Abstract

This paper introduces LLM-jp, a cross-organizational project for the research and development of Japanese large language models (LLMs). LLM-jp aims to develop open-source and strong Japanese LLMs, and as of this writing, more than 1,500 participants from academia and industry are working together for this purpose. This paper presents the background of the establishment of LLM-jp, summaries of its activities, and technical reports on the LLMs developed by LLM-jp. 2. Evaluating Structural Generalization in Neural Machine Translation

Abstract

Compositional generalization refers to the ability to generalize to novel combinations of previously observed words and syntactic structures. Since it is regarded as a desired property of neural models, recent work has assessed compositional generalization in machine translation as well as semantic parsing. However, previous evaluations with machine translation have focused mostly on lexical generalization (i.e., generalization to unseen combinations of known words). Thus, it remains unclear to what extent models can translate sentences that require structural generalization (i.e., generalization to different sorts of syntactic structures). To address this question, we construct SGET, a machine translation dataset covering various types of compositional generalization with control of words and sentence structures. We evaluate neural machine translation models on SGET and show that they struggle more in structural generalization than in lexical generalization. We also find different performance trends in semantic parsing and machine translation, which indicates the importance of evaluations across various tasks., Comment: To appear at ACL 2024 findings

3. Analyzing Social Biases in Japanese Large Language Models

Abstract

With the development of Large Language Models (LLMs), social biases in the LLMs have become a crucial issue. While various benchmarks for social biases have been provided across languages, the extent to which Japanese LLMs exhibit social biases has not been fully investigated. In this study, we construct the Japanese Bias Benchmark dataset for Question Answering (JBBQ) based on the English bias benchmark BBQ, and analyze social biases in Japanese LLMs. The results show that while current open Japanese LLMs improve their accuracies on JBBQ by setting larger parameters, their bias scores become larger. In addition, prompts with warnings about social biases and Chain-of-Thought prompting reduce the effect of biases in model outputs, but there is room for improvement in the consistency of reasoning.

We prove that the Krull-Schmidt decomposition of the Galois module of the $p$-adic completion of algebraic units is controlled by the primes that are ramified in the Galois extension and the $S$-ideal class group. 8. Molecular identification of hyaluronate lyase, not hyaluronidase, as an intrinsic hyaluronan-degrading enzyme in Clostridium perfringens strain ATCC 13124

Abstract

Abstract Clostridium perfringens, an opportunistic pathogen, produces mu-toxin hyaluronidases including endo-β-N-acetylglucosaminidases (Nags) as a virulence invasion factor. To clarify an intrinsic factor for degradation of host extracellular hyaluronan, we focused on hyaluronate lyase (HysA), distinct from endo-β-N-acetylglucosaminidases. C. perfringens strain ATCC 13124 was found to assimilate host-derived extracellular mucosubstances, hyaluronan and mucin, which induced expression of the hyaluronan-related genetic cluster, including hyaluronate lyase gene (hysA), but repressed endo-β-N-acetylglucosaminidase genes. This genetic cluster is conserved in some strains of C. perfringens. The recombinant strain ATCC 13124 hyaluronate lyase HysA showed an hyaluronan-degrading activity through β-elimination reaction. The hyaluronan-degrading enzyme in the culture supernatant of strain ATCC 13124 exhibited the lyase activity and was identical to the recombinant hyaluronate lyase on the native-PAGE gel, followed by activity straining. These results demonstrated that the intrinsic hyaluronan-degrading enzyme of C. perfringens strain ATCC 13124 is hyaluronate lyase HysA, but not hyaluronidases NagH, NagJ, and NagK.

Abstract

Antipsychotic treatment is vital for patients with schizophrenia even in the perinatal period, but the impact at the molecular biological level on offspring is unclear. The aim of the present study was to investigate the effects of intraperitoneal haloperidol injection to pregnant mice on glutamate and GABA receptors in the brain of offspring mice. Eight-week-old pregnant mice were treated with either intraperitoneal haloperidol or normal saline injection, and their offspring were defined as F1 mice. In addition, eight-week-old male mice were used as acute mice that were intraperitoneally injected with haloperidol or normal saline for 20 days. mRNA expression levels were measured by RT-qPCR. Western blotting was performed of the frontal lobes of F1 mice. In the hippocampi of F1 mice, Grik3 (p = 0.023) and Gabra3 (p = 0.004) mRNA expression levels were significantly higher in the haloperidol group than in the control group, whereas Gria2 (p

Abstract

In this study, we analyze the operational process of nanodiamond (ND) quantum thermometry based on wide-field detection of optically detected magnetic resonance (ODMR) of nitrogen vacancy centers, and compare its performance with that of confocal ODMR detection. We found that (1) the thermometry results are significantly affected by the shape and size of the camera region of interest (ROI) surrounding the target NDs and that (2) by properly managing the ROI and acquisition parameters of the camera, a temperature precision comparable to confocal detection in living cells can be obtained by wide-field ODMR. Abstract

The requirements placed on the foundry industry are mainly related to the need to maintain an appropriate quality–price ratio of the product and to maintain an appropriate production regime that takes into account modern legal solutions related to environmental protection. This work is part of the trend of searching for new or previously unused materials from renewable sources in moulding sand technology. This article concerns the possibility of using barley malt in moulding material technology as a binding material. The presented work contains the results of research on the release of gases during casting, where high temperatures cause the decomposition of chemical substances. The tests confirmed that moulding sands with barley malt as a binder do not cause excessive emissions of harmful gaseous products when pouring moulds with liquid casting alloy. The volume of gases formed was smaller than that of commonly used moulding sands with other binders. The total volume of gases produced and the kinetics of their release indicate that casting in this type of moulding sand did not cause any technological inconvenience or casting defects of gaseous origin. This was confirmed by research involving obtaining iron castings in industrial conditions using barley malt as a binder in moulding sand technology, which were then subjected to visual assessment and roughness analysis.

Abstract

Syphilitic infection is usually observed in young patients, and the first stage of the disease (primary syphilis) is characterized by painless cutaneous and lymph node lesions. Herein, we describe a 71-year-old Japanese man with primary syphilis that presented as unilateral inguinal lymphadenopathy without skin lesions. Originally, an incarcerated hernia was clinically suspected. The lymph node pathologically showed suppurative lymphadenitis with vague granulomas; immunohistochemistry was highly suggestive of a syphilitic infection. Primary syphilis was confirmed by positive serological tests. Syphilitic infection should be included in the clinicopathological differential diagnoses for patients with inguinal lymphadenopathy, even in elderly patients without skin lesions.

Abstract

In this paper, we propose a fabrication method of a high temperature-resistant ultrasonic transducer for non-destructive tests. In typical pipe non-destructive wall thickness measurements, couplant material and backing material which do not have heat resistance are necessary. Therefore, there are restrictions for the practical online pipe thickness monitoring in chemical and power plants. To avoid a limitation of these non-heat-resistant material, we propose a couplant-less transducer fabrication method by attaching a piezoelectric film directly on the tested object with the spray coating method. Our method is based on the sol-gel spray technique to fabricate a piezoelectric ceramic film. With the sensor fabricated with this method, we measured the wall thickness under high temperature (100, 200, 300, 400, 500, 600°C) conditions. From the results of the high-temperature experiment, it was found that the sensitivity decrease due to temperature rise was limited to 3dB and the difference of frequency response was also limited to 2 MHz at the second peak frequency at the maximum. According to the results, the performance deterioration due to high temperature was within an acceptable range. Additionally, the wall thickness could be calculated from reflected pulse waves. We concluded that sufficient availability of the piezoelectric film under high temperature were confirmed. This paper describes the effectiveness of piezoelectric film attached directly on the tested object.

Abstract

Abstract Background We experienced a nosocomial outbreak of coronavirus disease 2019 (COVID-19) from November 2020 to February 2021, during the third wave of the pandemic in Japan. Methods We retrospectively assessed the characteristics and data of 20 inpatients undergoing hemodialysis who were hospitalized for treatment of diseases other than COVID-19 during the COVID-19 nosocomial outbreak (“inpatient,” IP), and of 10 outpatients undergoing hemodialysis who were hospitalized for the care of COVID-19 under outpatient visits (“outpatient,” OP). Results Eleven patients in the IP group (55%) and one in the OP group (10%) died. Kaplan–Meier analysis showed that the IP group died more rapidly than the OP group (p = 0.02). Multivariate analysis among all hemodialysis patients showed that the IP group was not at risk of mortality independently; however, the activity of daily life (ADL) dependency was found to be an independent factor in increasing the risk of mortality (hazard ratio: 7.618). Conclusion Our findings show that the nosocomial infected group has a worse prognosis, although it is not an independent predictor for the risk of mortality. ADL dependency could predict the risk of mortality in all hemodialysis patients with COVID-19 during the third wave pandemic in Japan.

Abstract Objective The detection and monitoring of DNA methylation status in circulating tumor cell DNA (ctDNA) provides critical insights into cancer diagnosis and progression. The methylation status of the Dickkopf-related protein 3 (DKK3) promoter region is correlated with the metastasis and recurrence of multiple cancers. Thus, detecting the methylation status via non-invasive methods is essential for the diagnosis and prognosis of cancers. Using a droplet digital polymerase chain reaction approach, we have developed a highly sensitive and quantitative measurement of methylated and unmethylated DKK3 derived from circulating cell-free DNA (ccfDNA). Results We confirmed the specificity of droplet digital methylation specific polymerase chain reaction (ddMSP). We selected the optimal bisulfite conversion method using commercially available kits. We validated the ddMSP analysis system by analyzing the methylation status of genomic DNA extracted from cultured mesothelioma cells and mesothelial cells. Our system quantified approximately 30 copies of cell-free DNA per 4 mL, which is sufficient for detecting ctDNA. Finally, we quantified methylated and unmethylated DKK3 copies in ccfDNA from 21 patients with malignant mesothelioma.

Abstract Background Cancer stem cells (CSCs) are generated under irregular microenvironment in vivo, of which mimic is quite difficult due to the lack of enough information of the factors responsible for cancer initiation. Here, we demonstrated that mouse induced pluripotent cells (miPSCs) reprogrammed from normal embryonic fibroblasts were susceptible to the microenvironment affected by cancer cells to convert into CSCs in vivo. Methods Three different pancreatic cancer line cells, BxPC3, PANC1, and PK8 cells were mixed with miPSCs and subcutaneously injected into immunodeficient mice. Tumors were evaluated by histological analysis and cells derived from iPSCs were isolated and selected from tumors. The isolated cells were characterized for cancer stem cell characters in vitro and in vivo as well as their responses to anticancer drugs. The impact of co-injection of iPSCs with cancer cells on transcriptome and signaling pathways of iPSCs was investigated. Results The injection of miPSCs mixed with human pancreatic cancer cells into immunodeficient mice maintained the stemness of miPSCs and changed their phenotype. The miPSCs acquired CSC characteristics of tumorigenicity and self-renewal. The drug responses and the metastatic ability of CSCs converted from miPSCs varied depending on the microenvironment of cancer cells. Interestingly, transcriptome profiles of these cells indicated that the pathways related with aggressiveness and energy production were upregulated from the levels of miPSCs. Conclusions Our result suggests that cancer-inducing microenvironment in vivo could rewire the cell signaling and metabolic pathways to convert normal stem cells into CSCs.

El despertar actual en este ámbito tiene que ver con el poder vinculador de Internet y de las redes sociales por una parte y, por la otra, con la pasión, la imaginación y la visión de las nuevas generaciones de profesorado. Pero este despertar no debe ocultar ni llevar a menospreciar las iniciativas asociativas anteriores, pues aprendiendo de los errores y aciertos del pasado es como las nuevas asociaciones marfileñas de ELE se enfrentarán con éxito a los retos del presente y del futuro.

Abstract

Omidenepag isopropyl (OMDI) is an intraocular pressure (IOP)-lowering drug used to treat glaucoma. The active form of OMDI, omidenepag (OMD), lowers elevated IOP, the main risk factor for glaucoma, by increasing the aqueous humor outflow; however, a detailed understanding of this mechanism is lacking. To clarify the IOP-lowering mechanism of OMDI, the effects of OMD on the mRNA expression of the extracellular matrix, matrix metalloproteinases (MMPs), and tissue inhibitors of metalloproteinases (TIMPs) were evaluated in human trabecular meshwork cells. Under 2D culture conditions, the mRNA expression of FN1, COL1A1, COL1A2, COL12A1, and COL13A1 decreased in a concentration-dependent manner after 6 or 24 h treatment with 10 nM, 100 nM, and 1 μM OMD, while that of COL18A1 decreased after 6 h treatment with 1 μM OMD. Significant changes in expression were observed for many MMP and TIMP genes. Under 3D culture conditions, the extracellular matrix-related genes COL12A1 and COL13A1 were downregulated by OMD treatment at all three concentrations. Under both 2D and 3D culture conditions, COL12A1 and COL13A1 were downregulated following OMD treatment. Reduction in the extracellular matrix contributes to the decrease in outflow resistance, suggesting that the downregulation of the two related genes may be one of the factors influencing the IOP-lowering effect of OMDI. Our findings provide insights for the use of OMDI in clinical practice.
