Discharge produced plasma (DPP) is a promising tool to generate effective extreme ultraviolet (EUV) light. A hydrogen-like nitrogen ion emits light at 13.4 nm with an optical allowed atomic transition from n = 3 to 2, where n stands for a principal quantum number. We focus on the properties of population inversion between n =3 and 2 in recombining phase. Population inversion is calculated in the framework of population kinetics, and a gain coefficient G is estimated. In this study, dielectronic-capture ladderlike (DL) processes are included. At high density and low temperature, the DL deexcitations make a large contribution to the population inversion.

The gonadal sex differentiation in red sea bream,Pagrus major, which is one of the most important species for aquaculture in Japan, was revealed histologically. The suitable conditions for induction of all-male groups in the fish were investigated, and functional males were induced by the conditions of oral administration of 17a-methyltestosterone. Introduction: Cancer has become a major source of morbidity and mortality globally. Despite the threat that cancer poses to public health in sub-Saharan Africa (SSA), few countries in this region have data on cancer incidence. In this paper, we present estimates of cancer incidence in Nigeria based on data from 2 population-based cancer registries (PBCR) that are part of the Nigerian national cancer registry program., Materials and Methods: We analyzed data from 2 population based cancer registries in Nigeria, the Ibadan Population Based Cancer Registry (IBCR) and the Abuja Population Based Cancer Registry (ABCR) covering a 2 year period 2009-2010. Data are reported by registry, gender and in age groups. We present data on the age specific incidence rates of all invasive cancers and report age standardized rates of the most common cancers stratified by gender in both registries., Results: The age standardized incidence rate for all invasive cancers from the IBCR was 66.4 per 100000 men and 130.6 per 100000 women. In ABCR it was 58.3 per 100000 for men and 138.6 per 100000 for women. A total of 3393 cancer cases were reported by the IBCR. Of these cases, 34% (1155) were seen among males and 66% (2238) in females. In Abuja over the same period, 1128 invasive cancers were reported. 33.6% (389) of these cases were in males and 66.4% (768) in females. Mean age of diagnosis of all cancers in men for Ibadan and Abuja were 51.1 and 49.9 years respectively. For women, mean age of diagnosis of all cancers in Ibadan and Abuja were 49.1 and 45.4 respectively. Breast and cervical cancer were the commonest cancers among women and prostate cancer the most common among men. Breast cancer age standardized incidence rate (ASR) at the IBCR was 52.0 per 100000 in IBCR and 64.6 per 100000 in ABCR. Cervical cancer ASR at the IBCR was 36.0 per 100000 and 30.3 per 100000 at the ABCR. The observed differences in incidence rates of breast, cervical and prostate cancer between Ibadan and Abuja, were not statistically significant., Conclusion: Cancer incidence data from two population based cancer registries in Nigeria suggests substantial increase in incidence of breast cancer in recent times. A detailed paleomagnetic study conducted in the Sangiran area, Java, has provided a reliable age constraint on hominid fossil-bearing formations. A reverse-to-normal polarity transition marks a 7-m thick section across the Upper Tuff in the Bapang Formation. The transition has three short reversal episodes and is overlain by a thick normal polarity magnetozone that was fission-track dated to the Brunhes chron. This pattern closely resembles another high-resolution Matuyama-Brunhes (MB) transition record in an Osaka Bay marine core. In the Sangiran sediments, four successive transitional polarity fields lie just below the presumed main MB boundary. Their virtual geomagnetic poles cluster in the western South Pacific, partly overlapping the transitional virtual geomagnetic poles from Hawaiian and Canary Islands' lavas, which have a mean (40)Ar/(39)Ar age of 776 ± 2 ka. Thus, the polarity transition is unambiguously the MB boundary. A revised correlation of tuff layers in the Bapang Formation reveals that the hominid last occurrence and the tektite level in the Sangiran area are nearly coincident, just below the Upper Middle Tuff, which underlies the MB transition. The stratigraphic relationship of the tektite level to the MB transition in the Sangiran area is consistent with deep-sea core data that show that the meteorite impact preceded the MB reversal by about 12 ka. The MB boundary currently defines the uppermost horizon yielding Homo erectus fossils in the Sangiran area.

Thyroid hormones (THs) are essential for the embryonic and post-embryonic development of fish. We studied the role of THs during the early, post-embryonic, development of Pacific bluefin tuna. Embryos were treated with L-thyroxine (T(4)) or the anti-thyroid drug methimazole (MMI), and reared in microtitre plates for 3 days. Immersion in MMI, but not T(4), led to retardation of retinal pigment epithelium (RPE) pigmentation 3 days post-hatching (dph). Concurrent immersion in T(4) and MMI had no effect of RPE pigmentation. We also measured the expression of TRalphaA, TRalphaB, and TRbeta mRNA using real-time RT-PCR. Treatment with MMI significantly reduced TRbeta mRNA expression. Taken together these results suggest that the development of RPE pigmentation is mediated by TH, most likely via TRbeta.

We studied the profiles of 3,5,3'-l-triiodothyronine (T3), thyroxine (T4), and thyroid hormone receptors (TRs) in Pacific bluefin tuna (Thunnus orientalis) during embryonic and post-embryonic development. Both T3 and T4 were detected in embryos just before hatching, and it was found that the levels of both were increased in postflexion fish. The thyroid follicles were increased in both size and number in postflexion fish compared with preflexion fish. A TRbeta cDNA clone was generated by RACE. Two TRalpha cDNA clones were also partially identified and analyzed by real-time RT-PCR in this study. The TR mRNA levels in embryos were determined, and these were found to be lower than those in preflexion fish. Therefore, we considered that thyroid hormones function during early post-embryonic development as well as during embryonic development. Moreover, there was a peak in the TR mRNA level during postflexion stages, as seen during metamorphosis in Japanese flounder and Japanese conger eel. It is possible that thyroid hormones control the early development of scombrid fish through TRs, as they do for Pluronectiformes and Anguilliformes.

Background: Accumulating evidence indicates that interleukin-8 (IL-8) plays a major role in the mucosal inflammation caused by Helicobacter pylori infection. The purpose of the present study was to examine whether Lactobacillus gasseri OLL2716 (LG21) can inhibit the H. pylori-induced production of IL-8., Methods: A coculture system including MKN45 cells, H. pylori, and LG21 was established for an in vitro analysis. Biopsy specimens were obtained from H. pylori-infected human subjects consisting of 19 men and six women., Results: When LG21 was 1/100 less than H. pylori in a coculture system, LG21 significantly suppressed both the IL-8 mRNA and protein generation in the coculture. Live, but not heat- or UV-treated LG21, could exert the suppressive effect. However, this amount of LG21 could not suppress either the adhesion of H. pylori to the cell surface or the IL-8 production by tumor necrosis factor-alpha, which induces IL-8 generation through the activation of the transcription. These results thus suggest that LG21 suppresses an event leading to IL-8 production, which is specific for H. pylori-induced IL-8 generation, and this event is located upstream from the IL-8 transcription but downstream from the adhesion. The measurement of the IL-8 level using gastric biopsy specimens from H. pylori-infected subjects demonstrated that LG21 also suppresses the production of IL-8 in the gastric mucosa., Conclusions: Live LG21 were found to suppress H. pylori-induced IL-8 production in both a gastric cell line and within gastric mucosa.

A cDNA encoding transthyretin was cloned from the Pacific bluefin tuna (Thunnus orientalis). This cDNA contains a complete open reading frame encoding 151 amino acid residues. The deduced amino acid sequence is 81% and 55% identical to the gilthead seabream and common carp forms, respectively, and 33-39% to mammalian, reptilian, and amphibian forms. A 1.0-kb transcript was found in the the liver and ovary; the liver is the main source of this protein. Analysis of triiodo-L-thyronine (T(3)) and L-thyroxine (T(4)) binding demonstrated that both T(3) and T(4) bind to bluefin transthyretin. The binding activity of T(3) for bluefin transthyretin is higher than that of T(4). These results indicate that bluefin transthyretin acts as a transporter of thyroid hormones (THs) in the plasma, and plays an important role in the function of THs in target cells.

Background: Midkine is a heparin-binding growth factor preferentially expressed in tumor cells. The present study was performed to utilize anti-midkine antibody for tumor therapy., Methods: A monoclonal antibody to midkine was raised by immunizing mice deficient in the midkine gene. The binding site of the antibody was studied by using N-terminal half and C-terminal half of midkine, both of which were chemically synthesized. Doxorubicin (DOX)-conjugate of the antibody was produced by chemical conjugation. The effects of the antibody and the conjugate on cell growth were examined using a midkine-secreting tumor cell, i.e. human hepatocellular carcinoma cell (HepG2)., Results: The monoclonal antibody bound to the N-terminal half of midkine. The antibody did not inhibit the growth of HepG2 cells probably because the active domain of midkine is in the C-terminal half. We produced the antibody conjugated with DOX with the hope that the conjugate would be internalized accompanied with midkine. Indeed, the antibody-DOX conjugate significantly inhibited the growth of HepG2 cells compared with DOX-conjugated control IgG., Conclusion: The result raises the possibility of using anti-midkine antibody conjugated with DOX for cancer therapy.

Human melanocytes respond to UV irradiation by increasing the synthesis of melanin. While much is now understood of the pathways governing this process and the nature of the melanin synthesized, little is known of melanins produced by lower vertebrates and their capacity to respond to UV. Here we report that a fish, red seabream, can undergo 'suntanning'. Histological, colorimetric and chemical assays were performed for suntanned red seabream fish bred in net cages to analyse the melanins and compared with shaded or wild red seabream fish. For color evaluation, the L* values of suntanned fish were dramatically lower than those in the other two groups. Pyrrole-2,3,5-tricarboxylic acid (PTCA), an indicator of eumelanin, was detected in suntanned fish at five times higher levels than in shaded or wild fish while 4-amino-3-hydroxyphenyl-alanine (4-AHP), a marker for pheomelanin, could not be detected in any of the samples. Histological analysis showed that melanocytes in the suntanned skin enlarged and increased in number to form a monolayer at the surface of the skin. Analysis of L* values and PTCA levels showed quite a high correlation coefficient (r = -0.843). When comparing shaded and wild red seabream fish, the scores were closer but some significant differences were still found in some body areas. These results indicate that eumelanin accumulates in suntanned fish during the increase in skin color, which is induced by sunlight, presumably by ultraviolet radiation.

Midkine (MK) is one of a family of heparin-binding growth factors, and increased MK expression is reported in various types of human carcinomas. To clarify the association between serum MK (S-MK) concentrations and gastric cancer, we examined S-MK concentrations of gastric cancer patients (n=275) and healthy controls (n=275). S-MK concentrations of all subjects were measured by enzyme-linked immunosorbent assay (elisa). The medians (25th and 75th percentiles) of S-MK were 192 (123 and 314) pg/mL in the cases and 170 (81 and 273) pg/mL in the controls (P <0.01). We also compared S-MK concentrations in each group divided by the progression stage or histological type of cancer. A difference was observed in the median S-MK concentrations between early and advanced cancers [182 (105 and 301) pg/mL vs 203 (139 and 331) pg/mL, P=0.07], but not between intestinal and diffuse type cancers [185 (121 and 306) pg/mL vs 198 (127 and 323) pg/mL, P=0.51]. We found that those progression stages affect S-MK concentration more strongly than the histological types in gastric cancer patients. Because S-MK seems to reflect the progression stage of gastric cancer, it may serve as a useful marker in the clinical follow-up of gastric cancer patients.

Bovine liver catalase derivatives possessing diverse tissue distribution properties were synthesized, and their effects on hepatic metastasis of colon carcinoma cells were examined in mice. An intraportal injection of 1 x 10(5) colon 26 cells resulted in the formation of more than 50 metastatic colonies on the surface of the liver at 14 days after injection. An intravenous injection of catalase (CAT; 35000 units/kg of body weight) significantly (P < 0.001) reduced the number of the colonies in the liver. Galactosylated (Gal-), mannosylated (Man-) and succinylated (Suc-) CAT were also tested in the same system. Of these derivatives, Gal-CAT showed the greatest inhibitory effect on hepatic metastasis, and the number of colonies was significantly (P < 0.001) smaller than following treatment with catalase. High activities of matrix metalloproteinases (MMPs), especially MMP-9, were detected in the liver of mice bearing metastatic tumor tissues, which was significantly (P < 0.05) reduced by Gal-CAT. These results, combined with our previous finding that Gal-CAT can be efficiently delivered to hepatocytes, indicate that the targeted delivery of catalase to the liver by galactosylation is a promising approach to suppress hepatic metastasis. Decreased MMP activity by catalase delivery seems to be involved in its anti-metastatic effect.

Approximately 260 mg/l of authentic recombinant human pleiotrophin (rhPTN) was expressed into the medium of high-cell density fermentation using a Pichia pastoris protein expression system. The prepro-sequence of yeast alpha-mating factor was used successfully. The recombinant hPTN was efficiently recovered from the medium by expanded bed adsorption, and purified using successive column chromatography steps. In the purified rhPTN preparation, modified rhPTN were scarcely detected. Circular dichroism measurement of the purified PTN showed the presence of the characteristic beta-structures in the protein.

Midkine (MK) is a heparin-binding growth factor, which promotes growth, migration, and survival of various cells, and MK expression is increased in many human carcinomas. We determined the urinary MK level by enzyme-linked immunoassay. Taking 311pg/mg creatinine as a cut-off level, 70% of patients with various carcinomas (n=142) gave positive values, while only 5.5% of healthy volunteers (n=330) did. In case of gastric carcinoma, 17 out of 21 patients with stage 1 tumor were positive. Urinary MK levels are expected to become a convenient marker as an aid in detection of tumors.

In a previous study, we showed that targeted delivery of bovine liver catalase to hepatocytes by direct galactosylation augmented the inhibitory effect of the enzyme on experimental hepatic metastasis of colon carcinoma cells (unpublished data). Here, we examined the ability of catalase to inhibit tumor metastasis to the lung by controlling its biodistribution. Four types of catalase derivative, Gal-CAT, Man-CAT, Suc-CAT and PEG-CAT, were synthesized. Experimental pulmonary metastasis was induced in mice by i.v. injection of 1 x 10(5) colon 26 tumor cells. An i.v. injection of catalase (35,000 units/kg) partially, but significantly, decreased the number of colonies in the lung 2 weeks after tumor injection, from 93 +/- 29 (saline injection) to 63 +/- 23 (p < 0.01). Suc-CAT, Man-CAT and Gal-CAT showed effects similar to those of catalase on the number of colonies. However, PEG-CAT greatly inhibited pulmonary metastasis to 22 +/- 11 (p < 0.001). Furthermore, s.c. injection of catalase also greatly inhibited metastasis (11 +/- 6, p < 0.001). Neither inactivated catalase nor BSA showed any effects on the number of metastatic colonies, indicating that the enzymatic activity of catalase to detoxify H(2)O(2) is the critical factor inhibiting metastasis. (111)In-PEG-CAT showed a sustained concentration in plasma, whereas s.c.-injected (111)In-catalase was slowly absorbed from the injection site. Cytopathic effects of adenovirus-infected human cells consist of rounding and detachment from the substrate at late times postinfection. It is not known, however, whether any changes in cell adhesion are induced by adenoviruses before cytopathic effects become evident. We show here that attachment and spreading of human adenovirus type 2 (Ad2)-infected KB cells on fibronectin (FN) are, unexpectedly, promoted at intermediate times postinfection. The promotion of spreading by Ad2 is not only on fibronectin but also on laminin and vitronectin. In contrast, Ad2 or Ad5 mutants defective in the E1b 19-kDa function did not lead to a promotion in cell attachment but they led to a promotion in cell spreading. Inhibition of spreading of Ad2-infected KB cells on FN by the peptide Gly-Arg-Gly-Asp-Ser-Pro is partial, although it is complete in uninfected KB cells. We found, however, that Ad2 infection does not significantly change the levels of expression of beta 1 integrin mRNA and FN receptor polypeptides. Our results thus suggest that adenovirus infection leads to a promotion in adhesion through influencing integrin molecules without altering the quantity and/or through activation in the cytoskeleton or some other pathway.

A croaker (Nibea mitsukurii) has a high incidence of the skin melanoma, chromatophoroma, in a Pacific coastal area in Japan. A sea catfish (Plotosus anguillaris) bearing skin melanosis is also found in the same area. For elucidation of a correlation between these pigment cell neoplasms of the skin and environmental contaminants, an epidemiological survey was conducted to determine the distribution and prevalence of tumor-bearing fish. Based upon observations of a high prevalence of skin neoplasms near the discharge point for kraft pulp mills, experiments were conducted to determine the neoplastic induction efficiency of the effluent on the croaker and sea catfish species. Isolation and identification of mutagens in effluent extracts were carried out using the Ames test, followed by mass spectral analysis of mutagenic fractions. The effluent induced a chromatophoroma on one croaker of the 100 tested, and it induced pigment cell hyperplasia on 70 to 100% of the sea catfish. These skin neoplasms were grossly similar to those observed in the field. Five chloroacetones were identified from the Ames-positive fractions of the effluent, and tetrachlorocyclopentene-1,3-dione and two alpha-dicarbonyl compounds were also detected as mutagens. The above experiments indicate that the mutagenic contaminants found in kraft mill effluent may play an important role in the induction of skin neoplastic disease in fish.

Chromatophoromas in the croaker nibe, Nibea mitsukurii, are common neoplasms in feral fish which inhabit the shallow water in a unique geographic distribution along the Pacific coast of Japan. We undertook surveys of the epizootiology of tumor-bearing fish at 25 sites. The highest tumor incidence occurred at the station near the mouth of the Kumano river and was 47% (1,415 of 2,991). The incidence at 2 adjacent survey stations located approximately 18 and 30 km away were 2.7 and 2.5%, respectively. At the other survey stations, no tumor incidences were recorded or they were less than 5%. During the course of experimental studies on the chromatophoromas using tank-reared nibe, we found that nifurpirinol (NP), a drug used for the treatment of fish diseases, might also induce the chromatophoromas in the fish, as well as N-methyl-N'-nitro-N-nitrosoguanidine and 7,12-dimethylbenz[alpha]anthracene. Therefore, tank-reared nibe 5 months of age were divided into 4 groups of about 50 each and exposed to water containing 0, 0.5, 1, or 2 ppm NP, respectively, fourteen times for 1 hour each time. Three hundred nibe were kept as untreated controls. The incidences of chromatophore hyperplasia or neoplasia per group at 13 months were as follows: the untreated: 2.9% (6 of 204), 0 ppm: 5.3% (2 of 38), 0.5 ppm: 73% (36 of 49), 1 ppm: 87% (20 of 23), and 2 ppm: 100% (2 of 2). These and other results led us to believe that 1) NP is a carcinogen, 2) nibe have a high susceptibility to induction of chromatophoromas by chemical carcinogens, and 3) some environmental chemicals are causal factors in the hyperendemic occurrences of the tumors in wild nibe.

Chromatophoroma in the croaker (Nibea mitsukurii) showed a unique geographic distribution. The contribution of environmental chemicals to the cause of chromatophoroma in the feral croaker is considered likely on the basis of the following results in our studies. 1) Chromatophoroma was induced in tank-reared N. mitsukurii by administration of certain kinds of known carcinogens such as 7,12-dimethyl-benz(a)anthracene, N-methyl-N'-nitro-N-nitrosoguanidine, and nifurpirinol. 2) Local accumulation of pigment-cell hyperplasia in the catfish (Protosus anguillaris) showed similar tendencies to those of chromatophoroma in N. mitsukurii. 3) Removal of contaminated sediment from the harbor and the river appeared to reduce the incidence from 47% in 1973-1983 to about 20% in 1985-1987. 4) Waste water from a factory located at the station where the incidence of the neoplasm was the highest contained mutagenic substances such as chloroacetones and glyoxals [5]. Exposure of catfish to the waste water induced pigment-cell hyperplasia on the skin.
