The equatorial orientation of reproductive structures is known in some columnar cacti from extratropical deserts. It has been hypothesised that photosynthetically active radiation (PAR) interception is the main reason for this orientation, because of its key effect on nocturnal CO2 uptake. However, there are no studies addressing both the effect of PAR and its consequence, carbon gain, on fruit orientation. Accordingly, we tested whether PAR and carbon gain could explain the southern fruit orientation of Myrtillocactus geometrizans, an inter-tropical columnar cactus., We studied three populations of M. geometrizans in Mexico. For each population, azimuth of fruits, total daily PAR, nocturnal acid accumulation (NAA) and fruit production were measured. The relationships between rib orientation and number of fruits, as well as total daily PAR, were evaluated using periodic regressions. The effect of total daily PAR and NAA on number of fruits was assessed using generalised linear models., During spring, mean fruit orientation had a south azimuth for three populations. Likewise, rib orientation had a significant effect on fruit production, with the south-facing ribs having the maximum number of fruits. Total daily PAR was highest in the south-facing ribs, at least for those in the northern and central populations. Furthermore, during spring, there was a significant positive effect of total daily PAR and NAA on fruit production., Our results provide strong evidence that the higher carbon gain in equatorial ribs, through a highest interception of PAR, would be the responsible factor for equatorial orientation of fruits in an inter-tropical columnar cactus. Introduction: The self-incompatibility system in sweet cherry ( Prunus avium L.) prevents fertilization with own or genetically related pollen, and is genetically determined by the multi-allelic S -locus. Therefore, determining S -alleles is crucial for plant breeding and fruit production, as it enables the selection of compatible combinations of S -genotypes for successful pollination., Methods: In this study, S -alleles were identified in a total of 260 genotypes from the Caucasian region, the species' center of origin. S -allele genotyping was conducted using PCR fragment length analysis with the standard marker PaConsI-F/R2 and reference genotypes, complemented by sequence analysis through amplicon deep sequencing., Results and Discussion: The genotypes collected from Azerbaijan and Turkey exhibit a high allelic richness at the S -locus, particularly compared to modern sweet cherry cultivars worldwide. Nine previously undescribed S -alleles were identified and designated as S 45 , S 46 , S 47 , S 48 , S 49 , S 50 , S 51 , S 52 and S 53 . Given the expected high diversity for other traits, this plant material represents a valuable resource for further breeding research and introgression of new traits in future breeding programs. Furthermore, our results underscore that fragment length alone may not be sufficient for unambiguous assignment of S -alleles due to minimal length differences between different alleles. To address this issue, an S -allele reference ladder was developed using the rich diversity for precise assignment of the S -alleles. This tool can be applied in future experiments as a robust and cost-effective method for accurate S -genotyping across different runs and laboratories. Periglischrus calcariflexus new species, was found on the Greater Long-tongued bat Leptonycteris nivalis (Saussure) in Mexico. The female, male, deutonymphs, and protonymph are described and illustrated. Additionally, we evaluate the morphological variability of this species based on its geographical distribution. Considering this new species, the number of species in the vargasi species group within the genus Periglischrus increases to five; therefore, herein we include a key for the known species in the vargasi species group for the Neotropics., (© The Author(s) 2022. High CO 2 is able to ameliorate some negative effects due to climate change and intensify others. This study involves the sweet cherry (Prunus avium) cultivar 'Burlat' grafted on the 'Mariana 2624', 'Adara' and 'LC 52' rootstocks. In a climate chamber at two CO 2 concentrations, ambient (400 µmol mol -1 ) and elevated (800 µmol mol -1 ), the plants were submitted to waterlogging for 7 d, followed by 7 d of recovery after drainage. Waterlogging drastically decreased the rate of photosynthesis, significantly endangering plant survival, particularly for the 'LC 52' and 'Adara' rootstocks. 'Mariana 2624' was also clearly affected by waterlogging that increased lipid peroxidation and the Cl - and SO 4 2- concentrations in all the studied plants. Nevertheless, CO 2 was able to overcome this reduction in photosynthesis, augmenting growth, increasing soluble sugars and starch, raising turgor and regulating the concentrations of Cl - and SO 4 2- , while lowering the NO 3 - concentration in leaves of all the studied rootstocks. In concordance with these results, the proline levels indicated a more intense stress at control CO 2 than at high CO 2 for waterlogged plants. The increase in the ambient concentration of CO 2 and other greenhouse gases is producing climate events that can compromise crop survival. However, high CO 2 concentrations are sometimes able to mitigate certain stresses such as salinity or drought. In this experiment, the effects of waterlogging and CO 2 are studied in combination to elucidate the eventual response in sweet cherry trees. For this purpose, four sweet cherry cultivars ('Burlat', 'Cashmere', 'Lapins and 'New Star') were grafted on a typically hypoxia-tolerant rootstock (Mariana 2624) and submitted to waterlogging for 7 days at either ambient CO 2 concentration (400 µmol mol -1 ) or at elevated CO 2 (800 µmol mol -1 ). Waterlogging affected plants drastically, by decreasing photosynthesis, stomatal conductance, transpiration, chlorophyll fluorescence and growth. It also brought about the accumulation of proline, chloride and sulfate. Nonetheless, raising the CO 2 supply not only mitigated all these effects but also induced the accumulation of soluble sugars and starch in the leaf. Release of bud dormancy in perennial woody plants is a temperature-dependent process and thus flowering in these species is heavily affected by climate change. The lack of cold winters in temperate growing regions often results in reduced flowering and low fruit yields. This is likely to decrease the availability of fruits and nuts of the Prunus spp. in the near future. In order to maintain high yields, it is crucial to gain detailed knowledge on the molecular mechanisms controlling the release of bud dormancy. Here, we studied these mechanisms using sweet cherry ( Prunus avium L.), a crop where the agrochemical hydrogen cyanamide (HC) is routinely used to compensate for the lack of cold winter temperatures and to induce flower opening. In this work, dormant flower buds were sprayed with hydrogen cyanamide followed by deep RNA sequencing, identifying three main expression patterns in response to HC. These transcript level results were validated by quantitative real time polymerase chain reaction and supported further by phytohormone profiling (ABA, SA, IAA, CK, ethylene, JA). Using these approaches, we identified the most up-regulated pathways: the cytokinin pathway, as well as the jasmonate and the hydrogen cyanide pathway. Our results strongly suggest an inductive effect of these metabolites in bud dormancy release and provide a stepping stone for the characterization of key genes in bud dormancy release.

Climate change is submitting countries of the Mediterranean arc to periods of drought alternating with heavy rain and waterlogging. Eventual floods along with the rising CO 2 in the atmosphere present an unpredictable scenario that affects crop survival. The effect of both stresses combined has been studied in sweet cherry plants. 'Gisela 5' and 'Gisela 6' were evaluated as rootstocks of the sweet cherry cultivar 'Burlat'. Plants were placed in a controlled-climate chamber for 7days, then they were submitted to waterlogging for another 7days and the response to this stress and the subsequent recovery were studied (7 more days). The experiment was carried out at 400μmolmol -1 CO 2 (ambient CO 2 ) and 800μmolmol -1 CO 2 , at 26°C, and plant water status and growth, net CO 2 assimilation, transpiration, stomatal conductance, water potential, chlorophyll fluorescence, relative water content, anions content, proline, lipid peroxidation, soluble sugars, and starch were measured. Differences in the response and in its intensity were detected in both rootstocks. Some parameters - such as photosynthesis, soluble sugars, starch, TBARS, and NO 3 - - varied depending on the CO 2 conditions and the waterlogging effect. Historic demography changes of plant species adapted to New World arid environments could be consistent with either the Glacial Refugium Hypothesis (GRH), which posits that populations contracted to refuges during the cold-dry glacial and expanded in warm-humid interglacial periods, or with the Interglacial Refugium Hypothesis (IRH), which suggests that populations contracted during interglacials and expanded in glacial times. These contrasting hypotheses are developed in the present study for the giant columnar cactus Cephalocereus columna-trajani in the intertropical Mexican drylands where the effects of Late Quaternary climatic changes on phylogeography of cacti remain largely unknown. In order to determine if the historic demography and phylogeographic structure of the species are consistent with either hypothesis, sequences of the chloroplast regions psbA-trnH and trnT-trnL from 110 individuals from 10 populations comprising the full distribution range of this species were analysed. Standard estimators of genetic diversity and structure were calculated. The historic demography was analysed using a Bayesian approach and the palaeodistribution was derived from ecological niche modelling to determine if, in the arid environments of south-central Mexico, glacial-interglacial cycles drove the genetic divergence and diversification of this species. Results reveal low but statistically significant population differentiation (FST = 0.124, P < 0.001), although very clear geographic clusters are not formed. Genetic diversity, haplotype network and Approximate Bayesian Computation (ABC) demographic analyses suggest a population expansion estimated to have taken place in the Last Interglacial (123.04 kya, 95% CI 115.3-130.03). The species palaeodistribution is consistent with the ABC analyses and indicates that the potential area of palaedistribution and climatic suitability were larger during the Last Interglacial and Holocene than in the Last Glacial Maximum. Overall, these results suggest that C. columna-trajani experienced an expansion following the warm conditions of interglacials, in accordance with the GRH.

Professional and scientific networks built around the production of sweet cherry (Prunus avium L.) led to the collection of phenology data for a wide range of cultivars grown in experimental sites characterized by highly contrasted climatic conditions. We present a dataset of flowering and maturity dates, recorded each year for one tree when available, or the average of several trees for each cultivar, over a period of 37 years (1978-2015). Such a dataset is extremely valuable for characterizing the phenological response to climate change, and the plasticity of the different cultivars' behaviour under different environmental conditions. In addition, this dataset will support the development of predictive models for sweet cherry phenology exploitable at the continental scale, and will help anticipate breeding strategies in order to maintain and improve sweet cherry production in Europe., Competing Interests: The authors declare no competing financial interests.

The endemic Mexican cactus, Mammillaria pectinifera, shows low dispersal capabilities and isolated populations within the highly dissected landscape of Tehuacán Valley. These characteristics can restrict gene flow and act upon the genetic divergence and speciation in arid plants. We conducted a phylogeographic study to determine if the origin, current distribution, and genetic structure of M. pectinifera were driven by Quaternary geomorphic processes. Sequences of the plastids psbA-trnH and trnT-trnL obtained from 66 individuals from seven populations were used to estimate genetic diversity. Population differentiation was assessed by an analysis of molecular variance. We applied a stepwise phylogenetic calibration test to determine whether species origin and genetic divergence among haplotypes were temporally concordant with recognizable episodes of geomorphic evolution. The combination of plastid markers yielded six haplotypes, with high levels of haplotype diversity (h = 0.622) and low nucleotide diversity (π = 0.00085). The populations were found to be genetically structured (F(ST) = 0.682; P < 0.00001), indicating that geographic isolation and limited dispersal were the primary causes of genetic population differentiation. The estimated origin and divergence time among haplotypes were 0.017-2.39 and 0.019-1.237 mya, respectively, which correlates with Pleistocene tectonics and erosion events, supporting a hypothesis of geomorphically-driven geographical isolation. Based on a Bayesian skyline plot, these populations showed long term demographic stability, indicating that persistence in confined habitats has been the main response of this species to landscape changes. We conclude that the origin and haplotype divergence of M. pectinifera were a response to local Quaternary geomorphic evolution.

The mammary gland has long drawn the attention of the scientific community due to the limited knowledge of some fundamental aspects involved in the control of its function. Myotis velifer, a microchiropteran species, provides an interesting model to study some of the regulatory factors involved in the control of the mammary gland cycle. Having an asynchronous, monoestrous reproductive pattern, female M. velifer bats undergo drastic morphological changes of the breast during the reproductive cycle. Current research on non-chiropteran mammals indicates that serotonin (5-HT) plays a major role in the intraluminal volume homeostasis of the mammary gland during lactation; however, an analysis of both the expression and localization of the main components of the serotonergic system in the bat mammary gland is lacking. Thus, the objectives of the present study were: to describe the gross and histological anatomy of the mammary gland of M. velifer to establish the lactation period for this species; to analyze the distribution and expression of the main serotonergic components in the mammary tissues of these bats under the physiological conditions of lactation, involution and the resting phase; and to provide information on the involvement of 5-HT in the regulation of the physiological function of this organ. To assess the expression and localization of serotonergic components, multiple immunofluorescence, Western blot and HPLC methods were used. 5-HT and the enzyme that catalyzes its synthesis (TPH) were located in both myoepithelial and luminal epithelial cells, while the enzyme responsible for the catabolism of this neurohormone (MAO A) was found in luminal epithelial cells as well as in secreted products. We also found an increased expression of serotonergic components during lactation, indicating that elements of the serotonergic system may play an important role in lactation in this species of bat in a way similar to that of other mammal species.

Peucetia viridans (Hentz) lives almost exclusively on Croton ciliatoglandulifer (Ortega) in the dry forests of western Mexico. This spider is usually found on pubescent shrubs. Within their host plants, P. viridans has been associated with plant height and cover, as well as with number of flowers or inflorescences in anthesis. Flowers can be used as cues of good habitat conditions or because they attract prey detected by the spider. In this study, we assessed the importance of flowers, plant cover, and plant exposure (sun/shade) on the spider distribution in five 50-plant transects. In a field experiment, we also compared the number of spiders between controls, plants from which inflorescences were removed, and plants with artificial inflorescences. The results from the transects indicate that, when the number of flowers per spider is high, spiders were more abundant in exposed locations, which presumably offer better microclimatic conditions; when flowers become scarce, food may be more difficult to find and the spider distribution become strongly associated with the number of flowers, where they are more likely to find prey. Spider abundances on the experimental plot decreased on plants from which flowers were removed in comparison to control plants. Spider abundance increased on those in which artificial inflorescences were added. The similarity between plants with natural and artificial inflorescences suggests that spiders use flowers as cues of good microhabitats instead of prey visitors, which are significantly less abundant on artificial inflorescences.
