A membrane-bound hydrogenase from Desulfovibrio vulgaris Miyazaki F is a metalloenzyme that contains a binuclear Ni–Fe complex in its active site and mainly catalyzes the oxidation of molecular hydrogen to generate a proton gradient in the bacterium. The active-site Ni–Fe complex of the aerobically purified enzyme shows its inactive oxidized form, which can be reactivated through reduction by hydrogen. Here, in order to understand how the oxidized form is reactivated by hydrogen and further to directly evaluate the bridging of a hydride ligand in the reduced form of the Ni–Fe complex, a neutron structure determination was undertaken on single crystals grown in a hydrogen atmosphere. Cryogenic crystallography is being introduced into the neutron diffraction research field as it enables the trapping of short-lived intermediates and the collection of diffraction data to higher resolution. To optimize the cooling of large crystals under anaerobic conditions, the effects on crystal quality were evaluated by X-rays using two typical methods, the use of a cold nitrogen gas stream and plunge-cooling into liquid nitrogen, and the former was found to be more effective in cooling the crystals uniformly than the latter. Neutron diffraction data for the reactivated enzyme were collected at the Japan Photon Accelerator Research Complex under cryogenic conditions, where the crystal diffracted to a resolution of 2.0 angstroms. A neutron diffraction experiment on the reduced form was carried out at Oak Ridge National Laboratory under cryogenic conditions and showed diffraction peaks to a resolution of 2.4 angstroms.

NADH-シトクロムb5還元酵素（b5R）はFADを補欠分子族として結合する酸化還元酵素で、2電子供与体であるNADHから1電子受容体であるシトクロムb5（b5）への電子伝達反応を担っている。電子伝達反応の過程においてb5Rは二電子還元型、一電子還元型など複数の酸化還元状態を取ることが知られている。このb5Rの酸化還元反応サイクルにおいてはヒドリドイオン（H-）とプロトン（H+）の移動を伴っている。そのため、水素原子を含めた高分解能立体構造情報を取得することは、b5Rの酸化還元反応サイクルの理解に必要不可欠である。本研究では、b5Rにおける水素移動経路を理解するため酸化型については中性子構造解析を行い、還元型については1 Å分解能を超える高分解能X線構造解析を行った。酸化型b5Rは2種類のpH条件で作製した結晶を用い中性子構造解析を行った結果、FAD周辺の水素結合ネットワークに立体構造変化が見られた。還元型b5Rについては、野生型と一電子還元型を安定化する変異体の結晶を作製しX線構造解析を行った結果、FADおよびNADH周辺に立体構造変化が観測された。以上の中性子構造解析とX線構造解析の結果から、水素移動反応に重要であると考えられる立体構造情報を取得することができた。, 日本結晶学会令和2年度年会

膜タンパク質やタンパク質複合体などの立体構造に基づいたタンパク質間相互作用の解明は、現代の生命科学において重要な研究領域である。これらのタンパク質は分子量が大きく、得られる結晶の格子長も大きいことが多い。高格子結晶からのブラッグ反射分離には冷中性子の利用が適している。JRR-3はビームホールに3本の冷中性子ビームラインを有し、高強度化のため中性子導管のスーパーミラー化がこれまで進められてきた。特に、昨年度は炉心に最も近く高強度化に大きく寄与する生体遮蔽体内導管が更新された。本高度化で得られる強度利得に対する評価も計算機シミュレーションにより進められてきている[1]。 そこで、試料結晶の格子長に応じて波長可変な機構を導入した冷中性子ビームラインでの回折装置設置の検討を行っている。現在は、中性子イメージングプレート検出器を用いたBIX-3,4をJRR-3炉室で運用しているが、ビームホールはバックグラウンドが低く抑えられる点で有利である。但し、単結晶回折では有効利用可能な角度分散は限定される。よって、本高強度化において、検討装置が利用する角度分散の利得成分を求めなければならない。そこで、C1-3ビームポートを設置位置と想定し、McStasを用いて"角度分散= ±0.5° 以内の中性子ビームをシミュレーションした。その結果、図1で示すような中性子束スペクトルが得られた（全角度分散の場合も併せて示す）。本発表では、さらに炉室中性子束との比較、モノクロメータを用い単色化した中性子による試料位置での中性子束を報告し、高格子結晶に対する本回折装置の測定性能を評価する。 [1] 研究炉加速器管理部編, JAEA-Review 2014-047, 94-97, (2014)., 日本中性子科学会第20回年会

NADH-cytochrome b5 reductase (b5R) on endoplasmic reticulum membrane in mammalian liver cell plays a variety of roles concerning lipid unsaturation and xenobiotic metabolism. b5R transfers electrons from two-electron carrier of NADH to one-electron donor of cytochrome b5 (b5). In the redox cycle of b5R, a hydride transfer from NADH to oxidized FAD and deprotonation from the reduced FAD (FADH) take place in b5R. Therefore, high-resolution structure analyses including the information about hydrogen atoms and valence electron densities are required for understanding molecular mechanisms of the b5R redox reaction. High-resolution X-ray crystal structures were previously determined using the oxidized form of b5R from porcine liver (M. Yamada et al., J. Mol. Biol., 2013; K. Takaba et al., Sci. Rep., 2017). In this work, the neutron crystal structures of the oxidized form of b5R were determined including hydrogen atoms of solvent molecules, and the X-ray crystal structures of the reduced form of b5R were determined including hydrogen atoms of the NADH cofactor. The neutron structure analysis of the oxidized form of b5R clearly shows the hydrogen-bonding network from the FAD cofactor to the protein surface. The X-ray structure analysis of the reduced form of b5R reveals the NAD+ and NADH bound states using wild type and T66V mutant. Information about hydrogen atoms and valence electrons is important to understand functions of electron transfer proteins, because they are directly connected with the reactivity of redox reactions. In the redox proteins, the possibility of quantum tunneling has been discussed on the hydrogen and electron transfer reactions. Therefore, high-resolution structural information is required to detect small structural changes due to the redox reactions. The NADH-cytochrome b5 reductase (b5R) and cytochrome b5 (b5) redox system is involved in various electron transfer reactions, such as lipid unsaturation, cholesterol synthesis and drug metabolism. The X-ray crystal structure of oxidized form of b5R at 0.78 Å resolution clearly visualized valence electron densities of the FAD cofactor. The X-ray crystal structures of the oxidized and reduced forms of b5 shows small structural changes between two redox states. We have recently determined the neutron crystal structures of the oxidized form of b5R at 1.4 Å resolution. In addition, we have determined high-resolution X-ray crystal structures of the reduced form of b5R. 哺乳類の小胞体膜に存在するNADH-シトクロムb5還元酵素（b5R）は、脂肪酸の代謝やコレステロール合成などに関与する酸化還元タンパク質である。b5Rの酸化還元反応サイクルにおいては、2電子供与体であるNADHから1電子供与体であるシトクロムb5への電子伝達が行われる。またその過程では、b5Rに結合するFADとNADH間におけるヒドリドイオン（H-）とプロトン（H+）の移動が伴うと報告されている。そのため、外殻電子や水素原子を含めた高分解能の立体構造情報を得ることは、b5R酸化還元反応系における分子機能の理解に必要不可欠である。 これまでブタ肝臓由来のb5Rおよびb5において、高分解能のX線結晶構造が報告されている（M. Yamada et al., 2013; K. Takaba et al., 2017; Y. Hirano et al., 2015）。本研究では、酸化型b5Rについては溶媒の水素原子を含めた立体構造情報を取得するために中性子構造解析を行い、還元型b5Rについてはより高分解能での構造解析を目指してX線構造解析を行った。その中で酸化型b5Rの中性子構造解析においては、FAD周辺の水分子の水素原子を含めた水素結合ネットワークが明らかとなった。還元型b5Rについては、大型放射光施設SPring-8のBL44XUにおいてX線回折データ収集を行った結果、1 Å分解能を超える回折点を観測することに成功した。, 第19回日本蛋白質科学会年会 第71回日本細胞生物学会大会 合同年次大会

タンパク質において水素原子は触媒反応や酸化還元反応、外殻電子は原子間の結合や電子伝達反応などに直接関与している。したがって、タンパク質の高分解能立体構造解析により、水素原子や外殻電子密度の情報を高精度で明らかにすることができれば、タンパク質の分子機能の理解に新しい可能性を提示することができる。そこで本研究では、哺乳類の小胞体において脂質の合成や薬物の代謝等に関与するNADHシトクロムb5還元酵素（b5R）を用い、中性子回折およびX線回折を相補的に組み合わせた高分解能の立体構造解析を行い、タンパク質中の水素移動反応に重要な立体構造情報の取得を試みた。, 量子生命科学会第１回大会
