This paper proposes a virtual try-on system forapparel shopping that generates high-resolution virtualization without pixel disruption. The system employs a Parser Free Appearance Flow Network, which simultaneously warps clothes and generates segmentation while exchanging information. The proposed methodologyoutperforms existing virtual fitting methods at 192 x 256 resolution, as demonstrated by the Fréchet inception distance (FID) performance metric. The system's technical specifications, software and hardware requirements, and user interface design are presented in detail. The proposed methodology achieves realistic try-on images and high- quality virtual try-on images in real-time. Future research areas include the incorporation of personalized measurements and the development of more accurate bodymodels.

Myeloproliferative neoplasms (MPNs) are caused by somatic pathogenic variants that stimulate increased production and clonal expansion of CD34 multipotent hematopoietic stem cells. Recent World Health Organization (WHO) diagnostic criteria for the diagnosis of Philadelphia chromosome (Ph) negative MPNs includes detection of mutations in the Janus Kinase 2 (), myeloproliferative leukemia (), and calreticulin () genes. The purpose of this study was to demonstrate the clinical utility of an in-house next-generation sequencing (NGS) assay targeting only these canonical genes for the molecular diagnosis of patients with Ph-negative MPNs. We tested 274 samples of patients clinically suspected of having Ph-negative MPNs using an in-house developed NGS panel. The assay consists of two parts, a multiplexed PCR and a highly multiplexed NGS workflow capable of handling diverse samples. The assay is capable of simultaneously detecting mutations in exons 12 and 14, exon 9, and exon 10. Of the 274 samples tested, 49 samples harbored mutations in the gene (48 for the V617F and 1 for exon 12), 31 harbored mutations in the gene, and two harbored mutations in the gene. One sample harbored a mutation each in the and genes. Here, we present the distribution of mutations in an Indian cohort of 274 patients from India with Ph-negative MPNs. Further sub divided into four groups based on cephalometric mandibular plane inclination to anterior cranial base (SN-GoGn) and Frankfort’s mandibular plane (FMA) angle in hypodivergent and hyperdivergent patterns. The parameters lower airway space, mandibular body, mandibular ramus, chin thickness and chin throat length were measured. Result: Results showed maximum lower airway space, mandibular body, mandibular ramus, chin thickness, and chin throat length in the hypodivergent skeletal Class I group. The minimum lower airway space, mandibular body, mandibular ramus, chin thickness, and chin throat length observed in the hyperdivergent skeletal Class II group. The inter-group comparison of all samples indicates that there was a statistically significant difference between various groups and the measures of the hypodivergent samples are more than the hyperdivergent samples irrespective of the skeletal pattern. Conclusion: The study concludes that the lower airway, mandibular ramus, mandibular body, soft tissue chin thickness, chin throat length was less in hyperdivergent skeletal Class I and skeletal Class II samples than the skeletal Class I and skeletal Class II hypodivergent. Generally, these faults occur in the long transmission line system such as single line to ground fault (L-G), double line to ground (2L-G), triple line to ground fault (3LG) and Line to line fault (L-L). Fault analysis for different kinds of faults has been done and it impacts are appeared in simulation results such as current and voltage. This results into the MATLAB software in which transmission line model is simulated. MATLAB can manipulate and invert large matrices and can be used in many several applications. MATLAB is more convenient with a recent addition called Simulink, a program which is normally used in the analysis of modern control systems. Beta thalassemia major is a common genetic disorder characterized by the reduced production or absence of beta globin, a product of the haemoglobin subunit beta (HBB) gene. Every year, approximately 10,000–12,000 children with thalassemia major are born in India. Molecular methodologies like ARMS (amplification-refractory mutation system)-PCR (polymerase chain reaction) and capillary sequencing are used to detect HBB gene mutations. It is, however, challenging to achieve comprehensive coverage of the HBB gene by these methods. Next-generation sequencing (NGS) can be used to circumvent these problems. Commercial NGS panels are prohibitively expensive and hence are not routinely implemented in most laboratories. We have developed a cost-effective, highly sensitive and specific, indigenous targeted NGS assay for detecting mutations in the HBB gene. Using this custom NGS assay, we processed 1530 samples (3017 alleles), in which we detected a spectrum of 48 pathogenic/likely pathogenic variants (mutations); IVS-I-5 (c.92+5G>C) was the most common mutation detected (allele frequency (AF) 44.55%), followed by the 619-bp deletion (AF 10.74%), c.92+1G>T (AF 6.99%), c.27_28insG (AF 6.23%), c.47G>A (AF 5.77%) and c.126_129delCTTT (AF 4.71%). Additionally, we discovered a novel mutation (c.7delC) that was submitted to the HbVar database (HbVar ID 3193) as a variant of unknown significance (VUS), probably pathogenic. Philadelphia-negative myeloproliferative neoplasms (MPNs) are a group of clonal disorders that are characterized by excessive proliferation of abnormal myeloid precursors and mature cells. Somatic driver mutations in the JAK2, CALR, and MPL genes serve as major diagnostic criteria in the classification of the MPNs, namely polycythemia vera (PV), essential thrombocythemia (ET), and primary myelofibrosis (PMF). Although initially thought to be mutually exclusive, recent studies have reported the co-existence of JAK2, MPL, and CALR mutations. In this case report, we describe a case of a Philadelphia-negative myeloproliferative neoplasm harboring mutations in the CALR [NM_004343.3:c.1092_1143del52 (NP_004334.1:p.Leu367Thrfs)] and MPL [NM_005373.2:c.1543T>A (NP_005364.1:p.Try515Arg)] genes. Given the rarity of documented co-occurrence of driver mutations in these two genes and the concomitant paucity of data regarding management of patients harboring mutations in both these genes simultaneously, there are no clear guidelines for the treatment of patients with these mutation patterns and hence it is difficult to assess the true relevance of this genotype. Background In severe alcoholic hepatitis(SAH), we aimed to characterize significant bacterial communities associated with clinical events(CE) and define bacterial relationships associated with specific CE and outcomes at baseline and on corticosteroids. Specific bacteria were found to be associated with HE [Bifidobacteriaceae,Coriobacteriaceae], sepsis [Veillonellaceae,Prevotellaceae], CTP>10 [Bifidobacteriaceae, Synergistaceae], MELD>25 [Dehalobacteriaceae,Turicibacteraceae] and death [Enterobacteriaceae,Peptococcaceae]. Significantly higher relative abundance(RA) of Lachnobacterium, Catenibacterium associated with HE at-admission while orally-represented bacteria were associated with infections at admission. Propionibacterium, Fusobacteria were associated with DF≥65 while Eubacterium, Capnocytophaga were associated with CTP. Enhydrobacter and Pediococcus were preferentially abundant post-steroid-therapy. Aerococcus was associated with post-treatment death. Prevotella was associated with survival post steroid. Upregulation of phenylpropanoid-biosynthesis(innate-immunity) in those without follow-up infections and glycerophospholipid-metabolism(cellular-integrity) in those who died were significant. Co-occurrence between Christensenella, Prevotella and mutual-exclusion between Megamonas, Citrobacter was associated with HE at admission. Mutual-exclusion between Coprococcus eutactus, Catenibacterium and Megamonas was associated with infections at admission while Enterococcus cecorum, Acinetobacter schindleri, Mitsuokella were associated with AKI at admission (figure 1). Conclusions Specific gut-microbiota, their interactions and metabolites are associated with complications of SAH as well as outcomes with steroid-therapy. Advanced metagenomics-based precision-medicine as add-on treatments may be a novel therapeutic area for improving disease outcomes.

Bacterial diversity, significant taxa and respective metabolic-profiling using QIIME®, PICRUSt™ and linear-discriminant-analysis-effect-size-method were performed based on sub-group comparisons [sepsis, no-sepsis/sites of sepsis/interleukin-6 cut-offs( 1000)/episodes of infections/death or survival at same admission/overall survival]. Results Proteobacteria, Cyanobacteria and Actinobacteria related-genera associated with pathogenicity in conditions of immune-exhaustion(Corynebacterium, Lautropia) predominated in patients with sepsis. Metabolic-pathways associated with oxidative-stress(riboflavin, glutathione metabolism) and endotoxemia (lipopolysaccharide(LPS) synthesis, bacterial chemotaxis, sulfur-relay) were significantly upregulated in sepsis. Specific-taxa were found to be associated with sites of infection in DC patients. Eggerthella (associated with blood-stream infections with piperacillin-tazobactum use) was significant in bacteremic-DC patients in the presence of upregulated MAP-K signalling pathways(nutritional deprivation and cell-wall stress) while Acinetobacter, Priopionibacterium were significantly associated with biliary infections. Protective oxidant-pathways of cysteine/methionine metabolism that increase glutathione were upregulated in no-sepsis-group. Significant representation by Gammaproteobacteria-family of sulphur-metabolizing bacteria, exaggeration of orally predominant pathogens(Prevotella) and increase in pentose-phosphate-pathway(upregulated in severe LPS-related hyperinflammatory-stress) were notable in those with interleukin-6 >1000 (figure 1). Identifying beneficial strains that reduce immune exhaustion and supplementation of favourable metabolites could improve therapeutics in DC and sepsis, for which large population-based studies remain an unmet need.

Four children with novel likely pathogenic variants in the MC4R and LEP genes are reported. Genetic analysis is crucial in children with early onset obesity and should be considered.

Background Zika, dengue, and chikungunya are three mosquito-borne viruses having overlapping transmission vectors. Methods Procedures are reported that directly test urine and plasma (for patient diagnostics) or crushed mosquito carcasses (for environmental surveillance). Carcasses are captured on paper samples carrying quaternary ammonium groups (Q-paper), which may be directly introduced into the assay. To avoid the time and instrumentation requirements of PCR, the procedure uses loop-mediated isothermal amplification (LAMP). Downstream detection is done in sealed tubes, with dTTP-dUTP mixtures in the LAMP with a thermolabile uracil DNA glycosylase (UDG); this offers a second mechanism to prevent forward contamination. Reverse transcription LAMP (RT-LAMP) reagents are distributed dry without requiring a continuous chain of refrigeration. Results The tests detect viral RNA in unprocessed urine and other biological samples, distinguishing Zika, chikungunya, and dengue in urine and in mosquitoes infected with live Zika and chikungunya viruses. Conclusions A kit, complete with a visualization device, is now available for point-of-sampling detection of Zika, chikungunya, and dengue. The assay output is read in ca. 30 min by visualizing (human eye) three-color coded fluorescence signals. Assay in dried format allows it to be run in low-resource environments. Ocular surface squamous neoplasia (OSSN) is relatively rare with an incidence of 0.13–1.9/100,000 population. OSSN includes dysplastic lesions involving the squamous epithelium of the conjunctiva or cornea. Epibulbar squamous cell carcinoma and epithelioma have been noted commonly, but cases in which the tumor is primary on the cornea are sufficiently rare to warrant reporting in each instance. We describe a rare case of noninvasive OSSN involving the entire cornea in a human immunodeficiency virus‑negative patient. Rhabdomyosarcoma is a malignant neoplasm of primitive mesenchyme exhibiting skeletal muscle differentiation. Rhabdomyosarcoma, which can histopathologically be of embryonal, alveolar, botryoid and pleomorphic types; is a rare tumor in children, with an annual incidence of 4.3 cases per million. Pleomorphic rhabdomyosarcomas is a rare variant, occurring more commonly in adults. We report a rare case of ocular pleomorphic rhabdomyosarcoma in an 18-month-old child with atypical presentation mimicking orbital cellulitis. A provisional diagnosis of orbital cellulitis was made based on clinical and radiological examination and was planned for surgery. Histopathology confirmed the diagnosis of pleomorphic rhabdomyosarcoma. This case shows that pleomorphic rhabdomyosarcoma may also occur in children, it may mimic symptoms of orbital cellulitis, and any fast growing swelling should be carefully examined with a high degree of suspicion of this entity.

The current product development environment for mechatronic systems is characterized by tight budgets, reduced development times and immense complexity across the industries - aerospace, automotive, commercial vehicles, etc. A huge emphasis is being given to increase the efficiency in product testing and to make the process more productive in increasing quality, while being cost effective. Companies are seeking software testing tools that offer a comprehensive solution that helps in achieving this goal. Automated software testing for both hardware and the software components is one of the ways the industries are trying to gain efficiency in testing. Given the fact that current mechatronic systems are quite complex, have large numbers and types of IO channels, and are implementing distributed real-time control with multiple communication protocols, a test automation tool that incorporates and tests all of the system functionality is highly desirable. In particular, due to the focus on software quality, as more and more functions are added to the network of Electronic Control Units (ECU), a greater emphasis is laid on the requirements for a comprehensive automated testing tool. The efficiency in development and planned increase in reuse of test assets is an important consideration for efficiency gain. Bringing clarity to the test description would improve readability and maintainability of these test assets. This paper shows the lessons learned from a similar evolution in Model-Based Design, moving from handwritten code to model-based graphical executable representation, can be applied to the testing arena to develop better and more efficient tests. To make the testing process efficient, the test tool should be able to not just execute tests, but organize the test results and present them in a manner easily understood by the end user. The user should be easily able to select tests to be executed and visualize execution results of those tests without need of external data analysis and reporting tools. A user should also be able to gather data related to test execution for reporting on overall software quality metrics. In the case of real-time embedded systems, the traditional approach for software testing of test execution, data capture, analysis and reporting creates a lot of inefficiency and repeated testing. A new approach to run test evaluation in parallel to test execution, using the so-called real-time testing solution, eliminates these inefficiencies and results in significant gains in testing time and improving utilization of the test infrastructure. Globalized, distributed development environments may lead to organizations adopting a variety of tool chains and test platforms. Therefore, to gain efficiency, it is desired that the test automation solution should offer easy integration with other third-party test platforms and be independent of the hardware to protect the investment in the long run. Basing such integration interface on established standards gives the user freedom to make choices. Some standards, such as HIL API from ASAM, are being proposed for test systems in the automotive industry. The paper reviews applicability and benefits of such standards. Various industry standards, such as automotive functional safety standard ISO 26262, IEC 61508, IEC 62304 and DO-178C, have addressed requirements for software testing and test tools. It is therefore critical that the selected test automation tool provide support for the prescribed methods and support the standard compliant development process. Tool qualification is a critical topic addressed by these standards and is discussed in this paper. Further, increasingly important requirements such as integration with the overall software development process, management of tests and test data, test reusability, hardware platform independence, test development and execution efficiency, etc. are discussed.

We have developed a highly targeted custom next generation sequencing-based test targeting JAK2 exons 12 and 14, CALR exon 9, and MPL exon 10, which are implicated in Philadelphia-negative chronic myeloproliferative neoplasms. The assay is capable of ultra-high sequencing depth and producing mutational detection sensitivities of 0.5% and below. We have validated the performance of the test through orthogonal testing and demonstrated a high degree of multiplexing with up to 50 samples in a single run. We show the clinical utility of this test through the description of a couple of cases with myeloproliferative disorders with Type-II and Type-II-like mutations in exon 9 of the CALR gene.

Optical transparency of the intraocular lens (IOLs) is paramount in maintaining visual improvement after cataract extraction. There have been reports of the IOL developing opacity after years of cataract surgery. IOL opacities have been reported in silicone, acrylic lens, and very rarely in polymethylmethacrylate (PMMA) IOLs. Though the exact cause is not known, long-term exposure to ultraviolet light can be one of the most probably causes, leading to disruption of PMMA and release of gaseous (nitrogen) within the PMMA substance. We report one such rare case of PMMA IOL opacity in a 75-year-old female.

Present work investigates development of the vehicle design in the Texas Tech University EcoCAR program with industry standard vehicle development process (VDP). The three different architectures considered are fuel cell, GM two-mode hybrid and belt alternator/starter system (BAS+). The design process started from the architecture selection with the use of PSAT software. Based on results, best choice of vehicle architecture among the three considered is two-mode hybrid. With Matlab/Simulink environment vehicle model is developed. The plant model and controller model is tested with software-in-loop (SIL) and hardware-in-loop (HIL) simulations. The testing of this stage is done with NI PXI and dSPACE MicroAutoBox.

This paper presents Hardware-in loop (HIL) testing for GM two-mode hybrid. First Modeling and simulation of GM two-mode hybrid using model based design (MBD) process is performed. HIL testing is performed using dSpace MicroAutoBox (MABX) and National Instruments PXI. The control strategy compiled code is uploaded to MABX using ControlDesk software and the corresponding plant model is transferred to PXI using NI Veristand software. This work is part of EcoCAR competition by the Texas Tech University. EcoCAR: The Next Challenge is a vehicle engineering competition organized by the US Department of Energy (DOE) and General Motors (GM).

Axiomatic design (AD) techniques have not previously been applied in designing the overall architecture of an automobile. The present work investigates use of axiomatic design concepts for vehicle design in the Texas Tech University Eco-CAR program. The three different architectures considered are fuel cell, two-mode hybrid, and belt alternator/starter system (BAS+). The objective in using axiomatic design methods is to choose an architecture for the EcoCAR competition vehicle that follows the principles of axiomatic design, and in turn, should prove to be the best choice of vehicle architecture among the three considered. Function means trees (FMT) and design matrices (DM) are constructed for each of the architectures and are used in deciding whether the architecture is a coupled, uncoupled, or decoupled design per the independence axiom. The choice is supported by means of simulation results obtained for each architecture. Finally, a two-mode hybrid architecture is selected based on the use of axiomatic design and the simulation results.Copyright © 2010 by ASME
