Background: Alzheimer's disease (AD) is a neurodegenerative disease characterized by beta-amyloid (Aβ) deposition and increased acetylcholinesterase (AchE) enzyme activities. Indole 3 carbinol (I3C) and diindolylmethane (DIM) are reported to have neuroprotective activities against various neurological diseases, including ischemic stroke, Parkinson's disease, neonatal asphyxia, depression, stress, neuroinflammation, and excitotoxicity, except for AD. In the present study, we have investigated the anti-AD effects of I3C and DIM. Methods: Docking and molecular dynamic studies against AchE enzyme and network pharmacological studies were conducted for I3C and DIM. I3C and DIM's neuroprotective effects against self and AchE-induced Aβ aggregation were investigated. The neuroprotective effects of I3C and DIM against Aβ-induced neurotoxicity were assessed in SH-S5Y5 cells by observing cell viability and ROS. Results: Docking studies against AchE enzyme with I3C and DIM show binding efficiency of −7.0 and −10.3, respectively, and molecular dynamics studies revealed a better interaction and stability between I3C and AchE and DIM and AchE. Network pharmacological studies indicated that I3C and DIM interacted with several proteins involved in the pathophysiology of AD. Further, I3C and DIM significantly inhibited the AchE (IC50: I3C (18.98 µM) and DIM (11.84 µM)) and self-induced Aβ aggregation. Both compounds enhanced the viability of SH-S5Y5 cells that are exposed to Aβ and reduced ROS. Further, I3C and DIM show equipotential neuroprotection when compared to donepezil. Conclusions: Our findings indicate that both I3C and DIM show anti-AD effects by inhibiting the Aβ induced neurotoxicity and AchE activities. [ABSTRACT FROM AUTHOR]

Aliphatic-aromatic biodegradable random copolyester, poly (butylene sebacate-co-butylene isophthalate) (PBSeI) was synthesized by a two step direct melt polycondensation method. This copolyester was characterized by solubility studies, viscosity measurements, infrared, ¹H-NMR, 13C-NMR and thermal data. However, to be a real alternative to classical synthetic polymers and find applications, the thermal properties of biodegradable polymers have to be improved. Synthesis of nano-biocomposites, which are obtained by incorporation of nanofillers into a biomatrix, is an interesting way to create polymers with characteristic properties. In the present study, the nano-biocomposites were synthesized by dispersing different weight percentages (1, 3 and 5%) of organo-modified montmorillonite clay (OMMT) into the biodegradable polyester matrix by the solvent intercalation method. Structural characterization and thermal analysis were carried out to better understand, the relationship between the structuring of the nanofillers and the properties of nano-biocomposites. The X-ray diffraction patterns obtained for the systems confirmed the nanodispersion of the OMMT-clay in the polyester networks. The scanning electron microscopy and high resolution transmission electron microscopy analyses showed that there was no phase separation between the two components which confirmed the compatibility of polyester with the organo-modified clay system. The results show that clay incorporation improved the thermal properties of PBSeI due to the exfoliated structure of the nanocomposites formed, and thus may increase the applicability of this biodegradable polymer in different fields. New low bandgap small molecules based on a squaraine (SQ) chromophore, bis[4-(2,6-di-tert-butyl)vinylpyrylium]squaraine (TBU-SQ), bis[2,6-di-tert-butyl-4-(prop-1-en-2-yl)pyrylium]squaraine (MeTBU-SQ) and bis[4-(but-1-en-2-yl)-2,6-di-tert-butylpyrylium]squaraine (EtTBU-SQ), were synthesized and used as electron donors along with PC70BM for their application in solution processed organic bulk-heterojunction (OBHJ) solar cell (SC). The long wavelength of these SQ dyes are located in between 650-750 nm in thin films and the optical bandgaps are about 1.64, 1.52 and 1.48 eV, respectively. The electrochemical properties of these SQ dyes indicate that they are well suited for the fabrication of OBHJSCs as electron donors along with fullerene derivatives as electron acceptors. The OBHJ photovoltaic (PV) devices fabricated with the blend of TBU-SQ:PC70BM, MeTBU-SQ:PC70BM and EtTBU-SQ:PC70BM cast from chloroform (CF) solvent exhibited a power conversion efficiency (PCE) of 1.71%, 2.15%, and 1.89%, respectively. The PCE of the OBHJSCs based on MeTBU-SQ:PC70BM blends cast from DIO-THF (DIO = 1,8-diiodooctane) additive solvent and cast from DIO-THF with subsequent thermal annealing have been further improved up to 2.73% and 3.14%, respectively. This enhancement in the PCE is attributed to the improvement in the crystalline nature of the blend and more balanced charge transport resulting from the higher hole mobility. All these results have been supported by the quantum chemical calculations.
