2. Efficacy of IFN-γ, sCD40L, and Poly(I:C) Treated Bone Marrow-Derived Macrophages in Murine Mammary Carcinoma.

Abstract

Introduction: Here, we explored methods to generate anti-tumor bone marrow-derived macrophages (BMDM) and how delivery of the BMDM at early tumor sites could impact disease progression. Methods: BMDM treated with IFN-γ, sCD40L, poly(I:C), and a combination of the three were assessed. Results: Treatment with sCD40L had no significant impact on the BMDM. Treating BMDM with IFN-γ impacted IL-1β, MHC Class II, and CD80 expression. While poly(I:C) treatment had a greater impact on the BMDM than IFN-γ when assessed by the in vitro assays, the BMDM treated with poly (I:C) had mixed results in vivo where they decreased growth of the EMT6 tumor, did not impact growth of the 168 tumor, and enhanced growth of the 4T1 tumor. The combination of poly(I:C), IFN-γ, and sCD40L had the greatest impact on the BMDM in vitro and in vivo. Treatment with all three agonists resulted in increased IL-1β, TNF-α, and IL-12 expression, decreased expression of arginase and mrc, increased phagocytic activity, nitrite production, and MHC Class II and CD80 expression, and significantly impacted growth of the EMT6 and 168 murine mammary carcinoma models. Discussion: Collectively, these data show that treating BMDM with poly(I:C), IFN-γ, and sCD40L generates BMDM with more consistent anti-tumor activity than BMDM generated with the individual agonists. 24. Evaluation des Portfolio-Konzepts in den Kindergärten der Wiener Kinderfreunde

Abstract

Die vorliegende Arbeit setzt sich mit den Interaktionsmustern der Pädagoginnen in den Kindergärten der Wiener Kinderfreunde, die in den verschiedenen Lehrsituationen der Portfoliobearbeitung und des Morgenkreises auftreten, auseinander. Hierzu wurden die Qualitäten der Rückmeldungen, der Feinfühligkeit und das Auftreten der Joint Attention an adaptierten Versionen der Beobachtungssysteme von Dreßler (1999) und Aigner (2004) erhoben. Darüber hinaus wurden die von den Pädagoginnen geäußerten Entwicklungsbedenken bezüglich der von ihnen betreuten Zielkinder durch eine deutschsprachige Version des Parents’ Evaluation of Developmental Status (PEDS, Glascoe, 1999, 2003) erfasst. Ziel der durchgeführten Untersuchung war das Aufzeigen von Differenzen in den pädagogischen Interaktionsmustern der beiden Lehrsituationen und die Ermittlung einer Verbesserung des Entwicklungsstandes der Kinder gemäß den Einschätzungen der Pädagoginnen während des Beobachtungszeitraums. Der Literatur nachfolgend wurden die Häufigkeiten an geäußerten Bedenken für verschiedene, laut Literatur unterschiedsträchtige, Gruppen überprüft. Die Pädagoginnen unterschieden sich nicht in ihrer zur Schau gestellten Feinfühligkeit über die beiden Lehrsituationen hinweg, aber in den Frequenzen, in denen sie in diesen beiden Situationen positive und korrigierende Rückmeldungen gaben. Zudem zeigte sich ein Unterschied in der Häufung der Joint-Attention-Episoden, die ebenso wie die positiven und korrigierenden Rückmeldungen im Rahmen der Portfoliobearbeitung erhöht waren. In Bezug auf die geäußerten Entwicklungsbedenken durch die Pädagogin zeigte sich keine Veränderung während des Untersuchungszeitraums. Auch für die gebildeten Gruppen von Kindern mit deutscher und nicht deutscher Muttersprache, jüngeren und älteren Kindern und Kindern, Kindern, die schon länger oder erst seit kurzem den Kindergarten besuchten, zeigten sich keine Differenzen in den artikulierten Häufigkeiten von Bedenken. Einzig in den das Geschlecht als Trennvariable ansehenden Gruppen fand sich zu beiden Erhebungszeitpunkten ein Unterschied für den Bedenkenbereich „Umgang mit anderen“ mit einer erhöhten Nennung von Bedenken bei Jungen.

Abstract

Abstract Real-world data sources, including electronic health records (EHRs) and personal digital device data, are increasingly available, but are often siloed and cannot be easily integrated for clinical, research, or regulatory purposes. We conducted a prospective cohort study of 60 patients undergoing bariatric surgery or catheter-based atrial fibrillation ablation at two U.S. tertiary care hospitals, testing the feasibility of using a patient-centered health-data-sharing platform to obtain and aggregate health data from multiple sources. We successfully obtained EHR data for all patients at both hospitals, as well as from ten additional health systems, which were successfully aggregated with pharmacy data obtained for patients using CVS or Walgreens pharmacies; personal digital device data from activity monitors, digital weight scales, and single-lead ECGs, and patient-reported outcome measure data obtained through surveys to assess post-procedure recovery and disease-specific symptoms. A patient-centered health-data-sharing platform successfully aggregated data from multiple sources.

38. Targeted Disruption of Tumor-Derived Chemokine Synthesis and Reversal of Tumor-Induced Immune Suppression

Abstract

Chemokines play a pivotal role in the maturation of the immune system, and in the initiation, and maintenance of an immune response. Because of their key role in the immune response, the aberrant expression of chemokines can have a profound effect on the ability of T cells to respond to antigen. We have found that several breast cancer cell lines produced chemokines capable of recruiting T cells. However, instead of increasing anti-tumor immunity, the tumor-derived chemokines may have prevented an effective immune response by desensitizing T-cell chemokine receptors. Our hypothesis is that disrupting the synthesis of tumor-derived chemokines (using anti-sense technology) will remove tumor- induced immune suppression and enhance the immunogenicity of the tumor. In order to test this hypothesis we generated stable clones that lack CCL2 and CCL5 production compared to the parental tumor cell line. Using these tumors we found that both CCL2 and CCL5 impair the T cell response to the murine mammary carcinoma 4Tl. Moreover, tumor-derived CCL5 impaired T cell chemotactic activity and enhanced growth of the tumor in vivo. In addition, we found that tumor- derived CXCL 1 acts as an autocrine growth factor and angiogenesis factor for 4T 1.

Tumors that lack MCP-1 or RANTES elicited a better tumor-specific T cell response. Studies are underway to determine the extent of tumor immunity generated to a tumor that lacks both of these chemokines.
