Abstract

The incorporation of L-lysine by the marcfortine A producing Penicillium strain is described. The L-lysines were individually labeled with 15N at the alpha and epsilon positions and were analyzed by FAB-MS and NMR spectroscopy. The relative enrichment of the other nitrogens to Nbeta was determined by analyzing the HMBC spectra. The results showed that the L-lysine is metabolized to the pipecolate moiety by losing the alpha-amino group and not the epsilon-amino group.

Abstract

The screening UC 11136, a cholesteryl ester transfer protein inhibitor (CETP) reveals the existence of U-106305, a novel CETP. The NMR structural analysis reveals that the novel compound has a N-isobutyl-all-trans-4,5:6,7:8,9:10,11:12,13:16,17-hexamethylene-(E,E)-2,14- octadecadienamide structure. Analysis of the biosynthesis of U-106305 indicates that the nine acetates attached in a head-to-tail manner forms the backbone of the novel protein inhibitor whereas, the methyl group of L-methionine produce the cyclopropyl methylene carbons.
