With the development and approval of new proteasome inhibitors, proteasome inhibition is increasingly recognized in cancer therapy. Besides successful anti-cancer effects in hematological cancers, side effects such as cardiotoxicity are limiting effective treatment. In this study, we used a cardiomyocyte model to investigate the molecular cardiotoxic mechanisms of carfilzomib (CFZ) and ixazomib (IXZ) alone or in combination with the immunomodulatory drug dexamethasone (DEX) which is frequently used in combination therapies in the clinic. According to our findings, CFZ showed a higher cytotoxic effect at lower concentrations than IXZ. DEX combination attenuated the cytotoxicity for both proteasome inhibitors. All drug treatments caused a marked increase in K48 ubiquitination. Both CFZ and IXZ caused an upregulation in cellular and endoplasmic reticulum stress protein (HSP90, HSP70, GRP94, and GRP78) levels and DEX combination attenuated the increased stress protein levels. Importantly, IXZ and IXZ-DEX treatments caused upregulation of mitochondria fission and fusion gene expression levels higher than caused by CFZ and CFZ-DEX combination. The IXZ-DEX combination reduced the levels of OXPHOS proteins (Complex II–V) more than the CFZ-DEX combination. Reduced mitochondrial membrane potential and ATP production were detected with all drug treatments in cardiomyocytes. Our findings suggest that the cardiotoxic effect of proteasome inhibitors may be due to their class effect and stress response and mitochondrial dysfunction may be involved in the cardiotoxicity process.

Background/aim: The factors associated with fall-related self-efficacy should be addressed, especially in the elderly. The Modified Falls Efficacy Scale (MFES) is a simple instrument with good scaling properties and reliability. The aim of the present study was to determine the reliability and the validity of the Turkish version of the MFES from the viewpoint of balance. Materials and methods: In this study, 164 participants aged >65 years were included. The use of walking aids and assistive devices, history and frequency of falls in the previous year, living environment, and exercise habits were noted. Balance and risk of falling were assessed with the Berg Balance Scale (BBS). A forward-backward translation procedure was used for the Turkish version of the MFES. Results: None of the 14 items in the MFES were modified. The Turkish version of the MFES has excellent internal consistency (Cronbach's alpha, 0.978) and reliability (interclass correlation coefficient, 0.928-0.982), and its construct validity was supported by its ability to distinguish between the groups with respect to fall-risk factors and balance. According to the BBS scores, the high-fall-risk group had lower M ITS scores than the moderate- and low-fall-risk groups (chi(2) 34.153, P 0.001). Conclusion: The Turkish version of the MFES is a sensitive instrument for evaluation of fall-related confidence while carrying out indoor/ outdoor activities. It also predicts falls, reduced physical activity, balance and mobility problems, and restricted social participation and daily living activities. © 2017 EMO (Turkish Chamber of Electrical Enginners).The nonlinear function in Izhikevich neuron model (IzNM) makes difficult the digital hardware realizations of the model, so this parabolic function has been transformed to piecewise linear (PWL) functions in the literature. Some coefficients have been identified in the PWL functions by utilizing the classical step size method, but the values of these coefficients depend on the sensitivity of the step size considerably. In this study, the coefficients of the PWL functions in the modified IzNM are determined by using Genetic Algorithm (GA). After the parameter determination, the modified IzNM is simulated with the parameters, which are determined by both classical step size and GA. Also, the original and modified IzNMs exhibiting 'tonic spiking' and 'tonic bursting' behaviors are realized with digital programmable device, namely FPGA. Thus, it is tested the utility of the intelligent search algorithms in the neuronal structures and verified the adaptability of their results to the hardware implementations.

BACKGROUND: Multiple sclerosis (MS) involves impaired trunk control, leading to impaired upper-limb functionality, dexterity, and independence. Deciding early on a comprehensive approach pointing of functional disturbances and personal needs is essential for a multimodal, individualized, goal-oriented assessment and treatment program, recognizing the broad range of symptoms and disabilities associated with MS. In clinical practice, postural control of the trunk is purported to be an important contributor to voluntary upper-limb function, including motor control and dexterity. The objective of this study was to point out the impairments of and relationship between trunk control and comprehensive upper-limb functions in individuals with MS. METHODS: Tasks that were sought are optimal screening for deterioration in trunk control (Trunk Control Test [TCT] and Trunk Impairment Scale [TIS]) and upper-limb functionality by comparing them with the Expanded Disability Status Scale (EDSS), Nine Hole Peg Test (NHPT), Duruoz's Hand Index (DHI), and Functional Independence Measurement (FIM) results of 49 well-defined relapsing-remitting MS (RRMS) participants with those of 49 age-gender matched healthy subjects. RESULTS: Significant differences between the groups were evident across all tasks of the clinical tests studied (p < 0.05), except the TCT-balance in sitting position subscore. EDSS, NHPT, DHI, and FIM scores were highly correlated with the TCT subscores (rolling to weak side, sitting up from lying down) and TCT-total score, as well as TIS subscores (dynamic and coordination) and TIS total score (p ≤ 0.005). While TIS subscores were highly correlated with almost all parameters, just TIS-static subscore did not correlate with the DHI and FIM-cognitive scores. Also, DHI-hygiene subscore correlated poorly just with the TIS-coordination and TCT-coming to sitting position (p < 0.05). CONCLUSION: We found that patients with MS would present impaired upper-limb movement and decreased trunk control with high correlation between them, even in RRMS and in the very mild form of the disease. Identifying trunk control deficits provides better insight into dexterous difficulties of patients with MS and allows a more targeted neurorehabilitation focusing on upper limbs. In future studies, it would be of interest to examine the prognostic value of trunk control and upper-limb functionality in patients with MS using a longitudinal approach.

The present work aimed the study of rutin, protocatechuic, catechin, p- hydroxy benzoic, chlorogenic, caffeic, kuersetin, luteolin, kamferol, p-coumaric, vanilin and ferulic acids composition in different currant varieties in Turkey. Red and black currant varieties (Goliath, Red Lake, Rovada, Rosenthal, and Booskop Giant) were analyzed for phenolic acids by using reversed-phase high performance liquid chromatography (HPLC). Results showed that p-coumaric acid (1.66-0.04 µg g-1) was the predominant phenolic acid extracted from currant varieties. In addition, other acids, namely rutin (35.41-7.23 µg g-1), Protocatechuic (3.28-2.84 µg g-1), Chlorogenic (65.49-2.38 µg g-1), and kuersetin acids (2.29-1.50 µg g-1) were obtained in extracts from currant fruit. © 2017, Centenary University. Effect of two doses of calcium nitrate (2 and 4%), boric acid (1.5 and 3%) and GA3 (50 and 75 ppm) on yield, fruit characteristics, cracking and sunburn of pomegranate cv. Hicaznar was carried out in a commercial orchard. Calcium nitrate, boric acid and GA3 applications have been done during blossoming period and one month after blossoming. The fruit yield has been increased by both doses of calcium nitrate and the 3% boric acid dose in the first year while GA3’s 50 ppm dose had an improving effect in the second year. In the first year, all treatments increased the average fruit weight while in the second year only the 2% calcium nitrate and 3% boric acid were found to be effective. The 3% boric acid treatments reduced the rate of cracking of the fruits in first year and in the second year all treatments reduced cracking and the best results were obtained in the applications of GA3 and calcium nitrate. The 4% calcium nitrate application reduced the sunburn in the pomegranate fruits and 2% calcium nitrate application increased the amount of the total soluble solids. © Wydawnictwo Uniwersytetu Przyrodniczego w Lublime. Yüksek standartta bir spor eğitimi, nitelikli yetiştiriciler ile güçlü ve yetenekli sporculara gereksinim duyar. Araştırmacıların ortaya çıkardığı tüm verileri değerlendiren ve sporcularına yüksek standartta eğitimi vermeye çalışan antrenörler önemli konumdadır. Başarılı antrenörler genellikle dengeli, kendine hakim, sosyal bir kişiliğe sahiptirler. Bu araştırmanın amacı, Bursa ili voleybol antrenörlerinin sosyal tercihlerini ve mesleki yapılarını tespit ederek, bir eğitimcide bulunması gereken davranışları gösterip göstermediğini belirlemek, antrenörlerin işlevsel sorunlarını, mesleki beklentilerini ortaya koyarak ilgililere mesajlar ve öneriler sunmaktır. Araştırmamıza 109 erkek, 21 bayan voleybol antrenörü katılmıştır. Araştırmada elde edilen veriler, SPSS paket programı yardımıyla analiz edilmiştir. Analiz sonuçları %, frekans ve ki kare aracılığıyla tespit edilmiştir. Araştırmamıza katılan antrenörlerin öğrenim durumlarına bakıldığında % 76’sı üniversite mezunudur. Yine antrenörlerin % 61,5’i antrenörlüğü yan çalışma alanı olarak yapmaktadırlar. Antrenörlüğü özel çaba gerektiren bir meslek olarak gören antrenörler, bu mesleği yapmaktan memnun olduklarını belirtmişlerdir. Çalışma koşullarını önemli sorun olarak gören antrenörler, meslektaşlarıyla bazen çatışma yaşadıklarını belirtmişlerdir. Sporcuların eğitiminin niteliğini geliştirmek, antrenörlerin niteliğini geliştirmekle eşanlamlıdır. Antrenörün niteliği sporcunun, sporcunun niteliği, ülke sporunun niteliğinin yükselmesinde temel unsurlardır. Antrenörlerin hangi niteliklere sahip olması gerektiği konusundaki çalışmaların çoğalarak devam etmesi ve yapılan araştırmaların sonuçlarına göre, antrenör yetiştiren kurumlarda ve kademe eğitimlerinde öğrenim görenlerin yönlendirilerek, antrenör olacakların niteliklerinin bu ilkeler ve özellikler çerçevesinde yükseltilmesi ülke sporunun geleceği açısından büyük önem taşımaktadır.
