In today’s trauma surgery the treatment of critical sized bone defects can cause different problems, such as stability issues and impaired healing processes. The defects often have an irregular shape and result from inflammation, tumor resection or trauma. Additive manufacturing technologies can support the filling and healing process of such critical sized bone defects. With that in mind the aim of our project is, to build a biocompatible scaffold with a spongiosa like structure using Digital-Light-Processing (DLP) technology. This printing approach has a lot of advantages compared to other printing technologies. DLP with the printer used (Lumen X, Cellink) enables to print up to a resolution of 50 micrometer allowing to build scaffolds with high and biomimetic details. Further, this technology is useful to integrate cells directly into the printing process, because of the low cytotoxicity of the applied wavelength (405nm) in combination with biocompatible BioInk´s. In our study we established a biocompatible BioInk composed of Gelatine-Methacryloyl (GelMa) a photoinitiator and a photoabsorber. We were able to show the increased biocompatibility compared to commercially available alternatives by analyzing the cellular proliferation and differentiation of mesenchymal stem cells (MSC´s) and MG63 (osteosarcoma cell line) encapsulated into the BioInk. The scaffolds printed by the established BioInk had more than four times higher DNA content after 7 days compared to the commercial GelMa BioInk alternative, indicating cellular proliferation. Confocal laser scanning imaging of the different scaffolds underlined these results. Cells in the commercial alternative BioInk stayed round in their shape while cells in self-made BioInk showed an elongated morphology after staining with Hoechst and Phalloidin TRITC and survived up to over 2 weeks. The occurrence of neuroactive chemicals in the aquatic environment is on the rise and poses a potential threat to aquatic biota of currently unpredictable outcome. In particular, subtle changes caused by these chemicals to an organism's sensation or behavior are difficult to tackle with current test systems that focus on rodents or with in vitro test systems that omit whole-animal responses. In recent years, the zebrafish (Danio rerio) has become a popular model organism for toxicological studies and testing strategies, such as the standardized use of zebrafish early life stages in the Organisation for Economic Co-operation and Development's guideline 236. In terms of neurotoxicity, the zebrafish provides a powerful model to investigate changes to the nervous system from several different angles, offering the ability to tackle the mechanisms of action of chemicals in detail. The mechanistic understanding gained through the analysis of this model species provides a good basic knowledge of how neuroactive chemicals might interact with a teleost nervous system. Such information can help infer potential effects occurring to other species exposed to neuroactive chemicals in their aquatic environment and predicting potential risks of a chemical for the aquatic ecosystem. In the present article, we highlight approaches ranging from behavioral to structural, functional, and molecular analysis of the larval zebrafish nervous system, providing a holistic view of potential neurotoxic outcomes. Das humane Cytomegalovirus (HCMV) kann bei immunsupprimierten Patienten zu lebensbedrohlichen Organmanifestationen führen und ist weltweit die wichtigste kongenitale Virusinfektion. Die am häufigsten gegen HCMV eingesetzten Medikamente sind Ganciclovir, Cidofovir und Foscarnet. Durch eine frühe antivirale Therapie kann die Wahrscheinlichkeit einer klinisch manifesten Erkrankung reduziert werden, jedoch steigt mit der Therapiedauer auch die Wahrscheinlichkeit einer Virostatikaresistenzentwicklung. Die molekulare Ursache der Resistenzen gegen Ganciclovir sind Mutationen in der UL97- Serin-Threonin- Phosphotransferase, Mutationen in dem viralen Polymerasegen UL54 können zu Resistenzen gegen Ganciclovir, Cidofovir und Foscarnet führen. Heutzutage ist die Resistenztestung der Wahl die genotypische Empfindlichkeitstestung, da diese innerhalb von Stunden durchgeführt werden kann. Diese Methode erfordert allerdings, dass die Mutationen zuvor durch eine phänotypische Resistenztestung charakterisiert wurden, da nur so zwischen resistenzauslösenden Mutationen und einfachen Polymorphismen unterschieden werden kann. In dieser Arbeit wurden mittels der „en-passant“ BAC-Technologie (Tischer et al., 2006) die von Houldcroft et al. neu entdeckten Mutationen UL97 S512Stop, UL97 C480F, UL97 I429F und UL54 P522Q, Kombinationen aus Mutationen in beiden Genen, nämlich UL54 P522Q / UL97 C480F, sowie die von Sedlak et al. neu gefundene Kombination aus zwei schon zuvor einzeln charakterisierten Mutationen UL54 V715 / UL97 L595S generiert und anschließend funktionell charakterisiert (Houldcroft et al., 2016, Hall Sedlak et al., 2013). Eine Transfektion der verschiedenen Mutationen auf die Zellkultur war bei allen Mutationen erfolgreich, außer bei der UL97 Mutation S512Stop. Diese Mutation konnte auch bei dem ursprünglichen Patienten nur ein einziges Mal nachgewiesen werden (Houldcroft et al., 2016), sodass es wahrscheinlich ist, dass die Mutation UL97 S512Stop die Replikation von HCMV so stark einschränkt, dass ein weiteres Wachstum verhindert wird. Da das Wachstumsverhalten einen großen Einfluss auf die Behandlung von HCMV haben kann, wurden die Einzel- und Doppelmutationen mit Hilfe von Ein-Schritt-Wachstumszyklen untersucht. Hierbei zeigte sich eine mögliche Wachstumshemmung der UL97-Mutation I429F. Ob die einzelnen Mutationen und Doppelmutationen, die in dieser Arbeit charakterisiert werden sollten, resistenzauslösend sind, wurde über Plaquereduktionsassays bestimmt. Hierbei zeigten sich die Einzelmutationen UL54 P522Q und UL97 C480F jeweils sensibel gegenüber von Ganciclovir, Cidofovir und Foscarnet. Die Doppelmutation UL54 P522Q / UL97 C480F zeigte jedoch eine Ganciclovir- und möglicherweise auch eine Foscarnetresistenz. Dieses Phänomen sollte in weiteren Arbeiten genauer untersucht werden. Die UL97 Mutation I429F bewirkte eine Ganciclovirresistenz. Die Einzelmutation UL54 V715M wurde schon von Cihlar et al. als foscarnetresistent beschrieben (Cihlar et al., 1998). Bei UL97 L595S handelt es sich um eine der häufigsten Mutationen, die eine Ganciclovirresistenz verursachen (Wolf et al., 1995). In der Kombination wurden die beiden Mutationen in dieser Arbeit beschrieben und wie zu erwarten, zeigte auch die Doppelmutation eine Ganciclovir- und eine Foscarnetresistenz.
