Abstract

A literature review was conducted to determine whether there is a reduction of salivation with the use of antisialogogues. Several databases were used, including Cochrane Collaboration, PubMed, Scopus, EMBASE, and ISI Web of Knowledge. Findings indicate that the use of antisialogogues for dental procedures in general is questionable.

21. Effect of coral Goniopora Sp scaffold application on human osteoblast-like MG-63 cell activity in vitro.

Abstract

Background: Coral is an osteo-conductive biomaterial that can act as an alternative scaffold for osteogenesis. In this in vitro study we analyzed the activity of osteoblast-like cells after treatment with the coral Goniopora. Methods: Human osteoblast-like MG-63 cells were incubated in a-minimal essential medium supplemented with 10% fetal bovine serum and 300 ng/mL amphotericin B plus 1% penicillin-streptomycin and stored in a 5% CO2 incubator at 37°C. The Goniopora were smashed into size A (20 mesh), B (1-2 mm), and C (200 mesh) particles, sterilized using gamma radiation and applied to cells. Protein and alkaline phosphatase (ALP) concentrations were evaluated after incubation for 24 and 48 h. Results: The protein assay of 24 h and 48 h cultured osteoblasts illustrated that treated cells, whether with coral size A, B and C exhibited a lower mean value compared to the untreated cells. For ALP levels there were statistically significant differences at 48 h between B and C (p = 0.004), and A and C (p = 0.09). Conclusions: No significant differences in total protein concentrations were found among all groups after 24 and 48 h. Smaller coral size and longer incubation time tended to facilitate osteogenesis. Abstract

The mandibular condyle is a major growth site and is known to adapt to functional factors. Numerous studies have been performed on the effects of growth factors on the metabolism of primary cartilages, but only a few investigations have examined their action on primary and secondary cartilages. Therefore, the purpose of this study was to compare the effects of insulin-like growth factor-I (IGF-I), transforming growth factor-beta(1) (TGF-beta(1)), and fibroblast growth factor-2 (FGF-2) on the growth of secondary cartilage from the mandibular condyle and primary cartilage from the femoral head of new-born rats. In addition, synergy between these growth factors was investigated. The level of glycosaminoglycan (GAG) and DNA synthesis was analysed after 5 days in culture with the growth factors. The effects of TGF-beta(1) and FGF-2 on growth, tissue organization, and the GAG and collagen content were also evaluated.The stimulation of cell proliferation by the growth factors was higher in the mandibular condyles than in the femoral heads. The content of the matrix components was reduced more by FGF-2 in the mandibular condyles than in the femoral heads. Both TGF-beta(1) and FGF-2 antagonized the stimulatory effects of IGF-I on GAG synthesis in the two types of cartilage. In contrast, the total growth of mandibular condyles was not affected by TGF-beta(1) and FGF-2 while that of femoral heads was strongly reduced. This was mainly due to the inhibition of chondrocyte hypertrophy. These results show that in spite of the extensive effects of growth factors on the metabolism of mandibular condyles, their dimensional growth was not affected.

Abstract

Primary and secondary cartilages differ in embryonic origin and in histological organization, and are generally considered to have a different mode of growth. However, few studies have directly compared the two types of cartilage of the same animal at the same age. Therefore, we analysed several histological and biochemical differences between secondary cartilage of the mandibular condyle and primary cartilage of the femoral head of 4-d-old rats. We evaluated the tissue organization, the level of DNA and glycosaminoglycan (GAG) synthesis, and the GAG and collagen content. The expression of collagen types I, II and III and of receptors for insulin-like growth factor (IGF)-I, fibroblast growth factor (FGF), and transforming growth factor (TGF)-beta were investigated by immunohistochemistry. The ex vivo DNA and GAG synthesis as well as the GAG content of femoral heads were much higher than that of mandibular condyles. Mandibular condyles expressed both collagen types I and II, while femoral heads expressed only type II collagen. In the mandibular condyles, receptors for IGF-I, FGF, and TGF-beta were observed mainly in the superficial layers, whereas they were found throughout the entire femoral head. In conclusion, major differences were found between both types of cartilage, which might be related to their specific functional demands.
