampliamente distribuidos del mundo. Pese a su gran plasticidad ecológica, numerosos estudios muestran que su capacidad de resistir la sequía se está viendo superada en diversas zonas, especialmente en la cuenca mediterránea, donde se halla el límite meridional de su distribución. El presente artículo repasa una serie de trabajos recientes sobre los efectos directos e indirectos de la sequía sobre las poblaciones ibéricas de pino albar en un contexto de cambio climático. Específicamente, se tratan los siguientes aspectos: (1) ¿qué características ecofisiológicas explican la vulnerabilidad del pino albar a la sequía?; (2) ¿qué factores ambientales determinan sus patrones de crecimiento y cómo han variado en las últimas décadas?; (3) ¿qué factores ambientales explican la variabilidad espacial en las tasas demográficas de la especie (crecimiento, mortalidad, reclutamiento) a distintas escalas?; y (4) ¿cuáles son los impactos previsibles de un aumento de las condiciones de sequía y de los incendios forestales? El conjunto de los resultados registrados sugiere que una parte importante de las poblaciones ibéricas de pino albar podrían dejar de ser viables a medio plazo si se cumplen las proyecciones de cambio climático. Concluimos explorando hasta qué punto podemos utilizar la información anterior para identificar los individuos o poblaciones más vulnerables y el papel que la gestión podría tener a la hora de modular los impactos esperados.

Abstract

Northern corn leaf blight (NCLB), the most devastating leaf pathogen in maize (Zea mays L.), is caused by the heterothallic ascomycete Setosphaeria turcica. The pathogen population shows an extremely high genetic diversity in tropical and subtropical regions. Varietal resistance is the most efficient technique to control NCLB. Host resistance can be qualitative based on race-specific Ht genes or quantitative controlled by many genes with small effects. Quantitative resistance is moderately to highly effective and should be more durable combatting all races of the pathogen. Quantitative resistance must, however, be analyzed in many environments (= location × year combinations) to select stable resistances. In the tropical and subtropical environments, quantitative resistance is the preferred option to manage NCLB epidemics. Resistance level can be increased in practical breeding programs by several recurrent selection cycles based on disease severity rating and/or by genomic selection. This review aims to address two important aspects of the NCLB pathosystem: the genetics of the fungus S. turcica and the modes of inheritance of the host plant maize, including successful breeding strategies regarding NCLB resistance. Both drivers of this pathosystem, pathogen, and host, must be taken into account to result in more durable resistance.

Abstract

Maize white spot (MWS) is one of the most important foliar diseases in Brazil causing significant yield losses. Breeding genotypes with MWS resistance is the most sustainable alternative for managing such losses; however, their genetic control is poorly understood. Our objectives were to identify genomic regions controlling MWS resistance and to explore the presence of common regions controlling resistance to MWS, grey leaf spot (GLS) and northern corn leaf blight (NCLB). We performed a multi-parental QTL mapping for MWS and GLS resistance with a total of 474 testcrosses and phenotypic data collected in southern Brazil. Six QTLs for MWS resistance on bins 1.03, 1.04, 6.02, 8.05, 1.03, and 10.06 were detected. These findings were compared with previously reported QTLs for NCLB in the same populations, and a common QTL region (bin 8.05) controlling MWS and NCLB resistances was identified. Our findings contribute to a better understanding of MWS resistance by revealing three QTLs (bin 6.02, 1.03, and 10.06), to the best of our knowledge, not yet described in the literature, that are valuable for improving MWS resistance and one promising candidate region for multiple disease resistance. Abstract

El gres porcelánico es un material ampliamente utilizado en edificación. En los últimos años su uso ha experimentado un nuevo espectro de líneas de innovación e invención en sus aplicaciones en la arquitectura. En esta investigación de analiza la patente Panel de Acondicionamiento Térmico Cerámico, consistente en piezas de gres porcelánico de bajo espesor, que contienen tramas capilares a base de tubos de polipropileno de 3,5 mm de diámetro, e interfaz de pasta conductora. Estos sistemas trabajan con agua fría o caliente produciendo una climatización saludable por superficies radiantes. Tras una primera experiencia de prototipado y colocación de paneles en pared en un despacho de oficina, se han realizado simulaciones en el Museo de la Universidad de Alicante, colocando los paneles en pared, techo o tipo bafle, previa monitorización del estado actual del edificio. Se han analizado los parámetros de comportamiento térmico y se han comparado con otros materiales de acabado habituales. Se han obtenido resultados de confort térmico y ahorros energéticos de forma comparativa frente a sistemas todo-aire.

Key message High genetic variation in two European maize landraces can be harnessed to improve Gibberella ear rot resistance by integrated genomic tools. Abstract Fusarium graminearum (Fg) causes Gibberella ear rot (GER) in maize leading to yield reduction and contamination of grains with several mycotoxins. This study aimed to elucidate the molecular basis of GER resistance among 500 doubled haploid lines derived from two European maize landraces, “Kemater Landmais Gelb” (KE) and “Petkuser Ferdinand Rot” (PE). The two landraces were analyzed individually using genome-wide association studies and genomic selection (GS). The lines were genotyped with a 600-k maize array and phenotyped for GER severity, days to silking, plant height, and seed-set in four environments using artificial infection with a highly aggressive Fg isolate. High genotypic variances and broad-sense heritabilities were found for all traits. Genotype-environment interaction was important throughout. Abstract

Key message NCLB is the most devastating leaf disease in European maize, and the introduction of Brazilian resistance donors can efficiently increase the resistance levels of European maize germplasm. Abstract Northern corn leaf blight (NCLB) is one of the most devastating leaf pathogens in maize (Zea mays L.). Maize cultivars need to be equipped with broad and stable NCLB resistance to cope with production intensification and climate change. Brazilian germplasm is a great source to increase low NCLB resistance levels in European materials, but little is known about their effect in European environments. To investigate the usefulness of Brazilian germplasm as NCLB resistance donors, we conducted multi-parent QTL mapping, evaluated the potential of marker-assisted selection as well as genome-wide selection of 742 F1-derived DH lines. The line per se performance was evaluated in one location in Brazil and six location-by-year combinations (= environments) in Europe, while testcrosses were assessed in two locations in Brazil and further 10 environments in Europe. Jointly, we identified 17 QTL for NCLB resistance explaining 3.57–30.98% of the genotypic variance each. Two of these QTL were detected in both Brazilian and European environments indicating the stability of these QTL in contrasting ecosystems. We observed moderate to high genomic prediction accuracies between 0.58 and 0.83 depending on population and continent. Collectively, our study illustrates the potential use of tropical resistance sources to increase NCLB resistance level in applied European maize breeding programs.

Abstract

Maize production is on risk by Gibberella ear rot (GER) caused by Fusarium graminearum. This is one of the most important ear rot diseases in temperate zones as it leads to yield losses and production of harmful mycotoxins. We investigated, for the first time, the potential use of Brazilian tropical maize to increase resistance levels to GER in temperate European flint germplasm by analyzing six interconnected biparental populations. We assessed GER symptoms in Brazil and in Europe in up to six environments (= location × year combinations) during the growing seasons of 2018 and 2019. We conducted multi-parent QTL and biparental QTL mapping, and identified four QTLs with additive gene action, each explaining 5.4 to 21.8% of the total genotypic variance for GER resistance. Among them, QTL q1 was stable across test environments, populations, and between inbred lines and testcrosses. The accuracies of genomic prediction ranged from 0.50 to 0.59 depending on the resistance donor and prediction model. Abstract

This research aims to identify the intangible urban geometries of the physical-environmental, social-cultural, and political-economic components of Avenue 1 East, Caobos, San José de Cúcuta, Colombia. This in order to represent and quantify the connectivity trajectory of the intangible networks that are generated from the analysis of the meso and micro scale of the city. The applied methodology is based on the theory of the urban network, the study used a mixed approach, under a strategy called multi-methods or methodological triangulation. Within this research, physical concepts such as trajectory, displacement and space-time were assumed. This type of research allowed the study topics to be approached through layers through which the implicit dynamics in the sector under study were identified. As relevant results it is obtained that the use of a mathematical and physical logic strengthens the studies focused on city planning, the importance of hierarchically and quantitatively organizing the connection networks (meso and micro) in order to solve problems or phenomena typical of the daily life of the sector under study, guaranteeing a dynamic and real process for its growth.

9. Energy efficiency of flooded roofs. The University of Alicante Museum

Abstract

Inverted flat roofs were adopted as a common construction solution throughout the Modern Movement. They require greater protection of the waterproofing layer and sufficient thermal insulation. Depending on the climate conditions, a thicker or thinner thermal layer is installed and energy saving in keeping with the climatic conditions, regulations or national or regional energy policies is obtained. However, the common construction systems tend to be focused on minimising the construction costs of each project, using thermal insulation layers of extruded polystyrene, a modified bitumen waterproofing layer and a finishing layer of 5 cm of gravel. In the case of trafficable roofs, this layer is substituted with ceramic or hydraulic concrete paving. These solutions have given rise to an enormous reduction in the thermal mass of the roof layers located above the isolation layer and consequently, a lack of thermal inertia with which to cushion the effect of the thermal wave of the outdoor environment produced in the indoor climate of the building. Roofs flooded with water in climatologies which allow their application generate significant thermal inertia depending on the depth of the water. They reduce the amplitude of the thermal wave produced in the inside temperature of the buildings by the external thermal variations, generating a considerable increase in comfort and a reduction in energy consumption. This paper outlines the research carried out in the Alicante University Museum. The whole building and its flooded roof have been monitored over a full year cycle and data has been obtained regarding the dry interior temperatures of each layer of the walls, outdoor temperatures and solar radiation, relative humidities, etc. Through the use of simulation tools, comparative results of this roof with respect to inverted flat roofs have been obtained, such as the decrement factor and the heat wave.

10. Alzemon: estudio de seguimiento prospectivo del acetato de eslicarbacepina en monoterapia en pacientes con epilepsia de diagnóstico reciente

Abstract

Introduccion. El acetato de eslicarbacepina es un nuevo bloqueante de los canales de sodio en el tratamiento de las crisis de inicio focal. Los estudios prospectivos sobre su efectividad en monoterapia en pacientes con epilepsia parcial de reciente diagnostico en la practica clinica habitual son escasos. Objetivo. Evaluar la efectividad de la eslicarbacepina en monoterapia de inicio en pacientes con epilepsia parcial de reciente diagnostico en la practica clinica habitual. Pacientes y metodos. Estudio postautorizacion prospectivo y multicentrico. Se incluyo a pacientes con epilepsia parcial de reciente diagnostico de 18 anos o mas sin tratamiento previo. Las variables de eficacia fueron: porcentaje de pacientes libres de crisis, respondedores y reduccion en la frecuencia mensual de crisis. Las variables de seguridad analizan la tasa de retencion a los 12 meses y la aparicion de efectos adversos. Resultados. Se incluyo a 53 pacientes. La tasa de retencion fue del 77,4%. Al final del periodo de observacion, el 83% de los pacientes se encontraba libre de crisis y el 92,5% habia reducido en un 50% o mas su frecuencia basal. El 68% de los pacientes notifico algun efecto adverso y el 7,5% de ellos abandono el estudio por este motivo. El analisis de efectividad del subgrupo de 65 anos o mas no mostro diferencias respecto a la poblacion global. Conclusion. La eslicarbacepina en monoterapia en pacientes con epilepsia parcial de reciente diagnostico, tanto en la poblacion general como en la poblacion de mas de 65 anos, es eficaz y segura en la practica clinica habitual.

Abstract

Fusarium head blight (FHB) is one of the most serious diseases in small-grain cereals including triticale (× Triticosecale Wittmack). The disease reduces yield and accumulates mycotoxins which are harmful to human and animal health. Triticale grain is almost exclusively used on-farm in feed formulations for swine and other animals, and swine is the most susceptible farm animal to Fusarium mycotoxins. In order to evaluate the potential of genomics-assisted breeding to FHB, we performed the first genome-wide association study for FHB resistance in triticale. QTL for FHB resistance were identified on chromosomes 2A, 2B, 5B and 3R with an explained genotypic variance ranging from 0.28 to 30.23% and a total explained genetic variance of 56.64%. A QTL on chromosome 3R that explained 15.38% of the genotypic variance was identified for the first time. Association mapping was complemented by genome-wide prediction, which yielded a high prediction accuracy of 0.78 for FHB resistance when weighted genomic selection was performed. Collectively our findings highlight the potential of genomics-assisted approaches to improve Fusarium resistance in triticale in early generations.

Abstract

Purpose: Evaluate real-life experience with eslicarbazepine acetate (ESL) after first monotherapy failure in a large series of patients with focal epilepsy. Method: Multicentre, retrospective, 1-year, observational study in patients older than 18 years, with focal epilepsy, who had failed first antiepileptic drug monotherapy and who received ESL. Data from clinical records were analysed at baseline, 3, 6 and 12 months to assess effectiveness and tolerability. Results: Eslicarbazepine acetate was initiated in 253 patients. The 1-year retention rate was 92.9%, and the final median dose of ESL was 800 mg. At 12 months, 62.3% of patients had been seizure free for 6 months; 37.3% had been seizure free for 1 year. During follow-up, 31.6% of the patients reported ESL-related adverse events (AEs), most commonly somnolence (8.7%) and dizziness (5.1%), and 3.6% discontinued due to AEs. Hyponatraemia was observed in seven patients (2.8%). After starting ESL, 137 patients (54.2%) withdrew the prior monotherapy and converted to ESL monotherapy; 75.9% were seizure free, 87.6% were responders, 4.4% worsened, and 23.4% reported ESL-related AEs. Conclusion: Use of ESL after first monotherapy failure was associated with an optimal seizure control and tolerability profile. Over half of patients were converted to ESL monotherapy during follow-up.

15. Juan Bautista Antonelli: the first example of Alicante’s bastioned fortifications

Abstract

Juan Bautista Antonelli, one of the most important engineers of Felipe II’s reign, was the first engineer who proposed a bastioned fortification project for the city of Alicante. The reforms were motivated by the continual attacks from Turkish and Berber pirates. The walls of Charles V were soon obsolete by the development of artillery. The Antonelli’s fortification project was drafted in 1563 and paid attention to both attack by sea as a possible attack from the inside. The city was perfectly defended with four new fortified bastions and three demi-bastions. The old walled city concept had been forgotten. The project’s interpretation allows us to trace its hypothetical design and find the source of the XVII century proposals fortress. The project was never built and the city defence systems relied upon Santa Barbara’s castle for protection. One hundred and twenty five years later, in 1688, Ambrosio Borzano proposed a bastioned fortification project for the city of Alicante. As a result of his project only the bastion of San Carlos was built.

Abstract

A door-to-door survey of transient ischemic attack (TIA) and stroke was conducted in 3 towns of the Alcoi health area. The prevalence ratios found for TIA and stroke were 13/1,000 people (95% CI 8.8-17.9) and 21.5/1,000 (95% CI 15.8-27.2), respectively. The crude incidence rates for both TIA and stroke were 2.8/1,000 (95% CI 0.7-4.9). Methodological differences in the ascertainment of the cases may explain the variations in the rates between the present work and other studies in the literature.

Abstract

SiO2 thin films, deposited by the plasma‐enhanced chemical vapor deposition technique using tetraethylorthosilicate (TEOS) and O2 as precursors, exhibit electron‐paramagnetic‐resonance signals that are interpreted in terms of methyl‐ and dimethyl‐associated centers. The former are observed in as‐deposited films and in films thermally treated below 500 °C while the latter are observed in films that have been thermally treated between 700 and 950 °C. These hydrocarbon‐containing paramagnetic centers originate from TEOS’s ethyl constituents that are incorporated in the SiO2 film during growth.

Abstract

Matrix metalloproteinases belong to a growing family of proteases controlled by specific tissue inhibitors, involved in tissutal flogosis, wound healing, cancer invasion and metastasis. We developed an in vitro model to screen for potential toxic compounds in professional exposure. Human keratinocytes (HaCat) were used as target cells while matrix metalloproteinases (MMP) were selected as responders, developing and in vitro model of allergic dermatitis. The chemical agents: potassium dichromate and nickel sulphate as positive teste, because represent the main etiological agents of allergic dermatitis. Nickel contact at very low concentrations (10(-5), 10(-6) M) induced upregulation of MMP-2 and IL-8 mRNA production; chromium contact at very low concentrations killed all cells. Actually, our in vitro research is based on analysis of cytotoxic effects of xenobiotics on human lung fibroblasts; simultaneously we verified serum increasing in vivo of MMP-9, determinated in workers serum, exposed to anesthetic gas (fluorane). In only six exposed workers we observed MMP9 increasing over than normal range. Actually, we are continuing our research on a more representative sample.

Abstract

Objective: To analyze seizure control and treatment in pregnant women with epilepsy. Methods: Seizure control and treatment were recorded prospectively in 1,956 pregnancies of 1,882 women with epilepsy participating in EURAP, an international antiepileptic drugs (AEDs) and pregnancy registry. Results: Of all cases, 58.3% were seizure-free throughout pregnancy. Occurrence of any seizures was associated with localization-related epilepsy (OR: 2.5; 1.7 to 3.9) and polytherapy (OR: 9.0; 5.6 to 14.8) and for tonic-clonic seizures, with oxcarbazepine monotherapy (OR: 5.4; 1.6 to 17.1). Using first trimester as reference, seizure control remained unchanged throughout pregnancy in 63.6%, 92.7% of whom were seizure-free during the entire pregnancy. For those with a change in seizure frequency, 17.3% had an increase and 15.9% a decrease. Seizures occurred during delivery in 60 pregnancies (3.5%), more commonly in women with seizures during pregnancy (OR: 4.8; 2.3 to 10.0). There were 36 cases of status epilepticus (12 convulsive), which resulted in stillbirth in one case but no cases of miscarriage or maternal mortality. AED treatment remained unchanged in 62.7% of the pregnancies. The number or dosage of AEDs were more often increased in pregnancies with seizures (OR: 3.6; 2.8 to 4.7) and with monotherapy with lamotrigine (OR: 3.8; 2.1 to 6.9) or oxcarbazepine (OR: 3.7; 1.1 to 12.9). Conclusions: The majority of patients with epilepsy maintain seizure control during pregnancy. The apparently higher risk of seizures among women treated with oxcarbazepine and the more frequent increases in drug load in the oxcarbazepine and lamotrigine cohorts prompts further studies on relationships with pharmacokinetic changes. Risks associated with status epilepticus appear to be lower than previously reported.

Abstract

Objective: To evaluate the efficacy of a cognitive-motor program in patients with early Alzheimer disease (AD) who are treated with a cholinesterase inhibitor (ChEI).Methods: Patients with mild cognitive impairment (MCI) (12), mild AD (48), and moderate AD (24) (Global Deterioration Scale stages 3, 4, and 5) were randomized to receive psychosocial support plus cognitive-motor intervention (experimental group) or psychosocial support alone (control group). Cognitive-motor intervention (CMI) consisted of a 1-year structured program of 103 sessions of cognitive exercises, plus social and psychomotor activities. The primary efficacy measure was the cognitive subscale of the AD Assessment Scale (ADAS-cog). Secondary efficacy measures were the Mini-Mental State Examination, the Functional Activities Questionnaire, and the Geriatric Depression Scale. Evaluations were conducted at 1, 3, 6, and 12 months by blinded evaluators.Results: Patients in the CMI group maintained cognitive status at month 6, whereas patients in the control group had significantly declined at that time. Cognitive response was higher in the patients with fewer years of formal education. In addition, more patients in the experimental group maintained or improved their affective status at month 12 (experimental group, 75%; control group, 47%; p = 0.017).Conclusions: A long-term CMI in ChEI-treated early Alzheimer disease patients produced additional mood and cognitive benefits.

Abstract

The efficacy and tolerability of entacapone was investigated in a randomized, double-blind, placebo-controlled, 3-month study of 162 patients with Parkinson's disease (PD) treated with levodopa and a dopamine agonist and experiencing wearing-off motor fluctuations. Patients were randomized in a 3 : 2 ratio to entacapone 200 mg or placebo, administered with each dose of levodopa. Efficacy was judged on the improvement of “on” and “off” time while awake (Patient Diary and UPDRS part IV Item 39), Investigators' Global Assessment, the SF-36 Health Survey, and changes in levodopa dosages. Patients were monitored for adverse events, laboratory safety and vital signs throughout the study. Improvements in “on” time as assessed using patient diary data showed a trend in favour of entacapone, however these did not reach statistical significance. “Off” time while awake (UPDRS part IV Item 39) showed an improvement of at least one category in 36% of entacapone-treated patients, compared with 22% in the control group (p = 0.0038). The proportion of patients showing an improvement at the Investigators' Global Assessment was significantly higher (p = 0.0006) in the entacapone-treated group of patients. Also, the proportion of patients with a reduction in their daily levodopa dose was significantly higher (p = 0.02) in the entacapone group (28%) compared with placebo (13%). As expected, the most frequent adverse events were dopamine-mediated (dyskinesia: entacapone 31% versus placebo 13%), and harmless urinary discoloration. The modest increase in dyskinesias could be readily managed by levodopa down-adjustment, and, at study end there was no significant difference for the UPDRS “overall dyskinesia score” between entacapone and placebo. In conclusion, although the primary efficacy variable did not reach statistical significance, the present results demonstrate that entacapone provides additional antiparkinsonian benefits to levodopa therapy and is well tolerated in levodopa-treated PD patients experiencing wearing-off motor fluctuations despite adjunct dopamine agonist therapy.

Abstract

Pramipexol is a novel nonergot dopamine agonist which has high selectivity for intereacting with dopamine D2 receptors (especially with D3 receptor subtype). It has been effective in early Parkinson's disease as monotherapy and as adjunctive therapy with L-dopa in advanced stages of the disease. Clinical improvement can be observed after 3 or 4 weeks of treatment. The adverse events profile of pramipexol is similar, in general, to that of other dopamine receptor agonists, although it can be foreseen that pramipexol should not induce side effects related to the ergot chemical structure such as eritromelalgia, distal vasospasm, retroperitoneal fibrosis or pleural effusions. Nevertheless, the potential advantages of this promising dopamine agonist should be tested in well-designed prospective comparative studies with other available ergot and nonergot dopamine agonists.

Abstract

Different studies have shown that aspirin (AAS), in low doses, may lead to a considerable frequency of hemorrhagic complications when used in the long term.We compare the long-term occurrence of hemorrhagic complications with low doses of AAS and high doses of triflusal.Our series included 106 patients who took 900 mg triflusal per day (300 mg 3 times per day) and 111 who took AAS (330 mg/day once daily). The former were followed up for an average period of 48.3 months (20-94) and the latter for 46.3 months (2-84). The average follow-up period for the study was 47.3 months. The presence of hemorrhagic complications was evaluated, as was their frequency and follow-up curve.Compared with AAS, triflusal led to a 76% reduction in risk of hemorrhagic complications (2.8% against 10.8%; OR 0.24; IC 0.06-0.94). There was a slightly increased incidence of hemorrhages in the women's group. There were more hemorrhages than gastrointestinal hemorrhages (4.5% against 0.9%) and intracranial hemorrhages (1.8%-0.9%). The follow-up curve showed significant differences in the form of an increased risk of hemorrhagic complications with AAS.The risk of hemorrhage with AAS depended on the period of follow-up, in a similar manner to with oral anticoagulant agents, in patients with prophylaxis of cerebral infarct. On the other hand, this did not occur with triflusal, with which the risk was homogeneous and lower in the long term.

There are many affected who describe headaches brought on by stressful situations and events, although they also speak of the onset of their attacks during the period of calm immediately after such moments of stress. There are however few objective works in the literature which study the relationship between stress and migraine. In the present work we review that literature which does exist concerning such a relationship.

Abstract

Migraine is a paroxysmic abnormality in which asymtomatic periods alternate with the appearance of attacks. Such attacks are the end result of a chain of events leading on to the acute clinical syndrome. Amongst those phenomena which occur in the days prior to the attack starting, the factors which bring such attacks on have been widely studied by a great number of researchers. Identifying these initiating factors is a fundamental preventive element and looking into the behavioural mechanisms of such factors could prove useful in clarifying the pathogenic mechanisms of migraine. In the present study we review most of the works which have sought to identify these factors concerning the development of attacks and to work out their behavioural patterns.

Abstract

Throughout the many studies carried out on patients suffering from migraine, we find that a series of diseases or abnormalities with cardiovascular etiologies may be associated with the risk of migraine. In some studies published so far there is a greater prevalence of distinct cardiovascular diseases among those patients suffering from migraine. The shortage of case-control studies which might otherwise definitely establish a relationship between migraine and other cardiovascular diseases leads us to cautiously interpret the meaning of these associations. We studied family and personal history of cardiovascular disease in a group of patients with migraine, and we carried out a case-control study to try to establish the greatest risk of suffering from these diseases by our patients. We found that family antecedents of migraine with the next of kin was a risk factor for getting migraine and that Raynaud's disease was associated with a greater risk of suffering with this pathology.

Abstract

Migraine headache is a common, recurrent and often incapacitating disorder resulting in a substantial loss of work days and productivity, significant consumption of medicines, and considerable economic cost. We performed this study in order to determine the characteristic features of migrainous patients with regular or frequent intake of ergotamine preparations and non steroidal anti-inflammatory drugs (NSAIDs) and the existence of significant differences between both groups. 186 patients were included in the study, 36.6% suffering from migraine without aura and 63.4% from migraine with aura. 24.7% were males and 75.3% females. Mean age of the sample was 35.6 +/- 12.9 years. With regard to medication used for immediate relief of headache, 74.7% patients reported an intake of analgesics and 25.3% ergotamine preparations. In patients with analgesic intake the mean age was significantly lower than in patients with ergotamine for treatment of migraine. When clinic features of these two headache groups were examined, the duration of migraine attacks in the analgesic group was significantly higher (p = 0.0014) than in the ergotamine group, and the severity of migraine was lower (p0.05) than in this group. Among associated symptoms, nausea and vomiting were significantly less frequent (p = 0.024) in the analgesic group than in the group of patients with ergotamine intake.

Abstract

Knowing the epidemiology of migraine is important for a host of reasons. Migraine is one of the most prevalent types of headache. In addition, it constitutes a pathology which due to its cyclic, recurring and highly incapacitating characteristics has enormous repercussions on the quality of life of those affected; it is responsible for the loss of an important number of working days and of production; responsible too for the vast consumption of medicaments and consequently represents a considerable economic cost. An epidemiological study, however, of its pathology poses countless difficulties owing to its characteristics and its association with risk factors. In this present work we review the obstacles to carrying out migraine epidemiological studies and we also comment upon many such studies found in the literature. In each section we present the data obtained in such studies as were performed by our working group.

Abstract

A Research Design and Development Project was developed of a myoelectric prosthesis for a pediatric patient presenting congenital amputation of the left forearm below the elbow. A multidisciplinary work-team was formed for this goal, in order to solve the several (/various) aspects regarding this project (mechanical, ergonomics, electronics, physical). The prosthesis as an electromechanical device was divided in several blocks, trying to achieve a focused development for each stage, acording to requisites. A mechanical prototype of the prothesis was designed and built along with the circuitry needed for EMG aquisition, control logic and drivers. Having acomplished the previuos stages, the project is now dealing with the definitions of the interface between the prosthesis and the patient, with promising perspectives.

Abstract

Introduction Different studies have shown that aspirin (AAS), in low doses, may lead to a considerable frequency of hemorrhagic complications when used in the long term. Objective We compare the long-term occurrence of hemorrhagic complications with low doses of AAS and high doses of triflusal. Patients and methods Our series included 106 patients who took 900 mg triflusal per day (300 mg 3 times per day) and 111 who took AAS (330 mg/day once daily). The former were followed up for an average period of 48.3 months (20-94) and the latter for 46.3 months (2-84). The average follow-up period for the study was 47.3 months. The presence of hemorrhagic complications was evaluated, as was their frequency and follow-up curve. Results Compared with AAS, triflusal led to a 76% reduction in risk of hemorrhagic complications (2.8% against 10.8%; OR 0.24; IC 0.06-0.94). There was a slightly increased incidence of hemorrhages in the women's group. There were more hemorrhages than gastrointestinal hemorrhages (4.5% against 0.9%) and intracranial hemorrhages (1.8%-0.9%). The follow-up curve showed significant differences in the form of an increased risk of hemorrhagic complications with AAS. Conclusions The risk of hemorrhage with AAS depended on the period of follow-up, in a similar manner to with oral anticoagulant agents, in patients with prophylaxis of cerebral infarct. On the other hand, this did not occur with triflusal, with which the risk was homogeneous and lower in the long term.

Abstract

Forest vulnerability to drought is expected to increase under anthropogenic climate change, and drought-induced mortality and community dynamics following drought have major ecological and societal impacts. Here,we showthat tree mortality concomitant with drought has led to short-term (mean 5 y, range 1 to 23 y after mortality) vegetation-type conversion in multiple biomes across the world (131 sites). Self-replacement of the dominant tree species was only prevalent in 21% of the examined cases and forests and woodlands shifted to nonwoody vegetation in 10% of them. The ultimate temporal persistence of such changes remains unknown but, given the key role of biological legacies in long-term ecological succession, this emerging picture of postdrought ecological trajectories highlights the potential for major ecosystem reorganization in the coming decades. Community changes were less pronounced under wetter postmortality conditions. Replacement was also influenced by management intensity, and postdrought shrub dominance was higher when pathogens acted as codrivers of tree mortality. Early change in community composition indicates that forests dominated by mesic species generally shifted toward more xeric communities, with replacing tree and shrub species exhibiting drier bioclimatic optima and distribution ranges. However, shifts toward more mesic communities also occurred and multiple pathways of forest replacement were observed for some species. Drought characteristics, species-specific environmental preferences, plant traits, and ecosystem legacies govern postdrought species turnover and subsequent ecological trajectories, with potential far-reaching implications for forest biodiversity and ecosystem services.

Abstract

Mucopolysaccharidoses (MPSs) are childhood diseases caused by inherited deficiencies in glycosaminoglycan degradation. Most MPSs involve neurodegeneration, which to date is untreatable. Currently, most therapeutic strategies aim at correcting the primary genetic defect. Among these strategies, gene therapy has shown great potential, although its clinical application is challenging. We have shown previously in an MPS-IIIA mouse model that the molecular tweezer (MT) CLR01, a potent, broad-spectrum anti-amyloid small molecule, inhibits secondary amyloid storage, facilitates amyloid clearance, and protects against neurodegeneration. Here, we demonstrate that combining CLR01 with adeno-associated virus (AAV)-mediated gene therapy, targeting both the primary and secondary pathologic storage in MPS-IIIA mice, results in a synergistic effect that improves multiple therapeutic outcomes compared to each monotherapy. Moreover, we demonstrate that CLR01 is effective therapeutically in mouse models of other forms of neuronopathic MPS, MPS-I, and MPS-IIIC. Abstract

Pathogenic diseases frequently occur in drought-stressed trees. However, their contribution to the process of drought-induced mortality is poorly understood. We combined drought and stem inoculation treatments to study the physiological processes leading to drought-induced mortality in Norway spruce (Picea abies) and Scots pine (Pinus sylvestris) saplings infected with Heterobasidion annosum s.s. We analysed the saplings' water status, gas exchange, nonstructural carbohydrates (NSCs) and defence responses, and how they related to mortality. Saplings were followed for two growing seasons, including an artificially induced 3-month dormancy period. The combined drought and pathogen treatment significantly increased spruce mortality; however, no interaction between these stressors was observed in pine, although individually each stressor caused mortality. Our results suggest that pathogen infection decreased carbon reserves in spruce, reducing the capacity of saplings to cope with drought, resulting in increased mortality rates. Defoliation, relative water content and the starch concentration of needles were predictors of mortality in both species under drought and pathogen infection. Infection and drought stress create conflicting needs for carbon to compartmentalize the pathogen and to avoid turgor loss, respectively. Heterobasidion annosum reduces the functional sapwood area and shifts NSC allocation patterns, reducing the capacity of trees to cope with drought., (© 2022 The Authors. Thoracic aortic aneurysms represent a potentially life-threatening disease with increasing prevalence, probably related to the increasing mean age of the global population; the complications of thoracic aortic aneurysms can show multiple modes of presentation in the acute phase based on various factors, including the involvement of the structures with which the aneurysm is in close anatomical relationship. Abstract

Background: Rett syndrome (RTT) is a neurodevelopmental disorder caused by mutations in the X-linked methyl-CpG binding protein 2 (MeCP2) gene. While MeCP2 mutations are lethal in most males, females survive birth but show severe neurological defects. Because X-chromosome inactivation (XCI) is a random process, approximately 50% of the cells silence the wild-type (WT) copy of the MeCP2 gene. Thus, reactivating the silent WT copy of MeCP2 could provide therapeutic intervention for RTT., Methods: Toward this goal, we screened ~ 28,000 small-molecule compounds from several libraries using a MeCP2-luciferase reporter cell line and cortical neurons from a MeCP2-EGFP mouse model. We used gain/increase of luminescence or fluorescence as a readout of MeCP2 reactivation and tested the efficacy of these drugs under different drug regimens, conditions, and cellular contexts., Results: We identified inhibitors of the JAK/STAT pathway as XCI-reactivating agents, both by in vitro and ex vivo assays. In particular, we show that AG-490, a Janus Kinase 2 (JAK2) kinase inhibitor, and Jaki, a pan JAK/STAT inhibitor, are capable of reactivating MeCP2 from the inactive X chromosome, in different cellular contexts., Conclusions: Our results suggest that inhibition of the JAK/STAT pathway is a new potential pathway to reinstate MeCP2 gene expression as an efficient RTT treatment.

Abstract

Background: Phosphate is an essential plant macronutrient required to achieve maximum crop yield. Roots are able to uptake soil phosphate from the immediate root area, thus creating a nutrient depletion zone. Many plants are able to exploit phosphate from beyond this root nutrient depletion zone through symbiotic association with Arbuscular Mycorrhizal Fungi (AMF). Here we characterise the relationship between root architecture, AMF association and low phosphate tolerance in strawberries. The contrasting root architecture in the parental strawberry cultivars 'Redgauntlet' and 'Hapil' was studied through a mapping population of 168 progeny. Low phosphate tolerance and AMF association was quantified for each genotype to allow assessment of the phenotypic and genotypic relationships between traits., Results: A "phosphate scavenging" root phenotype where individuals exhibit a high proportion of surface lateral roots was associated with a reduction in root system size across genotypes. A genetic correlation between "root system size" traits was observed with a network of pleiotropic QTL found to represent five "root system size" traits. By contrast, average root diameter and the distribution of roots appeared to be under two discrete methods of genetic control. A total of 18 QTL were associated with plant traits, 4 of which were associated with solidity that explained 46% of the observed variation. Investigations into the relationship between AMF association and root architecture found that a higher root density was associated with greater AMF colonisation across genotypes. However, no phenotypic correlation or genotypic association was found between low phosphate tolerance and the propensity for AMF association, nor root architectural traits when plants are grown under optimal nutrient conditions., Conclusions: Understanding the genetic relationships underpinning phosphate capture can inform the breeding of strawberry varieties with better nutrient use efficiency. Solid root systems were associated with greater AMF colonisation. However, low P-tolerance was not phenotypically or genotypically associated with root architecture traits in strawberry plants. Furthermore, a trade-off was observed between root system size and root architecture type, highlighting the energetic costs associated with a "phosphate scavenging" root architecture.

49. Plasticity of Fine-Root Traits Under Long-Term Irrigation of a Water-Limited Scots Pine Forest.

Abstract

Trait-based approaches are increasingly used to investigate plant strategies for resource acquisition, growth, or competition between individual organisms or across species. However, the characterization of responses to environmental stimuli by fine-root systems of trees at the trait level is rather limited, particularly regarding the timing and degree of plasticity of the traits involved. These aspects become especially relevant under current climate-driven shifts in environmental conditions. In the present study, we examined the responses of the fine roots of Scots pines to increased soil water availability from long-term irrigation starting in the year 2003. The Scots pine forest is situated in a water-limited region in the central European Alps where increased tree mortality has been observed over the last two decades. The fine-root traits investigated include root system traits, root dynamic traits, architectural traits, and morphological traits. A first survey of fine-root traits in 2005 using ingrowth cores did not reveal any trait-based responses resulting from the irrigation treatment over a three-year period. Fine-root biomass, as periodically recorded by coring the topsoil from 2003 to 2016, showed a significant increase compared to the non-irrigated controls between three and nine years after the start of treatment. Overall, a maximum biomass increase due to the irrigation treatment was recorded in 2016 with about 80% higher biomass compared to controls. The analysis of fine-root traits revealed that irrigation significantly increased biomass, length, and production, but did not alter morphological and architectural traits, such as diameter, frequency of tips, specific root length (SRL), and root tissue density (RTD). In contrast, clear significant differences were found for all traits except for length when comparing the two root sampling methods, namely, ingrowth cores and soil coring. However, there were no interactions between the irrigation treatment and the sampling methods used and, therefore, the methods used did not affect the documented patterns, just the actual measured trait values.

Abstract

Background and Aims: The relationship between plant carbon economy and drought responses of co-occurring woody species can be assessed by comparing carbohydrate (C) dynamics following drought and rain periods, relating these dynamics to species' functional traits. We studied nine woody species coexisting in a continental Mediterranean shrubland that experienced severe drought effects followed by rain., Methods: We measured total non-structural carbohydrates (NSC) and soluble sugars (SS) in roots and stems during drought and after an autumn rain pulse in plants exhibiting leaf loss and in undefoliated ones. We explored whether their dynamics were related to foliage recovery and functional traits (height [H], specific leaf area [SLA], wood density [WD])., Key Results: During drought, NSC concentrations were overall lower in stems and roots of plants experiencing leaf loss, while SS decreases were smaller. Roots had higher NSC concentrations than stems. After the rain, NSC concentrations continued to decrease, while SS increased. Green foliage recovered after rain, particularly in plants previously experiencing higher leaf loss, independently of NSC concentrations during drought. Species with lower WD tended to have more SS during drought and lower SS increases after rain. In low-WD species, plants with severe leaf loss had lower NSC relative to undefoliated ones. No significant relationship was found between H or SLA and C content or dynamics., Conclusions: Our community-level study reveals that, while responses were species-specific, C stocks overall diminished in plants affected by prolonged drought and did not increase after a pulse of seasonal rain. Dynamics were faster for SS than NSC. We found limited depletion of SS, consistent with their role in basal metabolic, transport and signalling functions. In a scenario of increased drought under climate change, NSC stocks in woody plants are expected to decrease differentially in coexisting species, with potential implications for their adaptive abilities and community dynamics.
