Abstract Background We have analyzed the complete mitochondrial genomes of 22 Pan paniscus (bonobo, pygmy chimpanzee) individuals to assess the detailed mitochondrial DNA (mtDNA) phylogeny of this close relative of Homo sapiens. Results We identified three major clades among bonobos that separated approximately 540,000 years ago, as suggested by Bayesian analysis. Incidentally, we discovered that the current reference sequence for bonobo likely is a hybrid of the mitochondrial genomes of two distant individuals. When comparing spectra of polymorphic mtDNA sites in bonobos and humans, we observed two major differences: (i) Of all 31 bonobo mtDNA homoplasies, i.e. nucleotide changes that occurred independently on separate branches of the phylogenetic tree, 13 were not homoplasic in humans. This indicates that at least a part of the unstable sites of the mitochondrial genome is species-specific and difficult to be explained on the basis of a mutational hotspot concept. (ii) A comparison of the ratios of non-synonymous to synonymous changes (dN/dS) among polymorphic positions in bonobos and in 4902 Homo sapiens mitochondrial genomes revealed a remarkable difference in the strength of purifying selection in the mitochondrial genes of the F0F1-ATPase complex. While in bonobos this complex showed a similar low value as complexes I and IV, human haplogroups displayed 2.2 to 7.6 times increased dN/dS ratios when compared to bonobos. Conclusions Some variants of mitochondrially encoded subunits of the ATPase complex in humans very likely decrease the efficiency of energy conversion leading to production of extra heat. Thus, we hypothesize that the species-specific release of evolutionary constraints for the mitochondrial genes of the proton-translocating ATPase is a consequence of altered heat homeostasis in modern humans.

Recently, somatic recombination of human mitochondrial DNA (mtDNA) was discovered in skeletal muscle. To determine whether recombinant mtDNA molecules can be transmitted through the germ line, we investigated two families, each harboring two inherited heteroplasmic mtDNA mutations. Using allele-specific polymerase chain reaction and single-cell and single-molecule mutational analyses, we discovered, in both families, all four possible allelic combinations of the two heteroplasmic mutations (tetraplasmy), the hallmark of mtDNA recombination. We strongly suggest that these recombinant mtDNA molecules were inherited rather than de novo generated somatically, because they (1) are highly abundant and (2) are present in different tissues of maternally related family members, including young individuals. Moreover, the comparison of the complete mtDNA sequence of one of the families with database sequences revealed an irregular, nontreelike pattern of mutations, reminiscent of a reticulation. We therefore propose that certain reticulations of the human mtDNA phylogenetic tree might be explained by recombination of coexisting mtDNA molecules harboring multiple mutations.

Experimental evidence for human mitochondrial DNA (mtDNA) recombination was recently obtained in an individual with paternal inheritance of mtDNA and in an in vitro cell culture system. Whether mtDNA recombination is a common event in humans remained to be determined. To detect mtDNA recombination in human skeletal muscle, we analyzed the distribution of alleles in individuals with multiple mtDNA heteroplasmy using single-cell PCR and allele-specific PCR. In all ten individuals who carried a heteroplasmic D-loop mutation and a distantly located tRNA point mutation or a large deletion, we observed a mixture of four allelic combinations (tetraplasmy), a hallmark of recombination. Twelve of 14 individuals with closely located heteroplasmic D-loop mutation pairs contained a mixture of only three types of mitochondrial genomes (triplasmy), consistent with the absence of recombination between adjacent markers. These findings indicate that mtDNA recombination is common in human skeletal muscle.

In earlier studies it has been shown that stimulation of the median raphe nucleus (MR) in awake rabbits decreases the expression and frequency of oscillatory theta activity in the septohippocampal system, and the functional blockade of this nucleus evokes the regular and high-frequency theta rhythm. The present work was aimed at elucidation of serotoninergic influence of MR (which also contains cells of other chemical nature) to the septohippocampal system of theta activity. Serotonin reuptake blocker fluoxetine that increases brain serotonin level was applied. Hippocampal electroencephalogram was recorded in awake rabbits. Bilateral intracerebroventricular infusion of fluoxetine hydrochloride (Sigma, St. Louis; 15 micrograms in 5 microliters saline) in all cases reduced the rhythmic theta activity. In 15 of 18 (83.3%) of experiments the decrease in hippocampal theta oscillations was more than 50% of the control level. The theta band of the spectral density histogram decreased in the mean by 56 +/- 5.8% of the control level (from 10 to 93% in different experiments, p < 0.001). The mean latency of these changes was 3.5 +/- 0.11 minutes (2.9-4.1 min), the effect duration was 64 +/- 3.2 min (45.3-90 min). The mean frequency of the theta waves did not change as compared to the baseline and was equal to 5.25 +/- 0.5 Hz (4.5-6.5 Hz). The fluoxetine-induced reduction of the theta rhythm expression in hippocampus is the evidence of its inhibitory control by serotoninergic brain system. It is suggested that the increase of the frequency of hippocampal theta rhythm after the functional blockade of MR observed in our earlier experiments was the result of a release of the septohippocampal system from the influence of nonserotoninergic neurons (via glutamatergic reticular formation) and/or temporary cessation of the MR interaction with noradrenergic, dopaminergic and glutamate/aspartate systems.

Responses to sensory stimuli were analyzed in hippocampal CA1 neurons of unanesthetized rabbits in chronic experiments before and after reversible functional blockade of the median raphe nucleus (MR) and medial septal area (MS-DB) by local microinjections of anesthetic lidocaine. The MR blockade, which resulted in an enhancement of theta-modulation of the background activity of the hippocampal neurons, was followed by a depression of sensory responsiveness (only 46.7% of the neurons reactive before the blockade retained their responses). Reactions of all types were blocked, diminished, or inverted, but inhibitory responses were affected most severely. Lidocaine injection into the MS-DB, which blocked also all brain stem afferents ascending to the hippocampus via the MS-DB input, resulted in the total absence of theta-modulation; however, responsiveness to sensory stimuli remained relatively high (76.7% of the responses were preserved); on-effects were especially resistant to the MS-DB blockade. Comparison of evoked activity in two conditions of continuous theta rhythm generation (physostigmine injection and MR blockade) revealed striking similarities. This suggests that the theta-suppressing influence of the MR (presumably serotonergic) is primarily actualized through the control of cholinergic septo-hippocampal theta-generating mechanism. The results provide support for the view that the theta-rhythm acts like an active filter in information processing performed by hippocampal neurons.

The control of theta rhythm in neuronal activity of the medial septal area and hippocampal electroencephalogram by the brainstem structures was investigated in waking rabbits. In the first series of experiments stimulating electrodes were implanted into the midbrain reticular formation and median raphe nucleus. The standard frequency of theta-bursts in medial septal area neurons and in the electroencephalogram was uniformly and chronically decreased in all rabbits with electrodes implanted into the median raphe nucleus (4.7 +/- 0.5 Hz versus 5.2 +/- 0.19 Hz in animals without electrodes in median raphe nucleus). Weak electrical stimulation of the median raphe nucleus resulted in additional decrease of theta expression in the medial septal area neurons and its disappearance from the hippocampal electroencephalogram, where it was substituted by delta-waves and spindles. Stimulation of the reticular formation had the opposite effect, with an increase in theta frequency, regularity and expression in medial septal area neuronal activity and hippocampal electroencephalogram. In the second series of experiments reversible functional blockade of the median raphe nucleus by local microinjection of lidocaine was performed. This resulted in expression of theta-bursts in an additional group of medial septal area neurons, an increase in theta-burst frequency (by 0.5-2 Hz) and regularity with concomitant changes in the electroencephalogram. The effects of sensory stimuli on the background of increased theta activity were suppressed or significantly decreased. It is concluded that, in accordance with the data of other authors, the median raphe nucleus can be regarded as a functional antagonist of the reticular formation, powerfully suppressing theta-bursts of the medial septal area neurons and hippocampal theta rhythm. It is suggested that, in combination with the theta-enhancing influences of reticular formation, the median raphe nucleus may participate in termination of attention, its switching to other stimuli and stabilization of the effects of learning.

Spontaneous activity and responses to sensory stimuli were analysed in the hippocampal CA1 neurons of chronic unanesthetized rabbits before and after reversible functional blockade of the median raphe nucleus and medial septal area by local microinjections of anesthetic lidocaine. This evoked, correspondingly, persistent theta rhythm and its complete blockade for about 30 min. The results were compared to the neuronal data obtained earlier in the experiments with cholinergic drugs modulating expression of theta rhythm. Intra-median raphe nucleus injection of lidocaine evoked uniform increase of discharge rate in the hippocampal neurons with low and high spontaneous activity. Theta modulation of neuronal activity had increased regularity and frequency (by 0.5-2.0 Hz) and appeared in additional group of the neurons simultaneously with expression of persistent theta in the hippocampal electroencephalogram. Sensory responsiveness of the hippocampal neurons was drastically decreased (45% of the responses preserved). Reactions of all types were blocked, diminished, or inverted, but inhibitory responses were the most severely affected. Injection of lidocaine into medial septal area also blocked all brain stem afferents ascending to the hippocampus via medial septal area, thus, totally depriving hippocampus of brainstem-septal input. However, besides the total absence of theta modulation, spontaneous activity in majority of neurons was not significantly changed. Responsiveness to sensory stimuli also remained relatively high (77% of the responses preserved); on-effects were especially resistant to medial septal area blockade. Comparison of spontaneous and evoked activity in two theta states (physostigmine and median raphe nucleus blockade) revealed striking similarity of all characteristics, which suggested that theta-suppressing influences of median raphe nucleus (presumably serotonergic) are realized primarily through the control of cholinergic septo-hippocampal theta-generating mechanism. However, as the frequency of theta rhythm does not depend on it, an additional effect of disinhibition of activating reticular formation by the median raphe nucleus suppression is suggested. The data confirm that theta rhythm may be regarded as active filter in the information processing by the hippocampal neurons.

A method of controlled successive short-term (25-30 min) stabilization or suppression of the theta rhythm by local injections of lidocaine into median raphe nucleus or medical septal area (MS-DB) was tested for further analysis of the functional significance of theta-modulation in activity of hippocampal neurons. Extracellular recording of their activity (area CAl) was performed in unanaesthetized rabbits. Injection of lidocaine into MS-DS resulted in total suppression of theta modulation in neuronal activity and EEG of the hippocampus. Mean frequency of the background activity was not changed in the majority of the units but was significantly increased in a limited group of high-frequency neurons (putative inhibitory cells). Injection into medial raphe, the source of serotonergic afferents to MS-DB and hippocampus, was followed by an increase in regularity and frequency of theta modulation, by appearance of additional group of neurons with rhythmic modulation, and by expression of continuous theta in the hippocampal EEG. Mean frequency of discharge and its regularity were significantly increased in the majority of the cells. The data confirm the presence of tonic inhibitory control of theta-generating septo-hippocampal system by the medial raphe nucleus, which can be regarded as an antagonist of the midbrain reticular formation in this respect.
