Active matter is characterized by its ability to induce motion by self-generated stress. In the case of a solid, such motion can lead to shape transformations. The stress-generating components can be anisotropic endowing the material with mesoscopic orientational order. It is currently unknown how the specific postions and orientations of these active constituents influence morphological changes. We study theoretically the effects of imposing topological point defects in the arrangements of the stress-generating components on the morphology of elastic active nematic shells. We show that topological defects of charge +1 are uniquely capable of increasing, reducing or maintaining the intrinsic curvature of the shell. These changes depend on the nature of the active stress and the phase angle of the defect. We apply our theory to experiments conducted on contracting actomyosin sheets. By combining defects of different charges, we can generate shells with arbitrary complexity. We confirm this flexibility by reproducing the shape of the freshwater polyp Hydra, in which topological defects have been associated with morphological features of the animal. The actin cortex is a thin layer of actin filaments and myosin motors beneath the outer membrane of animal cells. It determines the cells' mechanical properties and forms important morphological structures. Physical descriptions of the cortex as a contractile active gel suggest that these structures can result from dynamic instabilities. However, in these analyses the cortex is described as a two-dimensional layer. Here, we show that the dynamics of the cortex is qualitatively different when gel fluxes in the direction perpendicular to the membrane are taken into account. In particular, an isotropic cortex is then stable for arbitrarily large active stresses. During cell division, sister chromatids are segregated by the mitotic spindle, a bipolar assembly of interdigitating antiparallel polar filaments called microtubules. Establishing a stable overlap region is essential for maintenance of bipolarity, but the underlying mechanisms are poorly understood. Using a particle-based stochastic model, we find that the interplay of motors and passive cross linkers can robustly generate partial overlaps between antiparallel filaments. Our analysis shows that motors reduce the overlap in a length-dependent manner, whereas passive cross linkers increase it independently of the length. The distributions of many proteins in rod-shaped bacteria are far from homogenous. Often they accumulate at the cell poles or in the cell center. At the same time, the copy number of proteins in a single cell is relatively small making the patterns noisy. To explore limits to protein patterns due to molecular noise, we studied a generic mechanism for spontaneous polar protein assemblies in rod-shaped bacteria, which is based on cooperative binding of proteins to the cytoplasmic membrane. For mono-polar assemblies, we find that the switching time between the two poles increases exponentially with the cell length and with the protein number., Comment: 13 pages, 6 figures

In the bacterium Escherichia coli, selection of the division site involves pole-to-pole oscillations of the proteins MinD and MinE. Different oscillation mechanisms based on cooperative effects between Min-proteins and on the exchange of Min-proteins between the cytoplasm and the cytoplasmic membrane have been proposed. The parameters characterizing the dynamics of the Min-proteins in vivo are not known. It has therefore been difficult to compare the models quantitatively with experiments. Here, we present in vivo measurements of the mobility of MinD and MinE using fluorescence correlation spectroscopy. Two distinct time-scales are clearly visible in the correlation curves. While the faster time-scale can be attributed to cytoplasmic diffusion, the slower time-scale could result from diffusion of membrane-bound proteins or from protein exchange between the cytoplasm and the membrane. We determine the diffusion constant of cytoplasmic MinD to be approximately 16\mu^{2}/s, while for MinE we find about 10\mu^{2}/s, independently of the processes responsible for the slower time-scale. We present a general theoretical framework to discuss mechanisms of morphogen transport and gradient formation in a cell layer. Trafficking events on the cellular scale lead to transport on larger scales. We discuss in particular the case of transcytosis where morphogens undergo repeated rounds of internalization into cells and recycling. Based on a description on the cellular scale, we derive effective nonlinear transport equations in one and two dimensions which are valid on larger scales. We derive analytic expressions for the concentration dependence of the effective diffusion coefficient and the effective degradation rate. We discuss the effects of a directional bias on morphogen transport and those of the coupling of the morphogen and receptor kinetics. Furthermore, we discuss general properties of cellular transport processes such as the robustness of gradients and relate our results to recent experiments on the morphogen Decapentaplegic (Dpp) that acts in the fruit fly Drosophila.

The cytoskeleton is an important subsystem of cells that is involved for example in cell division and locomotion. It consists of filaments that are cross-linked by molecular motors that can induce relative sliding between filaments and generate stresses in the network. In order to study the effects of fluctuations on the dynamics of such a system we introduce here a new class of driven diffusive systems mimicking the dynamics of active filament bundles where the filaments are aligned with respect to a common axis. After introducing the model class we first analyze an exactly solvable case and find condensation. For the general case we perform a mean-field analysis and study the behavior on large length scales by coarse-graining. We determine conditions for condensation and establish a relation between the hopping rates and the tension generated in the bundle., Comment: 10 pages, 4 eps figures

During division it is of primary importance for a cell to correctly determine the site of cleavage. The bacterium Escherichia coli divides in the center, producing two daughter cells of equal size. Selection of the center as the correct division site is in part achieved by the Min-proteins. They oscillate between the two cell poles and thereby prevent division at these locations. Here, a phenomenological description for these oscillations is presented, where lateral interactions between proteins on the cell membrane play a key role. Solutions to the dynamic equations are compared to experimental findings. In particular, the temporal period of the oscillations is measured as a function of the cell length and found to be compatible with the theoretical prediction., Comment: 17 pages, 5 figures. We discuss the formation of graded morphogen profiles in a cell layer by nonlinear transport phenomena, important for patterning developing organisms. We focus on a process termed transcytosis, where morphogen transport results from binding of ligands to receptors on the cell surface, incorporation into the cell and subsequent externalization. Starting from a microscopic model, we derive effective transport equations. We show that, in contrast to morphogen transport by extracellular diffusion, transcytosis leads to robust ligand profiles which are insensitive to the rate of ligand production.

We find that a 2D periodic potential with different modulation amplitudes in x- and y-direction and a perpendicular magnetic field may lead to a transition to electron transport along the direction of stronger modulation and to localization in the direction of weaker modulation. Based on a thorough numerical analysis of the spectrum of Harper's operator, which describes, e.g., an electron on a two-dimensional lattice subjected to a magnetic field perpendicular to the lattice plane, we make the following conjecture: For any value of the incommensurability parameter sigma of the operator its spectrum can be covered by the bands of the spectrum for every rational approximant of sigma after stretching them by factors with a common upper bound. We show that this conjecture has the following important consequences: For all irrational values of sigma the spectrum is (i) a zero measure Cantor set and has (ii) a Hausdorff dimension less or equal to 1/2. We show that the time evolution operator of kicked quantum systems, although a full matrix of size NxN, can be diagonalized with the help of a new method based on a suitable combination of fast Fourier transform and Lanczos algorithm in just N^2 ln(N) operations. It allows the diagonalization of matrizes of sizes up to N\approx 10^6 going far beyond the possibilities of standard diagonalization techniques which need O(N^3) operations. We show that avoided crossings of energy bands may give rise to a variety of phenomena such as transitions from metal to insulator and vice versa, changes in localization lengths, and changes in the fractal dimension of energy spectra. The multifractal dimensions D2^mu and D2^psi of the energy spectrum and eigenfunctions, resp., are shown to determine the asymptotic scaling of the width of a spreading wave packet. For systems where the shape of the wave packet is preserved the k-th moment increases as t^(k*beta) with beta=D2^mu/D2^psi, while in general t^(k*beta) is an optimal lower bound.
