Abstract There is increasing evidence for blood–brain barrier (BBB) alterations in Parkinson’s disease (PD), the second most common neurodegenerative disorder with rapidly rising prevalence. Altered tight junction and transporter protein levels, accumulation of α-synuclein and increase in inflammatory processes lead to extravasation of blood molecules and vessel degeneration. This could result in a self-perpetuating pathophysiology of inflammation and BBB alteration, which contribute to neurodegeneration. Toxin exposure or α-synuclein over-expression in animal models has been shown to initiate similar pathologies, providing a platform to study underlying mechanisms and therapeutic interventions. Here we provide a comprehensive review of the current knowledge on BBB alterations in PD patients and how rodent models that replicate some of these changes can be used to study disease mechanisms. Specific challenges in assessing the BBB in patients and in healthy controls are discussed. Finally, a potential role of BBB alterations in disease pathogenesis and possible implications for therapy are explored. The interference of BBB alterations with current and novel therapeutic strategies requires more attention. Brain region-specific BBB alterations could also open up novel opportunities to target specifically vulnerable neuronal subpopulations.

Increasing evidence points to infectious diseases as contributor to the pathogenesis of neurodegeneration in Parkinson's disease (PD), probably driven by a peripheral and CNS inflammatory response together with alpha-synuclein (aSyn) pathology. Pro-inflammatory lipopolysaccharide (LPS) endotoxin is suggested as a risk factor, and LPS shedding gram-negative bacteria are more prevalent in the gut-microbiome of PD patients. Here, we investigated whether LPS could contribute to the neurodegenerative disease progression via neuroinflammation, especially under conditions of aSyn pathology. To investigate this, we created a double-hit model based on the Thy1-aSyn mouse line (line 61), an established aSyn-overexpression model of PD, exposed to a single intraperitoneal injection of LPS at a dose of 0.8 mg/kg (equivalent to approximately 1,200,000 EU/kg). Clinical parameters, flow cytometry of blood and immune cells in the brain, brain immunohistology and motor behavior were evaluated over time. As expected, the LPS dosage induced transient acute symptoms and mild weight loss in mice, with full recovery after 7 days. In aSyn over-expressing mice, this single low dose of LPS was sufficient to alter the expression of specific markers on blood and brain immune cells and induced brain region-specific microgliosis that were present at 7 days post LPS injection. At 14 days post injection of LPS, aSyn expression was reduced in wild-type mice, indicating a specific response of the endogenous protein to the endotoxin. At this early time point, motor behavior is not yet robustly impacted by the observed pathological alterations. In conclusion, aSyn pathology renders the peripheral and central immune response more sensitive to a single low dose of bacterial endotoxin, which mimics a transient dysbiosis or gut infection. Thus, this data suggests that such peripheral triggers should be monitored in PD patients for instance by blood immune cell response as biomarkers. Furthermore, results from this study lend further support to the development of treatments aiming to reduce the impact of bacterial dysbiosis as a promising strategy to mitigate PD progression.

Dysfunction of the blood-brain barrier (BBB) is suggested to play a critical role in the pathological mechanisms of Parkinson's disease (PD). PD-related pathology such as alpha-synuclein accumulation and inflammatory processes potentially affect the integrity of the BBB early in disease progression, which in turn may alter the crosstalk of the central and peripheral immune response. Importantly, BBB dysfunction could also affect drug response in PD. Here we analyzed microvascular changes in isolated brain capillaries and brain sections on a cellular and molecular level during disease progression in an established PD mouse model that overexpresses human wild-type alpha-synuclein (Thy1-aSyn, line 61). BBB alterations observed in Thy1-aSyn mice included reduced vessel density, reduced aquaporin-4 coverage, reduced P-glycoprotein expression, increased low-density lipoprotein receptor-related protein 1 expression, increased pS129-alpha-synuclein deposition, and increased adhesion protein and matrix metalloprotease expression together with alterations in tight junction proteins. Striatal capillaries presented with more dysregulated BBB integrity markers compared to cortical capillaries. These alterations of BBB integrity lead, however, not to an overt IgG leakage in brain parenchyma. Our data reveals intricate alterations in key proteins of BBB function together with histological evidence for altered structure of the brain vasculature. Thy1-aSyn mice represent a useful model to investigate therapeutic targeting of BBB alterations in synucleinopathies.

Leafy vegetables, mainly lettuces, are currently the main crop cultivated in controlled environment agriculture (CEA), including vertical farming and plant factories. There is a rising demand to expand this portfolio with a wider variety of underutilized edible plants containing various bioactive compounds and sensory properties seeking to enrich human diets. However, the optimal cultivation conditions for these underutilized plants significantly differ from those optimized for lettuce, basil, and other popular CEA crops. Therefore, this study aims to explore the impacts of light-emitting diode (LED) lighting intensity (photosynthetic photon flux density, PPFD) on green leaf Perilla frutescens cultivated in CEA. Plants were grown under four levels of LED lighting PPFDs from 150 to 300 µmol m−2s−1 for 4 weeks. Plant biomass productivity, soluble sugar contents, antioxidant properties (DPPH, ABTS free radical scavenging activities, FRAP antioxidant power), and total contents of phenolic compounds in leaves were evaluated at harvesting time. Further, harvested plant material was stored in the dark, at +6 °C, and the water content, water loss and transpiration rate, leaf sugar contents, and antioxidant properties were monitored 1, 3, and 5 days after harvesting. The summarized data suggest that higher cultivation lighting PPFD results in better harvest quality preservation during post-harvest storage.

Agricultural crop production practices are being developed for organic, sustainable, and environmentally friendly farming systems. Developing efficient and resourceful crop fertilizers is significantly important for future agriculture. Various biofertilizers, such as animal manures, composts, and vegetable byproducts, have been successfully applied in agriculture. Anaerobic digestate, organic matter obtained from animal or plant waste processing during anaerobic digestion into biomass, has become popular due to its versatility, multiple purposes, and facile application methods. Digestate has recently been widely used in agriculture to enrich the soil with nutrients and thus increase crop yields. Several studies have shown that anaerobic digestate is a valuable fertilizer that can be used as a biofertilizer in field and greenhouse horticulture. Also, research has been carried out on the use of digestate in hydroponic horticulture. This review presents the research results and discusses the possibilities of using anaerobic digestate in greenhouse horticulture. Its objective is to provide a comprehensive understanding of the application of digestate from various sources and its impact on the growth, progress, yield, and quality of greenhouse-grown vegetables.

The prevalence of apples as the most widely consumed fruit globally does not exempt them from storage-related issues, resulting in substantial harvest losses. A prominent concern is the development of rot due to various factors during storage. This research endeavors to examine the influence of agrotechnological methods on the longevity of apples and the incidence of rot throughout storage. Apple trees (Malus domestica Borkh. cv. Rubin) grafted on dwarfing rootstocks P60 were planted in 2010 in single rows with a spacing of 1.25 m between trees and 3.5 m between rows. Eight combinations of different growth control measures (manual, mechanical pruning, spraying, trunk cutting) were selected for the experiment. The implementation of mechanical pruning, in conjunction with trunk cutting and Ca-prohexadione spraying, as well as summer pruning, detrimentally impacted the shelf life of apples. Examination of the storage period revealed a loss of 33–40% of the crop due to rot. Conversely, manual pruning sustained a consistent level of phenolic compounds throughout the storage period. Other pruning methods resulted in a notable increase in phenolic compounds, ranging from 67% to a two-fold rise compared to the compounds present at harvest. However, the integration of mechanical pruning with subsequent manual pruning not only significantly augmented the yield of apples but also yielded a shelf life akin to that of manually pruned apples. Following the analysis of the results, it is advisable to conduct mechanical pruning of the apples intended for storage along with supplementary manual pruning.

Microgreens are vegetable greens that are harvested early while they are still immature and have just developed cotyledons. One of the disadvantages and a challenge in production is that they exhibit a short shelf life and may be damaged easily. In seeking to prolong the shelf life, some pre- and postharvest interventions have been investigated. Here, kale and mustard microgreens were grown in a controlled-environment walk-in chamber at +21/17 °C, with ~65% relative air humidity, while maintaining the spectral composition of deep red 61%, blue 20%, white 15%, and far red 4% (150, 200, and 250 µmol m−2 s−1 photosynthetic photon flux density (PPFD)). Both microgreens seemed to exhibit specific and species-dependent responses. Higher PPFD during growth and storage in light conditions resulted in increased contents of TPC in both microgreens on D5. Additionally, 150 and 250 PPFD irradiation affected the α-tocopherol content by increasing it during postharvest storage in kale. On D0 150 for kale and 200 PPFD for mustard microgreens, β-carotene content increased. D5 for kale showed insignificant differences, while mustard responded with the highest β-carotene content, under 150 PPFD. Our findings suggest that both microgreens show beneficial outcomes when stored in light compared to dark and that mild photostress is a promising tool for nutritional value improvement and shelf-life prolongation.

Horticultural plant material offers several advantages for isolating exosomes and other natural plant-derived exosome-like nanoparticles (PDENs) due to the accessibility and affordability of plant material for widespread applications. This study aims to explore the impacts of the tomato genotype (‘Admiro’, ‘Roma’, ‘Brooklyn’, ‘Marmande’ and ‘Betalux’) and the main cultivation parameters in controlled environment agriculture on the yield and properties of their PDENs for pharmaceutical and cosmeceutical applications. The PDEN yield, size distribution, and antioxidative properties of young tomato seedlings were evaluated. The ‘Betalux’ tomato was distinguished by a remarkably higher nanoparticle concentration and a uniform size distribution and was selected for further experiments. The impact of cultivation temperature (18, 22, and 26 °C), nitrogen nutrition (0, 250, and 500 mg L−1), and the lighting photosynthetic photon flux density (PPFD; 150, 250, and 450 µmol m−2 s−1) on nanoparticle properties was investigated. Optimal conditions consisting of a temperature of 22 °C, 250 mg L−1 nitrogen nutrition, and 250 µmol m−2 s−1 lighting PPFD were used as a reference. Optimal temperature, nitrogen nutrition, and lighting intensity resulted in the highest nanoparticle yield, the most uniform particle distribution, and the highest impact of PDEN preparations on keratinocyte metabolic activity. Deviation from optimal cultivation conditions reduced the tomato biomass and the PDEN protein and yield.

BackgroundThe association between assisted reproductive technologies (ARTs) and the body mass index (BMI) of children remains controversial. Confounding by morbidity and other factors associated with parental infertility may have biased studies comparing children born after ART with children born after no treatment. We investigated the associations between different fertility treatments and BMI in children at age 5 to 8 years, adjusting for and stratifying by causes of parental infertility.Methods and findingsThis Danish cohort study included 327,301 children born between 2007 and 2012 (51% males, median age at follow-up 7 years). Of these, 13,675 were born after ART, 7,728 were born after ovulation induction with or without intrauterine insemination [OI/IUI], and 305,898 were born after no fertility treatments. Using the International Obesity Task Force (IOTF) standards, we defined overweight (BMI ≥ IOTF-25) and obesity (BMI ≥ IOTF-30). We compared children born after ART versus OI/IUI; intracytoplasmic sperm injection (ICSI) versus conventional in vitro fertilization (IVF); and frozen-thawed versus fresh embryo transfer and estimated crude and adjusted prevalences of children with overweight or obesity at age 5 to 8 years, prevalence odds ratios (PORs), and differences in mean BMI z-scores. Adjustment was performed using stabilized inverse probability of treatment weights, including parity, year of conception, parental causes of infertility, age, educational level, comorbidities, maternal country of origin, BMI, and smoking as covariates. The crude prevalence of obesity was 1.9% in children born after ART, 2.0% in those born after OI/IUI, and 2.7% in those born after no fertility treatment. After adjustment, children born after ART and OI/IUI had the same prevalence of being overweight (11%; POR 1.00, 95% confidence interval [CI] 0.91 to 1.11; p = 0.95) or obese (1.9%; POR 1.01, 95% CI 0.79 to 1.29; p = 0.94). Comparison of ICSI with conventional IVF yielded similar pattern (POR 0.95, 95% CI 0.83 to 1.07; p = 0.39 for overweight and POR 1.16, 95% CI 0.84 to 1.61; p = 0.36 for obesity). Obesity was more prevalent after frozen-thawed (2.7%) than fresh embryo transfer (1.8%) (POR 1.54, 95% CI 1.09 to 2.17; p = 0.01). The associations between fertility treatments and BMI were only modestly different in subgroups defined by the cause of infertility. Study limitations include potential residual confounding, restriction to live births, and lack of detailed technical information about the IVF procedures.ConclusionsWe found no association with BMI at age 5 to 8 years when comparing ART versus OI/IUI or when comparing ICSI versus conventional IVF. However, use of frozen-thawed embryo transfer was associated with a 1.5-fold increased risk of obesity compared to fresh embryo transfer. Despite an elevated relative risk, the absolute risk difference was low.

Introduction Comorbidities are common in patients with chronic obstructive pulmonary disease (COPD). Estimates of prevalence, incidence and prognostic impact of comorbidities provide foundational knowledge of COPD epidemiology. We examined the prevalence, incidence and prognostic impact of 21 comorbidities among patients with COPD compared with the Danish general population.Methods We conducted a nationwide, population-based cohort study based on longitudinal Danish registry data, covering all Danish hospitals (2010–2021). The cohorts comprised 142 973 patients with a first-time hospital-based diagnosis of COPD and 428 917 age-matched and sex-matched comparators from the general population. During follow-up, we estimated the 5-year risk and risk difference, using competing risk methods when applicable.Results At time of diagnosis, the comorbidities with the highest prevalence were mood, stress-related or anxiety disorders (25.2% for patients with COPD vs 13.1% for comparators), osteoporosis/hip fractures (17.4% vs 9.9%), diabetes (15.6% vs 10.5%), peripheral arterial disease (13.5% vs 4.9%) and heart failure (13.3% vs 4.0%). During follow-up, the risk of most incident comorbidities was markedly elevated among patients with COPD. The five comorbidities associated with the highest 5-year absolute risk difference with respect to the risk in the general population were mood, stress-related or anxiety disorders (5.7%), osteoporosis/hip fractures (5.6%), heart failure (4.2%), smoking-related cancers (2.8%) and peripheral arterial disease (2.7%). The 5-year mortality risk was 43% vs 17.7%. Among patients with COPD, the 5-year mortality risk markedly increased with the number of comorbidities present.Conclusions Our population-based findings underscore the importance of considering comorbidities in the management of COPD.

Anaerobic digestate has been commonly used for the cultivation of vegetable seedlings and as one of the measures for improving peat substrate. Studies have shown that there has been further research conducted on the effect of anaerobic digestate on greenhouse vegetable seedlings. The main objective was to investigate the effect of the additional insertion of different rates of solid grain waste digestate into peat substrate on tomato seedling quality. The results showed that 10% of solid grain waste digestate (peat digestate) application with transplanted seedlings had better biometrical measures and photosynthetic parameters for tomato seedlings compared with the control variant.

Plant extracts are getting attention for their sterilizing, growth-promoting properties in the agricultural field. No exception exists when it comes to the treatment of propagating material with these natural components. Plant extracts may have complex compositions which affect plants or seeds in a positive way. However, they could also cause negative effects, like decreased germination, secondary metabolite contents or biomass gain. As the agricultural field requires sustainable techniques for plant growth and quality assurance in production, it is important to evaluate the potential effects of every alternative natural compound that shows promise for future plant treatment. Garlic (Allium sativum L.) is a popular spice crop grown in various regions of the world. Therefore, our study focused on an investigation of the early physiological and biochemical changes in garlic cloves (cv. ‘Jarus’ and ‘Vasariai’) treated with Thymus vulgaris essential oil and Syzygium aromaticum extract. Experiments were carried out in controlled climate conditions, in which the treated cloves were kept for four weeks. Results show that S. aromaticum extract increased germination in cv. ‘Jarus’ compared to untreated garlic cloves. Meanwhile, T. vulgaris caused lower germination of both cultivars compared to untreated cloves. None of the extracts increased biomass gain in garlic cloves. ABTS antioxidant activity was decreased by both extracts in cv. ‘Jarus’ (~7–47%) and ‘Vasariai’ (~22–32%) compared to untreated garlic cloves. Antioxidant DPPH and FRAP activities were ~6–11% and ~14–15% higher after ‘Vasariai’ treatment with extracts. Meanwhile, in ‘Jarus’ only, S. aromaticum extract caused an increase in DPPH and FRAP antioxidant activities. The elemental content was the highest in garlic cloves treated with T. vulgaris essential oil. Total phenolic compounds (TPC) and flavonoids (TF) were 1.2 times higher after S. aromaticum treatment of ‘Jarus’ compared to untreated cloves, while slightly increased TPC and two times higher TF values were found after treatment with T. vulgaris. Around 10% higher TPC and 1.5–3 times lower TF values were observed after the plant extract treatment of ‘Vasariai’ garlic cloves. Our study demonstrates initial changes in garlic cloves dependent on the plant extract treatment and cultivar. This provides important information for the future utilization of such technology, either alone or in combination with others.

Essential oils and extracts are investigated in sustainable plant protection area lately. Alternative antifungal substances are especially relevant for major economic-relevance pathogens, like Botrytis cinerea (causal agent of strawberry grey mold), control. However, the reaction of plants to alternative protection with plant-origin products is currently unknown. Induced stress in plants causes changes in antioxidant and photosynthetic systems. The aim of the research was to determine the defense response of strawberry plants under application of coriander seed products. In the first step of the research, we determined coriander seed (Coriandrum sativum), black seed (Nigella sativa) and peppermint leaf (Menta × piperita) products’ antifungal activity against B. cinerea in vitro. Secondly, we continued evaluation of antifungal activity under controlled environment on strawberry plants of the most effective coriander seed products. Additionally, we evaluated the antioxidant and photosynthetic parameters in strawberries, to examine the response of plants. Antifungal activity on strawberries was determined based on grey mold incidence and severity after application of coriander products. Impact on photosynthetic system was examined measuring photosynthetic rate, transpiration rate, stomatal conductance, and intercellular to ambient CO2 concentration. Strawberry leaves were collected at the end of the experiment to analyze the antioxidant response. The highest antifungal activity both in vitro and on strawberries had coriander seed essential oil, which decreased grey mold severity. Coriander extract increased the photosynthetic capacity and antioxidant response of strawberry plants, however had negative effect on suppression of grey mold. In most cases, the essential oil activated antioxidant response of strawberry plants lower than extract. Our study results provide no direct impact of increased photosynthetic capacity values and antifungal effect after treatment with natural oils. The highest concentrations of coriander essential oil and extract potentially demonstrated a phytotoxic effect.

This study aims to evaluate the impacts of the lighting photosynthetic photon flux density (PPFD) on the growth, photosynthesis, and antioxidant response of common purslane (Portulaca oleracea) cultivars to determine energy-efficient lighting strategies for CEA. Green and golden purslane cultivars were cultivated in CEA chambers and four experimental treatments consisting of PPFDs of 150, 200, 250, and 300 ± 10 µmol m−2s−1 were performed, representing daily light integrals (DLIs) of 8.64–17.28 mol m−2d−1 throughout a 16 h photoperiod. The results show that photoresponses to light PPFDs are cultivar-specific. The green cultivar accumulates 174% more dry weight at 300 PPFD compared to the golden cultivar, and also has a higher LUE, but a lower ETR. Dry weight accumulation, plant height, and leaf area dependence on light intensity do not highlight the economic significance of light PPFD/DLI. The derivative parameter (Δ fresh weight (%)/ΔDLI %) more efficiently explains how the percentage increase in DLI due to an increased PPFD affects the percentage of biomass gain between these PPFD treatments. For both cultivars, the relative fresh weight gain is maximal when the lighting PPFD increases from 200 to 250 µmol m−2s−1 and declines with PPFD increases from 250 to 300.

Over the past ten years, particular attention has been paid to Artemisia L. genus plants and their chemical composition. Artemisia abrotanum L. and Artemisia absinthium L., which are traditional medicinal, aromatic plants with a strong digestive activity that is determined by the various phytochemicals. The research aim was to determine the variation in polyphenols in the samples of different vegetation stages of two Artemisia species and to evaluate the antioxidant activity of methanolic extracts in vitro. Phytochemical analysis of the Artemisia samples was carried out using spectrophotometric methods and HPLC-PDA techniques, while antioxidant activity was determined using DPPH, ABTS, FRAP, and CUPRAC assays. Significant differences in the diversity of phenolic compounds were found among Artemisia species during vegetation stages. Chlorogenic acid, 3,5-dicaffeoylquinic acid, and rutin were predominant among the phenolic compounds. The study provides valuable insights into the composition of phenolic compounds and flavonoids in Artemisia plants at different growth stages, shedding light on their potential pharmacological effects and antioxidant activities. These findings contribute to the knowledge of the quality and safety of herbal materials derived from Artemisia species.

Controlled environment agricultural (CEA) systems create technological opportunities for the higher nutritional value of vegetables and herbs. It was hypothesized that UV-A light, supplementing basal light emitting diode (LED) illumination in CEA, would enhance growth and nutritional value (nutraceutical compounds and mineral element contents) in purple and green basil in a UV-A wavelength-specific manner. Therefore, blue (452 nm) and red (662 nm) 1:10 basal LED lighting (250 μmol m−2 s−1, 16 h) was supplemented with 1 mW cm−2 of 343, 366, 386, or 402 nm UV-A LED light for green ‘Italiano classico’ and purple ‘Red rubin’ basil cultivation. Different wavelengths have specific impacts for two basil genotypes, and certain light wavelengths should be selected to boost growth or to alter the contents of specific nutraceutical compounds. UV-A/violet 402 nm light enhanced growth, chicoric acid, β carotene, lutein, and zeaxanthin contents in green basil, while 343 nm UV-A light increased fresh weight, ascorbic acid, and carotenoid content in purple basil. UV-A light of 386 nm has the most negligible impact on reducing mineral element (P, Ca, Fe, K, Mg, Mn, and Zn) contents in basil. Understanding the wavelength dependence of plant responses to UV-A is essential for optimizing quality preservation and improving basil cultivation in controlled environment systems.

The age of seedlings affects not only the quality of the seedlings but also the yield. The age of seedlings of sweet peppers grown in a greenhouse and their cultivation method has been little studied. Therefore, the aim of this research was to determine the effect of agrotechnological tools (direct seeding or transplanting) on sweet pepper (Capsicum annuum L.) seedling quality and crop yield. The research was carried out in a greenhouse covered with double polymeric film at the Institute of Horticulture, Lithuanian Research Centre for Agriculture and Forestry. Two factors were investigated: seedling establishment method (direct seeding and transplanting) and their different age (60, 50, and 40 days). The 60-day-old seedlings (both sown directly and grown by transplanting) were taller, and had more leaves, a larger leaf area, and a thicker stem, than the 50- and 40-day-old seedlings. The highest content of dry matter was found in the leaves of 60-day-old seedlings that were grown by transplanting. The chlorophyll index was also highest in the leaves of these seedlings. The highest photosynthetic parameters (photosynthetic rate, stomatal conductance, intercellular CO2, transpiration rate) were found in the leaves of 40-day-old seedlings (both sown directly and grown by transplanting). Higher yields were demonstrated in sweet peppers whose seedlings were planted in the greenhouse at the age of 60 days and which were sown directly in cups.

Background: Different asthma phenotypes are driven by molecular endotypes. A Th1-high phenotype is linked to severe, therapy-refractory asthma, subclinical infections and neutrophil inflammation. Previously, we found neutrophil granulocytes (NGs) from asthmatics exhibit decreased chemotaxis towards leukotriene B4 (LTB4), a chemoattractant involved in inflammation response. We hypothesized that this pattern is driven by asthma in general and aggravated in a Th1-high phenotype.Methods: NGs from asthmatic nd healthy children were stimulated with 10 nM LTB4/100 nM N-formylmethionine-leucyl-phenylalanine and neutrophil migration was documented following our prior SiMA (simplified migration assay) workflow, capturing morphologic and dynamic parameters from single-cell tracking in the images. Demographic, clinical and serum cytokine data were determined in the ALLIANCE cohort.Results: A reduced chemotactic response towards LTB4 was confirmed in asthmatic donors regardless of inhaled corticosteroid (ICS) treatment. By contrast, only NGs from ICS-treated asthmatic children migrate similarly to controls with the exception of Th1-high donors, whose NGs presented a reduced and less directed migration towards the chemokines. ICS-treated and Th1-high asthmatic donors present an altered surface receptor profile, which partly correlates with migration.Conclusions: Neutrophil migration in vitro may be affected by ICS-therapy or a Th1-high phenotype. This may be explained by alteration of receptor expression and could be used as a tool to monitor asthma treatment.

The precise management of the nutrient solution pH is substantial for optimizing the growth and yield of hydroponically cultivated crops. Nutrient solution pH affects the solubility and availability of essential nutrients; however, during hydroponic cultivation, the pH of nutrient solution fluctuates due to unbalanced ion absorption by plants. Nutrient solution pH fluctuations might be passively controlled using a supplementary buffering agent. Nutrient solution characteristics such as pH and buffering agent molarity must be addressed simultaneously. Therefore, in this study, we aimed to evaluate hydroponically cultivated lettuce’s physiological response to minor nutrient solution pH differences and the impact of supplementary buffering agent molarity on its growth, antioxidant activity, and mineral nutrient uptake. Green lettuce (Lactuca sativa (L.)) ‘Hugin’ was cultivated in deep water culture (DWC) hydroponics in a controlled environment replicating common vertical farming conditions, and nutrient solution pH impact within the range of 5.0–6.5 was evaluated every 0.5 pH unit. Initial nutrient solution buffer capacity was enhanced by adding MES (2-(N-morpholino) ethanesulfonic acid) at 0, 1, 3, and 5 mM concentrations. Results show that even small nutrient pH differences are reflected in plant physiological responses, indicating the significance of precise pH management. Even though dry weight accumulation is not affected by the pH 5.0–5.5 treatment, it increases root antioxidant response according to ABTS (2,2-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) radical scavenging activity. An initial nutrient solution buffer capacity increment with 3 mM of MES (2-(N-morpholino) ethanesulfonic acid) supplementary buffering agent resulted in a 17% yield increase compared to a nutrient solution without an additional buffering agent. MES buffer can provide adequate, passive, and precise nutrient solution pH control within the pH range of 6.0–6.5.

Constantly changing environments often negatively affect yield potential. Phytohormone-based biostimulants are known for their ability to control plant development and reduce the influences of negative environmental impacts and facilitate more efficient usage of resources. The aim of this study was to evaluate the effect of phytohormone-based biostimulants on lettuce (Lactuca sativa L.) antioxidant and photosynthetic responses and biomass formation. Lettuce was grown in a greenhouse with supplemental lighting; a 16 h photoperiod was maintained. Ten combinations of kinetin, indole-3-acetic acid, gibberellic acid, abscisic acid and salicylic acid were applied at 12–13 BBCH. The results thereof have shown that combining growth and stress phytohormones resulted in higher biomass formation; additionally, combining two growth or two stress hormones led to antagonistic effects and reduced photosynthetic rates. Furthermore, the application of gibberellic and salicylic acid had the most positive effect on lettuce productivity. The perspective offered by this work has shown that with the manipulation of hormone concentrations, photosynthetic and antioxidant systems can be controlled, thus enabling control of yield and quality.

This study aims to expand the artificial lighting potential of controlled environment cultivations systems by introducing UV-A (~315–400 nm) wavelengths into the traditional, visible spectrum lighting, seeking to improve the nutritional and sensory value of cultivated leafy vegetables. The experiment was conducted in a closed climate-controlled chamber, maintaining 21/17 °C day/night temperature, ~55% relative humidity, and a 16 h photo/thermo period. Several genotypes of leafy vegetables, red and green leaf lettuce cultivars, mustard, and kale were cultivated under 250 µmol m−2s−1 basal LED lighting, supplemented by 385 nm UV-A or 405 UV-A/violet wavelengths for 1.1 mW cm−2 for 12 h photoperiod for the whole cultivation cycle. The results show that UV-A/violet light impacts on leafy vegetable growth, free radical scavenging activity, sugar, and phytochemical (α tocopherol, α + β carotenes, epicatechin, rosmarinic and chicoric acid contents) are species-specific, and do not correlate with untrained consumer’s sensory evaluation scores. The 405 nm light is preferable for higher antioxidant and/or sensory properties of kale, mustard, and green leaf lettuces, but both UV-A wavelengths reduce growth parameters in red leaf lettuce.

Physics-informed neural networks (PINNs) were developed to overcome the limitations associated with the acquisition of large training data sets that are commonly encountered when using purely data-driven machine learning methods. This paper proposes a PINN surrogate modeling methodology for steady-state integrated thermofluid systems modeling based on the mass, energy, and momentum balance equations, combined with the relevant component characteristics and fluid property relationships. The methodology is applied to two thermofluid systems that encapsulate the important phenomena typically encountered, namely: (i) a heat exchanger network with two different fluid streams and components linked in series and parallel; and (ii) a recuperated closed Brayton cycle with various turbomachines and heat exchangers. The results generated with the PINN models were compared to benchmark solutions generated via conventional, physics-based thermofluid process models. The largest average relative errors are 0.17% and 0.93% for the heat exchanger network and Brayton cycle, respectively. It was shown that the use of a hybrid Adam-TNC optimizer requires between 180 and 690 fewer iterations during the training process, thus providing a significant computational advantage over a pure Adam optimization approach. The resulting PINN models can make predictions 75 to 88 times faster than their respective conventional process models. This highlights the potential for PINN surrogate models as a valuable engineering tool in component and system design and optimization, as well as in real-time simulation for anomaly detection, diagnosis, and forecasting.

Medicinal and agricultural plants contain numerous phytochemical compounds with pronounced biological effects on human health. They are known to encapsulate most of their characteristic bioactive compounds within membranous elements of intercellular communication known as exosomes. These nanovesicles serve as capsules protecting their biological activity and improving their penetration into the tissue. Therefore, the application of plant exosome preparations holds considerable potential for cosmetics and pharmacy, but the quality and consistency of plant material for exosome isolation is of critical importance. Therefore, in this study, we aimed to evaluate yield, size distribution patterns, and antioxidant properties between nanovesicle preparations of the following portfolio of medicinal plants: Kalanchoe daigremontiana, Artemisia absinthium, Hypericum perforatum, Silybum marianum, Chelidonium majus, and Scutellaria baicalensis. Results showed that nanoparticle yield, size distribution, and antioxidant activities were specific to plant species. Compared to other plants, nanoparticle preparations from Artemisia absinthium were distinguished by remarkably higher yield and concentration, while the highest antioxidant activity of plant-derived nanoparticle preparations per weight and per particle was determined to occur in Chelidonium majus and Hypericum perforatum samples. Results showed no significant correlation in DPPH (2-diphenyl-1-picrylhydrazyl) free radical scavenging activity and FRAP (ferric reducing antioxidant power) between plant material and nanoparticle preparations. More detailed biochemical analysis of exosome preparations is necessary to validate their biological activity and its relation to source plant cells.

BackgroundLong-term complications of venous thromboembolism (VTE) hamper physical function and impair quality of life; still, it remains unclear whether VTE is associated with risk of permanent work-related disability. We aimed to assess the association between VTE and the risk of receiving a permanent work-related disability pension and to assess whether this association was explained by comorbidities such as cancer and arterial cardiovascular disease.Methods and findingsA Danish nationwide population-based cohort study consisting of 43,769 individuals aged 25 to 66 years with incident VTE during 1995 to 2016 and 218,845 birth year-, sex-, and calendar year-matched individuals from the general population, among whom 45.9% (N = 120,540) were women, was established using Danish national registries. The cohorts were followed throughout 2016, with permanent work-related disability pension as the outcome. Hazard ratios (HRs) with 95% confidence intervals (CIs) for disability pension were computed and stratified by sex and age groups (25 to 34, 35 to 44, 45 to 54, and 55 to 66 years of age) and adjusted for comorbidities and socioeconomic variables. Permanent work-related disability pensions were granted to 4,415 individuals with VTE and 9,237 comparison cohort members (incidence rates = 17.8 and 6.2 per 1,000 person-years, respectively). VTE was associated with a 3-fold (HR 3.0, 95% CI: 2.8 to 3.1) higher risk of receiving a disability pension. Adjustments for socioeconomic status and comorbidities such as cancer and cardiovascular diseases reduced the estimate (HR 2.3, 95% CI: 2.2 to 2.4). The risk of disability pension receipt was slightly higher in men than in women (HR 2.5, 95% CI: 2.3 to 2.6 versus HR 2.1, 95% CI: 2.0 to 2.3). As this study is based on medical and administrative registers, information on post-VTE care, individual health behavior, and workplace factors linked to disability pension in the general population are lacking. Furthermore, as disability pension schemes vary, our results might not be directly generalizable to other countries or time periods.ConclusionsIn this study, incident VTE was associated with increased risk of subsequent permanent work-related disability, and this association was still observed after accounting for comorbidities such as cancer and cardiovascular diseases. Our results emphasize the social consequences of VTE and may help occupational and healthcare professionals to identify vulnerable individuals at risk of permanent exclusion from the labor market after a VTE event.

This study evaluates the effect of end-of-day blue (EOD B) light on the physiological response of lettuce (Lactuca sativa, Lobjoits Green Cos) at different phenological development stages. Plants were grown in a controlled environment growth chamber (day/night temperature 21 ± 2 °C; relative air humidity 60 ± 5%) under the light of light-emitting diodes (LEDs) consisting of 5% blue (B; 450 nm), 85% red (R; 660 nm), and 10% green (G; 530 nm) photosynthetic photon flux density (PPFD) at 200 µmol m−2 s−1 for 16 h d−1 (BRG, control) for 8, 15, and 25 days (BBCH 12, BBCH 14, and BBCH 18, respectively). For the EOD B treatments, lettuce plants were additionally illuminated with 100% of B light at 30 and 60 µmol m−2 s−1 PPFD for 4 h d−1 (B30 and B60, respectively). The results show that EOD B light caused the elevated shoot elongation of lettuce plants regardless of their growth stages. However, leaf width increased only in more developed lettuce plants (BBCH 18). EOD B light negatively affected the development of new leaves and fresh weight, except for seedlings (BBCH 12). Most photosynthetic and spectral leaf indices also decreased when lettuce was treated with EOD B light, especially under the PPFD level of 60 µmol m−2 s−1. Moreover, the changes in metabolic parameters such as DPPH free radical activity, free proline content, and H+-ATPase activity in lettuce showed a plant response to unfavorable conditions to EOD B light.

Cucumbers productivity and fruit quality depend on seedlings’ quality. The success of seedling cultivation largely depends on the choice of a suitable substrate. Therefore the aim of this research is to determine the effect of peat-vermicompost substrates on cucumber seedling quality and crop yield. The research was carried out in a greenhouse covered with double polymeric film in the Institute of Horticulture, Lithuanian Research Centre for Agriculture and Forestry. Cucumber seedlings were grown in different substrates: peat, peat + 10% vermicompost, peat + 20% vermicompost, and peat + 30% vermicompost. The growth of cucumber seedlings in peat and vermicompost substrates was induced. They were 1.9–18.6% taller, and the leaf area of this seedlings was 1.2–1.4 time larger. Furthermore, the fresh leaves mass of these seedlings was 22.7–33.1%, and the fresh root mass was 1.1–1.5 time bigger. The addition of vermicompost to peat substrate has a positive effect on the physiological parameters in the leaves of cucumber seedlings. The total yield of cucumbers grown in peat-vermicompost substrates was 7.4–11.1% higher than that of plants whose seedlings grew in peat substrate.

Optimal pre-analytical conditions for blood sample processing and isolation of selected cell populations for subsequent transcriptomic and epigenomic studies are required to obtain robust and reproducible results. This pilot study was conducted to investigate the potential effects of timing of CD4+ T-cell processing from peripheral blood of atopic and non-atopic adults on their transcriptomic and epigenetic profiles. Two heparinized blood samples were drawn from each of three atopic and three healthy individuals. For each individual, CD4+ T-cells were isolated from the first blood sample within 2 h (immediate) or from the second blood sample after 24 h storage (delayed). RNA sequencing (RNA-Seq) and histone H3K27 acetylation chromatin immunoprecipitation sequencing (ChIP-Seq) analyses were performed. A multiplicity of genes was shown to be differentially expressed in immediately processed CD4+ T-cells from atopic versus healthy subjects. These differences disappeared when comparing delayed processed cells due to a drastic change in expression levels of atopy-related genes in delayed processed CD4+ T-cells from atopic donors. This finding was further validated on the epigenomic level by examining H3K27 acetylation profiles. In contrast, transcriptomic and epigenomic profiles of blood CD4+ T-cells of healthy donors remained rather unaffected. Taken together, for successful transcriptomics and epigenomics studies, detailed standard operation procedures developed on the basis of samples from both healthy and disease conditions are implicitly recommended.

In order to ensure sufficient food resources for a constantly growing human population, new technologies (e.g., cold plasma technologies) are being developed for increasing the germination and seedling growth without negative effects on the environment. Pinaceae species are considered a natural source of antioxidant compounds and are valued for their pharmaceutical and nutraceutical properties. In this study, the seeds of seven different Norway spruce half-sib families were processed for one or two minutes with cold plasma (CP) using dielectric barrier discharge (DBD) plasma equipment. At the end of the second vegetation season, the total flavonoid content (TFC), DPPH (2,2- diphenyl-1-picryl-hydrazyl-hydrate), and ABTS (2,2’-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)) antioxidant activity, and the amounts of six organic acids (folic, malic, citric, oxalic, succinic, and ascorbic) were determined in the needles of different half-sib families of Norway spruce seedlings. The results show that the TFC, antioxidant activity, and amounts of organic acids in the seedling needles depended on both the treatment duration and the genetic family. The strongest positive effect on the TFC was determined in the seedlings of the 477, 599, and 541 half-sib families after seed treatment with CP for 1 min (CP1). The TFC in these families increased from 118.06 mg g−1 to 312.6 mg g−1 compared to the control. Moreover, seed treatment with CP1 resulted in the strongest increase in the antioxidant activity of the needles of the 541 half-sib family seedlings; the antioxidant activity, determined by DPPH and ABTS tests, increased by 30 and 23%, respectively, compared to the control. The obtained results indicate that the CP effect on the amount of organic acids in the needles was dependent on the half-sib family. It was determined that treatment with CP1 increased the amount of five organic acids in the needles of the 541 half-sib family seedlings. The presented results show future possibilities for using cold plasma seed treatment in the food and pharmacy industries.

This study aimed to evaluate the effect of dynamic red and blue light parameters on the physiological responses and key metabolites in lettuce and also the subsequent impact of varying light spectra on nutritive value. We explored the metabolic changes in carotenes, xanthophylls, soluble sugars, organic acids, and antioxidants; the response of photosynthetic indices [photosynthetic (Pr) and transpiration (Tr) rates]; and the intracellular to ambient CO2 concentration ratios (Ci/Ca) in lettuce (Lactuca sativa L. “Lobjoits Green Cos”). They were cultivated under constant (con) or parabolic (dyn) blue (B, 452 nm) and/or red (R, 662 nm) light-emitting diode (LED) photosynthetic photon flux densities (PPFDs) at 12, 16, and 20 h photoperiods, maintaining consistent daily light integrals (DLIs) for each light component in all treatments, at 2.3 and 9.2 mol m–2 per day for blue and red light, respectively. The obtained results and principal component analysis (PCA) confirmed a significant impact of the light spectrum, photoperiod, and parabolic profiles of PPFD on the physiological response of lettuce. The 16 h photoperiod resulted in significantly higher content of xanthophylls (neoxanthin, violaxanthin, lutein, and zeaxanthin) in lettuce leaves under both constant and parabolic blue light treatments (BconRdyn 16 h and BdynRdyn 16 h, respectively). Lower PPFD levels under a 20 h photoperiod (BdynRdyn 20 h) as well as higher PPFD levels under a 12 h photoperiod (BdynRdyn 12 h) had a pronounced impact on leaf gas exchange indices (Pr, Tr, Ci/Ca), xanthophylls, soluble sugar contents, and antioxidant properties of lettuce leaves. The parabolic PPFD lighting profile over a 16 h photoperiod (BdynRdyn 16 h) led to a significant decrease in Ci/Ca, which resulted in decreased Pr and Tr, compared with constant blue or red light treatments with the same photoperiod (BconRdyn and BdynRcon 16 h). Additionally, constant blue lighting produced higher α + β-carotene and anthocyanin (ARI) content and increased carotenoid to chlorophyll ratio (CRI) but decreased biomass accumulation and antioxidant activity.

In the study, we cultivated basil, beet, and mustard microgreens under different lighting treatments from light-emitting diodes (LEDs) and evaluated the contents of mineral nutrients. Microgreens grew under blue 447, red 638 and 665, far-red 731 nm LEDs, or the same spectrum but with partial substitution of 638 nm red with green 520 (BRG), yellow 595 (BRY), or orange 622 nm (BRO) LEDs (16 h photoperiod; total photon flux density of 300 μmol m −2 s −1). BRG, BRY, or BRO lighting had distinct effects on mineral contents among the microgreen species. BRG increased the content of mineral nutrients, especially in mustard and beet. In all microgreens, Ca and P were associated with BRG; in beet and mustard, Zn and Mg were associated with BRG; in basil, Zn was associated with BRY and Mg with BRO treatments. A broader photon spectrum increased Fe (up to 2.9–fold), K:Ca, P:Mg, and P:Zn in basil, and Fe:Zn in microgreens. We conclude that the partial replacement of red with green light was the most effective at enhancing the mineral nutrient content of microgreens, although responses varied among the crops studied.

Light and nutrients are among the most important factors for sustained plant production in agriculture. As one of the goals of the European Green Deal strategy is to reduce energy consumption, greenhouse growers focus on high-value crop cultivation with less-energy-demanding growing systems. This study aimed to evaluate the effect of fertilization at different light intensities on the growth of lettuce and basil and the activity of the antioxidant system. Sweet basil (Ocimum basilicum, ‘Opal’) and lettuce (Lactuca sativa, ‘Nikolaj’) were grown in a greenhouse supplementing natural light (~80 µmol m−2 s−1) with lighting at two photon flux densities (150 and 250 µmol m−2 s−1), 16 h photoperiod, and 20/16 °C day/night temperature in May (Lithuania, 55°60′ N, 23°48′ E). In each light regime treatment, half of the plants were grown without additional fertilization; the other half were fertilized twice a week with a complex fertilizer (NPK 3-1-3). The results showed that the antioxidant activity of basil was most affected by 150 µmol m−2 s−1 PPFD lighting and the absence of fertilization. Altered antioxidant activity in lettuce in the presence of 250 µmol m−2 s−1 PPFD additional light intensity and fertilization resulted in higher morphological parameters.

The study aimed to determine the changes in phenolic compounds content in lettuce (Lactuca sativa L. cv. Little Gem) depending on the preharvest short-term daytime or nighttime supplemental light-emitting diodes (LEDs) to high-pressure sodium lamps (HPS) lighting in a greenhouse during autumn and spring cultivation. Plants were grown in a greenhouse under HPS supplemented with 400 nm, 455 nm, 530 nm, 455 + 530 nm or 660 nm LEDs light for 4 h five days before harvest. Two experiments (EXP) were performed: EXP1—HPS, and LEDs treatment during daytime 6 PM–10 PM, and EXP2—LEDs treatment at nighttime during 10 AM–2 PM. LEDs’ photosynthetic photon flux density (PPFD) was 50 and HPS—90 ± 10 µmol m−2 s−1. The most pronounced positive effect on total phenolic compounds revealed supplemental 400 and 455 + 530 nm LEDs lighting, except its application during the daytime at spring cultivation, when all supplemental LEDs light had no impact on phenolics content variation. Supplemental 400 nm LEDs applied in the daytime increased chlorogenic acid during spring and chicoric acid during autumn cultivation. 400 nm LEDs used in nighttime enhanced chlorogenic acid accumulation and rutin during autumn. Chicoric and chlorogenic acid significantly increased under supplemental 455 + 530 nm LEDs applied at daytime in autumn and used at nighttime—in spring. Supplemental LEDs application in the nighttime resulted in higher phenolic compounds content during spring cultivation and the daytime during autumn cultivation.

In this study, we used apple tree (Malus domestica Borkh.) cv. Rubin grafts on dwarfing P60 rootstock. Our planting scheme was single rows with 1.25 m between trees and 3.5 m between rows. The aim of this study was to determine the impact of canopy training treatments, as a stress factor, on metabolic response to obtain key information on how to improve physiological behavior and the management of the growth and development of apple trees. The results indicated that all applied canopy training treatments significantly increased the total phenol and total starch contents in apple tree leaves. The total starch increased from 1.5- to almost 3-fold in all treatments, especially during the 2017 harvesting season, compared to the control. The fructose, sorbitol, and ratio of chlorophyll a to b in leaves also significantly increased. Higher precipitation levels induced changes in the accumulation of secondary metabolites in apple tree leaves and fruits during the 2017 harvesting season. The total phenol content significantly increased in apple tree leaves in all treatments, but the fructose content decreased. We observed the same tendencies in total phenolic content and glucose concentration in apple fruits. Therefore, the defense reaction might be a preferred option for apple tree cultivation and the optimization of its growth and development.

Modern apple orchard systems should guarantee homogeneity of fruit internal and external qualities and fruit maturity parameters. However, when orchards reach productive age, a variation of these parameters takes place and mostly it is related to uneven light distribution within the tree canopy. The aim of the study was to evaluate the canopy position’s effect on fruit internal and external quality parameters. This is the first study where all the main fruit quality and maturation parameters were evaluated on the same trees and were related to the light conditions and photosynthetic parameters. Four fruit positions were tested: top of the apple tree, lower inside part of the canopy, and east and west sides of the apple tree. Fruit quality variability was significant for fruit size, blush, colour indices, total sugar content, dry matter concentration, accumulation of secondary metabolites and radical scavenging activity. Fruit position in the canopy did not affect flesh firmness and fruit maturity parameters such as the starch index, Streif index and respiration rate. At the Lithuanian geographical location (55°60′ N), significantly, the highest fruit quality was achieved at the top of the apple tree. The tendency was established that apple fruits from the west side of the canopy have better fruit quality than from the east side and it could be related to better light conditions at the west side of the tree. Inside the canopy, fruits were distinguished only by the higher accumulation of triterpenic compounds and higher content of malic acid. Light is a main factor of fruit quality variation, thus all orchard management practices, including narrow two-dimensional tree canopies and reflecting ground covers which improve light penetration through the tree canopy, should be applied.

Global demand for food is increasing each year, but the area of land suitable for farming is limited. Thus, there is a need to grow not only larger quantities of food but also higher quality food products in the same area. This study aimed to evaluate the influence of rootstock and high-density orchards on cv. Auksis fruit quality. Two rootstocks were selected for this experiment, P 22 super dwarfing and P 60 dwarfing. Apple trees cv. Auksis were planted in the year 2001 in single rows spaced 1.00 m, 0.75 m, and 0.50 m, apart with 3 m between rows. High-density planting and rootstock combination was found to have no significant effect on sugar accumulation and most of the elements in apple fruits. However, super dwarfing P 22 rootstock accumulated significantly higher (up to 45%) content of organic acids and up to 33%–44% lower DPPH free radical scavering activity compared to P 60 dwarfing rootstock. After summarizing the obtained results, apples which accumulated the most antioxidants (according to the activity of phenolic compounds, DPPH• and ABTS•), magnesium, and potassium were collected from cv. Auksis apple trees which was grafted on super dwarfing P 22 rootstock and planted at 3 × 0.75 m distances.

The consumption of microgreens has increased due to their having higher levels of bioactive compounds and mineral nutrients than mature plants. The lighting conditions during the cultivation of microgreens, if optimally selected, can have a positive effect by further increasing their nutritional value. Thus, our study aimed to determine the changes in mineral nutrients contents of Brassicaceae microgreens depending on different blue–red (B:R) light ratios in light-emitting diode (LED) lighting and to evaluate their growth and nutritional value according to different indexes. Experiments were performed in controlled environment growth chambers at IH LRCAF, 2020. Microgreens of mustard (Brassica juncea ‘Red Lace’) and kale (Brassica napus ‘Red Russian’) were grown hydroponically under different B:R light ratios: 0%B:100%R, 10%B:90%R, 25%B:75%R, 50%B:50%R, 75%B:25%R, and 100%B:0%R. A 220 μmol m−2 s−1 total photon flux density (TPFD), 18 h photoperiod, 21/17 ± 2 °C temperature and 60% ± 5% relative humidity in the growth chamber were maintained during cultivation. We observed that an increasing percentage of blue light in the LED illumination spectrum during growth was associated with reduced elongation in the microgreens of both species and had a positive effect on the accumulation of mostly macro- and micronutrients. However, different B:R light ratios indicate a species-dependent response to changes in growth parameters such as leaf area, fresh and dry mass, and optical leaf indexes such as for chlorophyll, flavonol, anthocyanin, and carotenoid reflectance.

With the growing demand for quality food in the world, there is a new ambition to produce high-quality apples seeking reduced cultivation costs. The aim of this study was to evaluate the influence of agrotechnological tools on the quality of cv. Rubin apples during the harvest. The apple tree (Malus domestica Borkh.) cv. Rubin was grafted on dwarfing rootstocks P60, planted in single rows spaced 1.25 m between trees and 3.5 m between rows. Six agrotechnological tools were used—hand pruning, mechanical pruning, trunk incision, calcium-prohexadione, summer pruning and mechanical pruning one side, changing sides annually. The agrotechnical tools had no significant effect on accumulation of most sugars and elements, malic, folic and succinic acids in the fruits. Mechanical pruning resulted in significant accumulation of phenolic compounds, antioxidants, ascorbic acid, but reduced the amount of glucose compared to hand pruning. However, the trunk incision or spraying with ca-prohexadione together with mechanical pruning had no significant effect on sugar content but resulted in significantly higher amounts of phenols, antioxidants, ascorbic acid, Fe and Mn and reduced starch and citric acid.

Abstract: The Cancer and Leukemia Group B (CALGB), now part of the Alliance for Clinical Trials in Oncology, conducted a multicenter, single-arm, phase 2 study in patients ≥60 years with FMS-like tyrosine kinase 3 (FLT3)–mutated acute myeloid leukemia (AML). In this study, sorafenib was added to daunorubicin and cytarabine-based induction and consolidation chemotherapy and was also continued for 12 months of maintenance therapy. The primary end point of the study was overall survival (OS) at 1 year in the FLT3 internal tandem duplication (FLT3-ITD) cohort. Fifty-four patients with a median age of 67 years (range, 60.3-82.7 years) were enrolled; 39 were FLT3-ITD patients (71%) and 15 were FLT3-TKD (29%) patients. The observed 1-year OS (95% confidence interval [CI]) was 62% (45%-78%) for the FLT3-ITD patients (meeting the primary end point 62% vs 30% for a historical control group, P < .0001) and 71% (42%-92%) for the FLT3-TKD patients. The median disease-free survival and OS were 12.2 months (95% CI, 5-16.9) and 15.0 months (95% CI, 10.4-20.1), respectively, in the FLT3-ITD group and 9.6 (95% CI, 1.9 to not available [NA]) and 16.2 months (95% CI, 5.0 to NA) for the FLT3-TKD group. This study suggests that the addition of sorafenib to chemotherapy for FLT3-ITD AML is feasible and may improve the survival of older adults with FLT3-mutated AML. One of the most important factors affecting photosynthesis and metabolism is light absorbance by leaves and penetration through the canopy. The aim of this study was to evaluate the influence of planting density and tree development stages on photosynthetic activity, photosynthetic pigments, and carbohydrates in apple (Malus domestica Borkh.) trees in a combined way. The apple tree, Auksis, was grafted on dwarfing rootstock P 22. Space between rows was 3 m, trees were planted in 2001 in four distances: 0.25 m, 0.50 m, 0.75 m, and 1.00 m. Measurements and leaf samples were taken in the end of May (leaves fully expanded BBCH 20–25), in the middle of July (beginning of apple maturity BBCH 73–75) and at the end of August (harvest time BBCH 87–88) according BBCH—growth stages. Photosynthetic rate was significantly the lowest in the spring and tended to rise until fruit ripening, when it increased up to 19.4% compared to spring. Significantly the highest chlorophyll b and carotene α and β contents were found at the BBCH 73–75. The lowest levels of fructose and sorbitol in leaves were found at BBCH 73–75. The amount of starch accumulated in the leaves increased three times in summer compared to spring. Reduced distance between trees to four times (from 1 m to 0.25 m) showed clear competitive stress, as the decrease of photosynthetic rate (up to 36.4–38.6%) and total starch (up to 37–53%) was observed. The photosynthetic behaviour of apple trees was significantly affected by the development stage during the particular season which is related with physiological changes of metabolites transport and their distribution during fruit ripening and leaf senescence.

In this study, we sought to evaluate and compare the effects of constant and dynamic lighting on red and green leaf lettuce (Lactuca sativa L. Red Cos and Lobjoits Green cos) cultivated in a controlled environment. Plants were illuminated with the combination of red 662 and 638 nm, blue 452 nm, and far-red 737 nm at 16 h photoperiod and constant daily light integral (DLI) of each component. Five constant or dynamic lighting treatments were performed: (BR) constant flux of both B452 and R662; (B*R) constant flux of R662, but the DLI of B452 condensed in 8 h in the middle of photoperiod doubling the PPFD of blue light; (BR*) constant flux of B452, but the DLI of R662 light condensed in the middle of photoperiod; (BdynR) constant flux of R662, but the flux of B452 varies in the sinusoidal profile during 16 h photoperiod, imitating diurnal increase and decrease in lighting intensity; and (BRdyn) constant flux of B452, but the flux of R662 varies in sinusoidal profile. The lettuce’s response to dynamic lighting strategies was cultivar specific. Dynamic lighting strategies, mimicking natural lighting fluctuations, did not have a remarkable effect on photosynthesis and antioxidative parameters, but the dynamic flux of blue light component had a pronounced effect on higher macro and microelement contents in lettuce leaves.

The objective of this study was to evaluate how different UV-A wavelengths influence the morphology and photosynthetic behavior of red-leaf lettuce (Lactuca sativa L. cv. Maiko). In the experiments, the main photosynthetic photon flux consisted of red (R) and blue (B) light, supplemented with equal doses of different UV-A wavelengths (402, 387 and 367 nm). Treating the crops with low dosages of specific narrow-band UV-A radiation at key points in the life cycle initiated a cascade of responses in the above-ground biomass. According to the results, red-leaf lettuces acclimated to longer UV-A wavelengths by increasing biomass production, whereas different UV-A wavelengths had no significant effect on plant senescence reflectance, nor on the normalized difference vegetation index. A significant decrease in the maximum quantum yield of the PSII photochemistry of dark (Fv/Fm) and light (ΦPSII) adapted plants was observed. A lack of significant changes in non-photochemical fluorescence quenching indicates that photo-inhibition occurred under RBUV367, whereas the photosynthetic response under RB, RBUV402, and RBUV387 suggests that there was no damage to PSII. The correlation of the photosynthetic rate (Pr) with the stomatal conductance (gs) indicated that the increase in the Pr of lettuce under supplemental UV-A radiation was due to the increase of gs, instead of the ratio of the intracellular to ambient CO2 content (Ci/Ca) or stomatal limitations.

BackgroundBiochemical adrenal insufficiency induced by glucocorticoid treatment is prevalent, but data on the clinical implications are sparse. We investigated clinical consequences of glucocorticoid-induced adrenal insufficiency after oral glucocorticoid cessation.MethodsWe conducted a Danish population-based self-controlled case series utilizing medical registries. In this design each individual serves as their own control allowing event rates to be compared as a function of time and treatment. Clinical indicators of adrenal insufficiency were defined as diagnoses of gastrointestinal symptoms, hypotension, cardiovascular collapse, syncope, hyponatremia, and hypoglycaemia. We included 286,680 persons who discontinued long-term (≥ 3 months) oral glucocorticoid treatment. We defined five risk periods and a reference period (before treatment): period 0 (on treatment), withdrawal period (1 month before and after cessation), followed by three consecutive 2 month-risk periods after withdrawal (periods 2-4).ResultsMedian age at cessation was 69 years and 57% were female. Median treatment duration was 297 days and median cumulative dose was 3000 mg prednisolone equivalents. The incidence rates of hypotension, gastrointestinal symptoms, hypoglycemia and hyponatremia were increased in the withdrawal period compared to before treatment started (reference period). Incidence rate ratios comparing the withdrawal period with the reference period were 2.5 [95% confidence interval (CI): 1.4-4.3] for hypotension, 1.7 (95% CI: 1.6-1.9) for gastrointestinal symptoms, 2.2 (95% CI: 0.7-7.3) for hypoglycemia, and 1.5 (95% CI: 1.1-2.0) for hyponatremia. During 7 months of follow up, the rates of hypotension and gastrointestinal symptoms remained elevated compared to the reference period. Risk factors included use of antibiotics, increasing average daily dose of glucocorticoids, cumulative dose, and age.ConclusionOral glucocorticoid withdrawal was associated with adverse outcomes attributable to adrenal insufficiency. Our study underscores the need for future research to establish evidence-based clinical guidance on management of patients who discontinue oral glucocorticoids.

Creative professional service firms experience difficulties in establishing healthy and sustainable business models, as they must reconcile the often competing value systems upon which the models are based. They continuously negotiate between professional values and beliefs and the firm’s commercial goals, resulting in struggles between identity and strategy. Adopting a work lens, this study investigates the reciprocal tensions between identity and strategy in 17 business model design workshops with members of architectural firms. Observational data show that practitioners collaboratively construct their business models around professional values, thereby strengthening organizational identity, but constraining innovation in their business models. The research contributes to the body of literature on business model design processes by articulating how professional aspects of identity enable and constrain practitioners in shaping and being shaped by their strategic actions and decisions.

Background and Purpose. Inpatient rehabilitation in countries other than the United States (US) has been described as a time where patients are often not engaged in intensive physical activity. The purpose of this case report is to explore the amount and intensity of physical activity provided in inpatient rehabilitation after stroke in the US. Methods. This study presents a case report of a person admitted to an inpatient rehabilitation unit after sustaining a first stroke. A customized data collection tracked type of activity, activity intensity and social interaction every 5 minutes during the rehabilitation day. Results. 74 percent of the day was spent in low intensity, often seated, physical activity; 14% of the day was spent resting or sleeping. Only 2.91% the day was spent in moderate or high intensity activity with a mobility focus. Conclusions. Consistent with other studies, this case report suggests a relatively low physical demand to rehabilitation delivered in inpatient stroke rehabilitation. This case begins to raise questions about optimized rehabilitation parameters for acute stroke rehabilitation.

Background This two-stage, multi-institutional, randomized phase 2 trial assessed the toxicity and response rate associated with two treatment schedules of the histone deacetylase inhibitor, vorinostat (suberoylanilide hydroxamic acid; SAHA) in patients with relapsed acute myeloid leukemia and in selected untreated patients with high-risk acute myeloid leukemia.Design and Methods Patients with relapsed or untreated acute myeloid leukemia who were not candidates for chemotherapy entered one of the two treatment arms. In both arms a total dose of 8400 mg of vorinostat was delivered in each 21-day cycle of treatment: in arm A the dose regimen was 400 mg daily whereas in arm B the dose regimen was 200 mg three times daily for 14 days followed by 1 week rest.Results Data from all 37 patients were used for the analyses. In arm A (n=15), the confirmed complete remission rate was 0% (95% CI, 0% to 23%); this arm was closed at the planned interim analysis. In arm B (n=22), the confirmed complete remission rate was 4.5% (1 response; 95% CI, 0.4% to 24%), with a duration of response exceeding 398 days. The median time to treatment failure in arm A was 42 days (95% CI, 26 to 57); although a minimum of four cycles of treatment were planned, 11 patients (79%) received no more than two cycles. The median time to treatment failure in arm B was 46 days (95% CI, 20 to 71); 13 patients (59%) received no more than two cycles of treatment.Conclusions Vorinostat monotherapy demonstrated minimal activity in this group of patients with acute myeloid leukemia. Therapy was discontinued in many patients before the planned four cycles had been administered, either because of failure of vorinostat to control the leukocyte count or patients’ and physicians’ preference. Future studies of vorinostat in acute myeloid leukemia should focus on combinations with other drugs with which it might interact pharmacodynamically. ClinicalTrials.gov Identifier: NCT00305773.
