Background and Aim: Theileria annulata infection in cattle causes major economic losses in livestock production in many Central Asian countries, including the southern region of Kazakhstan. This study aimed to obtain a recombinant T. annulata surface protein (TaSP) and to investigate its possible use as an antigen in an indirect enzyme-linked immunosorbent assay (ELISA) for the serological diagnosis of bovine theileriosis. Materials and Methods: Recombinant TaSP was obtained by cloning a polymorphic region of the TaSP gene, expressing it in Escherichia coli strain BL21, and purifying it by metal chelating chromatography. An indirect ELISA using recombinant TaSP as an antigen was developed and evaluated for the detection of T. annulata-specific antibodies in plasma samples from 69 cows polymerase chain reaction (PCR)-positive or PCR-negative for T. annulata and/or Theileria orientalis from southern Kazakhstan. Results: The obtained recombinant protein had a molecular weight of 32 kDa, and mass spectrometry analysis of the purified protein identified it as a fragment of the surface protein of T. annulata. Initial testing of 69 field plasma samples from cattle showed that the results of indirect ELISA using TaSP as an antigen agreed substantially with those of T. annulata PCR (κ: 0.78). The relative sensitivity and specificity of indirect ELISA were 88.7% and 100%, respectively, using PCR as a reference. There was no evidence of cross-reaction with T. orientalis. Conclusion: Initial results using recombinant TaSP as an antigen in indirect ELISA are promising and support the widespread use of this assay for routine diagnosis and T. annulata seroprevalence studies in cattle in Kazakhstan and possibly neighboring countries.

Introduction Premature arrest of bone growth is the most common complication of bone fractures at the growth plate level. The purpose of the work was to evaluate the structural reorganization of metaepiphyseal cartilage following its direct injury with metal and biodegradable wires in an experiment. Materials and methods The metaepiphyseal cartilage of the distal femur of 18 lambs of both sexes was studied. The age of the animals at the beginning of the study was (43.92 ± 0.8) days, by 60 and 120 days (102.63 ± 0.82) and (161.1 ± 0.9) days, respectively. The animals underwent transphyseal insertion of wires/ pins: series 1 — Kirschner wires, series 2 — titanium wires, series 3 — poly-L-lactic acid pins. The duration of the experiment was 60 and 120 days. Clinical and radiographic studies were carried out. Histomorphometry was performed using an AxioScope.A1 microscope and Zenblue software (CarlZeissMicroImagingGmbH, Germany). Results Reactive changes in the growth plate at the interface with the wire were manifested by proliferation of chondrocytes in the zone of proliferating cartilage and in the reserve zone; the minimally expressed changes were noted in series 2, the most pronounced were in series 1. By the end of the experiment, at the interface with the wire in series 1, blood vessels penetrated into the metaepiphyseal cartilage; in series 3 the amount of the fibrous component was increased, which indicates further formation of “bone bridges” and “fibrous bridges,” respectively. In undamaged areas of the growth plate in all series, the zonal structure was preserved. By the end of the experiment, increased values of the thickness of the metaepiphyseal cartilage were noted (1.2 times higher than the control), differences between series were a tendency; in series 2 and 3 the ratio of metaepiphyseal cartilage zones was comparable to the control; in series 1 the proportion of the proliferating cartilage zone was increased by 4 %. Discussion The main problem with growth plate injuries is the formation of bone tissue or fibrosis, which affects the growing process. Currently, the question of choosing a treatment tactic for growth plate injury depending on the size of the “bone bridges” is debatable. Relevant are future comparative studies of the regeneration of metaepiphyseal cartilage defects after the use of fixators made from different materials. Conclusion Histomorphometric characteristics of the growth zone reliably showed that the insertion of wires, regardless of their material, was not accompanied by inhibition of the bone-forming function of the distal metaepiphyseal cartilage of the femur.

At present, the existing approaches to production of artificial isotopes are mostly based on the development experience from previous years. This work aims to develop an algorithm for selecting the most effective irradiation modes for target materials. The study is based on sequential modeling of irradiation of target isotopes by neutrons of different ‘single-group’ fluxes at the same neutron flux density within each energy group (BNAB-93). In this study, a flux density equal to 2×1015 n/(cm2×s) was used for each energy group. This approach will help ‘designing’ and selecting the actual neutron spectrum that has the highest efficiency compared to alternatives. The study modelled Co-60 and Lu-177g production for each energy group. The kinetics was analyzed in the most efficient groups in terms of specific activity. The maximum specific activity for Co-60 is reached in group 17 and is equal to 1 kCi/g. For the scheme of Lu-177g production through Lu-176 the maximum specific activity is reached in group 26 and is equal to 58.5 kCi/g. For the scheme of Lu-177g production through Yb-176, the maximum specific activity is reached in group 17 and is equal to 260 Ci/g, advantageous for production are groups 15–17 and 26.

Abstract Neuronal transmission relies on the regulated secretion of neurotransmitters, which are packed in synaptic vesicles (SVs). Hundreds of SVs accumulate at synaptic boutons. Despite being held together, SVs are highly mobile, so that they can be recruited to the plasma membrane for their rapid release during neuronal activity. However, how such confinement of SVs corroborates with their motility remains unclear. To bridge this gap, we employ ultrafast single-molecule tracking (SMT) in the reconstituted system of native SVs and in living neurons. SVs and synapsin 1, the most highly abundant synaptic protein, form condensates with liquid-like properties. In these condensates, synapsin 1 movement is slowed in both at short (i.e., 60-nm) and long (i.e., several hundred-nm) ranges, suggesting that the SV-synapsin 1 interaction raises the overall packing of the condensate. Furthermore, two-color SMT and super-resolution imaging in living axons demonstrate that synapsin 1 drives the accumulation of SVs in boutons. Even the short intrinsically-disordered fragment of synapsin 1 was sufficient to restore the native SV motility pattern in synapsin triple knock-out animals. Thus, synapsin 1 condensation is sufficient to guarantee reliable confinement and motility of SVs, allowing for the formation of mesoscale domains of SVs at synapses in vivo.

The classical Lotka–Volterra predator–prey model is globally stable and uniformly persistent. However, in real-life biosystems, the extinction of species due to stochastic effects is possible and may occur if the magnitudes of the stochastic effects are large enough. In this paper, we consider the classical Lotka–Volterra predator–prey model under stochastic perturbations. For this model, using an analytical technique based on the direct Lyapunov method and a development of the ideas of R.Z. Khasminskii, we find the precise sufficient conditions for the stochastic extinction of one and both species and, thus, the precise necessary conditions for the stochastic system’s persistence. The stochastic extinction occurs via a process known as the stabilization by noise of the Khasminskii type. Therefore, in order to establish the sufficient conditions for extinction, we found the conditions for this stabilization. The analytical results are illustrated by numerical simulations.

The study is devoted to modelling a sectoral ecosystem for the agro-industrial complex, incorporating into a single platform solution tools and mechanisms for the interaction between sectoral business structures in value chains as well as the proactive implementation of government functions for managing and supporting the industry. The ecosystem architecture has been developed and structured in terms of organizational, infrastructure, innovation, communication and logistics components. The role of the system actor and IT integrator is assigned to the Ministry of Agriculture of the Russian Federation. The main and additional functions of the digital platform are highlighted. A description of its product and service segments is provided. The options for implementing the process subsystem on the Gostech and GosOblako platforms as well as the Russian Agricultural Bank platform are assessed, the advantages of each solution presented. The importance of involving the research potential of departmental high education institutions and research institutes in the development of the ecosystem’s service segment is substantiated.

Iron ore pellets are reduced in a 50%H2–50%N2 1 atm gas mixture at 750, 800, 850, 900, and 950 °C while simultaneously documenting swelling (change in pellet radius) and weight change. Swelling increases with increasing temperature, with catastrophic swelling (>20% of reduction swelling index) observed at 850, 900, and 950 °C. As the pellet is reduced, the pellet radius increases until 40–50% reduction is achieved, followed by a decrease in diameter beyond 40–50% reduction at 750 and 850 °C. At 950 °C, the pellet radius continues to increase with additional pellet reduction without any subsequent decrease in diameter. Scanning electron microscopy (SEM) analysis shows that the neighboring grains inside the pellet sinter together at 750 and 850 °C, whereas the individual grains sinter internally at 950 °C. SEM analysis and observations suggest that the reduction process at 750 and 850 °C can be approximated as a topochemical reaction process, while the reduction process at 950 °C can no longer be approximated as a topochemical reaction process. An empirical equation for the radius of the pellet is derived with fitting parameters dependent on temperature and the degree of reduction of the pellet undergoing reduction based on the experimental data. In terms of nuclear raw materials, the issue of involving thorium in the fuel cycle is hardly very relevant. However, in view of the large-scale nuclear power development, the use of thorium seems to be quite natural and reasonable. The substitution of traditional uranium-plutonium fuel for uranium-thorium fuel in fast neutron reactors will significantly reduce the production of minor actinides, which will make it attractive for the transmutation of long-lived radioactive isotopes of americium, curium and neptunium that have already been and are still being accumulated. Due to the absence of uranium-233 in nature, the use of thorium in the nuclear power industry requires a closed fuel cycle. At the initial stage of developing the uranium-thorium cycle, it is proposed to use uranium-235 instead of uranium-233 as nuclear fuel. Studies have been carried out on the transmutation of minor actinides in a fast neutron reactor in which the uranium-thorium cycle is implemented. Several options for the structure of the core of such a reactor have been considered. It has been shown that heterogeneous placement of americium leads to higher rates of its transmutation than homogeneous placement does.

As various regions around the world implement carbon taxes, we assert that the competitiveness of steel products in the marketplace will shift according to individual manufacturers’ ability to reduce CO2 emissions as measured by cradle-to-gate Life Cycle Analysis (LCA). This study was performed by using LCA and cost estimate research to compare the CO2 emissions and the additional cost applied to the production of various decarbonized materials used in sheet for automotive industry applications using the bending stiffness-based weight reduction factor. The pre-pandemic year 2019 was used as a baseline for cost estimates. This paper discusses the future cost scenarios based on carbon taxes and hydrogen cost. The pathways to decarbonize steel and alternative materials such as aluminum and reinforced polymer composites were evaluated. Normalized global warming potential (nGWP) estimates were calculated assuming inputs from the current USA electricity grid, and a hypothetical renewables-based grid. For a current electricity grid mix in the US (with 61% fossil fuels, 19% nuclear, 20% renewables), the lowest nGWP was found to be secondary aluminum and 100% recycled scrap melting of steel. This is followed by the natural gas Direct Reduced Iron–Electric Arc Furnace (DRI-EAF) route with carbon capture and the Blast Furnace-Basic Oxygen Furnace (BF-BOF) route with carbon capture. From the cost point of view, the current cheapest decarbonized production route is natural gas DRI-EAF with Carbon Capture and Storage (CCS). For a renewable electricity grid (50% solar photovoltaic and 50% wind), the lowest GWP was found to be 100% recycled scrap melting of steel and secondary aluminum. This is followed by the hydrogen-based DRI-EAF route and natural gas DRI-EAF with carbon capture. The results indicate that, when applying technologies available today, decarbonized steel will remain competitive, at least in the context of automotive sheet selection compared to aluminum and composites.

Introduction Periarticular injuries in children include types I and II fractures according to the Salter-Harris classification and metaphyseal fractures. In most cases, conservative treatment is an effective method, but in some cases, surgical treatment, including external fixation, is the method of choice. Purpose Discussion of the principles of using the Ilizarov apparatus in the treatment of children with periarticular fractures of various locations, its advantages and disadvantages. Materials and methods We present the principles and features of the Ilizarov surgical techniques for treatment of children with periarticular fractures of the distal humerus, femur, radius and tibia. Discussion The methods of osteosynthesis for pediatric periarticular fractures imply transphyseal introduction of fixators that have a potential threat of iatrogenic damage to the growth plate in contrast to external fixation which performance implies that transosseous elements do not injure the growth zone, providing stable fixation of bone fragments in combination with early functional recovery of the damaged segment which is a key advantage over other methods. Conclusion The use of the Ilizarov apparatus enables to achieve the desired result in the treatment of pediatric periarticular fractures and has a number of advantages over other methods of surgical treatment.
