There is a growing focus on understanding the role of the male microbiome in fertility issues. Although research on the bacterial communities within the male reproductive system is in its initial phases, recent discoveries highlight notable variations in the microbiome's composition and abundance across distinct anatomical regions like the skin, foreskin, urethra, and coronary sulcus. To assess the relationship between male genitourinary microbiome and reproduction, we queried various databases, including MEDLINE (available via PubMed), SCOPUS, and Web of Science to obtain evidence-based data. The literature search was conducted using the following terms "gut/intestines microbiome," "genitourinary system microbiome," "microbiome and female/male infertility," "external genital tract microbiome," "internal genital tract microbiome," and "semen microbiome." Fifty-one relevant papers were analyzed, and eleven were strictly semen quality or male fertility related. The male microbiome, especially in the accessory glands like the prostate, seminal vesicles, and bulbourethral glands, has garnered significant interest because of its potential link to male fertility and reproduction. Studies have also found differences in bacterial diversity present in the testicular tissue of normozoospermic men compared to azoospermic suggesting a possible role of bacterial dysbiosis and reproduction. Correlation between the bacterial taxa in the genital microbiota of sexual partners has also been found, and sexual activity can influence the composition of the urogenital microbiota. Exploring the microbial world within the male reproductive system and its influence on fertility opens doors to developing ways to prevent, diagnose, and treat infertility. The present work emphasizes the importance of using consistent methods, conducting long-term studies, and deepening our understanding of how the reproductive tract microbiome works. Introduction. The human body is constantly exposed to an extremely low electromagnetic field (ELF-EMF), in particular at 50 Hz, emitted by power lines, domestic distribution lines, electrical appliances, etc. It is assumed that the increase in electromagnetic exposure may cause adverse effects upon human health, as well as raising concerns regarding the impact on human fertility. Objective. The aim of this in vitro study was to investigate the influence of ELF-EMF with a frequency of 50 Hz on the motility of human sperm. At the same time, the effectiveness of the dielectric screen constructed by ADR Technology® in absorbing the emitted radiation was examined. Materials and method. Semen samples of 20 patients were exposed to the influence of an extremely low electromagnetic field. After 5, 15 and 30 min., spermatozoa motility was analysed using a computer-assisted spermatozoa motility analysis system. The following sperm motility parameters were examined: 1) velocity straight linear motility; 2) cross-beat frequency; 3) lateral head displacement; 4) homogeneity of progressive motility velocity. Results. It was found that the ELF-EMF presented a negative effect on the motility of human spermatozoa. A significant decrease in spermatozoa motility speed and a significant increase in lateral head deviation values were observed under the influence of the electromagnetic field. ELF-EMF did not show an effect on either lateral head displacement or homogeneity of progressive motility velocity. Conclusions. A positive effect of the dielectric screen ADR Technology® was found. Idiopathic scoliosis (IS) etiology remains unclear, but strong genetic background is suggested. Previously reported TIMP2 study indicates an association of genic rs8179090 with IS progression in a Han Chinese population. However, there has been a lack of investigation into intragenic TIMP2 polymorphisms in IS patients. We recruited 100 Caucasian females with IS and 100 controls. Patients were subdivided accordingly to: progression rate, curve severity, joint mobility, and curve pattern. Allele‐specific‐polymerase chain reaction based on fluorescence resonance energy transfer was applied to evaluate nine TIMP2 polymorphisms. Distribution of genotype and allele frequency in only one polymorphism (rs11658743) differed in case–control study. Four of the polymorphisms (rs2277700, rs11077401, rs2376999, and rs4789934) showed non‐equal distributions either in genotype or/and allele distributions in the patients of different progression rates. The rs11077401 was related to curve severity patients distinction and the rs8179090 distinguished patients with different joint mobility level. Two polymorphisms either differed statistically in case of curve patterns subgrouping (rs8068674 and rs8179090) or showed a slight tendency toward significance in the recessive model of allele distributions (rs9916809 and rs8179090). The remaining two polymorphisms (rs2377005, rs11658743) showed no association with either clinical or radiographic IS characteristics. The influence of the G allele of the rs8179090 on the clinical course of IS has not yet been confirmed. We identified four TIMP2 polymorphisms (rs11077401, rs2376999, rs2277700, and rs4789934) that were associated with a higher risk of the progressive IS form. Further genetic association studies based on suggested clinical criteria would be necessary to validate TIMP2 polymorphisms associated with the curve progression. © 2019 Orthopaedic Research Society. Introduction. Human ghrelin is the endogenous ligand of the growth hormone secretagogue receptor type 1a (GHSR1a). It is suggested that ghrelin is involved in pituitary adenomas pathogenesis. There are inconsistent data regarding the effect of ghrelin on cell proliferation. In this study the outcome of ghrelin in the rat pituitary adenoma GH3 cell line on morphology and proliferation ratio was evaluated. The ghrelin receptor (Ghsr) mRNA expression in GH3 cell line was established as well, because it was found that heterogeneous expression pattern characterized physiological and pathological conditions of tissues of different origin. Material and Methods. Suitable experimental model pituitary tumor (rat GH3 cell line) was stimulated with ghrelin in the fi nal concentrations 10-12 M, 10-9 M and 10-6 M. Reverse transcription followed by real time polymerase chain reaction was used for ghrelin receptor gene transcript detection. The morphology as well as cell cycle of those cells were analyzed using Axio Vert. A1 Microscope (Zeiss) and BD FACSCalibur™ flow cytometer (Beckton Dickinson), respectively. The percentages of cells in the G0/G1, S, G2/M cycle phases were evaluated using the ModFit™ software (Verity Software, Inc., USA). Results. Ghsr mRNA presence was confi rmed in GH3 cells. Ghrelin did not affect conspicuously GH3 cells morphology, however the ghrelin-induced proliferation index increase was caused by both decline of G0/G1 phases cells count and increase those being in S+G2/M (p < 0.05). Conclusions. In conclusion, this study indicates that ghrelin stimulates GH3 cells proliferation and may play role in pituitary tumorigenesis via an autocrine/paracrine pathway. Deep paravertebral muscles and female sex hormones are potential elements participating in idiopathic scoliosis development. Estrogen acts through estrogen receptors: ESR1 and ESR2. There are no studies describing ESR2 expression in back muscles in girls with idiopathic scoliosis. The aim of this study was to evaluate ESR2 expression levels in back muscles on both sides of the spinal curve and correlation between the expression level and scoliosis parameters. Asymmetrical ESR2 expression in deep paravertebral muscles was found: 11 girls had higher expression level on the convex side and 5 girls had higher expression level on the concave side of the curvature. Patients with ESR2 (convex/concave) ratio ≥ 1 presented positive correlation between ESR2 ratio and Cobb angle. [ABSTRACT FROM AUTHOR]

Polymorphisms of these receptor genes have been implicated in male infertility in both Chinese and Caucasian populations. However, studies have produced inconsistent results. Spermatozoa defects that result in conception deficiencies could be related to estrogens, their receptors, or genes involved in estrogen-related pathways. This study aims to explore the potential association between the ESR1 and the ESR2 polymorphisms in relation to semen parameters of Caucasian males as well as fertilization success. A total of 116 males were included in this study. Forty couples underwent conventional in vitro fertilization, while 76 couples were treated by intracytoplasmic sperm injection. Standard semen analyses were performed according to the World Health Organization criteria. Polymerase chain reaction and restriction fragment length polymorphisms were used to determine genotype and allele distributions. A strong association between the ESR1 rs2234693 recognized by PvuII enzyme, genotype/allele distribution and fertilization success was shown. The T allele occurrence was significantly lower in the case of fertilization failure (p = 0.02). Additionally, the TT genotype was absent in the same group (p=0.02). In the case of the remaining analyzed polymorphisms, little to no interdependence of genotype/allele distribution and fertilization success was noted. Apart from ESR1 rs2234693, the study failed to demonstrate that fertilization success was associated with the selected polymorphisms. INTRODUCTION    Pituitary adenomas are heterogenous lesions commonly observed in the central nervous system. Signal transduction of ghrelin, an endogenous ligand specific for growth hormone secretagogue receptor (GHSR), has been reported to be involved in the development of endocrine tumors. However, there are limited data concerning the role of ghrelin and its functional receptor in pituitary adenomas. OBJECTIVES    The aim of the study was to establish the expression pattern of GHRL and its functional receptor GHSR1a in human pituitary adenomas. PATIENTS AND METHODS    Tissue specimens, including somatotropinomas (n = 20), prolactinomas (n = 5), and nonfunctioning adenomas (n = 52) were obtained from 77 patients. Thirteen normal pituitaries served as controls. The expression pattern of GHRL and GHSR1a mRNAs was established using reverse transcription followed by quantitative polymerase chain reaction. RESULTS    Ghrelin mRNA was detected in 92.2% of the samples including controls, while GHSR1a transcripts were detected in 54.4% of the cases. Significant differences were found among subgroups in the GHSR1a expression (P <0.0001) but not in that of GHRL (P = 0.7). The relative GHSR1a expression level was significantly lower for nonfunctioning tumors than for the control group or somatotropinomas. Controls revealed a strong positive correlation between the expression of both genes (r = 0.8; P <0.0001), unlike adenomas, which showed a weak negative correlation (r = -0.3; P >0.05). The maximum tumor diameter for nonfunctioning adenomas was higher than that for somatotropinomas (mean [SD], 31.4 [76] mm vs 24.8 [10.9] mm; P = 0.01). Neither the GHRL nor GHSR1a expression showed a significant correlation with tumor size in the subgroups. CONCLUSIONS    The presence of GHRL and GHSR1a in the neural system indicates their effect on pituitary function regulation and suggests their possible role in adenoma pathogenesis.

Introduction and Objectives: Motor and cognitive development of children with Down syndrome (DS) is delayed and inharmonic. Neuro-muscular abnormalities, such as hypotonia, retained primary reflexes, and slow performance of volitional reaction, result in difficulties with body balance. The aim of the presented study is to assess the global motor functions and body balance of children with DS in relation to age and mental development., Material and Methods: The study group consisted of 79 children with DS (42 boys, 37 girls), average age 6 years and 3 months ± 4 years and 6 months. Participants were divided according to age range into 3 groups: < 3 years old, 3-6 years old, > 6 years old. Children were assessed using Gross Motor Function Measure-88 (GMFM-88) and Paediatric Balance Scale (PBS). Psychological diagnosis served to determine the degree of mental development using the Brunet-Lezine Scale for children younger than 3 years old, and the Wechsler Intelligence Scale for Children (WISC) for those who are older than 3 years. Nine children in research group had not been diagnosed by psychologists, which is the reason why the analysis referring to mental development was performed in 70 children (34 girls, 36 boys), with an average age of 4 years and 6 months., Results: GMFM-88 scores were significantly lower in children with moderate psychomotor delay than in children with mild psychomotor delay, or normally developed children, p=0.043. GMFM-88 scores in children with profound mental impairment were lower than in children with mild or moderate mental impairment. There was a statistical significant correlation between GMFM-88 scores and the PBS scores, r= 0.7, p<0.0001., Conclusions: Motor development of children with Down syndrome from towns and villages in the Greater Poland region is associated with cognitive development, especially in the first three years of life, with the balance functions being closely related to motor skills.
