The study was performed according to the “AAMI/ESH/ISO Universal Standard (ISO 81060– 2:2018) protocol”. Subjects (n = 88) were recruited to fulfill the age, gender, BP and cuff distribution criteria according to the protocol requirements using the same arm sequential BP measurement method. Differences between observers’ mercury sphygmomanometer reference measurements and device BP values (test versus reference) and their standard deviation (SD) were calculated.Results: Ninety-one subjects were selected, 88 of whom were included. The mean BP differences between the simultaneous observers’ reference measurements were − 0.2 ± 1.7 mmHg for systolic BP (SBP) and − 0.2 ± 1.7 mmHg for diastolic BP (DBP). For validation criterion 1, the mean difference ± SD between the reference and device BP values were 3.6 ± 5.5 mmHg for SBP and 1.4 ± 4.5 mmHg for DBP. For criterion 2, the SD of the mean BP differences between the test device and reference BP per subject was 5/4.1 mmHg for SBP/DBP (≤ 5.9/6.8). These results fulfilled the protocol requirements.Conclusion: The Combei BP118A oscillometric device for home BP measurement fulfilled all of the requirements of the AAMI/ESH/ISO Universal Standard (ISO 81060– 2:2018) in general population and consequently can be recommended for home BP measurements.Keywords: blood pressure measurement, accuracy, validation, home blood pressure, oscillometric, Combei

Abstract

Although primary prostate cancer is largely curable, progression to metastatic disease is associated with very poor prognosis. E6AP is an E3 ubiquitin ligase and a transcriptional co-factor involved in normal prostate development. E6AP drives prostate cancer when overexpressed. Our study exposed a role for E6AP in the promotion of metastatic phenotype in prostate cells. We revealed that elevated levels of E6AP in primary prostate cancer correlate with regional metastasis and demonstrated that E6AP promotes acquisition of mesenchymal features, migration potential, and ability for anchorage-independent growth. We identified the metastasis suppressor NDRG1 as a target of E6AP and showed it is key in E6AP induction of mesenchymal phenotype. We showed that treatment of prostate cancer cells with pharmacological agents upregulated NDRG1 expression suppressed E6AP-induced cell migration. We propose that the E6AP-NDRG1 axis is an attractive therapeutic target for the treatment of E6AP-driven metastatic prostate cancer.

Abstract

BACKGROUND: The cyclin-dependent kinase inhibitor, p21, is well known for its role in cell cycle arrest. Novel anti-cancer agents that deplete iron pools demonstrate marked anti-tumor activity and are also active in regulating p21 expression. These agents induce p21 mRNA levels independently of the tumor suppressor, p53, and differentially regulate p21 protein expression depending on the cell-type. Several chelators, including an analogue of the potent anti-tumor agent, di-2-pyridylketone 4,4-dimethyl-3-thiosemicarbazone (Dp44mT), have entered clinical trials, and thus, their molecular mechanism of action is crucial to assess. Hence, this investigation examined how several iron chelators transcriptionally regulate p21. METHODS: Promoter-deletion constructs; luciferase assays; RT-PCR; western analysis; gene silencing; co-immunoprecipitation. RESULTS: The transcriptional regulation of the p21 promoter by iron chelators was demonstrated to be dependent on the chelator and cell-type examined. The potent anti-cancer chelator, Dp44mT, induced p21 promoter activity in SK-MEL-28 melanoma cells, but not in MCF-7 breast cancer cells. Further analysis of the p21 promoter identified a 50-bp region between -104 and -56-bp that was required for Dp44mT-induced activation in SK-MEL-28 cells. This region contained several Sp1-binding sites and mutational analysis of this region revealed the Sp1-3-binding site played a significant role in Dp44mT-induced activation of p21. Further, co-immunoprecipitation demonstrated that Dp44mT induced a marked increase in the interactions between Sp1 and the transcription factors, estrogen receptor-α and c-Jun. CONCLUSIONS AND GENERAL SIGNIFICANCE: Dp44mT-induced p21 promoter activation via the Sp1-3-binding site and increased Sp1/ER-α and Sp1/c-Jun complex formation in SK-MEL-28 cells, suggesting these complexes were involved in p21 promoter activation.

Abstract

Tumor metastasis is a critical clinical problem that requires urgent attention. Recently, there has been growing interest in the development of metastasis suppressor proteins as targets for anti-cancer therapy. N-myc downstream regulated protein 1 (NDRG1) is an important and well established metastasis suppressor protein that has shown promise as a significant therapeutic target to inhibit cancer metastasis. Notably, a novel series of thiosemicarbazone-based anti-cancer agents under development have shown the ability to up-regulate NDRG1. Therefore, further studies examining the effect of NDRG1 as a therapeutic target to prevent cancer metastasis are warranted.

Essential metals, such as iron and copper, play a critical role in a plethora of cellular processes including cell growth and proliferation. However, concomitantly, excess of these metal ions in the body can have deleterious effects due to their ability to generate cytotoxic reactive oxygen species (ROS). Thus, the human body has evolved a very well-orchestrated metabolic system that keeps tight control on the levels of these metal ions. Considering their very high proliferation rate, cancer cells require a high abundance of these metals compared to their normal counterparts. Interestingly, new anti-cancer agents that take advantage of the sensitivity of cancer cells to metal sequestration and their susceptibility to ROS have been developed. These ligands can avidly bind metal ions to form redox active metal complexes, which lead to generation of cytotoxic ROS. Furthermore, these agents also act as potent metastasis suppressors due to their ability to up-regulate the metastasis suppressor gene, N-myc downstream regulated gene 1. This review discusses the importance of iron and copper in the metabolism and progression of cancer, how they can be exploited to target tumors and the clinical translation of novel anti-cancer chemotherapeutics.

Abstract

The endoplasmic reticulum (ER) plays a major role in the synthesis, maturation and folding of proteins and is a critical calcium (Ca2+) reservoir. Cellular stresses lead to an overwhelming accumulation of misfolded proteins in the ER, leading to ER stress and the activation of the unfolded protein response (UPR). In the stressful tumor microenvironment, the UPR maintains ER homeostasis and enables tumor survival. Thus, a novel strategy for cancer therapeutics is to overcome chronically activated ER stress by triggering pro-apoptotic pathways of the UPR. Considering this, the mechanisms by which the novel anti-cancer agent, Dp44mT, can target the ER stress response pathways were investigated in multiple cell-types. Our results demonstrate that the cytotoxic chelator, Dp44mT, which forms redox-active metal complexes, significantly: (1) increased ER stress-associated pro-apoptotic signaling molecules (i.e., p-eIF2a, ATF4, CHOP); (2) increased IRE1a phosphorylation (p-IRE1a) and XBP1 mRNA splicing; (3) reduced expression of ER stress-associated cell survival signaling molecules (e.g., XBP1s and p58IPK); (4) increased cleavage of the transcription factor, ATF6, which enhances expression of its downstream targets (i.e., CHOP and BiP); and (5) increased phosphorylation of CaMKII that induces apoptosis. In contrast to Dp44mT, the iron chelator, DFO, which forms redox-inactive iron complexes, did not affect BiP, p-IRE1a, XBP1 or p58IPK levels. This study highlights the ability of a novel cancer therapeutic (i.e., Dp44mT) to target the pro-apoptotic functions of the UPR via cellular metal sequestration and redox stress. Assessment of ER stress-mediated apoptosis is fundamental to the understanding of the pharmacology of chelation for cancer treatment.

Abstract

Glutaminase activity in intact mitochondria from rat liver is activated by spermine, as indicated both by increased glutamate production from glutamine and by increased respiration with glutamine as sole substrate. Glutaminase activity assayed in membranes from frozen-thawed mitochondria, is activated by spermine about 6-fold at physiological concentrations of its other effectors (NH4+ at 0.7 mM, Pi 5 mM) and at pH 7.4. Spermine decreased the apparent Km for glutamine from 38 to 15 mM at 5 mM Pi, and increased the sensitivity of the enzyme for phosphate activation so that the concentration required for 50% stimulation decreased from 15 to 4 mM. Half-maximal spermine effects occurred at 0.15 mM, which is in the physiological range. Spermine was effective in the presence of physiological concentrations of Mg2+. We suggest that spermine may be a physiological activator of hepatic glutaminase.

Abstract

The study of iron chelators as anti-tumor agents is still in its infancy. Iron is important for cellular proliferation and this is demonstrated by observations that iron-depletion results in cell cycle arrest and also apoptosis. In addition, many iron chelators are known to inhibit ribonucleotide reductase, the iron-containing enzyme that is the rate-limiting step for DNA synthesis. Desferrioxamine is a well known chelator used for the treatment of iron-overload disease, but it has also been shown to possess anti-cancer activity. Another class of chelators, namely the thiosemicarbazones, have been shown to possess anti-cancer activity since the 1950's, although their mechanism(s) of action have only recently been more comprehensively elucidated. In fact, the redox activity of thiosemicarbazone iron complexes is thought to be important in mediating their potent cytotoxicity. Moreover, unlike typical iron chelators which simply act to deplete tumors of iron, several thiosemicarbazones (i.e., Bp44mT and Dp44mT) do not induce this effect, their anti-cancer efficacy being due to other mechanisms e.g., redox activity. Other reports have also shown that some thiosemicarbazones inhibit topoisomerase IIa, demonstrating that this class of agents have multiple molecular targets and act by various mechanisms. The most well characterized thiosemicarbazone iron chelator in terms of its assessment in humans is 3-aminopyridine-2-carboxaldehyde thiosemicarbazone (3-AP). Observations from these clinical trials highlight the less than optimal activity of this ligand and several side effects related to its use, including myelo-suppression, hypoxia and methemoglobinemia. The mechanisms responsible for these latter effects must be elucidated and the design of the ligand altered to minimize these problems and increase efficacy. This review discusses the development of chelators as unique agents for cancer treatment.

Abstract

Cancer is one of the leading causes of death worldwide and there is an increasing need for novel anti-tumor therapeutics with greater selectivity and potency. A new strategy in the treatment of cancer has focused on targeting an essential cell metabolite, iron (Fe). Iron is vital for cell growth and metabolism, forming a crucial component of the active site of ribonucleotide reductase (RR), the rate-limiting enzyme in DNA synthesis. Cancer cells in particular require large amounts of Fe to proliferate, making them more susceptible to the Fe deficiency caused by Fe chelators. Beginning with primordial siderophores, Fe chelators have since evolved to a new generation of potent and efficient anti-cancer agents. Recently, investigations have led to the generation of novel di-2-pyridylketone thiosemicarbazone (DpT) and 2-benzoylpyridine thiosemicarbazone (BpT) ligands that demonstrate marked and selective anti-tumor activity both in vitro and in vivo against a wide spectrum of tumors. The mechanism of action of these novel ligands includes alterations in the expression of key regulatory molecules as well as the generation of redox active Fe complexes. Interestingly, nonsynthetic Fe chelators including silybin and curcumin, both of which are derived from plants, also have a high potential in the treatment of cancer. This review explores the development of novel Fe chelators for the treatment of cancer and their mechanisms of action. © 2011 Bentham Science Publishers Ltd.

Abstract

Investigation of the metabolic status of cows in early lactation is the most important measure in assessing the health and productivity of cattle. Today, the analysis of metabolites, hormones and blood in the early lactation can be largely predicted health risk and of cows productivity. The daily work of veterinarians are available to a large number of markers in the blood that indicate the energy balance (NEFA, BHB, glucose), the balance of proteins (albumin, total protein, urea), the balance of ions (Ca, P, Mg), inflammatory response (blood pictures, inflammatory proteins), oxidative status (SOD, MDA), status of the liver (bilirubin, AST, ALT), hormonal (insulin, cortisol, IGF-I, STH, T3, T4) and hematology finding. In addition to the assessment of the metabolic status must be considered body condition of the cows and the amount of milk production. The aim of this study was to investigate the correlation between these parameters and to determine what parameters by whose determination can be seen overall metabolic status of cows. In this way we will make the extraction of information from large amounts of data, so we will focus only on those elements of the metabolic and endocrine profile that has the most reliable to describe the metabolic status of cows . In this way we will make the rationalization of laboratory testing metabolism cows. Blood was taken from 50 cows of the Holstein - Friesian breed. The concentration of the above- mentioned parameters were determined. They are made of the correlation matrix and then by extraction of the main factors using factor analysis. The results show that the extract may by three main factors. Factor 1 explains 61.7% of the variation, and it highly correlates with the values of insulin, IGF-I, STH, NEFA and glucose (metabolic stress). Factor 2 explains 20.2% of variation and on highly correlated with the BHB, AST, triglycerides and cholesterol (a liver). Factor 3, which explained 7% of the variation variables correlated with milk production in cows (productivity). [ABSTRACT FROM AUTHOR]
