Background The pathogenesis of inflammatory bowel diseases (IBD) such as Crohn’s disease (CD) and ulcerative colitis (UC) is still incompletely understood. However, despite having been described back in the 1980s and reported to be involved in chronic inflammatory arthritis, the role of interleukin 3 (IL-3) signaling via the IL-3 receptor (IL-3R) in the development and perpetuation of chronic intestinal inflammation remains largely unexplored. In this study, we therefore aimed to explore the role of IL-3 signaling via IL-3R in experimental colitis as well as human IBD. Methods We analysed IL-3 expression in lamina propria mononuclear cells (LPMCs) from patients with IBD and from mice with experimental colitis. We compared the development of T cell transfer colitis in Rag1-/- mice after transfer of Il3r-/- and Il3r+/+ naïve T cells and investigated the underlying mechanisms by immunofluorescence and functional cell trafficking assays. The mechanical properties of Il3r-/- and Il3r+/+ T cells were determined by real-time deformability cytometry (RT-DC) and atomic force microscopy (AFM) and cytoskeleton structure was analysed by scanning electron microscopy (SEM) and fluorescence recovery after photobleaching (FRAP). Results The expression of IL-3 and IL3+ T cells were increased in the gut of patients with IBD and IL-3 expression was associated to shorter flare-free survival. In vivo, experimental chronic colitis upon T cell transfer was exacerbated in the absence of IL-3 or IL-3R signalling. This was attributable to IL-3-induced changes in kinase signalling and actin cytoskeleton structure, resulting in increased mechanical deformability and enhanced egress of regulatory T (Treg) cells from the inflamed colon mucosa. Conclusion We uncover a crucial role of IL-3R signalling in regulating TReg mechanobiology and tissue egress. Together, our data demonstrate a central – probably counter-regulatory – role for IL-3 receptor signalling in Treg cells in the pathogenesis of murine and human chronic intestinal inflammation and therefore suggest that IL-3 might be a novel target for future therapeutic approaches in IBD.

COVID-19 and influenza have simultaneously emerged in many regions of the world. COVID-19 and influenza are similar in terms of transmission mode, clinical symptoms and other aspects. There are also similarities in the mechanism of influenza virus and novel coronavirus on cells. At the same time, it is feasible and significant to do a good job in the prevention and control of COVID-19 and influenza. To study the clinical features of myeloperoxidase(MPO) antineutrophil cytoplasmic antibody (ANCA) associated hypertrophic pachymeningitis (HP). Clinical data of 15 cases diagnosed with MPO-ANCA vasculitis complicated with HP were retrospectively analyzed. Nine cases were males and the other 6 were females, with an average age of (58±8) years. All cases presented with chronic headache. Contrast-enhanced magnetic resonance imaging (MRI) scan showed local or diffused thickening of cerebral and/or spinal dura matter while brain parenchyma were normal. Nine cases developed multiple cranial nerve paralysis, with trigeminal nerve and auditory nerve involved most commonly. The main clinical manifestations were facial pain, hearing loss and tinnitus. Two cases were complicated with hypertrophic spinal pachymeningitis (HSP) and 4 cases were complicated with pulmonary diseases. The isomer depletion of ^{93m}Mo was recently reported [Chiara et al., Nature (London) 554, 216 (2018)NATUAS0028-083610.1038/nature25483] as the first direct observation of nuclear excitation by electron capture (NEEC). However, the measured excitation probability of 1.0(3)% is far beyond the theoretical expectation. In order to understand the inconsistency between theory and experiment, we produce the ^{93m}Mo nuclei using the ^{12}C(^{86}Kr,5n) reaction at a beam energy of 559 MeV and transport the reaction residues to a detection station far away from the target area employing a secondary beam line. The isomer depletion is expected to occur during the slowdown process of the ions in the stopping material. In such a low γ-ray background environment, the signature of isomer depletion is not observed, and an upper limit of 2×10^{-5} is estimated for the excitation probability. This is consistent with the theoretical expectation. Our findings shed doubt on the previously reported NEEC phenomenon and highlight the necessity and feasibility of further experimental investigations for reexamining the isomer depletion under low γ-ray background.

We report on the design, construction, and performance of a custom apparatus built to measure the frequency- and temperature-dependent absorptivity of millimeter-wave light by cosmic analog dusts. We highlight the unique challenges faced as well as a few key innovations that are part of the instrument. Among those is an ultra-compact Fourier transform spectrometer. We have measured its effective frequency range and FWHM resolution to be 150–2100 GHz and ∼ 45 G H z , respectively. Another innovation is a cold sample positioner whose temperature can be controlled within the range of 3.7–50 K. The use of a pulse-tube cryocooler results in a pulse-synchronous signal that dominates the detector (bolometer) signal. Methods used to address that challenge are also presented.

To investigate the heterogeneity of decidual endothelial cells and their changes during delivery.Single-cell RNA sequencing was used to characterize the transcriptomes of decidual endothelial cells before and after the onset of labor.Decidual endothelial cells (9748 cells) were divided into five subgroups with different functions according to differences in the transcriptome. The functions of cluster 5 were enriched in vascular development and response to growth factors. After the onset of labor, the activities of each cluster were different, including the interleukin 17 pathway and regulation of ERK1 and ERK2 cascade. The downregulated genes were related to scavenger receptor (cluster 5), which may reflect the process of endothelial activation. In terms of genetic changes, cluster 5 may be more actively involved in labor than the other clusters.Peripartum decidual endothelial cells are heterogeneous and participate in labor to varying degrees. One of the five subtypes of endothelial cells may be more actively involved in labor onset. Our findings may enable the assessment of decidual endothelial cells and labor onset.

To investigate the influences of adiponectin (APN) on the liver injury in sepsis rats and to explore whether it exerts a therapeutic effect through the adenosine monophosphate-activated protein kinase (AMPK)/mammalian target of rapamycin (mTOR) pathway.A rat model of sepsis was established through cecal ligation and puncture (CLP) (CLP group), and APN treatment group (APN group) and control group were also set. The changes in the liver function-related indicators, serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST), were determined by automatic biochemistry analyzer, and the levels of tumor necrosis factor-α (TNF-α), interleukin (IL)-1, and IL-6 were measured via enzyme-linked immunosorbent assay (ELISA). Hematoxylin-eosin (HE) staining was employed to detect liver tissue injury, and the hepatocyte apoptosis and necrosis after intervention with APN were evaluated using in situ fluorescence staining. Moreover, the mRNA expression of APN in liver tissues was detected via quantitative Reverse Transcription-Polymerase Chain Reaction (qRT-PCR), and the expression levels of phosphorylated AMPK and mTOR proteins in liver tissue samples were determined using Western blotting.In terms of changes in liver function-related indicators, the concentrations of ALT and AST were substantially raised in the CLP group, and compared with those in the control group, the concentrations of the two indicators significantly declined in the APN group, showing statistically significant differences (p0.05). CLP and APN group had evidently higher levels of inflammatory factors than the control group, but their levels in APN group were notably lower than those in the CLP group (p0.05). It was found through the HE staining that the sepsis rats in CLP group had massive inflammatory cell infiltration, and that the inflammatory cells were remarkably decreased in the APN group after APN treatment. According to the in-situ fluorescence staining detection results, CLP group exhibited a notable increase in the cell apoptosis rate, and APN group had substantially reduced apoptotic cells (p0.05). The determination results of APN expression revealed that CLP group had a lowered level of APN, and that the level of APN in APN group was markedly higher than that in the control group. Based on the results of Western blotting, the level of phosphorylated AMPK was remarkably elevated, and that of phosphorylated mTOR was lowered in the CLP group compared with those in the control group, while in comparison with CLP group, APN group showed a considerable elevation of phosphorylated AMPK level and a distinct decline in the phosphorylated mTOR level.APN can activate the AMPK/mTOR pathway and reduce hepatocyte apoptosis to alleviate liver injury in sepsis rats.

The aim of this study was to investigate the effect of micro-ribonucleic acid-195 (miR-195) on myocardial fibrosis in hypertensive rats through the transforming growth factor beta 1 (TGFβ1)-Smad3 signaling pathway.Spontaneously hypertensive rats (SHRs) were selected in this study to establish the animal model. The content of miR-195 in the model group and control group was measured, respectively. Arterial blood pressure, liver function and myocardial function in the two groups were detected and examined. Pathological changes in rat myocardial tissues were detected via hematoxylin-eosin (HE) staining. After that, myocardial fibroblasts were collected and added with miRNA inhibitors and mimics to suppress and overexpress miR-195. Thereafter, Reverse Transcription-Polymerase Chain Reaction (RT-PCR) and Western blotting were employed to detect the mRNA and protein expression levels of checkpoint kinase 1 (Chek1) and alpha-smooth muscle actin (α-SMA) (important molecules for proliferation and differentiation of myocardial fibroblasts), as well as the related pathway TGFβ1-Smad3. Furthermore, the effects of miR-195 on myocardial fibrosis in hypertensive rats via the TGFβ1-Smad3 signaling pathway were comprehensively observed.Serum alkaline phosphatase (ALP), glutamic pyruvic aminotransferase (ALT) and creatine kinase (CK) levels in the SHR group were significantly higher than those of the normal group. Cardiac function examination showed that SHR group had significantly reduced fractional shortening (FS, %) and ejection fraction (EF, %) in comparison with the normal group. However, systolic blood pressure, diastolic blood pressure, left ventricular end-diastolic dimension (LVEDd) and left ventricular end-systolic dimension (LVESd) were markedly elevated in the SHR group. In addition, the miR-195 expression level was remarkably reduced in hypertensive rats. Histopathological changes in rat myocardial tissues were detected through HE staining. The results showed that the normal group had orderly arranged myocardial cells. However, SHR group showed disorderly arranged myocardial cells, thickened myocardial fibers and myocardial fibrosis. RT-PCR assay results revealed that the mRNA levels of Collagen, Chek1, α-SMA, TGFβ1 and Smad3 in rat myocardial fibroblasts were significantly reduced in Mimics group (p0.05) and increased in Inhibitors group (p0.05). Western blotting results demonstrated that, compared with the control group, the protein levels of α-SMA, TGFβ1 and Smad3 in rat myocardial cells decreased significantly in Mimics group (p0.05). Opposite results were observed in Inhibitors group (p0.05). The above results suggested that overexpression of miR-195 inhibited the expressions of TGFβ1-Smad3 signaling pathway and related molecules, further repressing myocardial fibrosis.MiR-195 participates in the development and progression of myocardial fibrosis in hypertensive rats through the TGFβ1-Smad3 signaling pathway. Furthermore, this can inhibit the development of myocardial fibrosis in hypertensive rats and prevent myocardial diseases.

Glioma is a common aggressive cancer and a major public health problem worldwide, with high incidence, recurrence, and mortality. Circular RNA (circRNA) has been reported to be involved in glioma, but the role of circ_0079593 in glioma is still unclear.The real-time quantitative polymerase chain reaction (RT-qPCR) was performed to quantify the expression levels of circ_0079593, miR-499a-5p, and karyopherin alpha 2 (KPNA2) in glioma tissues or cells. The protein expression level of KPNA2 was assessed by Western blot. 3-(4, 5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazol-3-ium bromide (MTT), flow cytometry, and transwell assays were conducted to evaluate proliferation, apoptosis, migration and invasion of glioma cells, respectively. The relationship between miR-499a-5p and circ_0079593 or KPNA2 was analyzed by bioinformatics database and confirmed by Dual-Luciferase reporter analyses, respectively. The effects of circ_0079593 silencing in vivo were measured by a xenograft experiment.Circ_0079593 and KPNA2 were elevated in glioma tissues and cells. Loss-of functional experiments revealed that knockdown of circ_0079593 hampered the progression of glioma by repressing proliferation, motility and inducing apoptosis in vitro and declining tumor growth in vivo. Similarly, suppression of KPNA2 impeded the process of glioma by inhibiting proliferation, motility and increasing apoptosis. MiR-499a-5p, interacting with KPNA2, was a target gene of circ_0079593. In addition, overexpression of KPNA2 could reverse the effects of circ_0079593 knockdown on proliferation, apoptosis, migration and invasion of glioma cells. Mechanistically, circ_0079593 mediated proliferation, motility and apoptosis of glioma cells by regulating KPNA2 expression via sponging miR-499a-5p.Circ_0079593 stimulated the pathological process of glioma via acting as competing endogenous RNA to sponge miR-499a-5p.

A novel Cu(I) coordination polymer [Cu2(MA)2Cl2]n (1, MA is melamine) is reported here. It is synthesized by hydrothermal reactions of CuCl and MA. Moreover, this compound shows an excellent photocatalytic effect for methyl orange and methyl blue degradation. The biological activity of the compound on the blood glucose regulation is evaluated. First of all, real time RT-PCR is exploited to measure the aldose reductase activity. Then, the blood glucose concentration released into the plasma is determined. The CCK-8 is used to measure the toxicity of the new compound on normal liver cells.

Endometrial cancer (EC) is one of the three most common gynecological cancers. Due to the lack of effective treatment for EC patients in an advanced stage, the mortality rate of EC is increasing rapidly. Hence, it is essential to seek for novel molecular therapeutic targets and biomarkers for EC. The aim of this study was to explore the role of miR-218 in the occurrence and development of EC and to investigate the possible underlying mechanism.The expression level of miR-218 in EC tissues and cell lines was detected by quantitative Real Time-Polymerase Chain Reaction (qRT-PCR). Wound healing assay and Matrigel assay were performed to determine the migration and invasion abilities of EC cells. Meanwhile, the potential targets of miR-218 were predicted by bioinformatics analysis and confirmed by Luciferase reporter gene assay. In addition, the protein expression level of Adducin 2 (ADD2) was assessed by Western blotting analysis.QRT-PCR results revealed that miR-218 was significantly downregulated in EC tissues and cell lines. Wound healing assay and Matrigel assay demonstrated that miR-218 suppressed the migration and invasion abilities of EC cells. Online prediction databases predicted that ADD2 was a direct target of miR-218, which was verified by Luciferase reporter gene assay. Rescue experiments further validated that miR-218 could serve as a carcinoma suppressor by negatively regulating ADD2 expression in EC.In the present study, we elucidated that miR-218 served as a tumor suppressor in EC by negatively regulating ADD2. This might bring a novel insight into new molecular therapeutic targets and biomarkers for EC.

目的 探讨重型地中海贫血（地贫）患者异基因造血干细胞移植（allo-HSCT）后并发自身免疫性溶血性贫血（AIHA）诊断、治疗及转归，以提高地贫患者造血干细胞移植疗效。 方法 回顾性分析2007年7月至2017年12月共计291例行allo-HSCT重型地贫患者的临床资料。 结果 重型地贫allo-HSCT后AIHA发生率为1.72%（5/291）；AIHA中位发生时间为移植后7（5~12）个月，5例移植后AIHA患者直接和间接Coombs试验均阳性，患者主要表现为头晕、乏力、面色苍白、皮肤巩膜黄染、酱油色尿。228例HLA相合同胞供者移植患者有1例（0.43%）移植后发生AIHA，而63例非亲缘供者移植患者有4例（6.36%）移植后发生AIHA。非亲缘供者移植患者AIHA发生率高于HLA相合同胞供者移植患者。1例患者单用泼尼松治疗死亡，4例患者采用甲泼尼龙联合利妥昔单抗治疗有效，目前生存良好，其中2例Coombs试验转阴。 结论 该组重型地贫患者allo-HSCT后AIHA发生率为1.72%，Coombs试验有助于诊断移植后AIHA，非亲缘供者移植病例移植后AIHA发生率高于HLA相合同胞供者移植组，利妥昔单抗联合糖皮质激素是治疗重型地贫allo-HSCT后AIHA的有效方法。

The coronal heating region is able to generate mirror mode structures by ion mirror instabilities. Linear magnetic holes are believed to be the remnants of mirror mode structures, thus they are believed to be messengers from the coronal heating region. They can be convected to ∼9 au with the solar wind flow, indicating that a stabilizing mechanism is necessary to make the magnetic holes survive for such a long time. Here, we investigate a magnetic hole with a size of ∼6.7 ρ i in the solar wind based on observations by the Magnetospheric Multiscale mission. The unprecedented high-resolution data enable us to reveal the existence of the ion vortex inside the structure for the first time. Such an ion vortex forms a ring-like current, which is consistent with the magnetic field depression. The self-consistent structure of the magnetic hole contributed by the ion vortex can help to further shed light on the mechanism of the long-term survival of magnetic holes in the astrophysical plasma.

Electron-scale magnetic holes filled with high-energy electrons can provide a seed population of electrons in the magnetosphere and might play an important role in the interaction between the magnetosphere and solar wind. Theoretical studies have investigated their generation mechanisms based on the 1D or 2D geometry of the structure. However, the generation mechanism is still unclear. Here we report on the 3D geometry of the electron-scale magnetic hole in the solar wind based on the Magnetospheric Multiscale mission. We find that the cross section of the magnetic hole with a size of ∼0.2–0.6 ρ i (ion gyroradius) is either circular or 2D sheet-like. Electron vortices exist in both kinds of cross sections. The ellipse is a possible candidate for the geometry of the magnetic hole in the plane including its axis. Surprisingly, such an elliptical geometry suggests that the axial lengths of all our selected magnetic holes are ∼1–2 ρ i. This 3D geometry might shed some light on the generation mechanism and role of the electron-scale magnetic hole in the astrophysical plasma.

Moving object detection based on color information is easily affected by illumination changes and shadows in complex scenes. Depth information can provide complementary information. In the paper, a novel method is presented by using color and depth information. Firstly, we improve the codebook algorithm by fusing the depth information as the fourth channel in the code word. Next, a compensation factor algorithm is presented to make the edges accurate. So the final detection result can be obtained by logic operation. Experiments adapt the public datasets, and experimental results show that the proposed method can successfully cope with the limitations of color-based or depth-based detection

To understand the value of high frequency ultrasound in the clinical screening of parathyroid gland, and to summarize the intrinsic relationship between primary hyperparathyroidism and recurrent urinary calculi.98 cases of urinary calculi were randomly selected, and the patients were admitted to our hospital from March 2014 to August 2017. A total of 100 healthy subjects were selected as group B in the same period. High frequency color Doppler ultrasonography scan recorded the results.Among the subjects in group A, 67 (68.37%) showed parathyroid gland, 14 cases (14.29%) had tumor mass in the parathyroid system, 40 cases more than those in group B (40.00%) and 2 cases (2.00%), (p0.05). There were 10 cases (10.20%) of primary hyperparathyroidism in group A and no cases of primary hyperparathyroidism in group B (p0.05). The occurrence of primary hyperparathyroidism was 26.92% (7/26) in the number of cases, with 3 and more cases of urinary calculi, which was higher than that in the first recurrent cases (3/72), (p0.005).One of the key causes of recurrent episodes of urinary calculi is primary hyperparathyroidism, which can be applied to high frequency ultrasonography to develop professional screening of parathyroid gland in cases of urinary calculi.

Study on CO2 flux of saline and alkali soil in arid area is of great significance to greenhouse gas control. At present, the main research contents at home and abroad focus on the ways of CO2 sequestration and migration and transformation in saline-alkali soils in arid areas. The influencing factors of CO2 flux in saline-alkali soil in arid area include soil temperature, humidity, pH, EC and soil physical and chemical properties. Inorganic carbon equilibrium process of CO2(g)—CO2 (aq)—HCO3- (aq)—CaCO3(s) is common in soil, its dynamic process dominates the inorganic carbon exchange at the ground-gas interface. This is of great significance to the study of soil-atmospheric carbon circulation system for exploring the principle and mechanism of greenhouse gas change in order to slow down global warming and strengthen the management and control of greenhouse gases. It contributes to China’s energy conservation and emission reduction planning and international carbon balance negotiations.

To calculate resting energy expenditure (REE) in patients receiving invasive mechanical ventilation and compare different predictive equations with indirect calorimetry(IC).A total of 60 patients in intensive care unit(ICU) were enrolled. Measure calculating daily REE in the first week included IC, Harris-Benedict formula, Penn State formula and Swinamer formula. 目的 通过剂量递增的Ⅰ期临床试验，评价重组人-鼠嵌合抗CD20单克隆抗体注射液IBI301单次给药在经治疗达客观缓解的CD20+ B细胞非霍奇金淋巴瘤患者中的耐受性、安全性及药效学特征。 方法 纳入9例受试者，按剂量递增原则设125、375和500 mg/m2三个剂量组，每组3例逐组完成试验。3组受试者均接受单次IBI301静脉输注，125、375、500 mg/m2组IBI301的中位暴露量分别为243、690、980 mg。观察IBI301给药后不良事件（AE）发生情况。采用流式细胞术检测外周血CD19+、CD20+ B细胞比例，采用速率散射比浊法检测血清IgG、IgM水平，进行药效学评价。 结果 9例受试者发生52例次AE，包括：125 mg/m2组18例次，375 mg/m2组14例次，500 mg/m2组20例次。其中不良反应共26例次，22例次与试验药物可能有关，4例次可能无关；对症处理后均消失或恢复至基线水平。常见的AE包括WBC降低、上呼吸感染、ANC降低、消化不良、高尿酸血症、感觉异常、口腔黏膜炎和头晕。研究中未出现AE导致的用药暂停、受试者退出或试验提前中止，未发生严重AE（SAE）及剂量限制性毒性（DLT）事件。所有受试者CD20+、CD19+ B细胞比例均有不同程度下降，而IgG和IgM水平无明显变化。 结论 IBI301耐受性良好，受试者出现的不良反应均恢复，未发生SAE及DLT事件。IBI301对CD20+ B细胞有较好的清除作用。
