Antioxidant capacity, cytotoxicity on two vital cell lines and the anti-cancer activity of methanolic Urtica dioica L. leaf extract (UDE) collected from Duzkoy, Giresun, Turkey were studied by determining safe concentration. The antioxidant capacity of the extract was expressed as vitamin C equivalency by spectrophotometric MTT assay. The cytotoxic concentration 50% was measured by the linearity between UDE concentrations (CC50) and the cell viability of non-cancer kidney cell lines (BHK-21, MDBK). The anti-cancer activity was conducted on human hepatocellular carcinoma cells (HepG2) by determining inhibition concentration (IC50) on cell proliferation. The vitamin C equivalence of UDE increased linearly by increasing the concentration. The cytotoxic and non-toxic concentrations of UDE were determined on BHK-21 and MDBK with 15.71 mg/ml and 5.14 mg/ml of CC50 respectively. The extract inhibited the proliferation of human hepatocellular carcinoma cells with a 2.46 mg/ml of IC50. In conclusion, the present study tried to explain in detail the dose-dependent activity of Urtica dioica L. leaf extract. The dose-response results showed that Urtica dioica L. leaf extract could have low cytotoxicity, but potential anti-cancer activity at safe concentrations. [ABSTRACT FROM AUTHOR]

Research background. The cytotoxicity on various vital cell lines and the activity on foodborne pathogens (murine norovirus 1 as a norovirus surrogate and Campylobacter jejuni) of methanolic Urtica dioica L. leaf extract (UDE) were studied. Experimental approach. The cytotoxic concentration of 50% (CC50) was measured by the linearity between UDE concentrations and cell viability. Antibacterial effects on C. jejuni were analyzed by the broth microdilution method with a spectrophotometer. The virucidal and antiviral activities of UDE were determined by the virus titration method on the host cell infectivity and expressed as the tissue cell infective dose of 50% using the method of Spearman-Karber. Results and conclusions. The CC50 of UDE was determined on macrophage as the virus host cell. MIC and MBC of UDE were determined as 5 mg/ml and 20 mg/ml for both C. jejuni isolated from poultry meat and the standard strain. UDE inhibited MNV-1 on three pathways of host cell infectivity at approximately the same 50% inhibitory concentration (1.45-1.87 mg/ml). In conclusion, the present study tried to explain in detail the dose-dependent activity of Urtica dioica L. leaf extract on two important foodborne pathogens causing outbreaks worldwide. Ionizing radiation has widespread use in medicine in the diagnosis and treatment of many medical conditions. Radiology technicians are one group that is occupationally exposed to low doses of radiation. There are questions regarding whether low dose exposure to radiation could have long-term health consequences. Assessing the effect of radiation on genetic material is essential for appraising long-term health results. Hereditary variations in DNA repair genes cause differentiation in individual responses to radiation related health effects. This study aimed to determine oxidative stress and DNA damage, and their relationship to XRCC1 (Arg399Gln) and XRCC3 (Thr241Met) polymorphisms in radiology technicians occupationally exposed to low dose radiation. Peripheral blood samples were collected from 45 radiology technicians and age-matched with 40 healthy control individuals working in office environments. Our results showed that radiology technicians had significantly greater oxidative stress and DNA damage than the control group, and women appeared more susceptible to occupational radiation exposure than men. Individuals with wild-type genotypes for XRCC1 (Arg/Arg) and XRCC3 (Thr/Thr) had less DNA damage. Lower DNA damage levels could be explained by the enhanced capacity to repair low dose radiation induced DNA damage. Further studies are needed to evaluate the role of DNA repair genes in individuals that are occupationally exposed to low dose radiation. [ABSTRACT FROM AUTHOR]
