The ways in which the accumulation of mutations might contribute to the process of aging in higher organisms or in individual clones of cells has been discussed at length. No corresponding treatment of the consequences of transcription errors in the translation of the DNA message into RNA and protein sequences seems to be available. Here I show that a consideration of the rate of accumulation of such errors leads to a paradox, the resolution of which may be relevant to the problem of aging, and that there are a number of simple experiments that should decide whether this is the case. We have used amino acids activated by carbonyldiimidazole to study the enantiospecificity of peptide elongation in aqueous solution. Peptide 'primers' Glu10 and Ala3Glu10 were elongated with the enantiomers of arginine, glutamic acid, asparagine, phenylalanine, serine and valine. The homochiral addition was always the more efficient reaction; the enantiospecificity was large in some cases but very small in others. In every case Ala3Glu10 was elongated more efficiently than Glu10.

Monomeric cysteine residues attached to cysteine-containing peptides by disulfide bonds can be activated by carbonyldiimidazole. If two monomeric cysteine residues, attached to a 'scaffold' peptide Gly-Cys-Glyn-Cys-Glu10, (n = 0, 1, 2, 3) are activated, they react to form the dipeptide Cys-Cys. in 25-65% yield. Similarly, the activation of a cysteine residue attached to the 'scaffold' peptide Gly-Cys-Gly-Glu10 in the presence of Arg5 leads to the formation of Cys-Arg5 in 50% yield. The significance of these results for prebiotic chemistry is discussed.

Unlike glutamic acid, L-gamma-carboxyglutamic acid does not oligomerize efficiently when treated with carbonyldiimidazole in aqueous solution. However, divalent ions such as Mg2+ catalyze the reaction, and lead to the formation of oligomers in good yield. In the presence of hydroxylapatite, L-gamma-carboxyglutamic acid oligomerizes efficiently in a reaction that proceeds in the absence of divalent ions but is further catalyzed when they are present. After 'feeding' 50 times with activated amino acid in the presence of the Mg2+ ion, oligomers longer than the 20-mer could be detected. The effect of hydroxylapatite on peptide elongation is very sensitive to the nature of the activated amino acid and the acceptor peptide. Glutamic acid oligomerizes more efficiently than L-gamma-carboxyglutamic acid on hydroxylapatite and adds more efficiently to decaglutamic acid in solution. One might, therefore, expect that glutamic acid would add more efficiently than L-gamma-carboxyglutamic acid to decaglutamic acid on hydroxylapatite. The contrary is true--the addition of L-gamma-carboxyglutamic acid is substantially more efficient. This suggests that oligomerization on the surface of hydroxylapatite depends on the detailed match between the structure of the surface of the mineral and the structure of the oligomer.

Photooxidation and consequent cleavage of oligodeoxynucleotides by the uranyl ion is greatly enhanced in the presence of a terminal 5'- or 3'-phosphate group. This enhanced cleavage is confined to nearby phosphodiester bonds in the case of a 5'-phosphate, but is less localized in the case of a 3'-phosphate. Several attempts to use a complementary oligodeoxynucleotide to direct a uranyl "warhead" against an oligodeoxynucleotide target were unsuccessful. Our results are most easily explained if we suppose that uranyl ions form coordination complexes with terminal phosphates and that, on photoexcitation, coordinated uranyl ions extract a hydrogen atom from the CH bond of a nearby deoxyribose residue.

Molecular orbital theory has been used to describe the electronic structure of ferrocene and related compounds. It is shown that, provided the valence‐state promotion energies are considered, the thermochemistry and magnetic properties may be understood.

The electronic transitions observed in complexes of the transition‐metal ions are interpreted in terms of a slightly modified crystal‐field theory. Parameters of chemical interest are derived.

The distortions from cubic symmetry which occur in certain transition-metal oxides, particularly those with the spinel structure, are discussed in terms of crystal (ligand) field theory. It is shown that many such distortions, including all the large ones, are related in a simple fashion to the electronic configuration of the metal ion and may be considered to arise as a consequence of a Jahn-Teller type of distortion.

The spectrum of the hydrated manganous ion is interpreted in detail and the factors influencing the widths of the bands are deduced. The band widths in other transition‐metal spectra are shown to be explicable in the same way.

Detailed X-ray analysis of crystals of ferrocene by Dunitz, Orgel & Rich confirms that the molecule has a 'sandwich' structure with approximate D5d symmetry, and Fe-C, 2.045±0.01 A; C-C, 1.403±0.02 A.

Poly(C, G) random copolymer templates direct the oligomerization of 2-Me-ImpG and 2-MeImpC, resulting in the production of a variety of oligo(G, C)s. The efficiency of monomer incorporation into newly synthesized oligomers is greater for 2-MeImpG than for 2-MeImpC, and decreases for both monomers as the guanine content of the template increases. The relatively low efficiency of oligomerization on guanine-rich templates is largely a consequence of intra- and intermolecular template self-structure. The problem of template self-structure is clearly a major obstacle to the development of a system of self-replicating polynucleotides. The distribution of oligomeric products can be characterized in detail using high-pressure liquid chromatography on an RPC-5 column. Oligomers are separated on the basis of chain length, base composition and phosphodiester-linkage isomerism. Oligomers up to about the 12-mer, with base composition Gn, Gn-1C and Gn-2C2, have been identified. The 3' to 5' regiospecificity of the products is high, particularly for oligomers with base composition Gn.

Poly(C,A) random copolymer templates direct the oligomerization of 2-MeImpG (2-MeImpX is the 5'-phospho-2-methylimidazolide of the nucleoside X) and 2-MeImpU, resulting in the production of a variety of oligo (G,U)s. This reaction is less efficient than comparable reactions involving poly(C,U) or poly(C,G) templates. The efficiency of monomer incorporation into newly synthesized oligomers is lower for 2-MeImpU than 2-MeImpG, and cannot be improved by increasing the concentration of 2-MeImpU relative to 2-MeImpG. This suggests that RNA templates containing runs of consecutive adenine residues would not be suitable for use in a chemical self-replicating system. The distribution of oligomeric products can be characterized in detail using high-pressure liquid chromatography on an RPC-5 column. Oligomers are separated on the basis of chain length, base composition, and phosphodiester-linkage isomerism. Oligomers up to about the 13-mer, with base composition Gn, Gn-1, U, and Gn-2, U2, have been identified.

The relatively poor incorporation of adenine is partly due to an intrinsically less efficient incorporation reaction, and partly due to the masking of uracil sites by G X U non-complementary pairing. The efficiency of adenine incorporation can be improved by decreasing the concentration of 2-MeImpG and increasing the concentration of 2-MeImpA in the reaction mixture. The oligomeric product distribution can be characterized in detail using high-pressure liquid chromatography on an RPC-5 column. Oligomers are separated on the basis of chain length, base composition, and phospho-diester-linkage isomerism. The 3'----5' regiospecificity of monomer addition to template-bound oligomers is lower for 2-MeImpA than for 2-MeImpG. The presence of an adenine residue at the 2'(3') terminus of the acceptor strand lowers the regiospecificity of 2-MeImpA addition even further.

THE striking shift in the position of the visible absorption bands of dicyano bis (1 : 10 phenanthroline) iron(II) and the corresponding 2-2′-dipyridyl complex caused by the addition of acid was first noted by Schilt1, who has since shown that these compounds react reversibly to give mono- and di-protonated ions2. Schilt suggested that the hydrogen is attached directly to the metal in the protonated species. A second possibility is that the protonated compounds are ‘isocyanides’.
