1. Ketone 3-hydroxybutyrate: a biomarker of aggression?

Abstract

Aggression is a complex behavior broadly involving many body systems, commonly investigated at the biological level within specific pathways. Studying metabolites in relation to aggression can shed light on the biology of aggression in a broader context of various biological pathways affecting behavior. Associations between metabolites and aggression are not well known. Here, using a selected set of 11 low molecular weight metabolites including amino acids and ketones, we aimed to identify metabolites that associate with adolescent aggression. Our dataset is a sub-sample of a population-based, longitudinal study of Finnish twins born in 1983-1987 with available plasma samples at age 22 and aggression levels rated in adolescence (N=725) by parents of the twins (at age 12), their teachers (at ages 12 & 14), or the twins and their co-twins (at ages 14 & 17). Linear regression models, adjusted for age, sex, body mass index and the clustered twin data, were used with the metabolite as the outcome and aggression levels as predictors. Five metabolites showed unadjusted correlations greater than 0.1 with aggression. However, we found that aggression showed consistent negative associations only with 3-hydroxybutyrate, a ketone body produced in the fasting state. Effect sizes for different single raters were generally similar in magnitude, while age 12 teacher-rated aggression and age 14 self-rated aggression were both significant predictors of 3-hydroxybutyrate in multi-rater modeling. These exploratory results indicate ketone metabolism as a possible new pathway to investigate further to better understand the biology of aggression.

Abstract

The SwissFEL Injector Test Facility operated at the Paul Scherrer Institute between 2010 and 2014, serving as a pilot plant and testbed for the development and realization of SwissFEL, the X-ray Free-Electron Laser facility under construction at the same institute. The test facility consisted of a laser-driven rf electron gun followed by an S-band booster linac, a magnetic bunch compression chicane and a diagnostic section including a transverse deflecting rf cavity. It delivered electron bunches of up to 200 pC charge and up to 250 MeV beam energy at a repetition rate of 10 Hz. The measurements performed at the test facility not only demonstrated the beam parameters required to drive the first stage of an FEL facility, but also led to significant advances in instrumentation technologies, beam characterization methods and the generation, transport and compression of ultra-low-emittance beams. We give a comprehensive overview of the commissioning experience of the principal subsystems and the beam physics measurements performed during the operation of the test facility, including the results of the test of an in-vacuum undulator prototype generating radiation in the vacuum ultraviolet and optical range., Comment: 60 pages, 70 figures; final version as published, except with table of contents and Fig. 68 in full resolution

Abstract

Cannabis is the most widely produced and consumed illicit psychoactive substance worldwide. Occasional cannabis use can progress to frequent use, abuse and dependence with all known adverse physical, psychological and social consequences. Individual differences in cannabis initiation are heritable (40-48%). The International Cannabis Consortium was established with the aim to identify genetic risk variants of cannabis use. We conducted a meta-analysis of genome-wide association data of 13 cohorts (N=32 330) and four replication samples (N=5627). In addition, we performed a gene-based test of association, estimated single-nucleotide polymorphism (SNP)-based heritability and explored the genetic correlation between lifetime cannabis use and cigarette use using LD score regression. No individual SNPs reached genome-wide significance. Nonetheless, gene-based tests identified four genes significantly associated with lifetime cannabis use: NCAM1, CADM2, SCOC and KCNT2. Previous studies reported associations of NCAM1 with cigarette smoking and other substance use, and those of CADM2 with body mass index, processing speed and autism disorders, which are phenotypes previously reported to be associated with cannabis use. Abstract

In this work we investigate the effects of the electron-electron interaction between a molecular junction and the metallic leads in time-dependent quantum transport. We employ the recently developed embedded Kadanoff-Baym method [Phys. Rev. B 80, 115107 (2009)] and show that the molecule-lead interaction changes substantially the transient and steady-state transport properties. We first show that the mean-field Hartree-Fock (HF) approximation does not capture the polarization effects responsible for the renormalization of the molecular levels neither in nor out of equilibrium. Furthermore, due to the time-local nature of the HF self-energy there exists a region in parameter space for which the system does not relax after the switch-on of a bias voltage. These and other artifacts of the HF approximation disappear when including correlations at the second-Born or GW levels. Both these approximations contain polarization diagrams which correctly account for the screening of the charged molecule. We find that by changing the molecule-lead interaction the ratio between the screening and relaxation time changes, an effect which must be properly taken into account in any realistic time-dependent simulation. Another important finding is that while in equilibrium the molecule-lead interaction is responsible for a reduction of the HOMO-LUMO gap and for a substantial redistribution of the spectral weight between the main spectral peaks and the induced satellite spectrum, in the biased system it can have the opposite effect, i.e., it sharpens the spectral peaks and opens the HOMO-LUMO gap., Comment: 18 pages, 26 figures

Abstract

The gap between students' formal, school-based learning and informal, social learning has been widely acknowledged. To overcome this gap, academic service-learning (AS-L) pedagogy involves deliberate efforts to interconnect formal and informal learning by enhancing students' transitions across multiple authentic settings and fostering their participation in various social roles. The aim of this study was to examine how AS-L is practiced among Finnish primary and secondary schools, what kind of stakeholder networks are involved, what the experienced outcomes are, and which factors enable the AS-L pedagogy to be embedded into the Finnish formal school context. The data consisted of semi-structured educator interviews describing nine cases of implementing AS-L practices in diverse Finnish school settings. Empirically driven, thematic data-analysis generated an overall description of the AS-L pedagogies examined. The findings confirm that AS-L pedagogy fulfiled the curricular aims, assisted in meeting the learning objectives and affirmed the fundamental role of external stakeholders in AS-L pedagogy. However, results also revealed challenges related to schools' operational infrastructures, lesson frameworks, and teacher collaboration hindering the interdisciplinary implementation of AS-L pedagogy, preventing synergies with subject-specific learning contents, and limiting stakeholder networks. Innovative teacher activity, in turn, was identified as a success factor in applying AS-L pedagogy. The results indicate that interconnecting formal and informal learning requires schools' systemic change, and furthermore, legitimising teacher's transformative efforts fosters development and implementation of novel pedagogical practices. 10. Insertion Device Controls at the Swiss Light Source

Abstract

In the initial phase of the Swiss Light Source, four insertion devices will be installed. The control system for all the ID's follows an uniform concept to allow a rapid installation schedule, while at the same time allowing for a variety in operational requirements. The components of the control system include the gap drive system with encoders, temperature monitoring, beam position monitoring, power supply controllers for corrector magnets [1] and a timing module [2]. The gap drive system requirements range from micron-level accuracy to driving double undulators in synchronism with each other and also with the other beamline elements. The local synchronism is achieved with a tightly coupled system having an intelligent motor controller and the global synchronism by extending the machine timing distribution to the insertion devices and the beamlines with the capability to add beamline-specific timing events. Abstract

Primary central nervous system lymphoma (PCNSL) is an aggressive brain disease where lymphocytes invade along perivascular spaces of arteries and veins. The invasion markedly changes (peri)vascular structures but its effect on physiological brain pulsations has not been previously studied. Using physiological magnetic resonance encephalography (MREGBOLD) scanning, this study aims to quantify the extent to which (peri)vascular PCNSL involvement alters the stability of physiological brain pulsations mediated by cerebral vasculature. Clinical implications and relevance were explored. In this study, 21 PCNSL patients (median 67y; 38% females) and 30 healthy age-matched controls (median 63y; 73% females) were scanned for MREGBOLD signal during 2018–2021. Motion effects were removed. Voxel-by-voxel Coefficient of Variation (CV) maps of MREGBOLD signal was calculated to examine the stability of physiological brain pulsations. Group-level differences in CV were examined using nonparametric covariate-adjusted tests. Subject-level CV alterations were examined against control population Z-score maps wherein clusters of increased CV values were detected. Spatial distributions of clusters and findings from routine clinical neuroimaging were compared [contrast-enhanced, diffusion-weighted, fluid-attenuated inversion recovery (FLAIR) data]. Whole-brain mean CV was linked to short-term mortality with 100% sensitivity and 100% specificity, as all deceased patients revealed higher values (n = 5, median 0.055) than surviving patients (n = 16, median 0.028) (p < .0001). After adjusting for medication, head motion, and age, patients revealed higher CV values (group median 0.035) than healthy controls (group median 0.024) around arterial territories (p ≤ .001). Abnormal clusters (median 1.10 × 10₅mm₃) extended spatially beyond FLAIR lesions (median 0.62 × 10₅mm₃) with differences in volumes (p = .0055).

Abstract

Background/Objectives: The glycaemic index (GI) is used to describe the blood glucose-raising potential of carbohydrate-containing foods. Only a few descriptions of the addition of GI values to national food composition databases (FCDBs) exist. We tested whether the value documentation framework established within the European Food Information Resource (EuroFIR) Network could be used for GI values when adding them to the Finnish FCDB. Methods: The list of foods requiring GI values was based on the National FINDIET 2007 Survey data and extended with foods encoded in a food-frequency questionnaire used in other nationally representative studies. The minimum quality of GI measurements was verified when gathering values from various sources, using earlier defined criteria. If a measured GI value for a food was directly available, or could be imputed or estimated, the value was added to the Finnish FCDB and documented using core standard vocabularies of EuroFIR. The GI values of composite foods were calculated using recipe calculation software. Results: A total of 2210 foods required a GI value. GI values for 1322 foods were available and added to the FCDB. The remaining 888 foods were composite foods and received a GI value through recipe calculation. The standard vocabularies describing the origin of the GI values, the methods used in their derivation and their qualitative characteristics were suitable for GI values. Conclusions: GI values can be added to FCDBs and documented using terms similar to those used for traditional food composition data. Standardised value documentation may provide transparency for GI database compilation processes. Abstract

BACKGROUND A trial involving adults 50 years of age or older (ZOE-50) showed that the herpes zoster subunit vaccine (HZ/su) containing recombinant varicella-zoster virus glycoprotein E and the AS01(B) adjuvant system was associated with a risk of herpes zoster that was 97.2% lower than that associated with placebo. A second trial was performed concurrently at the same sites and examined the safety and efficacy of HZ/su in adults 70 years of age or older (ZOE-70). METHODS This randomized, placebo-controlled, phase 3 trial was conducted in 18 countries and involved adults 70 years of age or older. Participants received two doses of HZ/su or placebo (assigned in a 1: 1 ratio) administered intramuscularly 2 months apart. Vaccine efficacy against herpes zoster and postherpetic neuralgia was assessed in participants from ZOE-70 and in participants pooled from ZOE-70 and ZOE-50. RESULTS In ZOE-70, 13,900 participants who could be evaluated (mean age, 75.6 years) received either HZ/su (6950 participants) or placebo (6950 participants). During a mean follow-up period of 3.7 years, herpes zoster occurred in 23 HZ/su recipients and in 223 placebo recipients (0.9 vs. 9.2 per 1000 person-years).
