Abstract

Baorangia albostipitata sp. nov. from India is proposed based on morphological and molecular phylogenetic data. The species is characterized by a pinkish pileus with yellowish pileal margin, whitish pileal context that changes to pale blue very slowly on bruising or cut, yellowish hymenophore without any color change on bruising or cut, whitish stipe that changes to bright orange when bruised, and subglobose to ellipsoid basidiospores. Abstract

Abstract

Wood is a major pool of organic carbon that is highly resistant to decay, owing largely to the presence of lignin. The only organisms capable of substantial lignin decay are white rot fungi in the Agaricomycetes, which also contains non?lignin-degrading brown rot and ectomycorrhizal species. Comparative analyses of 31 fungal genomes (12 generated for this study) suggest that lignin-degrading peroxidases expanded in the lineage leading to the ancestor of the Agaricomycetes, which is reconstructed as a white rot species, and then contracted in parallel lineages leading to brown rot and mycorrhizal species. Molecular clock analyses suggest that the origin of lignin degradation might have coincided with the sharp decrease in the rate of organic carbon burial around the end of the Carboniferous period.

Abstract

Clavaria viriditincta Krishnapriya & T.K.A. Kumar, sp. nov. (Fig. 1) MycoBank no:—MB846541 Type:— INDIA. Kerala State, University of Calicut campus (11.11 oN, 75.89 oE, altitude 32 m), 06 June 2022, Krishnapriya K., (holotype, ZGCKP247). Etymology:—‘ viriditincta’ refers to the green colour of the basidiomata. Diagnosis:— Clavaria viriditincta differs from other Clavaria species by its large, distinclty green basidiomata, ellipsoid basidiospores (6–7 × 4–5 µm), and inflated hyphae with ampulliform septal swellings. Description:— Basidiomata 30–80 × 3–5 mm, simple, unbranched, cylindrical, solid when young, becoming fistulose with age, terete in cross section, apex acute to subacute, glabrous, fertile throughout, deep green, darker at the extreme apex, no colour change on bruising, context fleshy, without any distinct odour, no colour reaction in Fe 3 Cl and KOH. Basidiospores 6–7 × 4–5 µm (Q =1.2–1.7 µm, Q m =1.4 µm), ellipsoid, smooth, thin-walled, hyaline, guttulate, apiculus prominent (up to 1 µm long), inamyloid, cyanophilic in cotton blue. Basidia 30–40 × 7–8 µm, agguttulate, clavate, without basal clamp-connection, sterigmata 4 (up to 5 µm long). Hymenium 20–30 µm wide. Subhymenium 40–50 µm wide. Context composed of generative hyphae, 4–10 µm wide, inflated up to 20 µm wide, interwoven, septate, with ampulliform septal swellings, hyaline, thick-walled (up to 1 µm, inamyloid, cyanophilic in cotton blue. Hyphal clamp-connections absent. Habitat and distribution:—on soil, among leaf litter, gregarious. This species was collected from an area dominated by Hevea brasiliensis. Abstract

Tremella poilkavensis A. Thomas & T.K.A. Kumar Fig.1: A–P. MycoBank: 847007 Etymology:—Refers to the collection locality, Poilkave sacred grove. Diagnosis:— T. poilkavensis differs from other Tremella species by their medium-sized, amber brown to blackish brown basidiocarp, pyriform to capitate basidia, ellipsoid, subcylindrical, subglobose or amygdaliform basidiospores, conidiospores budding off from basidia, growth on ascomata of Biscogniauxia species and presence of host hyphae within the basidiocarp. Holotype:— INDIA. Kerala State, Kozhikode District, Poilkave sacred grove, on ascomata of a Biscogniauxia sp. growing on dead and decaying wood, 14 July 2021, Anjitha Thomas, ZGCAT243. Basidiocarp medium-sized, 55–75 × 7–26 mm, gelatinous to cartilaginous, cerebriform, hollow, sessile, broadly attached to the substratum, amber brown to blackish brown when fresh, turning more blackish when dry. Spore print pale yellow. Hymenium pale yellow to brownish yellow in water, dikaryophyses absent. Basidia 19–31 × 9–13.3 µm (including stalk), pyriform to capitate, four-celled, oblique or longitudinally septate, cruciate, thick-walled, guttulate, pale yellow to brownish yellow, with basal clamp connection. Stalk of basidia 5–21 × 2.5–4.5 µm. Sterigma 21–38 × 2–3 µm. Basidia budding off subglobose to ellipsoid conidiospores. Basidiospores 6–9.8 × 4–6.2 µm (Q=1.06–1.8 µm, Qm=1.43 µm), ellipsoid, subcylindrical, subglobose or amygdaliform, hyaline, thin-walled, smooth, apiculate, guttulate. Direct germination, secondary spore production and microconidia formation observed. Microconidia 2–3 × 1.5–2 µm, subglobose to ellipsoid. Terminal swollen cells present within the trama, 7.7–13.3 × 4–7.6 µm, broadly fusiform, ellipsoid, lemoniform, oblong, capitate or clavate in shape, sometimes with 1–3 × 1–1.53 µm apical protuberances. Tramal hyphae mixed with host hyphae, 2–4 µm wide, thick-walled, branched, smooth, guttulate, with clamp connections. Haustoria abundantly present throughout the trama, frequently branching, with basal clampconnection, often attached to the host hyphae. Additional specimens examined:— INDIA. Abstract

A Dacryopinax species that was cultured in Costa Rica and fruited in the laboratory provided DNA for the first sequenced genome for the Dacrymycetes. Here we characterize the isolate morphologically and cytologically and name it D. primogenitus. Molecular sequences from the nuclear large subunit gene and internal transcribed spacer indicated that it is closely related to the South American D. indacocheae with which it agrees structurally. Both species form conidia on the basidiocarp, and D. primogenitus also forms them on the mycelium. Unlike previous reports for the Dacrymycetales postmeiotic nuclear division results in uninucleate basidiospores and six residual nuclei in the basidium after basidiospore discharge. Ultrastructural analysis shows the characteristic septalpore apparatus for the class and endogenous origin of the epibasidia/sterigmata, which may be a common occurrence in Dacrymycetes and the early diverging orders of its sister class, the Agaricomycetes. Abstract

Neolecta represents the earliest derived extant ascomycete lineage (Taphrinomycotina) to produce ascomata. For this reason the genus has been of interest with regard to ascoma evolution in ascomycetes. However, the evidence is equivocal regarding whether the Neolecta ascoma is homologous or analogous to ascomata produced in the later derived ascomycete lineages (Pezizomycotina). We investigated phylogenetically informative septal pore ultrastructure of Neolecta vitellina to compare with Pezizomycotina. We found that crystalline bodies that block nonascogenous septal pores in Neolecta differ from Woronin bodies, a synapomorphy for the Pezizomycotina, in three ways: (i) vacuolar origin, (ii) associated material and (iii) being loosely membrane bound. We also observed a unique type of membranous material within the septal pore, as well as distant from the septal pore, that appears to be associated with the endoplasmic reticulum. The vacuolar crystals and membranous material might have a function analagous to septal pore structures (e.g. Woronin bodies, lamellate structures) in the Pezizomycotina. Abstract

Molecular phylogenetic analyses indicate that the monophyletic classes Orbiliomycetes and Pezizomycetes are among the earliest diverging branches of Pezizomycotina, the largest subphylum of the Ascomycota. Although Orbiliomycetes is resolved as the most basal lineage in some analyses, molecular support for the node resolving the relationships between the two classes is low and topologies are trustable. We provide ultrastructural evidence to inform the placement of Orbiliomycetes by studying an Orbilia, a member of the only order (Orbiliales) of the class. The truncate ascus apex in the Orbilia is thin-walled except at the margin, and an irregular wall rupture of the apex permits ascospore discharge. Ascus, ascogenous and non-ascogenous hyphae were simple septate, with septal pores plugged by unelaborated electron-dense, non-membranous occlusions. Globose Woronin bodies were located on both sides of the septum. Nuclear division was characterized by the retention of an intact nuclear envelope, and a two-layered disk-shaped spindle pole body. The less differentiated nature of the spore discharge apparatus and septal pore organization supports an earliest diverging position of Orbiliomycetes within the subphylum, while the closed nuclear division and disk-shaped spindle pole body are interpreted as ancestral state characters for Ascomycota. Abstract

Wallemia (Wallemiales, Wallemiomycetes) is a genus of xerophilic Fungi of uncertain phylogenetic position within Basidiomycota. Most commonly found as food contaminants, species of Wallemia have also been isolated from hypersaline environments. The ability to tolerate environments with reduced water activity is rare in Basidiomycota. We sequenced the genome of W. sebi in order to understand its adaptations for surviving in osmotically challenging environments, and we performed phylogenomic and ultrastructural analyses to address its systematic placement and reproductive biology. W. sebi has a compact genome (9.8 Mb), with few repeats and the largest fraction of genes with functional domains compared with other Basidiomycota. We applied several approaches to searching for osmotic stress-related proteins. In silico analyses identified 93 putative osmotic stress proteins; homology searches showed the HOG (High Osmolarity Glycerol) pathway to be mostly conserved. Despite the seemingly reduced genome, several gene family expansions and a high number of transporters (549) were found that also provide clues to the ability of W. sebi to colonize harsh environments. Phylogenetic analyses of a 71-protein dataset support the position of Wallemia as the earliest diverging lineage of Agaricomycotina, which is confirmed by septal pore ultrastructure that shows the septal pore apparatus as a variant of the Tremella-type. Mating type gene homologs were identified although we found no evidence of meiosis during conidiogenesis, suggesting there may be aspects of the life cycle of W. sebi that remain cryptic., (Copyright © 2012 Elsevier Inc. Abstract

The phylogeny of Auriculoscypha anacardiicola, an associate of scale insects in India, is investigated using subcellular characters and MP and Bayesian analyses of combined nuLSU-rDNA, nuSSU-rDNA and 5.8S rDNA sequence data. It has simple septa with a pulley-wheel-shaped pore plug, which is diagnostic of phytoparasitic members of the Pucciniomycetes, and hyphal wall break on branching, a phenomenon unique to some simple septate heterobasidiomycetes. The septal ultrastructure of A. anacardiicola is similar to that of the genus Septobasidium. The close relationship to Septobasidium is also confirmed by rDNA sequence analyses. The polyphyletic nature of the order Platygloeales, noted in earlier studies, is evident from the present molecular analysis as well. The placement of Auriculoscypha in the Platygloeales can no longer be justified and both ultrastructural and molecular evidence strongly support the placement of Auriculoscypha in the Septobasidiales.
