Full Text, Thesis (PhD Doctorate), Doctor of Philosophy (PhD), School of Environment and Sc, Science, Environment, Engineering and Technology, This thesis describes the novel use of multiple steroid hormone analysis to investigate and gain understanding of previously unknown aspects of the endocrinology and reproductive biology of the Southern Hemisphere humpback whale (Megaptera novaeangliae). Steroid hormones are a class of structurally related molecules that function as messengers in a variety of important physiological processes (e.g. salt homeostasis, immune system, sexual development and reproductive cycles). A number of endogenous steroid hormones have been identified as critical for reproduction (e.g. androgens, progestogens, oestrogens) and stress response (e.g. corticosteroids) in mammals. Understanding the species-specific roles and baseline levels of endogenous steroid hormones enable these compounds to be used as biomarkers of reproductive status and stress exposure. The use of steroid biomarkers for these purposes promises to be extremely advantageous in cetacean species, since other approaches often require increased time and resources. However, the collection and interpretation of steroid hormone data in the humpback whale is currently limited by methodological issues (e.g. challenging sampling, difficult comparison between sample types, analytical approach restricted to single hormone analysis), and by a general lack of detailed knowledge of the humpback whale endocrine system. Collection of biological samples from cetaceans is inherently difficult. In particular, free-swimming individuals of baleen whale species, including humpback whales, cannot be kept in captivity, nor they can be immobilised for sampling. Contemporary studies have demonstrated that steroid hormones analysis is viable in baleen species through the use of remotely collected tissues and fluids (i.e. faeces, blow, blubber). The seasonal migration of the humpback whales from the Antarctic feeding areas to the tropical breeding grounds, however, imposes further restrictions in regard to sample types and timing of collection

Abstract

BACKGROUND: The obesity pandemic is associated with multiple major health concerns. In addition to diet and lifestyle, there is increasing evidence that environmental exposures to chemicals known as obesogens also may promote obesity. OBJECTIVES: We investigated the massive environmental contamination resulting from the Deepwater Horizon (DWH) oil spill, including the use of the oil dispersant COREXIT in remediation efforts, to determine whether obesogens were released into the environment during this incident. We also sought to improve the sensitivity of obesogen detection methods in order to guide post-toxicological chemical assessments. METHODS: Peroxisome proliferator-activated receptor gamma (PPARγ) transactivation assays were used to identify putative obesogens. Solid-phase extraction (SPE) was used to sub-fractionate the water-accommodated fraction generated by mixing COREXIT, cell culture media, and DWH oil (CWAF). Liquid chromatography-mass spectrometry (LC-MS) was used to identify components of fractionated CWAF. PPAR response element (PPRE) activity was measured in PPRE-luciferase transgenic mice. Ligand-binding assays were used to quantitate ligand affinity. Murine 3T3-L1 preadipocytes were used to assess adipogenic induction. RESULTS: Serum-free conditions greatly enhanced the sensitivity of PPARγ transactivation assays. CWAF and COREXIT had significant dose-dependent PPARγ transactivation activities. From SPE, the 50:50 water:ethanol volume fraction of CWAF contained this activity, and LC-MS indicated that major components of COREXIT contribute to PPARγ transactivation in the CWAF. Molecular modeling predicted several components of COREXIT might be PPARγ ligands. We classified dioctyl sodium sulfosuccinate (DOSS), a major component of COREXIT, as a probable obesogen by PPARγ transactivation assays, PPAR-driven luciferase induction in vivo, PPARγ binding assays (affinity comparable to pioglitazone and arachidonic acid), and in vitro murine adipocyte differentiation. Abstract

Lake superior's food web was evaluated in 1994 with regards to the presence of hydrophobic organochlorine contaminants (OCs), among them toxaphene and chlordane and metabolites. Because of its concentrations that were two to 15 times higher than total PCBs, toxaphene was the major OC in the food web. There was no significant relationship between the accumulation of OCs in the food web and the log octanol/water partition effect. Because of this, deep water organisms in Lake Superior view settling particles as important sources of OCs.

Abstract

Distributions of organochlorine pesticides and polychlorinated biphenyls (PCBs) in Lake Baikal, Russia, were determined by measuring the concentrations of these compounds from air and water samples collected in June 1991. Air concentrations of chlorinated bornanes, chlordanes and hexachlorobenzene were similar to those measured in the Arctic regions, but air concentrations of hexachlorocyclohexanes, DDTs and PCBs were similar to those in the Great Lakes region. Measurements from air samples in nearby Irkutsk were higher than those for Lake Baikal, suggesting regional transport and deposition of organochlorine pesticides and PCBs.

45. A review of halogenated natural products in Arctic, Subarctic and Nordic ecosystems

Abstract

Halogenated natural products (HNPs) are organic compounds containing bromine, chlorine, iodine, and rarely fluorine. HNPs comprise many classes of compounds, ranging in complexity from halocarbons to higher molecular weight compounds, which often contain oxygen and/or nitrogen atoms in addition to halogens. Many HNPs are biosynthesized by marine bacteria, macroalgae, phytoplankton, tunicates, corals, worms, sponges and other invertebrates. This paper reviews HNPs in Arctic, Subarctic and Nordic ecosystems and is based on sections of Chapter 2.16 in the Arctic Monitoring and Assessment Program (AMAP) assessment Chemicals of Emerging Arctic Concern (AMAP, 2017) which deal with the higher molecular weight HNPs. Material is updated and expanded to include more Nordic examples. Much of the chapter is devoted to “bromophenolic” HNPs, viz bromophenols (BPs) and transformation products bromoanisoles (BAs), hydroxylated and methoxylated bromodiphenyl ethers (OH-BDEs, MeO-BDEs) and polybrominated dibenzo-p-dioxins (PBDDs), since these HNPs are most frequently reported. Others discussed are 2,2′ -dimethoxy-3,3′ ,5,5′ -tetrabromobiphenyl (2,2′ -dimethoxy-BB80), polyhalogenated 1′- methyl-1,2′-bipyrroles (PMBPs), polyhalogenated 1,1′ -dimethyl-2,2′ -bipyrroles (PDBPs), polyhalogenated N-methylpyrroles (PMPs), polyhalogenated N-methylindoles (PMIs), bromoheptyl- and bromooctyl pyrroles, (1R,2S,4R,5R,1′E)-2-bromo-1-bromomethyl-1,4-dichloro-5-(2′-chloroethenyl)-5- methylcyclohexane (mixed halogenated compound MHC-1), polybrominated hexahydroxanthene derivatives (PBHDs) and polyhalogenated carbazoles (PHCs). Aspects of HNPs covered are physicochemical properties, sources and production, transformation processes, concentrations and trends in the physical environment and biota (marine and freshwater). Toxic properties of some HNPs and a discussion of how climate change might affect HNPs production and distribution are also included. The review concludes with a summary of research need
