ABSTRACT Sustainability-focused research networks and communities of practice have emerged as a key response and strategy to build capacity and knowledge to support transformation towards more sustainable, just and equitable futures. This paper synthesises insights from the development of a community of practice on social-ecological systems (SES) research in southern Africa over the past decade, linked to the international Programme on Ecosystem Change and Society (PECS). This community consists of a network of researchers who carry out place-based SES research in the southern African region. They interact through various cross-cutting working groups and also host a variety of public colloquia and student and practitioner training events. Known as the Southern African Program on Ecosystem Change and Society (SAPECS), its core objectives are to: (1) derive new approaches and empirical insights on SES dynamics in the southern African context; (2) have a tangible impact by mainstreaming knowledge into policy and practice; and (3) grow the community of practice engaged in SES research and governance, including researchers, students and practitioners. This paper reflects on experiences in building the SAPECS community, with the aim of supporting the development of similar networks elsewhere in the world, particularly in the Global South.

Social-ecological systems (SES) research has emerged as an important area of sustainability science, informing and supporting pressing issues of transformation towards more sustainable, just and equitable futures. To date, much SES research has been done in or from the Global North, where the challenges and contexts for supporting sustainability transformations are substantially different from the Global South. This paper synthesises emerging insights on SES dynamics that can inform actions and advance research to support sustainability transformations specifically in the southern African context. The paper draws on work linked to members of the Southern African Program on Ecosystem Change and Society (SAPECS), a leading SES research network in the region, synthesizing key insights with respect to the five core themes of SAPECS: (i) transdisciplinary and engaged research, (ii) ecosystem services and human well-being, (iii) governance institutions and management practices, (iv) spatial relationships and cross-scale connections, and (v) regime shifts, traps and transformations. For each theme, we focus on insights that are particularly novel, interesting or important in the southern African context, and reflect on key research gaps and emerging frontiers for SES research in the region going forward. Such place-based insights are important for understanding the variation in SES dynamics around the world, and are crucial for informing a context-sensitive global agenda to foster sustainability transformations at local to global scales.

Responsiveness in metal-organic frameworks involving amorphous phases remains poorly understood. Here, the authors demonstrate MOFs that reversibly switch between well-defined crystalline and structurally degenerate amorphous states mediated by competing intra-framework forces.

The formation of polysubstituted cyclopropane derivatives in the gold(I)-catalyzed reaction of olefins and propargylic esters is a potentially useful transformation to generate diversity, therefore any method in which its stereoselectivity could be controlled is of significant interest. We prepared and tested a series of chiral gold(I)-carbene complexes as a catalyst in this transformation. With a systematic optimization of the reaction conditions, we were able to achieve high enantioselectivity in the test reaction while the cis:trans selectivity of the transformation was independent of the catalyst. Using the optimized conditions, we reacted a series of various olefins and acetylene derivatives to find that, although the reactions proceeded smoothly and the products were usually isolated in good yield and with good to exclusive cis selectivity, the observed enantioselectivity varied greatly and was sometimes moderate at best. We were unable to establish any structure-property relationship, which suggests that for any given reagent combination, one has to identify individually the best catalyst.

Background:As science and modern technology have brought many advances, we have also come to overshoot planetary boundaries, while still falling short of development goals to eradicate poverty and inequality. A growing recognition of the complexity of development problems and contexts calls for new framings, including a new approach to monitoring and evaluation (ME) as one of the mechanisms by which modern societies aim to steer towards a more sustainable future. New approaches to ME mean new skills for the ME practitioner. Objectives: This article proposed a framing for ME skills, comprising of technical, relational and transformational (T-R-T) competences. Method: Adapted from the literature, this competence framework was tested in a broader learning needs assessment and then applied retrospectively to author’s experience in developmental evaluations in complex social–ecological contexts in southern Africa. Results: The emerging insights were that not only technical competence is needed, but also relational competence that goes beyond interpersonal skills, to enable the production and uptake of evaluation findings. In addition, the limitations of mainstream ME methods in the face of complexity seemed to create a need for ‘transformational’ competence, which included evaluators’ ability to develop credible ME alternatives. Conclusion: The T-R-T framework helped to advance the notions of ‘hard’ and ‘soft’ skills and expanded on existing ME competence frameworks. Recommendations included a call for innovative educational and professional development approaches to develop relational and transformational competencies, in addition to training for technical competence.

The possible internal dynamics of non‐isotope‐labeled small‐molecule ligands inside a target protein is inherently difficult to capture. Whereas high crystallographic temperature factors can denote either static disorder or motion, even moieties with very low B‐factors can be subject to vivid motion between symmetry‐related sites. Here we report the experimental identification of internal μs timescale dynamics of a high‐affinity, natural‐abundance ligand tightly bound to the enzyme human carbonic anhydrase II (hCAII) even within a crystalline lattice. The rotamer jumps of the ligand's benzene group manifest themselves both, in solution and fast magic‐angle spinning solid‐state NMR 1H R1ρ relaxation dispersion, for which we obtain further mechanistic insights from molecular‐dynamics (MD) simulations. The experimental confirmation of rotameric jumps in bound ligands within proteins in solution or the crystalline state may improve understanding of host‐guest interactions in biology and supra‐molecular chemistry and may facilitate medicinal chemistry for future drug campaigns. Die experimentelle Erfassung möglicher interner Dynamik eines Liganden ohne Isotopenmarkierung im proteingebundenen Zustand ist grundsätzlich schwierig. Hohe Temperaturfaktoren aus Kristallstrukturen können statische Heterogenität nicht von Bewegung unterscheiden, zudem können für symmetrische Bewegungen selbst bei niedrigsten B‐Faktoren schnelle Bewegungen vorhanden sein. Hier zeigen wir die sowohl in Lösung als auch im Kristall experimentell beobachtete interne μs‐Dynamik eines nicht‐isotopenmarkierten Liganden, während dieser mit hoher Affinität an das Enzym humane Carboanhydrase II (hCAII) gebunden ist. Der sprunghafte Wechsel zwischen den Rotameren der Benzolgruppe des Liganden verursacht sowohl in Lösungs‐ als auch Festkörper NMR mit schneller Rotation um den magischen Winkel 1H R1ρ Relaxationsdispersion, für die mittels Molecular Dynamics (MD)‐Simulationen mechanistische Einblicke erhalten werden. Die experimentelle Erfassung von konformationellem Austausch in gebundenen Liganden in Lösung und im Festkörper könnte einem besseren Verständnis von Wirt‐Gast‐Interaktionen in Biologie und supramolekularer und medizinischer Chemie dienen. Combining the selective targeting of tumor cells through antigen-directed recognition and potent cell-killing by cytotoxic payloads, antibody-drug conjugates (ADCs) have emerged in recent years as an efficient therapeutic approach for the treatment of various cancers. Besides a number of approved drugs already on the market, there is a formidable follow-up of ADC candidates in clinical development. While selection of the appropriate antibody (A) and drug payload (D) is dictated by the pharmacology of the targeted disease, one has a broader choice of the conjugating linker (C). In the present paper, we review the chemistry of ADCs with a particular emphasis on the medicinal chemistry perspective, focusing on the chemical methods that enable the efficient assembly of the ADC from its three components and the controlled release of the drug payload.
