39. Biopesticide production using Bacillus thuringiensiskurstaki by valorization of starch industry wastewater and effluent from aerobic, anaerobic digestion

Abstract

Introduction: Unconventional alternatives such as aerobic and anaerobic effluent from starch industry contain essential nutrients for Btk active ingredient synthesis. Effluent from starch industry is rich in carbon and nitrogen and can replace expensive feedstock used during the fermentation process. Objectives : The main objective of this study was to achieve a biopesticide formulation from starch industry wastewater (SIW) with high entomotoxicity (UI/ml) of larvae comparable to Foray 76B, which is a commercial biopesticide. Methods: Bacillus thuringiensisvar kurstaki HD1 (Btk) strain was cultivated and sub-cultured to aerobic, anaerobic digested effluent and SIW. Pre-treatment was carried on these different substrates to enhance the residual carbon required for Btk growth and delta endotoxin synthesis. After 48 hours of fermentation, cells count and delta-endotoxin were determined. A biopesticide formulation containing fermented broth and adjuvants was fed to larvae to determine larvae mortality. Results: Btk cell growth and sporulation profile in SIW media displayed a high total cell count and viable spores compared to btk growth in anaerobic or aerobic media after 48h fermentation. The maximum endotoxin concentration in the SIW medium was 435μg/mL, whereas, in anaerobic and aerobic effluent, the maximum concentrations were at 161 μg/mL and 136 μg/mL, respectively. When acidic treatment was performed at pH 2 for these substrates, entomotoxicity obtained from aerobic and anaerobic biopesticide formulations displayed significantly higher entomoxicity than the untreated ones. The entomotoxicity of SIW treated at pH 2 was equivalent to the standard Foray 76B which is 20,000 IU/μL. Conclusion : Anaerobic and aerobic effluent did not contain enough total organic carbon to augment Btk growth and entomotoxicity. Substrates pre-treated at pH 2 provided significant organic matter for Btk growth and resulted in larval mortality equivalent to the com ercial biopesticide Foray 76B.

Abstract

BACKGROUND: Crude glycerol is an industrial by‐product of biodiesel producing companies and requires a high cost for purification. In fact, it is a good carbon source and can be used for lipid production from oleaginous microbes. However, crude glycerol has several impurities that may impact the cellular metabolism for lipid production. RESULTS: In this study, crude glycerol from different sources was employed for lipid production and the effect of its impurities on the biomass and lipid production was investigated on Yarrowia lipolytica SKY7, which is a well‐known yeast for lipid and citric acid production. Growth inhibition was observed in BIOCARDEL, BIOLIQ, and ROTHSAY glycerol when compared with pure glycerol. This was due to high sodium concentrations in BIOCARDEL, high potassium concentrations in BIOLIQ, and high sulphur concentrations in ROTHSAY glycerol. Among three crude glycerol sources, the highest lipid concentration (14.78 g L−1) was obtained using BIOCARDEL glycerol at 96 h. However, the higher citric acid concentrations of 18.70 g L−1 in ROTHSAY glycerol and 12.00 g L−1 in BIOLIQ were obtained at 96 h when compared with 8.30 g L−1 in BIOCARDEL glycerol. CONCLUSION: A high potassium and sulphur concentration in glycerol medium inhibits cell growth and lipid production in Yarrowia lipolytica SKY7, while it favors citric acid production. © 2020 Society of Chemical Industry [ABSTRACT FROM AUTHOR]

Abstract

The aim of this study was to produce Bacillus thuringiensis-based biopesticide using starch-producing industry wastewater (SIW) fortified with soybean medium and optimize the formulated product using different adjuvants. This study was necessary as low endotoxin concentration is obtained in formulated biopesticide when SIW alone is used as fermentation medium. The fermentation runs were conducted using SIW alone and SIW fortified with 25% soybean (w/v) medium in 2000 L and 150 L bioreactor, respectively. SIW supplemented with soybean medium showed an increase in cell count (from 1.95 × 108 to 1.65 × 109 CFU mL–1), spore synthesis (from 1.5 × 108 to 1.35 × 109 CFU mL–1) and endotoxin concentration (from 436 to 1170 μg mL–1) when compared to SIW medium alone. The fermented broth was concentrated using continuous centrifugation and adjuvants were added for biopesticide formulation in order to enhance its resistance against UV rays and rainfastness. Entomotoxicity of the formulation produced using fermented broth of SIW fortified with soybean (38,000 IU μL–1) was higher than that obtained by SIW medium alone (21,000 IU μL–1), commercial biopesticide Foray 76B (20,000 IU μL–1) and Btk sander's (12,500 IU μL–1).
