We investigate the effects of an isolated meso-γ-scale surface heterogeneity for roughness and albedo on the atmospheric boundary-layer (ABL) height, with a case study at a semi-arid forest surrounded by sparse shrubland (forest area: 28km2, forest length in the main wind direction: 7 km). Doppler lidar and ceilometer measurements at this semi-arid forest show an increase in the ABL height over the forest compared with the shrubland on four out of eight days. The differences in the ABL height between shrubland and forest are explained for all days with a model that assumes a linear growth of the internal boundary layer of the forest through the convective ABL upwind of the forest followed by a square-root growth into the stable free atmosphere. For the environmental conditions that existed during our measurements, the increase in ABL height due to large sensible heat fluxes from the forest (600Wm-2 in summer) is subdued by stable stratification in the free atmosphere above the ABL, or reduced by high wind speeds in the mixed layer.

5. Turbulent transport of energy across a forest and a semiarid shrubland

The role of secondary circulations has recently been studied in the context of well-defined surface heterogeneity in a semiarid ecosystem where it was found that energy balance closure over a desert–forest system and the structure of the boundary layer was impacted by advection and flux divergence. As a part of the CliFF (Climate feedbacks and benefits of semi-arid forests, a collaboration between KIT, Germany, and the Weizmann Institute, Israel) campaign, we studied the boundary layer dynamics and turbulent transport of energy corresponding to this effect in Yatir Forest situated in the Negev Desert in Israel. The forest surrounded by small shrubs presents a distinct feature of surface heterogeneity, allowing us to study the differences between their interactions with the atmosphere above by conducting measurements with two eddy covariance (EC) stations and two Doppler lidars. As expected, the turbulence intensity and vertical fluxes of momentum and sensible heat are found to be higher above the forest compared to the shrubland. Turbulent statistics indicative of nonlocal motions are also found to differ over the forest and shrubland and also display a strong diurnal cycle. The production of turbulent kinetic energy (TKE) over the forest is strongly mechanical, while buoyancy effects generate most of the TKE over the shrubland. Overall TKE production is much higher above the forest compared to the shrubland. The forest is also found to be more efficient in dissipating TKE. The TKE budget appears to be balanced on average both for the forest and shrubland, although the imbalance of the TKE budget, which includes the role of TKE transport, is found to be quite different in terms of diurnal cycles for the forest and shrubland. The difference in turbulent quantities and the relationships between the components of TKE budget are used to infer the characteristics of the turbulent transport of energy between the desert and the forest.

Simulating the influence of heterogeneous surfaces on atmospheric flow using mesoscale models (MSM) remains a challenging task, as the resolution of these models usually prohibits resolving important scales of surface heterogeneity. However, surface heterogeneity impacts fluxes of momentum, heat, or moisture, which act as lower boundary conditions for MSM. Even though several approaches for representing subgrid-scale heterogeneities in MSM exist, many of these approaches rely on Monin–Obukhov similarity theory, preventing those models from resolving all scales of surface heterogeneity. To improve upon these residual heterogeneity scales, a novel heterogeneity parameterization is derived by linking the heterogeneous covariance function in spectral space to an associated homogeneous one. This covariance function approach is subsequently used to derive a parameterization of the aerodynamic resistance to heat transfer of the surface layer. Here, the effect of surface heterogeneity enters as a factor applied to the stability correction functions of the bulk similarity approach. To perform a first comparison of the covariance function approach against the conventional bulk similarity and tile approaches, large-eddy simulations (LESs) of distinct surface heterogeneities are conducted. The aerodynamic resistances from these three parameterizations are subsequently tested against the LES reference by resolving the surface heterogeneities with six different test-MSM grids of varying cell dimension. The results of these comparisons show that the covariance function approach proposed here yields the smallest deviations from the LES reference. In addition, the smallest deviation of the covariance function approach to the reference is observed for the LES with the largest surface heterogeneity, which illustrates the advantage of this novel parameterization.

Positron emission tomography (PET) and prompt gamma (PG) detection are promising proton therapy monitoring modalities. Fast calculation of the expected distributions is desirable for comparison to measurements and to develop/train algorithms for automatic treatment error detection. A filtering formalism was used for positron-emitter predictions and adapted to allow for its use for the beamline of any proton therapy centre. A novel approach based on a filtering formalism was developed for the prediction of energy-resolved PG distributions for arbitrary tissues. The method estimates PG yields and their energy spectra in the entire treatment field. Both approaches were implemented in a research version of the RayStation treatment planning system. The method was validated against PET monitoring data and Monte Carlo simulations for four patients treated with scanned proton beams. Longitudinal shifts between profiles from analytical and Monte Carlo calculations were within -1.7 and 0.9 mm, with maximum standard deviation of 0.9 mm and 1.1 mm, for positron-emitters and PG shifts, respectively. Normalized mean absolute errors were within 1.2 and 5.3%. When comparing measured and predicted PET data, the same more complex case yielded an average shift of 3 mm, while all other cases were below absolute average shifts of 1.1 mm. Normalized mean absolute errors were below 7.2% for all cases. A novel solution to predict positron-emitter and PG distributions in a treatment planning system is proposed, enabling calculation times of only a few seconds to minutes for entire patient cases, which is suitable for integration in daily clinical routine.

The role of secondary circulations has recently been studied in the context of well-defined surface heterogeneity in a semiarid ecosystem where it was found that energy balance closure over a desert–forest system and the structure of the boundary layer was impacted by advection and flux divergence. As a part of the CliFF (“Climate feedbacks and benefits of semi-arid forests”, a collaboration between KIT, Germany, and the Weizmann Institute, Israel) campaign, we studied the boundary layer dynamics and turbulent transport of energy corresponding to this effect in Yatir Forest situated in the Negev Desert in Israel. The forest surrounded by small shrubs presents a distinct feature of surface heterogeneity, allowing us to study the differences between their interactions with the atmosphere above by conducting measurements with two eddy covariance (EC) stations and two Doppler lidars. As expected, the turbulence intensity and vertical fluxes of momentum and sensible heat are found to be higher above the forest compared to the shrubland. Turbulent statistics indicative of nonlocal motions are also found to differ over the forest and shrubland and also display a strong diurnal cycle. The production of turbulent kinetic energy (TKE) over the forest is strongly mechanical, while buoyancy effects generate most of the TKE over the shrubland. Overall TKE production is much higher above the forest compared to the shrubland. The forest is also found to be more efficient in dissipating TKE. The TKE budget appears to be balanced on average both for the forest and shrubland, although the imbalance of the TKE budget, which includes the role of TKE transport, is found to be quite different in terms of diurnal cycles for the forest and shrubland. The difference in turbulent quantities and the relationships between the components of TKE budget are used to infer the characteristics of the turbulent transport of energy between the desert and the forest.

Der Einfluss von Oberflächenheterogenität auf die Wechselwirkungen zwischen Oberfläche und Atmosphäre ist nicht nur für Messungen des turbulenten Austausches, sondern auch für Wetter- und Klimasimulationen von signifikanter Bedeutung, da Oberflächenheterogenitäten mit der atmosphärischen Turbulenz auf verschiedenen Skalen wechselwirken. Während die Anzahl an numerischen Studien zu atmosphärischen Flüssen über heterogenen Oberflächen stetig zunimmt, existiert nur eine geringe Zahl an analytischen Theorien, was durch die nichtlineare Natur der zugrundeliegenden Gleichungen begründet ist. In dieser Dissertation wurden Wechselwirkungen von heterogenen Oberflächen mit der Atmosphäre unter Zuhilfenahme von analytischen und numerischen Methoden auf verschiedenen Skalen untersucht, wobei ,,large-eddy'' Simulationen (LES) für den numerischen Teil Verwendung fanden. Die erste Untersuchung bezog sich auf die mikro-$\gamma$ skalige Heterogenität von windzugewandten Vegetationskanten, die in einer analytische Lösung für den mittleren atmosphärischen Fluss durch eine solche Kante in neutraler Schichtung resultierte. Um die fehlenden Modellparameter (Integrationskonstanten) zu bestimmen, und um das Modell gegen LES zu testen, wurden mehrere Simulationen mit variierten Vegetationslängenskalen durchgeführt. Des Weiteren wurde das neu entwickelte Modell mit dem analytischen Modell von Belcher et al. [J. Fluid. Mech., 44:479-504, 2003] verglichen. Dieser Vergleich zeigte, dass das Modell von Belcher et al. (2003) hauptsächlich für Vegetation von geringer Dichte angewandt werden kann, während das neu entwickelte Modell die besten Ergebnisse für Vegetation erzielte, die dicht genug war um eine vollständige Anpassung des atmosphärischen Flusses zu bewirken. Neben atmosphärischen Wechselwirkungen von mikro-$\gamma$ skaligen Heterogenitäten, wurde mit dem semiariden Yatirwald auch eine ausgeprägte meso-$\gamma$ skalige Heterogenität unter Zuhilfenahme von detaillierten LES untersucht. Diese Simulationen fanden bei der Bestimmung von Auftreten, Position und Stärke von Sekundärzirkulationen Verwendung, welche eine entscheidende Rolle für den atmosphärischen Austausch des semiariden Ökosystems spielen könnten. Die numerische Studie resultierte in einem Auftreten von Sekundärzirkulationen in allen drei Fällen von unterschiedlicher atmosphärischer Stabilität (schwach konvektiver, leicht konvektiver und stark konvektiver Fall). Obwohl die horizontale Ausdehnung der Zirkulationen zu klein war um den ganzen Wald mit dem umgebenden Buschland zu koppeln, zeigten Untersuchungen des aerodynamischen Widerstandes für Wärmeaustausch, dass die Sekundärzirkulationen die atmosphärischen Wechselwirkungen in Regionen mit Aufwind verstärkten und in Regionen mit Abwind abschwächten. Da die zuvor genannten Mechanismen direkt mit der Heterogenität der Oberfläche verknüpft sind, können sie nicht durch homogenen Parametrisierungen beschrieben werden. Dennoch ist die Parametrisierung von heterogenen Oberflächen für atmosphärische Simulationen unter Verwendung von mesoskaligen Modellen wichtig, da die Rasterung dieser Modelle zu grob ist um viele wichtige Skalen der Oberflächenheterogenität aufzulösen. Um auch diese Skalen zu berücksichtigen, wurde eine Parametrisierung des aerodynamischen Widerstandes für heterogene Oberflächen entwickelt, was den dritten Teil dieser Dissertation darstellt. Die analytische Herleitung der Parametrisierung basiert auf der Verknüpfung der Kovarianzfunktion des eigentlichen, heterogenen, Szenarios mit der Kovarianzfunktion eines zugehörigen, homogenen, Szenarios im Spektralraum. Unter Annahmen über die Form der Turbulenzspektren des homogenen Szenarios, und unter Vernachlässigung von Advektion und dispersiven Flüssen, wurden Korrekturfaktoren für den aerodynamischen Bulkwiderstand bestimmt. Des weiteren zeigte ein Vergleich des Kovarianzfunktionsansatzes mit den konventionellen ,,bulk'' und ,,tile'' Methoden für drei Szenarien von idealisierten Oberflächenheterogenitäten, dass der Kovarianzfunktionsansatz die kleinsten Abweichungen von der LES Referenz besitzt. Dieses Ergebnis demonstriert den Vorteil einer Verwendung der neuen Methode in mesoskaligen Modellen.

The difference of turbulent quantities and the relationships between the components of TKE budget are used to infer the characteristics of turbulent transport of energy between the desert and the forest.

Henoch-Schoenlein purpura (HSP) is a generalized anaphylactoid "teucocytoclastic" vasculitis, which in more than 50% of patients is complicated by gastrointestinal symptoms such as vomiting, abdominal pain, melena and life-threatening bleedings. In five consecutive patients with HSP on admission factor XIII (F XIII) was decreased to 8-58% of normal. In acute attacks an enormous F XIII consumption takes place. Recurrent release of elastase from inflammatory cells (PMNL, monocytes, macrophages) can be detected by intermittent peaks of elastase-α 1-protease inhibitor (ELP-α1-PI) complex in plasma (peak values 450-950 ng/mi). Elastase is considered to cause specific F XIII degradation with bleeding into the intestinal wall as a result. High-dose F XIII substitution therapy is effective in controlling bleeding complications and abdominal pain. The benefit of F XIII therapy could be demonstrated in a rapid and marked shrinking of the intramural hematomas by raltrascanic scan. Key Words: Henoch-Schoenlein purpura-Factor XIII—Elastase.

Congenital factor XIII deficiency is a rare disease, but has provided valuable information on the physiological role of factor XIII and the benefit of factor XIII replacement therapy. It could be shown that not only homozygous patients but also heterozygotes are at risk for bleeding complications. Acquired factor XIII deficiency, however, is much more common, and preliminary studies suggest a lack of factor XIII to be an important feature of various diseases. In acute states and severe hemorrhages, replacement therapy with factor XIII concentrates is recommended. Recent progress in assay methods and future clinical studies should help to evaluate the therapeutic potential of factor XIII.

Seasonal changes of ascorbate peroxidase and monodehydroascorbate radical reductase activities were studied in foliar tissues of Norway spruce (Picea abies L.). In mature needles, APX activities did not show seasonal fluctuations and were similar to those found in resting buds. Monodehydroascorbate radical reductase activity was higher in needles than in buds and higher in winter than in summer. Maximum activities of both enzymes were found before bud break and minimum activities in newly formed needles. When spruce seedlings were exposed to an artifical frost event of - 5 deg C for one night in spring, ascorbate peroxidase activity declined in young needles before the onset of visible injury but corresponding to a sudden upsurge in lipid peroxidation. After one week, some shoots showed severe symptoms of injury, some were slightly injured and others did not show any visible injury. In lethally injured needles, antioxidative protection (ascorbate peroxidase, monodehydroascorbate radica l reductase, glutathione reductase, glutathione, ascorbate, superoxide dismutase) had collapsed. Surviving needles showed a coordinated increase in all components of the antioxidative system suggesting an efficient induction of defense systems. However, enhanced protection was observed only transiently. In fall, needles that had been exposed to frost in spring contained significantly less antioxidants than unstressed needles indicating that unseasonal frost caused memory effects.

Häufigster Auslöser für erworbene FXIII-Hemmkörper, die eine defekte Fibrinvernetzung verursachen, ist die Langzeitanwendung bestimmter Medikamente. Die Blutungssymptomatik zeigt das gesamte Spektrum des schweren FXIII-Mangels. Sofern ein Medikament als Ursache des Hemmkörpers in Frage kommt, ist dieses sofort abzusetzen. Weiterhin sollte versucht werden, durch hochdosierte Gabe eines FXIII-Konzentrates den Inhibitor zu überspielen und damit eine Blutstillung zu erzielen

ZusammenfassungNur wenige Autoren berichteten bislang von einer disseminierten intravasalen Gerinnung (DIC) als Komplikation einer Heparin-assoziierten Thrombozytopenie (HAT). In der Literatur werden bis 1990 nur 14 Patienten beschrieben, die meisten von ihnen verstarben. Die HAT II und ihre Komplikationen entstehen nur bei Patienten, die einen Antikörper gegen Plättchenfaktor 4 oder den Plättchenfaktor-4-Heparin-Komplex entwickeln. Während sich der Fc-Anteil des Antikörpers oder der Antikörper-Plättchenfaktor-4-Komplex an die Plättchen bindet, kann der Plättchenfaktor 4 oder der Plättchenfaktor-4-Antikörper-Komplex auch mit Heparansulfat auf der Oberfläche von Endothelzellen reagieren. Eine solche Bindung auf der Endothelzelle kann eine generalisierte Endothelzellschädigung auslösen, die die Freisetzung von von-Willebrand-Faktor (Ristocetin-Kofaktor), Gewebeplasminogenaktivator (t-PA) und Gewebsaktivator zur Folge hätte. Letzterer kann FVII aktivieren, eine intravasale Thrombinbildung auslösen und so zu einer DIC als Komplikation einer HAT führen.Deshalb wurden bei vier Patienten mit HAT Typ II Parameter einer EndothelzellAktivierung (t-PA; Ristofact.), einer Gerinnungsaktivierung (Thrombin-Antithrombin-lll-Komplex, TAT; Prothrombinfragment F1+2, Prf F1+2) und einer Aktivierung des Fibrinolysesystems (Plasmin-α2-Antiplasmin, PAP) gemessen, zusätzlich auch der Antithrombin-Illund der Plasminogen-Wert zu Beginn der Thrombozytopenie. Zu Beginn und während der Thrombozytopenie waren t-PA, Ristofact., TAT, Prf F1+2 und PAP erhöht, während Plasminogen erniedrigt war und AT III sich im unteren Normalbereich bewegte. Im Gegensatz dazu war der ElastaseProteinaseinhibitor-Komplex, mit einer Ausnahme, niedrig. Diese Ergebnisse verdeutlichen, daß der zirkulierende Antikörper gegen Plättchenfaktor 4 nicht nur einen erhöhten Plättchenumsatz bewirkt, sondern auch noch die Endothelzellen sowie das Gerinnungsund Fibrinolysesystem aktiviert.

Two isozymes of superoxide dismutase (SOD; EC 1.15.1.1) were purified from Norway spruce (Picea abies L.) needles to apparent electrophoretic homogeneity. Purification factors were 354 for SOD I and 265 for SOD II. The native molecular mass of both purified enzymes was approximately 33 kD, as determined by gel filtration. The subunit molecular weights, as estimated by sodium dodecyl sulfate polyacrylamide gel electrophoresis, were 20,000 for SOD I and 16,000 for SOD II in the presence of 2-mercaptoethanol, and 15,800 and 15,000, respectively, in its absence. These results indicate that the native enzymes were homodimers whose subunits contained intrachain disulfide bonds. Isoelectric points determined by nondenaturing isoelectric focusing were 4.5 and 5.5 for SOD I and II, respectively. NH2-terminal sequence analysis of the first 22 to 23 amino acids revealed 70 to 75% sequence identity with chloroplastic CuZn SODs from other plant species for SOD I, and 75% sequence identity with the cytosolic CuZn SOD from Scots pine for SOD II. SOD I was the major activity in needles and it was associated with chloroplasts. SOD II activity was dominant in roots. peerReviewed

Dabei bewegt sich die Arbeit an der Schnittstelle zwischen der Freiraumplanung und den Sozial- und Kulturwissenschaften. Anhand von Beispielen aus dem öffentlichen Raum und von Industriebrachen wird dargestellt, wie und vor welchem Hintergrund Events gegenwärtig zur Konstitution und Transformation von Orten gezielt eingesetzt werden und in welchem Verhältnis diese Strategien zur Freiraumplanung stehen. These days organised events and performances influence the appearance and use of open spaces in a considerable way. The central question of the thesis is how and why events and performances have effects on open spaces and give them social significance beyond their limited duration. Therefore the thesis is situated between landscape architecture and social sciences. With examples of public spaces and of industrial brownfields it is described how and why events are used as effective strategies for constitution and transformation of space. Further on these examples point out the relationship between the strategies of event management and landscape architecture in view of the development of places.

Purpose: We describe and demonstrate a fast analytical tool for prompt-gamma emission prediction based on filter functions applied on the depth dose profile. We present the implementation in a treatment planning system (TPS) of the same algorithm for positron emitter distributions. Methods: The prediction of the desired observable is based on the convolution of filter functions with the depth dose profile. For both prompt-gammas and positron emitters, the results of Monte Carlo simulations (MC) are compared with those of the analytical tool. For prompt-gamma emission from inelastic proton-induced reactions, homogeneous and inhomogeneous phantoms alongside with patient data are used as irradiation targets of mono-energetic proton pencil beams. The accuracy of the tool is assessed in terms of the shape of the analytically calculated depth profiles and their absolute yields, compared to MC. For the positron emitters, the method is implemented in a research RayStation TPS and compared to MC predictions. Digital phantoms and patient data are used and positron emitter spatial density distributions are analyzed. Results: Calculated prompt-gamma profiles agree with MC within 3 % in terms of absolute yield and reproduce the correct shape. Based on an arbitrary reference material and by means of 6 filter functions (one permore » chemical element), profiles in any other material composed of those elements can be predicted. The TPS implemented algorithm is accurate enough to enable, via the analytically calculated positron emitters profiles, detection of range differences between the TPS and MC with errors of the order of 1–2 mm. Conclusion: The proposed analytical method predicts prompt-gamma and positron emitter profiles which generally agree with the distributions obtained by a full MC. The implementation of the tool in a TPS shows that reliable profiles can be obtained directly from the dose calculated by the TPS, without the need of full MC simulation.« less

36. Turbulent transport of energy across a forest and a semi-arid shrubland.

- Abstract
The role of secondary circulations has recently been studied in the context of well defined surface heterogeneity in a semi-arid ecosystem where it was found that energy balance closure over a desert-forest system and the structure of the boundary layer was impacted by advection and flux divergence. As a part of the CliFF (Climate Feedbacks and benefits of semi-arid forests, a collaboration between KIT, Germany and the Weizmann Institute, Israel) campaign, we studied the boundary layer dynamics and turbulent transport of energy corresponding to this effect in the Yatir forest situated in the Negev desert in Israel. The forest surrounded by small shrubs presents a distinct feature of surface heterogeneity, allowing us to study the differences between their interactions with the atmosphere above by conducting measurements with two EC stations and two Doppler LiDARs. As expected, the turbulence intensity and vertical fluxes of momentum and sensible heat are found to be higher above the forest compared to the shrubland. Turbulent statistics indicative of nonlocal motions are also found to differ over the forest and shrubland and also display a strong diurnal cycle. The production of turbulent kinetic energy (TKE) over the forest is strongly mechanical, while buoyancy effects generate most of the TKE over the shrubland. Overall TKE production is much higher above the forest compared to the shrubland. The forest is also found to be more efficient in dissipating TKE. The TKE budget appears to be balanced on average both for the forest and shrubland, although the imbalance of the TKE budget, which contains the role of TKE transport, is found to be quite different in terms of their variation with atmospheric stability and diurnal cycles for the forest and shrubland. The effect of very large mesoscale motions is also directly quantified following a recent formulation by Banerjee and Katul, 2013, using the measured longitudinal velocity variances and boundary layer heights. The difference of turbulent quantities and the relationships between the components of TKE budget are used to infer the characteristics of turbulent transport of energy between the desert and the forest. Seasonal changes of ascorbate peroxidase and monodehydroascorbate radical reductase activities were studied in foliar tissues of Norway spruce (Picea abies L.). In mature needles, APX activities did not show seasonal fluctuations and were similar to those found in resting buds. Monodehydroascorbate radical reductase activity was higher in needles than in buds and higher in winter than in summer. Maximum activities of both enzymes were found before bud break and minimum activities in newly formed needles. When spruce seedlings were exposed to an artifical frost event of –5°C for one night in spring, ascorbate peroxidase activity declined in young needles before the onset of visible injury but corresponding to a sudden upsurge in lipid peroxidation. After one week, some shoots showed severe symptoms of injury, some were slightly injured and others did not show any visible injury. In lethally injured needles, antioxidative protection (ascorbate peroxidase, monodehydroascorbate radical reductase, glutathione reductase, glutathione, ascorbate, superoxide dismutase) had collapsed. Surviving needles showed a coordinated increase in all components of the antioxidative system suggesting an efficient induction of defense systems. However, enhanced protection was observed only transiently. In fall, needles that had been exposed to frost in spring contained significantly less antioxidants than unstressed needles indicating that unseasonal frost caused memory effects.
