The role of the hippocampus (Hp) in absence epileptic networks and the effect of endocannabinoid system on this network remain enigmatic. Here, using adapted nonlinear Granger causality, we compared the differences in network strength in four intervals (baseline or interictal, preictal, ictal and postictal) in two hours before (Epoch 1) and six hours (epochs 2, 3 and 4) after the administration of three different doses of the endocannabinoid agonist WIN55,212-2 (WIN) or solvent. Local field potentials were recorded for eight hours in 23 WAG/Rij rats in the Frontal (FC), Parietal PC), Occipital Cortex (OC) and in the hippocampus (Hp). The four intervals were visually marked by an expert neurophysiologist and the strength of couplings between electrode pairs were calculated in both directions. Ictally, a strong decrease in coupling strength was found between Hp and FC, as well as a large increase bidirectionally between PC and FC and unidirectionally from FC and PC to OC, and from FC to Hp over all epochs. The highest dose of WIN increased the couplings strength from FC to Hp and from OC to PC during 4 and 2 hr respectively in all intervals, and decreased the FC to PC coupling strength postictally in epoch 2. A single rat showed generalized convulsive seizures after the highest dose: this rat shared not only coupling changes with the other rats in the same condition, but showed many more. WIN reduced SWD number in epoch 2 and 3, their mean duration increased in epochs 3 and 4. Conclusions:during SWDs FC and PC are strongly coupled and drive OC, while at the same time the influence of Hp to FC is diminished. The first is in agreement with the cortical focus theory, the latter demonstrates an involvement of the hippocampus in SWD occurrence and that ictally the hippocampal control of the cortico-thalamo-cortical system is lost. WIN causes dramatic network changes which have major consequences for the decrease of SWDs, the occurrence of convulsive seizures, and the normal cortico-cortical and cortico-hippocampal interactions., (Copyright © 2023 Elsevier B.V. Purpose: Spike-and-wave discharges (SWDs) recorded in the cortical EEGs of WAG/Rij rats are the hallmark for absence epilepsy in this model. Although this type of epilepsy was long regarded as a form of primary generalized epilepsy, it is now recognized that there is an initiation zone - the perioral region of the somatosensory cortex. However, networks involved in spreading the seizure are not yet fully known. Previously, the dynamics of coupling between different layers of the perioral cortical region and between these zones and different thalamic nuclei was studied in time windows around the SWDs, using nonlinear Granger causality. The aim of the present study was to investigate, using the same method, the coupling dynamics between different regions of the cortex and between these regions and the hippocampus., Methods: Local field potentials were recorded in the frontal, parietal, and occipital cortices and in the hippocampus of 19 WAG/Rij rats. To detect changes in coupling reliably in a short time window, in order to provide a good temporal resolution, the innovative adapted time varying nonlinear Granger causality method was used. Mutual information function was calculated in addition to validate outcomes. Results of both approaches were tested for significance., Results: The SWD initiation process was revealed as an increase in intracortical interactions starting from 3.5s before the onset of electrographic seizure. The earliest preictal increase in coupling was directed from the frontal cortex to the parietal cortex. Then, the coupling became bidirectional, followed by the involvement of the occipital cortex (1.5s before SWD onset). There was no driving from any cortical region to hippocampus, but a slight increase in coupling from hippocampus to the frontoparietal cortex was observed just before SWD onset. After SWD onset, an abrupt drop in coupling in all studied pairs was observed. In most of the pairs, the decoupling rapidly disappeared, but driving force from hippocampus and occipital cortex to the frontoparietal cortex was reduced until the SWD termination., Conclusion: Involvement of multiple cortical regions in SWD initiation shows the fundamental role of corticocortical feedback loops, forming coupling architecture and triggering the generalized seizure. Frequency-temporal electroencephalogram (EEG) reactions to hypogravity were studied in 7 male subjects at the age of 20 to 27 years. The experiment was conducted using dry immersion (DI) as the best known method of simulating the space microgravity effects on the Earth. This hypogravity model reproduces hypokinesia, i.e. the weight-bearing and mechanic load removal, which is typical of microgravity. EEG was recorded by Neuroscan-2 (Compumedics) before the experiment (baseline data) and at the end of day 2 in DI. Comparative analysis of the EEG frequency-temporal structure was performed with the use of 2 techniques: Fourier transform and modified wavelet analysis. The Fourier transform elicited that after 2 days in DI the main shifts occurring to the EEG spectral composition are a decline in the alpha power and a slight though reliable growth of theta power. Similar frequency shifts were detected in the same records analyzed using the wavelet transform. According to wavelet analysis, during DI shifts in EEG frequency spectrum are accompanied by frequency desorganization of the EEG dominant rhythm and gross impairment of total stability of the electrical activity with time. Wavelet transform provides an opportunity to quantify changes in the frequency-temporal structure of the electrical activity of the brain. Quantitative evidence of frequency desorganization and temporal instability of EEG wavelet spectrograms may be the key to the understanding of mechanisms that drive functional disorders in the brain cortex in the conditions of hypogravity.

The effects of metoprine, an inhibitor of histamine N-methyltransferase, on open field activity and brain regional histamine (HA) content were examined in rats with mixed, absence and audiogenic, epilepsy (WAG/Rij-AGS), rats with audiogenic epilepsy (Wistar-AGS) and in non-epileptic control rats (Wistar-nAGS). HA content was increased by metoprine (20mg/kg, i.p.) in the cortex, striatum, thalamus, hypothalamus and hippocampus of the rats from all three tested groups. However, WAG/Rij rats showed a lower rate of metoprine-induced HA accumulation in the striatum and thalamus than Wistar rats. For the open field test, the main effect of metoprine (20mg/kg, i.p.) was a general increase of locomotor activity although distinctive features, such as hyperlocomotion and exaggerated sniffing, were characteristic for the epileptic rats (WAG/Rij-AGS and Wistar-AGS, respectively). Individual rats from all the groups showed stereotyped behavior of shuttle type and head bobbing. Electroencephalographic data obtained in WAG/Rij-AGS rats confirmed that metoprine-induced behavioral activation was accompanied by suppression of spike-wave discharges, the main hallmark of absence seizures. Taken together, these results show that inhibition of the histamine catabolism may induce motor activation of particular patterns in epileptic rats and provoke stereotyped behavior., (Copyright © 2012 Elsevier B.V. The effect of genotype in locus TAG 1A of gene receptor dopamine second type on characteristics of peak-and-wave EEG pattern in somatosensory, parietal and occipital areas of the cortex was studied in two groups of rats. Quantitative analysis showed that the peak-wave discharge of the first type in rats with A1/A1 genotype had a significantly longer duration, occurred more frequently leading to a significant increase in their peak-wave index. The results are of clinical interest creating a theoretical basis for improved diagnosis of absence epilepsy and selection of anticonvulsants.

Purpose: The WAG/Rij strain of rats, a well-established model for absence epilepsy, has comorbidity for depression. These rats exhibit depression-like behavioral symptoms such as increased immobility in the forced swimming test and decreased sucrose intake and preference (anhedonia). These depression-like behavioral symptoms are evident in WAG/Rij rats, both at 3-4 and 5-6 months of age, with a tendency to aggravate in parallel with an increase in seizure duration. Here we investigated whether the behavioral symptoms of depression could be prevented by the suppression of absence seizures., Methods: Ethosuximide (ETX; 300 mg/kg/day, in the drinking water) was chronically applied to WAG/Rij rats from postnatal day 21 until 5 months. Behavioral tests were done before the cessation of the treatment. Electroencephalography (EEG) recordings were made before and after cessation of treatment to measure seizure severity at serial time-points., Results: ETX-treated WAG/Rij rats exhibited no symptoms of depression-like behavior in contrast to untreated WAG/Rij rats of the same age. Moreover, treated WAG/Rij rats did not differ from control age-matched Wistar rats. ETX treatment led to almost complete suppression of spike-wave discharges (SWDs) in 5-6 month old WAG/Rij rats. Discontinuation of chronic treatment was accompanied by a gradual emergence of SWDs; however, a persistent reduction in seizure activity was still present 47 days after discontinuation of the chronic treatment., Discussion: The results suggest that seizure activity is necessary for the expression of depression-like behavioral symptoms and confirm that epileptogenesis can be prevented by early and chronic treatment.

Cortical spreading depression (SD) is thought to underlie the migraine aura but the mechanisms of triggering SD in the human cortex remain unknown. Because growing evidence points to a key role of brainstem circuits in initiating migraine attacks, the present study examined whether recurrent episodes of brainstem activation in rats could induce cortical SD. Explosive running elicited by sounds in rodents with inherited hypersensitivity to acoustic stimuli (reflex audiogenic epilepsy), is known to reflect a transient aberrant activation of the brainstem. Repeated induction of such audiogenic responses enhances the excitability of the cortex, culminating in its epileptic activation (audiogenic kindling). In Wistar rats with inherited hypersensitivity to sounds, 15 brief episodes of running were induced by sound stimulation, and slow potential shifts as well as the EEG were recorded in the cortex. Single unilateral SD began to occur in the cortex following a running episode after the 5th to 15th test (mean 9.4+/-1.2). Once appeared, SD was regularly recorded in subsequent tests. The side of the SD initiation closely correlated with the direction of running. Triggering SD was not associated with epileptic activation of the cortex in most rats. The present findings suggest that the sensory-induced brainstem excitation could be a potent trigger of SD in the hyperexcitable cortex, providing an experimental evidence of a possible causative role of the brainstem activation in initiating the migraine aura.

In this study 58 participants of the liquidation of the consequences of Chernobyl accident in 1986-1987 were investigated. All the patients complain of constant headaches, disorders of memory, general weakness, rapid fatigability, decreased sexual drive, emotional instability etc. The complex (comprehensive) modern methods of investigation were used to carry out the objective assessment of presented complains and of character of the central nervous system damage: complex computer quantitative analysis of mental capacity; analysis of personality traits by using the MMPI test; single photon emission tomography (with the drug of Ceretec); X-ray computer tomography; magnetic resonance computer tomography. The experimental study with examination of primates who were exposured in sum dose 1 Gy (by drop method) was carried out, too. The results of complex investigation of participants of liquidation of Chernobyl accident consequences enable to postulate the formation of premature aging of an organism in these persons. Data of the experimental study of primates irradiated in dose 1 Gy revealed formation of the brain atrophy in the remote period after low dose radiation exposure.

Spreading depression (SD), a self-propagating wave of reversible cellular depolarization, is thought to play an important role in brain pathophysiology. SD and seizures are closely related events but little is known about involvement of SD in chronic epileptogenesis. Here we show that cortical SD is the first and highly reproducible manifestation of audiogenic kindling induced by repeated sound stimulation of WAG/Rij rats with genetic audiogenic and absence epilepsy. Repetition of sound-induced running seizures in freely moving rats led to an appearance and gradual intensification of post-running facial and forelimb clonic convulsions coupled with afterdischarge in the fronto-parietal cortex. Before the development of these traditional manifestations of audiogenic kindling, an unilateral cortical SD wave began to be triggered by audiogenic seizures. Once cortical SD appeared, it became a permanent component of subsequent seizures. SD was always recorded in the hemisphere ipsilateral to the running direction. Only at the late stages of audiogenic kindling SD developed bilaterally. To estimate the contribution of SD in postictal effects of audiogenic seizures, we compared cortical activity after seizures induced SD or not. It was found that only seizures with cortical SD were followed by postictal suppression of spontaneous spike-wave discharges displayed by WAG/Rij rats. The results show that (1) cortical SD is readily triggered by brief sensory-induced seizures in awake animals; (2) SD may be responsible for postictal changes in cortical activity; (3) unilateral initiation of SD suggests asymmetrical recruitment of the cortex into seizure network during audiogenic kindling.

In rat model of absence epilepsy, abnormal complex movements of cervicothorathic part (jerks) were observed in the state of passive wakefulness. When a desynchronized low-amplitude electrical activity was recorded in the parietal and visual cortex, either SWD or jerks were observed. The SWD appearance prevented a jerk. If the discharge appeared between jerks, the interval between jerks increased. Particularly important is the fact that after subtraction of the discharge duration, the distribution of intervals between jerks became the same as in the absence of the SWD. This finding shows that an SWD just interrupts the development of a jerk. That is, when the discharge is over, the jerk develops from the level at which it was stopped by the discharge rather than from the initial level of excitation.

The brain serotoninergic (5HTergic) system of epileptic subjects can influence their vulnerability to stress. We studied the putative dependency of 5HT neurotransmission parameters on emotional stress, and the presence, types and severity of seizures using rats with genetic generalized (absence and/or audiogenic) epilepsy, of WAG/Rij and Wistar strains. The animals were stressed by exposure to a short aversive noise or left without sound stimulation. Tissue concentrations of 5HT, tryptophan (TRT) and 5-hydroxyindolacetic acid (5HIAA) were assessed by HPLC. The stressor activated the 5HTergic system within thalamus (5HIAA elevated), frontal cortex (5HT, TRT elevated), hypothalamus (increased TRT) in all rats. However, the normal (non-epileptic) rats displayed the highest response in the frontal cortex and the lowest one in the thalamus, as compared to the epileptic rats. Absence-epileptic rats exhibited higher thalamic 5HIAA increase than their controls. Significant correlations existed between propensity of absence epilepsy and 5HTergic parameters measured in the cortex and hypothalamus of absence-epileptic rats. No major difference was found between groups with and without audiogenic epilepsy. The results imply that the stress response depends on the presence of epileptic pathology and the seizure type and severity. The brain 5HT may be involved in the control of the paroxysms and behaviour in absence-epileptic subjects.

To study the role of the cortex and sub-cortical structures in the generation of epileptic spike-wave discharges in more detail, cortical and striatal activity was eliminated by the induction of spreading depression in a non-invasive way. EEG and DC potentials were recorded from the cortex and striatum of WAG/Rij rats. Several of these rats show two forms of generalised epilepsy: spontaneously occurring non-convulsive absence seizures, together with convulsive audiogenic seizures. The latter can be evoked by a brief sound stimulation, provoking a fit of wild running, which is regarded as the first phase of an audiogenic seizure. In a majority of fits the cortical DC potential does not show main changes, while the spontaneously occurring spike-wave discharges are briefly suppressed for some minutes. In a minority of fits, however, audiogenic seizures are associated with a spreading depression wave, clearly expressed in the cortical DC potential. This wave is bilaterally initiated in the cortex and propagates to the caudate nucleus of the striatum. In these cases spontaneously occurring spike-wave discharges are fully suppressed for about 1 h. It is suggested that cortical spreading depression, triggered by a short audiogenic seizure, induces a long-lasting suppression of spike-wave discharges. These results are in line with the concept that spike-wave discharges are originally initiated in the cortex, as proposed by the 'cortical focus' theory. The precise role of the striatum remains less clear, although this structure seems not to play a pivotal role in spike-wave generation.

Purpose: The effect of systemic administration of the gamma-aminobutyric acid (GABA)-transaminase inhibitor vigabatrin (VGB) on different components of convulsions was tested in the model of audiogenically kindled seizures, which consist of brainstem (running, tonus) and forebrain (clonus) elements., Methods: Audiogenically susceptible rats of Krushinsky-Molodkina (KM), Wistar, and WAG/Rij strains received repeated sound stimulation (60 dB, 10-80 kHz) until kindled audiogenic seizures were reliably elicited. Kindled audiogenic seizures consisted of running, tonic, and generalized clonic phases in KM rats (severe audiogenic seizures) and of running and Racine stage 5 facial/forelimb clonus in Wistar and WAG/Rij rats (moderate seizures). Vehicle, 100, or 200 mg/kg of VGB was intraperitoneally injected 2, 4 and 24 h before the induction of kindled audiogenic seizures., Results: At both doses, VGB did not change the seizure latency and the duration of running and tonic convulsions, but suppressed clonic ones in all rat strains. In KM rats, the mean duration of posttonic clonus was significantly reduced at 24 h after 100 mg/kg and from 4 h after 200 mg/kg. In Wistar and WAG/Rij rats, the mean duration of facial/forelimb clonus was reduced from 4 and 2 h after 100- and 200-mg/kg administration, respectively; 24 h after the high-dose injection, clonus was completely blocked in all rats of both strains. No difference in efficacy of VGB between Wistar and WAG/Rij rats was observed., Conclusions: VGB more effectively suppresses clonic convulsions than running and tonic ones in audiogenically kindled rats. It is supposed that this selective anticonvulsive effect of VGB results from different sensitivities of forebrain and brainstem epileptic networks to the presumed GABA enhancement.

Behavioral response to a new environment of Wistar and WAG/Rij rats with absence and/or audiogenic seizures (AGSs) was investigated. Behavior was observed in open-field (OF) and light-dark choice (LD) tests. Correlations of test performance with seizure parameters were evaluated. AGS-susceptible Wistar rats exhibited reduced exploration (rearing) in both tests and a tendency toward hyperlocomotion in the OF test. Genetically absence-epileptic WAG/Rij rats demonstrated agitation (increased vertical/horizontal locomotion, enhanced defecation/urination) in the LD test, whereas they exhibited reduced exploration, increased grooming, and hyperlocomotion in the OF test. Anxiety level, as estimated by grooming time in the OF test and latency to first "risk assessment" in the LD test, correlated positively with the propensity for absence seizures in WAG/Rij rats not susceptible to AGSs. It can be concluded that the behavioral response to novelty stress in epileptic subjects depends on the type and severity of seizures.

Mixed forms of epilepsy in patients are often refractory. Therefore, animal models of comorbid convulsive and nonconvulsive seizure are needed for experimental research. Susceptibility to audiogenic convulsions was studied in a large group of young and adult WAG/Rij rats with inherited absence epilepsy. In 30% of adult rats, sound stimulation provoked audiogenic seizures of moderate intensity. The seizures had two excitation periods separated by a remarkably stable "arrest" of paroxysmal movements. Up to 20% of young WAG/Rij rats were also susceptible to audiogenic seizures, with a longer latency, lower intensity, and more simple seizure patterns. No difference in manifestations of spike-wave discharges was observed between the WAG/Rij rats with and without audiogenic seizures. This subpopulation of WAG/Rij rats genetically predisposed to absence and audiogenic seizures is proposed as an animal model suitable for investigation of basal mechanisms and pharmacological profiles of this mixed form of epilepsy.

The effects of ketamine, a noncompetitive antagonist at the NMDA receptor, were studied on the EEG and in the open field in a genetic animal model of generalized absence epilepsy--the WAG/Rij rat strain. Animals of this strain display spontaneous occurring generalized spike-wave discharges (SWDs) in the EEG. Ketamine was systemically administered in a dose range from 3 to 30 mg/kg. Biphasic effects of ketamine were observed in the EEG. The first phase was a dose-dependent suppression of SWDs, followed by a second phase characterized by the facilitation of SWDs. This increase was expressed first as an increased number of SWDs, and later on as a significant prolongation of individual discharges and decrease in frequency of SWDs. An obvious amplitude modulation of the discharges was also found. During the period of suppression of SWDs, a new phenomenon was observed: quasi-periodic groups of spikes or wave spikes, with an internal frequency of 4-5 Hz and a periodicity of about 5 s. That quasi-periodic activity vanished a few minutes prior to the recovery of the classical SWDs. However, a specific 5-s amplitude modulation of SWDs remained present in the recovery period. The propensity of that specific ketamine-induced activity was found to be correlated with propensity of SWDs in background EEGs of drug-free animals. Ketamine also produced a dose-related initial behavioral excitation, a decrease of muscle tone in hind quarters, followed by front quarters and head, and an absence of locomotor activity. However, the time course of the behavioral changes cannot explain the effects on the EEG. It can be concluded that ketamine has more effects on the EEG than previously assumed which cannot be explained by a simple blockade of the NMDA receptor. It is proposed that the obtained specific dynamics of SWDs' frequency may be caused by changes in the activity of the thalamo-cortical pacemaker that is generating SWDs.

The remote effects of neonatal (on the 3d-to-9th postnatal days) ketamine injections (10 and 50 mg/kg in 20 microliters of distilled water, s.c.) were analyzed in adult Wistar, WAG/Rij, and KM (a strain with high audiogenic sensitivity) rats. Both ketamine and water injections increased pain sensitivity in adult rats. Neonatally injected water increased the mean score of seizures in Wistar and WAG/Rij, whereas ketamine water solution injected in the dose of 50 mg/kg did not change the seizure intensity (as compared to the intact control). Consequently, ketamine significantly reduced the mean score of the audiogenic seizure fit without change in its latency. In highly sensitive KM rats the neonatally injected ketamine (50 mg/kg) significantly shortened the mean latency of the fit onset, and fit stages developed faster. Thus, the neonatal ketamine injection increased the audiogenic seizure susceptibility of brain structures in KM rats.

Complex analysis of EEG and thermographic parameters carried out in 10 healthy subjects and 34 patients, Chernobyl clean-up participants revealed a correlation between EEG and brain temperature changes in the baseline state and during mental arithmetic. During cognitive activity the maximal increase in the average EEG coherence and temperature shifts in healthy subjects were observed in the left frontotemporal and right parietotemporal areas. In patients changes in both parameters under study were most pronounced, the interhemispheric relations were impaired. The visual analysis revealed "flat" and "hypersynchronous" EEG types in patients. The dominant pathologic activity in the betal range indicative of mediobasal and oral brainstem lesions was characteristic of the flat EEG. This type of activity was observed in 60% of patients. In these cases, a general decrease in EEG coherence and temperature was most pronounced in the left hemisphere. The hypersynchronou EEG type (40% patients) was characterized by paroxysmal activity in the theta and alpha ranges suggesting diencephalic brain lesions. In these cases, EEG coherence and temperature were more variable; changes in the right hemisphere were significant, be it increase or decrease. Our complex approach to investigation of brain activity in different aspects seems to be promising in estimation of the brain functional state both in healthy persons and patients in remote terms after exposure to radiation. The specific hemispheric temperature changes revealed in Chernobyl patients especially during cognitive activity can be the sequels of postradiation disorders of vascular neuro-circulation. The EEG findings suggest subcortical disorders at different levels (diencephalic or brainstem) and functional failure of the right or left hemispheres in remote terms after exposure to radiation.

Typological behavior reactions of WAG/Rij rats were studied from the standpoint of divergent modulatory integration hypothesis. This rat strain has a genetically determined dominant dysfunction of the benzodiazepine system of the thalamic nuclei. This disorder provokes an epileptiform disease such as absence epilepsy. It was suggested that the dysfunction of this system would result in a modification of the modulatory systems, which support the motivation states of escape and avoidance reactions as well as of the modulatory systems, which form the emotional states. Modifications of these states are the background of typological behavioral features of WAG/Rij rats. It was shown that WAG/Rij have the lower threshold of the development of haloperidol catalepsy, higher levels of fear and depression. On the first day of training in a shuttle box, WAG/Rij rats demonstrated better avoidance performance than Wistar rats. On the second and 28th days, the amnestic effect of the epileptiform disease was observed. The amnestic effect was also observed after passive avoidance conditioning. The results are discussed in terms of the modulatory integratin hypothesis.

Rats of the WAG/Rij strain are commonly seen as a genetic model for generalised absence epilepsy in man. Interestingly, generalised absence epilepsy shows, in addition to the fully generalised spike-wave discharges, a second type of spike-wave discharge, which lasts for a shorter time, has a lower frequency, and a lower incidence. The originally described distinction between the two types of spike-wave discharges was mainly based on the shape, polarity and duration of the discharges. In the present study other characteristics such as the spatial and temporal distribution of the spike and wave components of the two discharges and frequency spectra were found to differ between the two types. In addition, a reciprocal regulation of the two types of spike-wave discharges by drugs affecting the dopaminergic system (haloperidol and apomorphine) was observed. The results convincingly demonstrate the difference between the two phenomena and warrant the search for neurobiological mechanisms underlying both types of spike-wave discharges.

The possibility of correcting seizure activity with neural transplantation was studied in Wistar rats with audiogenic seizures and in Krushinskii-Molodkina rats with high level of audiogenic seizures. In Wistar rats seizures were absent during 24 weeks after combined bilateral transplantation of striatal and cerebellar tissue from newborn rats into the parietal cortex. The same transplantation performed in Krushinskii--Molodkina rats increased the latency of audiogenic seizures. In some rats the intensity of seizures decreased, but they did not completely disappeared. Suppression of seizure activity in Krushinskii-Molodkina rats was observed after transplantation of striatal and cerebellar tissue simultaneously into the parietal cortex and inferior colliculi.

The electrical activity of different cortical areas was recorded in 37 WAG/Rij rats under conditions of chronic experiment. The typical for absence epilepsy spike-wave discharges occur in EEG of all the animals of the strain. The average amplitude distribution maps were constructed based on the recording of two main components of the spike-wave discharge. The most frequently occurring discharges of 5-9 s in duration were taken for map construction. Separate maps were constructed for the beginning and for the end a discharge. It was shown that at the beginning of a discharge the maximum of spike amplitude was localized in the frontal cortical area in the site of localization of the eye, vibrissa and chewing movement centers. At the same time, the wave was better pronounced in the occipital cortical areas. At the end of a discharge spike amplitude diminished and the cortical square over which it was manifested was cut down. On the contrary, at the end of a discharge were amplitude increased and the area of its distinct recording spread to the frontal cortical areas. The wave amplitude maximum was observed over the cortical areas 17. The obtained evidence suggest the relatively independent spatio-temporal dynamics of the two main components of the spike-wave discharge complex.

Adult WAG/Rij rats are considered adequate genetic models for human generalized absence epilepsy. Rats of this strain of 8, 12, and 18 weeks old and age-matched control Wistar rats were exposed to sound stimulation. After offset of stimulation, all WAG/Rij rats showed cataleptic or even cataplexic reactions, which could persist for up to 20 min. Age effects could be demonstrated. None of the Wistar rats showed cataleptic reactions. Electroencephalographic studies in WAG/Rij rats of 21 weeks showed that spike-wave discharges were abundantly present in the background electroencephalogram prior to sound stimulation. Age-matched Wistar rats had almost no spike-wave discharges. Spike-wave discharges in WAG/Rij rats disappeared during sound stimulation and were then increased compared to the prestimulation and stimulation periods. The electroencephalogram during the cataleptic state was also characterized by the presence of large amplitude 2 Hz waves, interspersed with spike-wave discharges. The data suggest that the cataleptic state can be elicited in genetically epilepsy-prone rats. The youngest WAG/Rij rats showed no spike-wave discharges during the cataleptic state. In all, the data suggest that epilepsy-prone animals are sensitive for catalepsy at an age at which the EEG signs of generalized absence epilepsy are not yet manifest.

The effects of neurotransplantation on development of audiogenic seizures (AS) were studied in Krushinskiĭ-Molodkina (KM) rats genetically predisposed to AS and in Wistar rats especially selected for AS. Within 24 weeks after bilateral combined transplantation of the newborn striatum and cerebellum into the parietal cortex, the AS didn't develop in Wistar rats in 74% of tests. In control Wistar rats with cortical grafts or saline injections into the parietal cortex there were no significant changes in the AS development. In the experimental KM rats the grafting led to the 4-5-times increase in the AS latency. In 50% of rats it decreased the AS severity by 1-2. In the control KM rats these parameters didn't change. It was concluded that it was possible to suppress or decrease the AS development by the transplantation of the newborn striatal and cerebellar tissue into the cortex of rats predisposed to this kind of seizures.

Clinical, neuropsychologic, neurophysiologic as well as neuromorphologic (in legal cases) examinations of Chernobyl liquidators were performed. In addition, the study was made of the monkeys exposed to 1,5 Gy radiation. The results obtained testified to the organic character of cerebral damages. They may be characterised as chronic progressive discirculatory-hypoxic syndrome in which disorders of vascular permeability were expressed and dystrophic destructive irreversible alterations of nervous cells occurred.

The temperature topography of the cerebral cortex of rats following clinical death and resuscitation was investigated with the technique of thermoencephaloscopy. Complete restoration of the neurological status of the animals was achieved over the course of one to two days. Marked disturbances in the background thermal maps and the thermal reactions of the cerebral hemispheres induced by a stressor were identified in the remote post-resuscitation period (up to two months). A pathological mosaicism of the thermal characteristics was detected under the conditions of relative rest, as were disturbances in the dynamics of the temperature reactions of the brain and in the character of the interhemispheric asymmetries under stress. The individual character of the post-resuscitation pathology which is found both in the baseline thermal maps as well as following a functional load is emphasized. The results of the investigation point to the importance of an individual approach in the rehabilitation therapy of the post-resuscitation illness.

It was found out in experiments on monkeys that one of the most serious remote consequences of small and medium doses of ionizing radiation was the development of stable organic disorders in various cerebral structures. Clinical researches substantiate the necessity to make registration of post-radiation pathological changes in central nerve system of patients.

By thermoencephaloscopy method the temperature relief of the rats cerebral cortex was studied after the clinical death and reanimation. The rehabilitation of the animals neurological status was completed in 1-2 days. In the remote postresuscitation period (up to 2 months), expressed disturbances were revealed of the background thermomaps and of thermal reactions of the cerebral hemispheres, evoked by the stress influence. Pathologic mosaics of thermal characteristics was revealed in conditions of relative rest and disturbance of dynamics of the brain temperature reactions and the character of interhemispheric asymmetries at stress influence. Individual character is pointed out of postreanimative pathology, which is manifest both in the background thermomaps and after the functional load. The results of the studies suggest the importance of individual approach in rehabilitation therapy of postreanimative disease.

By the method of forced immobilization the rabbits were brought into the state of "animal hypnosis" (immobilization reflex), and their ECoG was recorded, which was further processed on the computer. It was found that during hypnosis a functional interhemispheric brain asymmetry was developed in rabbits with activity predominance in the right hemisphere. The "animal hypnosis" is a phasic process: in the ECoG of the rabbit under hypnosis a regular alternation of delta and theta activity takes place. Electrophysiological reconstructions in the rabbit brain during the change of its functional state correlate with the brain thermal reactions, revealed earlier.

The method of dynamic thermovision was used in white rats to investigate the distant reactions of the cerebral cortex arising during local chemical perturbation (2% KC1). In addition to the earlier-described temperature wave of propagating depression (T waves), local areas of warming (by 0.2-0.3 degrees) symmetrical to the site of stimulation were found in the opposite hemisphere. Latent excitation foci during local chemical action were found on the dynamic thermal maps of the brain surface in regions remote from the stimulation site in the same or contralateral hemisphere. A change in the site of T-wave origination and the typical sequence of its propagation through the cortex could be observed when two potassium foci were created, as well as the development of foci of prolonged temperature oscillations at the site of encounter of two T waves. On the basis of the obtained data several questions are discussed concerning the forms and mechanisms of distant effects in the CNS.

By means of dynamic thermovision method, distant reactions of the cerebral cortex were studied in white rats appearing under local chemical action (2% KCl). Besides the previously described temperature wave SD (T-wave), in these conditions in the contralateral hemisphere local areas of warming up (0.2-0.3 degrees) were observed symmetrical with the place of stimulation. Under local chemical action, latent foci of excitation were revealed on dynamic thermal maps of the brain surface in the areas of the same or contralateral hemisphere, distant from the places of stimulation. During creation of two K+ foci a change was observed of place of T-wave appearance and of the usual order of its propagation over the cortex and also the formation of a focus of prolonged temperature fluctuations in the place of two T-waves meeting. On the basis of the obtained data a number of questions is discussed on the forms and mechanisms of distant effects in the CNS.

A mathematical model is proposed to describe a heat wave accompanying the spreading cortical depression wave in the large brain. The model takes into account heat production of neurons, thermal transmission with thermoconductivity and volume thermal losses. The model fits well for the wave shape and permits one to estimate heat production which is about 4-20 J/g X min.

The resting EEGs of several brain structures (motor and visual cortex, caudate nucleus and intralaminar thalamic nuclei) were submitted to spectral and coherence computer analyses in two rat strains. Genetically predisposed to convulsive state KM rats were shown to differ from nonpredisposed Wistar rats in EEG spectral properties. KM rats EEG pattern was characterized by increase of low frequencies (1-2 Hz) power and decrease of faster activity (5-12 Hz) power in cortical spectrograms as well as by decrease of caudate nucleus EEG absolute power. The coherence value between cortical or subcortical structures at below 4 Hz was intensified in KM rats. Reinforcement of cortical auto-oscillating properties manifested by ECoG synchronization in cortical-thalamic resonance interaction as well as weakening of striatal inhibitory system may constitute neurophysiological mechanisms of enhanced convulsive readiness. The probable role of mediator imbalance in these mechanisms is discussed.

Conditioned switching-over was elaborated in white rats: in one situation an alimentary instrumental reflex to sound was elaborated, in another one--an electro-defensive avoidance reflex to the same sound, different time of the day and different chamber illumination used as the switching-over factor. One-time influence of spreading depression by means of bilateral application of 10% KCl solution for 1.5 min to dura mater brought to the disturbance of the conditioned switching-over for 2-3 days. Electrolytic destruction of posterior hypothalamus area was produced in some animals. After restoration of functions one-time application of spreading depression elicited deep and long-term disturbance of conditioned switching-over. Apparently more complex conditioned activity suffers greater at short-term elimination of cerebral cortex functions, especially in conditions of compensatory recovery after lesions of the central nervous system.

Dynamics of spatial distribution of temperature over the dorsal surface of white rat cerebral cortex was studied through the skull with the thermovision technics. Visual, somatosensory and acoustic stimulation revealed both diffuse and regionally specific thermoreactions of the brain as well as multiple local termoresponses. After injection of KCl into the cortex temperature waves spreading over the cortex were obtained. Mechanisms of brain patterned thermoreactions dealing with blood flow and neuronal metabolic thermoproduction are discussed.

Desynchronous (low voltage fast activity), synchronous (high voltage slow waves) as well as convulsive brain activities were stimulated by a computer model of neuronal population. Network excitatory and inhibitory elements possessed fundamental dynamic properties of real neurones. Being independent both of the excitability of elements and of external influence efficacy, synchronous (desynchronous) network activity resulted from the increase (decrease) of the average power of "neuronal" interconnections which imitated mutual and recurrent excitation and inhibition. The inhibition efficacy being reduced as compared with excitation, synchronization of elements became intensified. As a consequence, the rhythmic activity amplitude increased and the appearance of self-sustained oscillations simulating convulsive activity was facilitated. The probable mechanism of EEG activation by virtue of the reduction of mutual and recurrent excitation and inhibition efficacy as well as the significance of inhibitory mechanism deficiency for epileptogenesis are discussed.

Dynamics of spatial temperature distribution over the dorsal surface of the cerebral cortex was studied through the skull with the thermovision technique on immobilized white rats. Direct cortical stimulation revealed local thermoresponses (up to +0.2 degrees C) under electrodes, in the symmetrical point in the opposite hemisphere as well as in some other cortical zones. The response latency was within the limit of 160 ms, peak latency--2-5 s and relaxation time 2-3 min. Nembutal caused depression of thermoresponses, increase in their thresholds, limitation in size as well as slowing down of the development and prolongation in time.
