Breast cancer is a leading cause of death in women and has scope for improvement in treatment stratification. We report the use of high contrast UCNP staining to distinguish different levels of HER2 expression in HER2 analyte control and HER2-positive breast cancer tissue biopsies. A contrast of 40 was found as compared to the negative control and 25 as compared to conventional DAB staining.

SAbPred is a server that makes predictions of the properties of antibodies focusing on their structures. Antibody informatics tools can help improve our understanding of immune responses to disease and aid in the design and engineering of therapeutic molecules. SAbPred is a single platform containing multiple applications which can: number and align sequences; automatically generate antibody variable fragment homology models; annotate such models with estimated accuracy alongside sequence and structural properties including potential developability issues; predict paratope residues; and predict epitope patches on protein antigens. Antibodies are a class of proteins vital in mediating immune responses in vertebrates. Their binding site is highly malleable, allowing them to bind virtually any antigen. The versatility of antibody binding sites has received much attention from the pharmaceutical industry, marking them out as the most important category of biopharmaceuticals. The development of antibodies which bind to a specific antigen has thus far been achieved by costly and time-consuming experimental screening campaigns. However, in recent years computational approaches to antibody design have started to emerge, which offer an alternative. Computational antibody design techniques focus on determination of the binding site on the antibody, antibody-modelling, antibody-antigen docking and prediction of the binding site on the antigen. Here, we explore aspects of computational antibody design with the aim of gaining a better understanding of antibody-antigen interactions and improving existing artificial antibody design tools. We start by demonstrating our structural antibody database which has become a primary resource for antibody structural information. This is followed by a detailed analysis of the antibodyantigen interactions. The information gathered from this analysis allowed us to create an antibody contact site prediction tool, Antibody i-Patch. This tool was then employed to develop a local antibody-antigen docking pipeline, which used knowledge of the binding site of the antigen. We then tackled the global antibody-antigen docking problem by developing EpiPred, antigen binding site predictor which was employed in our global antibody-antigen docking pipeline.

Background: The immunomagnetic separation technique is the basis of monocyte isolation and further generation of monocyte-derived dendritic cells. Objective: To compare the efficiency of monocyte positive and negative separation, concentration of beads, and their impact on generated dendritic cells. Methods: Monocytes were obtained using monoclonal antibody-coated magnetic beads followed the Ficoll-Paque gradient separation of mononuclear cell fraction from the peripheral blood of 6 healthy volunteers. CD14 expression was analyzed by flow cytometry. Results: The percentage of MDDCs generated from monocytes obtained by positive and negative selection was comparable (51.8 ± 15.0 and 46.7 ± 3.4, respectively; p=0.885). The median values for the number of MDDCs obtained from monocytes after positive selection (3.9 × 106) and for MDDCs obtained from monocytes after negative selection (3.1 × 106) were comparable (p=0.194). The use of the recommended or half of the amount of beads for both types of separation had no significant influence on the percentage of isolated cells. Conclusions: Both types of magnetic separation including recommended and reduced concentrations of beads did not affect the yield and the purity of monocytes and their surface CD14 expression. However, DCs originated from the “positively” separated monocytes had noticeable higher expression of CD80.

Systematyczna aktywność fizyczna wpływa korzystnie na organizm i pomaga w uzyskiwaniu wartości potęgujących zdrowie. Wśród potencjalnych wartości wymieniane są np. wzrost wydolności fizycznej organizmu, wystąpienie objawów wagotonii takich jak zwolnienie spoczynkowej częstości skurczów serca, czy oszczędnej pracy układu oddechowego, utrzymanie pożądanych stężeń hemoglobiny we krwi, obniżanie nadmiernej masy ciała i zawartości podskórnej tkanki tłuszczowej, a także siły mięśni stabilizujących kręgosłup [Bouchard i wsp., 2007].

Szkoła wyższa jest niezależną jednostką, określającą program nauczania i zakres realizowanych zadań edukacyjnych. Decyduje także o rozwoju kultury fizycznej na uczelni, a do jej zadań należy stworzenie odpowiednich warunków. Aktualnie system szkolnictwa wyższego potwierdza konieczność podejmowania działań mających na celu ustawiczne podnoszenie jakości kształcenia. Wychodząc z takiego założenia Centrum Kultury Fizycznej wychodzi naprzeciw oczekiwań studentów i dokonuje zmian w organizacji zajęć wychowania fizycznego w Uniwersytecie Marii Curie Skłodowskiej w Lublinie przygotowując ofertę zajęć dydaktycznych.

Systematyczna aktywność fizyczna daje wiele korzyści zdrowotnych, zmniejsza ryzyko wystąpienia chorób układu krążenia, chorób metabolicznych i niektórych chorób nowotworowych, a także poprawia jakość życia [Bouchard i wsp., 2007; Chomistek i wsp., 2011; Haskell i wsp., 2007; Mitchell i Barlow, 2011]. Odpowiednia dawka aktywności fizycznej, oprócz zdrowego żywienia, jest niezbędna dla utrzymania optymalnych wielkości wskaźników fizjologicznych charakteryzujących zdrowie, zwłaszcza wydolności fizycznej, składu ciała i profilu lipidowego.

У статті проаналізовано інтенсивне дослідження творчості Лесі Українки на польському ґрунті. У полі зору науковців постає сакрум як потреба віднайдення сенсу буття. Доведено, що у творах Лесі Українки сакрум не пов’язаний з поняттям «божественний», а взяті до уваги ідеї розширеного сприйняття і розуміння цього світу. The intensive research work of Lesya Ukrainka on polish basis are analyzed in this article. In the field of view of scholars the sakrum arises as need to infer meaning of life. Are proved, that sakrum in Lesya Ukrainka’s creatives doesn’t connect with notion of “divine”, but are taken into account the ideas of advanced perception and understanding of this world.
