To prepare oncology, genetics and molecular medicine professionals for basic, translational and epidemiological cancer genetics research through advanced training in a novel specialized cancer genetics career development program (CGCDP).Established in 2001, the City of Hope CGCDP is the only program in the NCI R25T portfolio focused on interdisciplinary training of oncologists and geneticists as cancer genetics and cancer prevention research program leaders. Program infrastructure, curriculum, recruitment strategies, implementation, evaluation and outcome are described.During the initial four-year period, twelve highly qualified health care professionals were recruited for cancer genetics academic, experiential, and research training. All trainees presented abstracts, published peer-reviewed journal articles, and/or were awarded cancer genetics research grants. Most are now in academic/research venues, following a cancer genetics research career trajectory.The CGCDP is an NIH-funded program that has successfully trained doctoral and mid-level professionals in cancer genetics research. Program graduates are addressing the burden of cancer by translating cancer genetics investigations into individual risk assessment tools and effective cancer screening and prevention interventions.

The northern fowl mite, Ornithonyssus sylviarum, is an ectoparasite of birds and a poultry pest. The ability of northern fowl mites to orientate to a heat source is investigated with individual mites video-recorded in two-dimensional arenas and exposed to spatial or temporal heat gradients. Recorded tracks are digitally analysed for variation in linear velocity, mean direction of movement, and patterns in angular displacement. Mean direction of movement in a spatial gradient is significantly associated with the position of the heat source for 24/29 mites tested (P 0.1). Angular displacement that orientates a mite towards the heat source is positively correlated with the preceding deviation from that direction (P < 0.01). Angular displacement away from the heat source is random. The temporal heat gradient is such that no spatial reference to the heat source exists within the plane of the arena. Mites in an ambient (27 degrees C) to heated (30 degrees C) transition have angular displacement distributions similar to control mites (ambient to ambient transition). However, mites in a heated to ambient transition execute angular displacements approximately 25 degrees greater than mites in the other treatments (P < 0.03). Mites compare the shift in temperature over time and alter their direction of movement by a programmed (idiothetic) response to a decrease in temperature, rather than through detection of the spatial position of the gradient (allothetic).

Male Cadra cautella were presented with five heterogeneous pheromone clouds (created from source doses of 0, 0.01, 1, 100, and 10 000 ng) with and without superimposed plumes of either clean air or sex pheromone in a wind tunnel. Moths provided with the lowest doses of background clouds without a superimposed plume did not fly upwind. Moths provided with higher doses of background clouds, with or without superimposed air plumes, increased their track, course, and drift angles (i.e., their zigzags headed more towards crosswind) with increased dose, but slowed their velocity. No differences in flight track parameters were observed for moths provided with a superimposed pheromone plume, regardless of the background cloud dose. Moreover, moths were able to locate the source of superimposed air plumes in the highest background dose, and of superimposed pheromone plumes in any background dose. The significance of these results is discussed in the context of mating disruption.

It is thought that orientation by male moths along pheromone plumes is guided by interception of filaments of pheromone along that plume and that clean air gaps are required for upwind progress. Given that several investigations have determined that cells sensitive to pheromone can resolve only low rates of encounter with pheromone filaments, generally up to 10 pulses/s, it would appear that individual filaments encountered at higher rates would not be resolved by the insects' sensory system and therefore the stimulus would be perceived as a non-flickering signal. Behaviourally, this has been thought to be expressed as the cessation of upwind progress. Previous studies with Cadra cautella (Walker) (Lepidoptera: Pyralidae) demonstrated that upwind flight by these males is not inhibited in rapidly pulsed plumes. Therefore, to determine whether a flickering signal is required for upwind progress by C. cautella, males were introduced to homogeneous clouds of pheromone in a wind tunnel and their behaviour recorded. For comparison, Pectinophora gossypiella (Saunders) (Lepidoptera: Gelechiidae), a species from a long-diverged lineage, was also used. Upwind progress by C. cautella is not impeded by the constant olfactory signal provided by a homogeneous cloud of pheromone, but this is not true for P. gossypiella. Furthermore, although C. cautella directs its flight upwind in a homogeneous cloud, its heading is not always due upwind. Potential mechanisms are discussed. It is suggested that C. cautella does not require a flickering signal to progress upwind.

Airborne pheromone plumes in wind comprise filaments of odour interspersed with gaps of clean air. When flying moths intercept a filament, they have a tendency to surge upwind momentarily, and then fly crosswind until another filament is intercepted. Thus, the moment-to-moment contact with pheromone mediates the shape of a flight track along the plume. Within some range of favourable interception rates, flight tracks become straighter and are headed more due upwind. However, as the rate of interception increases, there comes a point at which the moth should not be able to discern discreet filaments but, rather, should perceive a 'fused signal'. At the extreme, homogeneous clouds of pheromone inhibit upwind progress by representative tortricids. In a wind tunnel, Cadra cautella (Walker) (Lepidoptera: Pyralidae) were presented with 10 ms pulses of pheromone at a repetition rate of 5, 10, 17 and 25/s and a continuous, internally turbulent plume. Pulse size and concentrations were verified with a miniature photoionization detector sampling surrogate odour, propylene, at 100 Hz. Male moths maintain upwind progress even at plumes of 25 filaments/s. Furthermore, moths exhibited greater velocities and headings more due upwind at 17 and 25 Hz than at the lower frequencies or with the continuous plume. It is hypothesized that either C. cautella possesses a versatile sensory system that allows the resolution of these rapidly pulsed pheromone plumes, or that this species does not require a 'flickering' signal to fly upwind.

The patterns of stimulus available to moths flying along pheromone plumes in a 3-m-long wind tunnel were characterized using a high frequency photoionization detector in conjunction with an inert tracer gas. Four contrasting flow regimes and source conditions were produced: odor released in pulses from a vertical and horizontal array of four sources, odor released continuously from a point source, and odor released continuously from a point source into an oscillatory wake. Although the four flow regimes produced plumes of intermittent and fluctuating concentration, there were considerable differences in the structure of the signal presented to the sensor. Pulses of tracer gas released at 10 Hz retained most of their longitudinal and lateral separation. The plume growing in the disturbed flow (`oscillatory'), was broader in its lateral extent than the plume growing in an undisturbed flow (`continuous'), and the concentrations in the former were lower at each downstream position. The signal recorded in the disturbed flow had higher intermittency, but the ratio between the peak concentration and the signal mean was lower than in the continuous plume. Time scales were typically longer in the tunnel than in a field setting, but length scales and the main features of intermittency and fluctuation were similar. Moths flying along plumes of pheromone in this and similar wind tunnels typically slow their velocity and narrow the lateral excursions of their flight track as they approach a pheromone source. Which features of the plumes measured in this study account for these behavioral reactions remains to be determined.

Three approaches were used to investigate effects of host plant epicuticular waxes on oviposition site selection by Plutella xylostella (L.). In the first approach, oviposition on canola (Brassica napus L.) that had epicuticular wax reduced by application of a carbamate herbicide (S-ethyl dipropylthiocarbamate) was compared with oviposition on untreated control plants. A second approach compared oviposition on sibling strains of B. napus with different wax blooms (glossy and waxy), and a third approach compared oviposition by P. xylostella on parafilm that had been applied to glossy and waxy B. napus strains for transfer of leaf components. Significantly more eggs were deposited on herbicide-treated plants (with reduced epicuticular wax) than on untreated controls. Similarly, more eggs were deposited on glossy than on waxy sibling strains of B. napus. In parafilm assays significantly more eggs were deposited on treated than on untreated parafilm. Several mechanisms could explain the differences in attractiveness of surfaces with varying wax content as oviposition sites for P. xylostella, including visual, chemical, and tactile differences between substrates. These mechanisms are discussed.

There is currently a debate about the role played by temporal patterns in neural activity in olfactory coding. An accurate analysis of this question, however, is only possible if the temporal properties of a stimulus itself are well defined. So far, no technique with sufficient temporal resolution has been available to accomplish this. Using a photoionization detector (PID), we show that theconfiguration of the odor delivery apparatus and the airflow settings greatly influence the integrity of a stimulus profile within an odor delivery apparatus. In a situation where pulsatile odor stimuli are applied to a stationary preparation, we tested the effect of1) axial and off-center location within the airstream,2) airflow of the odor delivery,3) exit tube length,4) exit tube diameter,5) orientation of the odor delivery device in relation to the exhaust flow, and 6) exhaust tube air speed.This has important implications for the study of time in olfaction; significant planning must be incorporated into the design of the experiment to provide a well-defined odor delivery system.

Trains of 20-ms-duration pulses of pheromone were delivered at rates of 1-33 Hz to antennal preparations of males of Bombyx mori and Lymantria dispar, two moth species with bipectinate antennae. Resolution of rapidly pulsed plumes of pheromone was not compromised by a complex antennal morphology or by moderate changes in wind speed (25-50 cm/s). Fourier analysis of the electroantennograms resolved the temporal structure of the signal at frequencies up to 25 Hz for B. mori and up to 5 Hz for L. dispar. The ability of these sensory structures to identify the original (unchanged) frequency of the pulse train is particularly noteworthy because air is slowed by about an order of magnitude as it passes through bipectinate antennae. Although an unchanging frequency in slowed airflow may be counterintuitive, this flow pattern, and its effects on odorant patch shape and spacing, is explained from fluid mechanical principles (i.e., the principle of continuity). An unchanging frequency suggests that as decelerating air passes through a bipectinate antenna, the slowed patches of odorant are stretched, thinned, and brought closer together by the same factor with which they are slowed.

Dynamic properties of pheromone plumes are behaviorally important in some moths for inducing upwind flight, but little is known about the time-dependent properties of odor transduction or the mechanisms that limit receptor dynamic sensitivity. We stimulated male antennae of two moth species, Cadra cautella and Spodoptera exigua, with pheromone plumes in a wind tunnel while recording electroantennograms (EAG) and concentration of a surrogate plume (propylene, which mimics a pheromone plume) using a photoionization detector (PID). Turbulent plumes were produced by mechanical baffles, creating broad frequency range dynamic concentration changes at the antennae. Frequency response functions and coherence functions between PID and EAG signals were used to measure the dynamic responses of the two species to pheromone blends and individual components. A single time constant filter fitted the responses of both species, but S. exigua was about three times faster than C. cautella. Responses to individual pheromone components were significantly different in S. exigua but not in C. cautella. We also fitted the data with a simple block-structured nonlinear cascade. This supported the simple filter model but also suggested that the response saturates at an early stage of chemotransduction.

Male antennae of Cadra cautella, Pectinophora gossypiella, and Spodoptera exigua were presented with 20-ms-duration pulses of their two-component pheromone at rates of 1 to 33 Hz. Fourier analyses of electroantennograms resolved the temporal structure of trains of pheromone filaments delivered at up to 33 Hz for C. cautella and S. exigua and 25 Hz for P. gossypiella. Pheromone components tested separately for each species were generally equivalent in filament resolution to complete blends. Ambient temperatures of 18, 23 and 28 degrees C affected filament resolution only slightly, with poorer ability to discriminate rapidly pulsed signals at 18 degrees C. The question of how, or indeed if, such frequencies are conserved beyond the peripheral nervous system, remains.
