Most plants grow and develop by taking up nutrients from the soil while continuously under threat from foraging animals. Carnivorous plants have turned the tables by capturing and consuming nutrient-rich animal prey, enabling them to thrive in nutrient-poor soil. To better understand the evolution of botanical carnivory, we compared the draft genome of the Venus flytrap (Dionaea muscipula) with that of its aquatic sister, the waterwheel plant Aldrovanda vesiculosa, and the sundew Drosera spatulata. We identified an early whole-genome duplication in the family as source for carnivory-associated genes. Recruitment of genes to the trap from the root especially was a major mechanism in the evolution of carnivory, supported by family-specific duplications. Still, these genomes belong to the gene poorest land plants sequenced thus far, suggesting reduction of selective pressure on different processes, including non-carnivorous nutrient acquisition. 症例は74歳女性．糖尿病性腎症による慢性腎不全で平成14年より維持血液透析（HD）を受けていた．HD間体重増加が中1日で平均3kg，中2日で平均5kgと多くHD時血圧低下が著明で1回のHDでは除水不十分なため，HD＋限外濾過（ECUM）を最大週5回施行し週末にドライウェイトに達する状態であった．そのため，除水補助目的で腹膜透析（PD）併用を開始した．PD処方はイコデキストリン2Lを2分割投与し平均1日800gの除水が得られた．PD併用後のHD間体重増加は中1日で平均1kg，中2日で平均1.5kgとなり週3回のHD（ECUM）でコントロールできるようになった．週あたりのHD（ECUM）時間は30時間から9時間へと著明に短縮し患者の生活の質も上昇した．血液データでは，小分子物質のみならず，β2マイクログロブリンの低下を認めた．一方，PD併用前は非HD時の血圧が高く心保護を考えアンジオテンシン受容体拮抗薬やβブロッカーを投与していたが，HD（ECUM）中の血圧低下を認めてからは投与を中止していた．しかし，PD併用後はHD（ECUM）中の血圧低下が改善し再開することができた．体重増加の著しいHD困難症患者では除水補助目的でのPD併用も選択肢の一つと考えられる．

A new species of a chlorarachniophyte alga, Bigelowiella longifila sp. nov., is described. It is classified as a member of Bigelowiella as flagellate cells constitute the main stage of the life cycle. However, this alga is different from the only described species of the genus, B. natans Moestrup, in having a unique amoeboid stage in the life cycle. We observed an interesting behavior of amoeboid daughter cells after cell division: One of the two daughter cells inherits the long filopodium of the parental cell, and it subsequently transports its cell contents through the filopodium to develop at its opposite end. The other daughter cell forms a new filopodium. This unequal behavior of daughter cells may have evolved before the chlorarachniophytes and some colorless cercozoans diverged.

We determined the complete nucleotide sequence of the chloroplast genome of Selaginella uncinata, a lycophyte belonging to the basal lineage of the vascular plants. The circular double-stranded DNA is 144,170 bp, with an inverted repeat of 25,578 bp separated by a large single copy region (LSC) of 77,706 bp and a small single copy region (SSC) of 40,886 bp. We assigned 81 protein-coding genes including four pseudogenes, four rRNA genes and only 12 tRNA genes. Four genes, rps15, rps16, rpl32 and ycf10, found in most chloroplast genomes in land plants were not present in S. uncinata. While gene order and arrangement of the chloroplast genome of another lycophyte, Hupertzia lucidula, are almost the same as those of bryophytes, those of S. uncinata differ considerably from the typical structure of bryophytes with respect to the presence of a unique 20 kb inversion within the LSC, transposition of two segments from the LSC to the SSC and many gene losses. Thus, the organization of the S. uncinata chloroplast genome provides a new insight into the evolution of lycophytes, which were separated from euphyllophytes approximately 400 million years ago.

胃癌の診断にて腹腔鏡補助下幽門側胃切除術を施行し, 切除標本から胃結核の合併を認めた1例を経験した. 症例は62歳の男性で, 主訴は食思不振. 来院1週間前から食思不振が出現し, 徐々に顔面蒼白となりタール便も見られるようになる. 来院時貧血を認め, 緊急内視鏡検査を施行したところ胃角部全体を覆う巨大潰瘍を認めた. 内科的治療を行ったが難治性であり, 経過観察中の生検にて胃癌の診断を得たため腹腔鏡補助下幽門側胃切除術を施行した. 術後は発熱が続き, 切除標本の病理組織学的検査結果より胃結核症の合併を認めた. 抗結核薬にて治療を行ったが, 全身状態は悪化し死亡退院となる. 原因は手術侵襲による免疫力低下による結核症の増悪と考えられた. 結核症は現在も散見され, 多彩な病像を呈することがあるため, 診断や治療に際しては慎重な対処が必要である.

症例は55歳の男性で, 発熱, 右側腹部の発赤, 疼痛を訴え来院した. 来院時, WBC 13,900/μl, CRP 27.7mg/dlと炎症反応が上昇し, CTにて右側腹部に10×6×5cmの内部が不均一な嚢胞性病変を認め, 周囲脂肪織への炎症の波及が疑われ入院となった. 入院翌日には自潰し多量に排膿があったが, 抗生剤にて炎症反応は改善し, 8日後に排膿は消失し退院となった. しかし, 13日後再度自潰し, 瘻孔造影にて小腸が造影されたため手術を施行した. 腹腔内にφ2-3mmの白色小結節とムチン状腹水, 右側腹部に癒着を認めた. 診断不能のためムチン状粘液と小結節を採取したのみで閉腹した. 病理組織学的検査にて結核菌は認めないが, 一部壊死を伴った類上皮細胞からなる結節性病変を認め, 腸結核を疑い大腸内視鏡を施行した. 回盲部から類上皮細胞肉芽腫, ラングハンス型巨細胞を認めたため腸結核の診断にて治療開始し, 約2週間後に瘻孔は閉鎖した. 腸結核は多彩な病像を呈することがあり注意を要する.

The phylogenetic position of Cystodium J. Sm. is studied here for the first time using DNA sequence data. Based on a broad sampling of leptosporangiate ferns and two plastid genes (rbcL and atpB), we show that Cystodium does not belong to the tree fern family Dicksoniaceae, as previously thought. Our results strongly support including Cystodium within the large polypod clade, and suggest its close relationship to the species-poor grade taxa at the base of the polypod topology (Sphenomeris and Lonchitis ,o rSaccoloma in this study). Further studies, with an expanded taxon sampling within polypods, are needed to fully understand the more precise phylogenetic relationships of Cystodium. Cystodium J. Sm., with its single species C. sorbifolium (Sm.) J. Sm., has traditionally been included in Dicksoniaceae (Christensen, 1938; Pichi Sermolli, 1977; Tryon and Tryon, 1982; Kramer, 1990; Stevenson and Loconte, 1996) and, more specifically, has been considered to be closely related to Dicksonia L'Her. (Hooker, 1844; Copeland, 1947; Holttum and Sen, 1961). Cystodium grows in lowland rainforests from Borneo to New Guinea and adjacent islands such as the Bismarck and Louisiade Archipelagos, and the Solomon Islands (Holttum, 1963; Croft, 1986; Kramer, 1990). It has a creeping, hairy, dictyostelic rhizome with large, bipinnate leaves that are two meters or more in length (Croft, 1986). The terminal sori are covered by a true indusium

Photosynthetic pigments were analyzed by HPLC for 27 samples of the Cladophorales (Ulvophyceae, Chlorophyta). The carotenoid compositions of the examined algae were classified into three types based on the final compound of biosynthesis of the α-carotene branch: lutein type, characterized by containing lutein as a major carotenoid and lacking loroxanthin and siphonaxanthin; loroxanthin type, characterized by containing loroxanthin and lacking siphonaxanthin; and siphonaxanthin type, characterized by containing siphonaxanthin. We constructed molecular phylogenetic tree of the species examined in the present study using 18S rRNA gene sequences and mapped the carotenoid types of the species onto the tree. The molecular phylogenetic analysis divided the Cladophorales into two major clades, clade 1 and Aegagropila-clade (clade 2), and divided clade 1 into subclade 1-1 and subclade 1-2. All the examined species positioned in the Aegagropila-clade and those of the subclade 1-1 belonged to the loroxanthin type, whereas both lutein type and siphonaxanthin type appeared only in the subclade 1-2. The clades delineated by molecular phylogenetic analysis were congruent with distribution of the carotenoid types, indicating that the carotenoid types are of taxonomic significance in the Cladophorales. Considering the distribution pattern of these carotenoid types and minimum state changes in the Cladophorales, we concluded that the loroxanthin type was the primitive (plesiomorphic) state and the siphonaxanthin type and lutein type appeared in the subclade 1-2 as advanced (apomorphic) state within this order and suggested that the cladophoralean siphonaxanthin type would have been secondarily acquired.

twenty-seven individuals of Batrachospermum helminthosum Bory were collected from various sites in Japan, from temperate Iwate Prefecture to subtropical Okinawa Prefecture. The chloroplast-encoded rbcL gene was sequenced from each sample. Phylogenetic analyses were performed using these and previously published sequences of 12 samples from North America. There were five haplotypes among the Japanese samples. Haplotype 1 was collected throughout a large geographical area corresponding to most of the sampling range in this study and was represented by most individuals (21 individuals). The other haplotypes were represented by one to three individuals, respectively. These distributions of haplotypes suggest the occurrence of one large metapopulation and/or recent colonization over a wide geographical area in Japan. Phylogenetic analysis showed two well-supported clades within B. helminthosum, one clade containing four haplotypes (1-4) from Japan and the other clade comprising five North American haplotypes. Such differences between Japanese and North American haplotypes might be a result of continental drift and subsequent isolation in the Mesozoic era. The position of haplotype 5 from Okinawa in Japan is poorly resolved, but it is not closely related to the other four Japanese haplotypes. It is suggested that several taxonomic characteristics (sexuality, point of origin of carpogonium-bearing branches, and trichogynes with or without basal knobs or branching) is not fixed genetically but is affected by environmental conditions. Based on the results of the present study, B. coerulescens Sirodot and B. elegans Sirodot should be placed in synonymy with B. helminthosum.

The Red Data Book of Japanese Vascular Plants is based on their risk of extinction. In order to construct the list, 2000 taxa were evaluated using population data and rates of decline for approximately 4400 grids, each of approximately 100 km(2). This database can be used to estimate the impact of human activity on a threatened plant's risk of extinction. In order to evaluate extinction risks and apply the evaluation to conservation actions, the discount mean time to extinction is defined as a measure of extinction risk, where the present value of a species' persistence in the future decreases exponentially. The rate of decrease has to be much less than the rate of economic discounting, in order to realize intergenerational sustainability. Increases of the inverse, and logarithm, of the discount mean time to extinction are considered measures of the extinction risk. We applied these measures to an environmental impact assessment for the Japanese World Exposition that is to be held in 2005. Development will have a greater impact on threatened Salvia species than it will on star magnolia, Magnolia tomentosa, which has been conserved by changing the site plan.

The phylogenetic position of a freshwater green alga, Aegagropila linnaei (Cladophorales, Ulvophyceae), was investigated using nuclear 18S rRNA gene sequences. This alga has usually been called Cladophora aegagropila (L.) Rabenhorst or Cladophora sauteri (Nees ex Kutz.) Kutz. Based on morphology, it was formerly classified into the section Aegagropila or into the subgenus Aegagropila, together with several marine species of the genus Cladophora. This classification is not supported by the present phylogenetic analyses in which two very distinct Cladophorales clades are recognized. Aegagropila linnaei groups together in a well-supported clade with Cladophora sp., Pithophora sp., Chaetomorpha okamurae, Arnoldiella conchophila, Wittrockiella lyallii, and Cladophora conchopheria. Aegagropila linnaei and its closely related species share some ultrastructural and biochemical characteristics, like pyrenoid structure, carotenoid composition, and cell wall composition. Freshwater species, included in the analysis, were located in two distantly related lineages, indicating that adaptation from a marine to a freshwater habitat has happened at least twice independently in the Cladophorales.

CpDNA variation in Japanese beech, Fagus crenata (Fagaceae), was studied in 45 populations distributed throughout the species' range. Two cpDNA regions were sequenced: the non-coding region between the trnL (UAA) 5′exon and trnF (GAA), and the trnK region (including matK). Thirteen distinct cpDNA haplotypes were recognized and each haplotype was found to be geographically structured. Two major clades (I and II+III) were revealed in phylogenetic analyses among the haplotypes using F. sylvatica as an outgroup. The haplotypes of Clade I were distributed mainly along the Japan Sea side of the Japanese Archipelago, while those of Clade II+III occurred chiefly along the Pacific Ocean side. Consequently, the distribution of the two major cpDNA clades suggests that there were two migration routes in the history of F. crenata; one along the Japan Sea and the other along the Pacific Ocean side of the Japanese Islands.

The morphological variation among reproductive organs of extant seed plants makes assessment of organ homology difficult. Comparisons of expression patterns of homeotic genes that control organ development will yield new information about the homology of organs to assess inferences deduced from previous morphological studies. In angiosperms, the FLORICAULA/LEAFY (FLO/LFY) genes convert a vegetative shoot meristem to a floral meristem by inducing floral homeotic genes, most of which belong to the MADS‐box gene family. To provide insights into the evolution of reproductive organs in seed plants, a FLO/LFY homologue (GpLFY) was cloned from Gnetum parvifolium. GpLFY mRNA was expressed in both the vegetative shoot apex and the female strobilus. The GpLFY mRNA signal was detected in early developmental stages of the collar and the ovule primordium, including the nucellus and three envelopes. Some MADS-box genes function as floral homeotic genes. The Arabidopsis LFY gene is a positive regulator of floral homeotic genes, and homologs of the FLO/LFY gene family in other angiosperms and gymnosperms are likely to have a similar function. To investigate the origin of the floral homeotic gene regulatory cascade involving the FLO/LFY gene, FLO/LFY homologs were cloned from a leptosporangiate fern (Ceratopteris richardii), two eusporangiate ferns (Angiopteris lygodiifolia and Botrychium multifidum var. robustum), three fern allies (Psilotum nudum, Equisetum arvense, and Isoetes asiatica), and a moss (Physcomitrella patens). The FLO/LFY gene phylogenetic tree indicates that both duplication and loss of FLO/LFY homologs occurred during the course of vascular plant evolution. The expression patterns of the Ceratopteris LFY genes (CrLFY1 and 2) were assessed. CrLFY1 expression was prominent in tissues including shoot tips and circinate reproductive leaves, but very weak in other tissues examined. Expression of CrLFY2 was also prominent in tissues, including shoot tips and circinate reproductive leaves. These patterns of expression are dissimilar to that of any Ceratopteris MADS-box gene previously reported, suggesting that the induction of MADS-box genes by FLO/LFY is not established at the stage of ferns.

Based on comprehensive comparisons of the inserted positions, lengths of introns and so on, the presence of at least three kinds of introns, which probably have independent origins, was suggested in Caulerpales.

金沢大学環日本海域環境研究センター生物多様性研究部門, We investigated the nucleotide sequences of cDNA fragments coding calcitonin from ultimobranchial glands in 2 species of urodelans (1 salamander and 1 newt) and 4 species of anurans (1 toad and 3 frogs) by the reverse transcription polymerase chain reaction (RT-PCR) method and rapid amplification of cDNA ends (RACE)-PCR method. The salamander and newt calcitonins were each 97% and 94% similar to the lungfish and caiman calcitonins that we have already reported, in the amino acid sequences. However, anuran calcitonins were not only dissimilar (63-81%) to the lungfish and caiman calcitonins but also diversified (59-91%) even among anurans. The sequence identity of toad calcitonin was always low (59-66%) among anurans.

Mitochondrial DNA restriction fragment length polymorphisms were used to examine cytoplasmic diversity within a relic-like population of Fagus hayatae, located in northern Taiwan. Fifteen trees were surveyed for three restriction endonucleases (BamHI, EcoRI and HindIII) and five mitochondrial gene probes (atpA, atp6, atp9, coxI and coxII). The analysis failed to reveal any polymorphisms, an observation that suggests cytoplasmic uniformity in the F. hayatae population examined. It is also interesting to note that the chondriome type of our F. hayatae samples is very close to that characteristic of F. crenata populations in the southernmost area of Japan.

species exhibit a paternal chloroplast inheritance and a maternal mitochondrial inheritance. The levels and patterns of cpDNA and mtDNA introgression between the two pine species, P. pumila and P. parviflora var. pentaphylla, were examined at three mountain sites in Japan. The pine species were examined by using PCR-based diagnostic genetic markers of cpDNA and mtDNA. The survey which was carried out in multiple hybrid zones demonstrated a generality in the uni-directional pattern of cytoplasmic gene flow between the two pine species, i.e. paternal cpDNA flowed from P. parviflora var. pentaphylla to P. pumila, and in contrast, maternal mtDNA flowed from P. pumila to P. parviflora var. pentaphylla. Whenever plants which had a non-native combination of cpDNA and mtDNA were observed, they always had the cpDNA haplotype of P. parviflora var. pentaphylla and the mtDNA haplotype of P. pumila. The existence of only this type of cytoplasmic chimera may suggest that F1 hybrids are successfully produced only in the crossing of P. pumila as the maternal parent and P. parviflora var. pentaphylla as the paternal parent. The present study also detected extensive mtDNA capture in populations of P. parviflora var. pentaphylla located in the southern and middle parts of the Ohu Mountains, Tohoku, Japan. In that area, nearly all of the plants examined had the mtDNA haplotype of P. pumila. The extensive mtDNA introgression suggests that seed flow could be an effective medium for interspecific gene exchange.

Using the polymerase chain reaction method, we amplified calcitonin genes from the cDNA of the ultimobranchial glands or from the genomic DNA of the blood cells and liver of various fishes. The fishes examined were primitive bony fishes (lungfish, polypterus, sturgeon, and gar), 16 species of teleosts, and cartilaginous fishes (stingray). Sequenced calcitonin genes were compared among fishes and with those of reptiles and chickens. The similarity of the calcitonin genes was the highest between the reptile and chicken groups and the primitive bony fishes (sequence similarity of the nucleotides 81–90%). The values between teleosts and the primitive bony fishes were 70–81%, with the exception of eels. Eel calcitonin genes were very similar to those of primitive bony fishes (83–88%). Stingray calcitonin genes were relatively more similar to those of the primitive bony fishes (74–78%) than to teleosts (63–73%). In goldfish and sardine, two types of calcitonin genes were found. We concluded that the genes of primitive bony fish are placed at a fundamental position in this hormone, at least among these vertebrates.

Nucleotide sequences fromrbcL were used to infer relationships of Lophosoriaceae and Hymenophyllopsidaceae. The phylogenetic positions of these two monotypic fern families have been debated, and neither group had been included in recent molecular systematic studies of ferns. Maximum parsimony analysis of our data supported a sister relationship betweenLophosoria andDicksonia, and also betweenHymenophyllopsis andCyathea. Thus, both newly-examined families appear to be part of a previously characterized and well-supported clade of tree ferns. The inferred relationships ofLophosoria are consistent with most (but not all) recent treatments. However,Hymenophyllopsis includes only small delicate plants superficially similar to filmy ferns (Hymenophyllaceae), very different from the large arborescent taxa. Nevertheless, some synapomorphic characteristics are shared with the tree fern clade. Further studies on gametophytes ofHymenophyllopsis are needed to test these hypotheses of relationship.

The amount and distribution of mitochondrial (mt) DNA restriction fragment length polymorphism was determined among individual tree samples of two Japanese beech species, Fagus crenata and F.japonica. Individual plants were collected from 16 F. crenata populations throughout the range of the species, and from three F. japonica populations. We detected enough variation to characterize eleven and three chondriome types in F. crenata and F.japonica, respectively. The grouping of beech chondriome types based upon the cladistic analysis of mtDNA polymorphism allowed us to recognize the apparent geographical patterns of mtDIMA diversity: the resulting three main groups occupied distinct geographic areas. This geographic differentiation is likely to reflect the history of the Japanese beech forests after the last glacial period of the Pleistocene. In addition, the mtDNA polymorphism encountered within F. crenata encompassed all the variation observed in F.japonica. Our result suggests the need for re-evaluation of their phylogenetic relationships.

Genetic variation in an endangered tree species, Chionanthus retusus, was examined using enzyme electrophoresis. This species occurs in East Asia from Japan to Taiwan and southern China. In Japan, however, it occurs only in two restricted areas: at the northernmost tip of Tsushima Island between Japan and Korea, and in the eastern Gifu Prefecture (Tono region) of central Honshu. In the Tono region, the habitat of C. retusus is becoming fragmented under the influence of human activity and each stand contains a few individuals of C. retusus, while the population on Tsushima consists of approximately 1000 individuals. From the fossil evidence, C. retusus has been distributed continuously including the both two regions, and the present distribution is apparently relic in Japan. In contrast to the Tono population, the population of Tsushima seems to be stable. Comparing the genetic diversity and differentiation of the populations between these two regions should shed light on how a relic woody long-lived plant keeps and/or looses its genetic diversity, and how a population differentiates from other disjunct populations. Allozyme electrophoresis of 18 loci for the Tono region indicated that 22.2% of the loci are polymorphic (P) with a gene diversity of (h) 0.087. The population of Tsushima has 38.9% polymorphic loci and a gene diversity of 0.149. We could not find any spatial genetic structure of C. retusus in the Tono region by spatial autocorrelation analysis. These results suggest that considerable genetic variation is still present in the Tono region and the influence of the recent habitat fragmentation seems not so large, yet. Because C. retusus is a long-lived tree, the population history may hinder the effect of the fragmentation at present. The genetic diversity (GST) between the populations of the Tono and Tsushima is 0.280, showing the genetic differentiation between these two regions has occurred.

On the basis of 1,290 bp sequences of the chloroplast generbcL, a molecular phylogeny of seven of nine genera of the Celtidaceae and four of six genera of the Ulmaceae was produced. These data were analyzed together with some other urticalean genera using three methods (i.e., maximum parsimony, maximum likelihood, and neighbor joining methods). Maximum likelihood topology among 18 trees obtained indicated that the Urticales are monophyletic with its common clade splitting basally into two: one leading to a line comprisingAmpelocera (traditionally placed in Celtidaceae) and Ulmaceae, and the other leading to a line comprising the remaining genera of Celtidaceae, Moraceae, and other Urticales. Ulmaceae, to whichAmpelocera is a sister group, are monophyletic, as supported by many lines of morphological evidence. In contrast to Ulmaceae, the monophyly of Celtidaceae (excludingAmpelocera) was not supported, and resolution of relationships of Celtidaceae with other Urticales, as well as of those within the family, is left for future study.

Cyanidioschyzon merolae andCyanidium caldarium are representative species among of the most primitive algae, although the two species are distinctly different in various morphological traits. We determined the nucleotide sequence of therbcL gene and a flanking 8-kb region in the plastid genome of each of these algae. In both algae, 12 genes were identified in this region, in an identical order. This gene order is not conserved in the plastid genomes of other species of the kingdom Plantae that have been sequenced to data. An additional unidentified open reading frame was also found in the two algae that we analyzed, which has not been described in any other species of algae includingPorphyra purpurea. Comparison of the amino acid sequences of selected genes also supported the conclusion thatCyanidioschyzon merolae andCyanidium caldarium are closely related and that they are distinct from other rhodophytes.

Classification of the genusAconitum (Ranunculaceae) has long been considered quite difficult because its species show high levels of morphological and ecological variability. The molecular phylogeny of Asian aconites,Aconitum subgenusAconitum was, therefore, studied based on RFLP and sequences of the intergenic spacer between thetrnL (UAA) 3′ exon andtrnF (GAA), and of thetrnL intron, of the chloroplast DNA. UsingAconitum subgenusLycoctonum as an outgroup, we obtained a statistically reliable molecular tree composed of six clades branched radiatively at the very base. There are three clades of Japanese aconites, a single clade of the species of Yunnan and Himalayas, and two clades of Siberian plants. All the tetraploid taxa of Japan we studied did not show any difference based on the molecular characters analyzed, though they have been classified into many taxa. Evolution and phytogeography of the Asian aconites as well as the phylogeny are discussed.

Eleven species belonging to seven genera in the Lardizabalaceae were analyzed in terms of restriction fragment length polymorphism (RFLP) of chloroplast DNA and the sequence of the chloroplast gene,rbcL, of Lardizabalaceae and its related families. Phylogenetic trees inferred from parsimony, neighbor joining and maximum likelihood methods based on RFLP data showed that two South American genera,Boquila andLardizabala, and three East Asian genera,Akebia, Holboellia andStauntonia are closely related to each other, respectively. On the other hand, the parsimony, neighbor joining and maximum likelihood trees constructed using sequence data of therbcL gene showed thatAkebia, Stauntonia, Boquila andLardizabala clustered as(((Akebia, Stauntonia), Boquila), Lardizabala). This difference may be attributable to fewer informative sites inrbcL genes than in RFLP in this family.Decaisnea diverges at the very base of the Lardizabalaceae.

Arisaema serratum is a perennial herb capable of changing sex expression from year to year. We developed five polymorphic microsatellite markers for A. serratum to estimate male reproductive success. Variability at these loci was examined in two populations, one at Horigane and one at Kanazawa Japan; the number of alleles per locus ranged from five to 35 (the mean 21.4) in Horigane and from three to 36 (22.0) in Kanazawa. This high allelic diversity indicates that our markers are suitable for the study of male reproductive success in A. serratum.

Arisaema serratum possesses a pitfall-trap flower pollination system. However, little is known about the efficiency and pattern of pollen movement in A. serratum. Thus, the aims of this study are to (1) determine the paternal parents of the seeds and (2) elucidate pollen movement in a natural population. Paternity analysis using microsatellite markers was performed. Seeds were collected from a natural population of A. serratum in 2001 at Horigane, Japan. Small midges became trapped in female spathe tubes during the flowering period. We found that (1) seeds in a fruit were fertilized by multiple sires; (2) seeds sired by a paternal parent were either clumped, exclusively, or randomly distributed on the spadix, depending on the parent; (3) to a great extent, a few males contributed as sires; (4) distance from a female was not a factor in the inequality of reproductive success among males; (5) male reproductive success was not correlated with its size. We conclude that pollen carryover and the trap-flower pollination system are likely to result in multiple paternity and inequality in male success.

Part of the large subunit of the ribulose-1,5-bisphosphate carboxylase/oxygenase (rubisco) gene (rbcL) was sequenced from three fern species: Adiantum capillus-veneris, Botrypus strictus, and Osmunda cinnamomea var. fokiensis. This region included 1,333 base pairs, about 90% of the gene. Maximum likelihood analysis of the deduced amino acid sequences indicated that (1) Botrypus (Ophioglossaceae) clustered monophyletically with other ferns (Adiantum, Angiopteris, Osmunda); the closest relative to Botrypus among the three species was Osmunda, which did not support the hypothesis that the Ophioglossaceae are linked to the progymnosperm-seed plant lineage. (2) Eusporangiate ferns containing Botrypus (Ophioglossaceae) and Angiopteris (Marattiaceae) were a paraphyletic group. (3) Seed plants and the four fern species examined formed a monophyletic group, but ferns and bryophytes (liverwort) did not. Variations in rates of substitution for synonymous and nonsynonymous codons were found in fern lineages.

The chromosome number of Viola pilosa was counted as 2n = 20. This number as well as other characteristics such as acaulescent stoloniferous habit, adaptations to myrmecochorous dispersal and slightly uncinate styles suggest close similarity of V. pilosa and its relatives to V. ser. Viola. On the other hand, they contrast in chromosome number, fruit characters and style morphology with other species traditionally placed in V. ser. Serpentes (W. Becker) Steenis. We thus propose that V. pilosa, including the type of V. ser. Serpentes, and its relatives be transferred to V. ser. Viola. A new name is given to what is left of Becker's "Serpentes group": V. ser. Australasiaticae Okamoto.

Partial nucleotide sequences of the large subunit of ribulose-1,5-bisphosphate carboxylase (rubisco) gene (1333 base pairs: about 90% of the gene) from several seed plants were determined. Phylogenetic trees based on amino acid sequences were inferred by using the neighbor joining and maximum likelihood methods. The results indicate (1) monophyly of gnetum group (Ephedra, Gnetum, Welwitschia), (2) monophyly of extant gymnosperms containing gnetum group, which contradicts the results of morphological data.

The acute effects of nifedipine (20 mg) on left ventricular diastolic function were investigated in 16 patients with chronic coronary artery disease by measuring left ventricular pressure with a manometer-tipped catheter and by measuring volume with cineangiography. Heart rates were maintained by right atrial pacing. Left ventricular peak systolic pressure (-15%; p less than 0.01 vs control) decreased significantly. With afterload reduction, left ventricular ejection fraction (+11%; p less than 0.01) increased. There was no significant change in left ventricular end-diastolic pressure. The diastolic peak filling rate of left ventricular volume significantly increased (+36%; p less than 0.05), whereas the time from end-systole to the peak filling rate remained unchanged. Administration of nifedipine did not improve left ventricular relaxation as assessed by the isovolumic pressure decay. There was also no significant change in the left ventricular diastolic pressure-volume relationship. We conclude that nifedipine improves left ventricular systolic function with afterload reduction but has little or no effect on left ventricular diastolic properties in patients with chronic coronary artery disease.

Opinions on the basal relationship of land plants vary considerably and no phylogenetic tree with significant statistical support has been obtained. Here, we report phylogenetic analyses using 51 genes from the entire chloroplast genome sequences of 20 representative green plant species. The analyses, using translated amino acid sequences, indicated that extant bryophytes (mosses, liverworts, and hornworts) form a monophyletic group with high statistical confidence and that extant bryophytes are likely sisters to extant vascular plants, although the support for monophyletic vascular plants was not strong. Analyses at the nucleotide level could not resolve the basal relationship with statistical confidence. Bryophyte monophyly inferred using amino acid sequences has a good statistical foundation and is not rejected statistically by other data sets. We propose bryophyte monophyly as the currently best hypothesis.

MADS-box genes encode transcription factors involved in various important aspects of development and differentiation in land plants, metazoans, and other organisms. Three types of land plant MADS-box genes have been reported. MIKC C -and MIKC*-type genes both contain conserved MADS and K domains but have different exon/intron structures. M-type genes lack a K domain. Most MADS-box genes previously analyzed in land plants are expressed in the sporophyte (diploid plant body); few are expressed in the gametophyte (haploid plant body). Land plants are believed to have evolved from a gametophyte (haploid)-dominant ancestor without a multicellular sporophyte (diploid plant body); most genes expressed in the sporophyte probably originated from those used in the gametophyte during the evolution of land plants. To analyze the evolution and diversification of MADS-box genes in land plants, gametophytic MADS-box genes were screened using macroarray analyses for 105 MADS-box genes found in the Arabidopsis genome. Eight MADS-box genes were predominantly expressed in pollen, the male gametophyte; all but one of their expression patterns was confirmed by Northern analyses. Analyses of the exon/intron structure of these seven genes revealed that they included two MIKC -type, one M-type, and four MIKC*-type MADS-box genes. Previously, MIKC*-type genes have been reported only from a moss and a club moss, and this is the first record in seed plants. These genes can be used to investigate the unknown ancestral functions of MADS-box genes in land plants. The macroarray analyses did not detect expression of 56 of 61 M-type MADS-box genes in any tissues examined. A phylogenetic tree including all three types of Arabidopsis MADS-box genes with representative genes from other organisms showed that M-type genes were polyphyletic and that their branch lengths were much longer than for the other genes. This finding suggests that most M-type genes are pseudogenes, although further experiments are necessary to confirm this possibility. Our global phylogenetic analyses of MADS-box genes did not support the previous classification of MADS-box genes into type I and II groups, based on smaller scale analyses. An evolutionary scenario for the evolution of MADS-box genes in land plants is discussed.

We describe an autopsy case of severe intracranial hemorrhage which occurred during the infusion of tissue plasminogen activator (t-PA) for acute myocardial infarction. A 75-year-old man was admitted with substernal chest pain of 3-h duration and electrocardiographic changes consistent with an acute inferior myocardial infarction. Physical examination was unremarkable, except for an initial blood pressure reading of 160/96 mmHg. The patient received 3,000 IU intravenous heparin followed by a 2.4 x 10(6) IU bolus dose of tissue plasminogen activator (t-PA) (Alteplase). This was followed by a drip infusion of 21.6 x 10(6) IU of t-PA over 1 h (total dose 41 mg). Thirty minutes after the infusion of t-PA was initiated, the patient suddenly lost consciousness and began to have violent convulsions, followed by cardiac arrest. Autopsy revealed massive hemorrhage in the bilateral cerebrum and brain stem. To our knowledge, this is the first case of sudden death during t-PA infusion therapy.

SUMMARY Gnetales, one of the extant gymnosperm orders, has traditionally been recognized to be most closely related to flowering plants, because the reproductive organ of Gnetales has some morphological characteristics similar to flowering plants. Most recent molecular phylogenetic studies do not support the sister relationship of the Gnetales and flowering plants, but instead support a close relationship between Gnetales and other extant gymnosperms. The MADS genes are transcription factors, some of which are involved in reproductive organ development in flowering plants. To resolve the discrepancy in phylogenetic inferences, and to provide insights into the evolution of reproductive organs in seed plants, four MADS genes (GpMADS1–4) were cloned from Gnetum parvifolium. GpMADS2 is likely to be a pseudogene and the other three genes were characterized. A MADS gene tree based on partial amino acid sequences showed that GpMADS3 is included in the AGL6 group, but the other two genes do not cluster with any previously reported MADS gene. The three GpMADS genes were expressed during the early stage of ovule development in the differentiating nucellus and three envelopes. A comparison of MADS gene expression among conifers, Gnetum, and flowering plants suggests that the comparable reproductive organs in Gnetum and flowering plants evolved in parallel, and is likely to support the homology between the ovule-ovuliferous scale complex of conifers and the Gnetum ovules, including the three envelopes.

A 55-year-old man had rhabdomyolysis and myocardial damage induced by palytoxin. Weakness and myalgia of four extremities occurred five hours after eating a fish. Rhabdomyolysis developed and the serum creatine phosphokinase (CK) was elevated to 40, 000 IU/ι on the 3rd day. Gastric lavage with activated charcoal and forced mannitol-alkaline diuresis therapy were performed. The patient recovered with no complication such as renal failure. In this case, palytoxin was suggested to induce myocardial damage which was demonstrated by an elevation of the myosin light chain level and a change in electrocardiogram.(Internal Medicine 37: 330-333, 1998)

Podostemaceae are aquatic herbs and are famous for their anomalous vegetative morphology, occurring only in rapids. Because of its peculiar morphology, there were no established theory nor accepted opinion on the phylogenetic position. Especially, Cusset and Cusset (1988) proposed the new class, Podostemopsida beside the Magnollopsida and Liliopsida. We, therefore, consequences extensive and detailled analyses using nucleotide sequences ofrbcL genes for two genera and three species of the family together with many representatives of the families of flowering plants to determine the closest ally. The conclusion was that the Crassulaceae is a sister group to the Podostemaceae.

Clinical and experimental observations have suggested that newly developed collaterals usually remain even after successful revascularization. We present a patient in whom coronary collateral regression was angiographically demonstrated within about 1 month after percutaneous transluminal coronary angioplasty, which led to the development of acute myocardial infarction. This case suggests that there may be a possibility of unexplained clinically important anatomical or functional regression of collaterals after reperfusion.
