Abstract

Qualia are private conscious experiences of which the associated feelings can be reported to other people. Whether qualia are amenable to scientific exploration has often been questioned, which is challenged by the present article. The following arguments are given: 1. the configuration of the brain changes continuously and irreversibly, because of genetic and environmental influences and interhuman communication; 2. qualia and consciousness are processes, rather than states; 3. private feelings, including those associated with qualia, should be positioned in the context of a personal brain as being developed during life; 4. consciousness and qualia should be understood in the context of general system theory, thus concluding that isolated, in vitro, properties of neurons and other brain constituents might marginally contribute to the understanding of higher brain functions, mind or qualia; 5. current in vivo approaches have too little resolution power - in terms of space and time - to delineate individual and subjective brain processes.

Abstract

An on-line in vitro perfusion technique is described that allows the continuous quantification of cellular glucose metabolism in vitro. Using biosensor technology, we measure glucose and lactate metabolism at a minute-to-minute time resolution for periods up to several days. The application of our perfusion--detection technique for in vitro monitoring is demonstrated in a wide variety of cells, including primary neuronal and astroglia cultures, yeast cells, and human lymphocytes. The method shows that variations in oxygen delivery or exposure to a noncompetitive pseudosubstrate (here 2-deoxyglucose) affects normal glucose metabolism. The innovative advantage of the present system is that, in contrast to other devices including a recently described system, metabolism per cell can be quantified. The potential of in vitro on-line monitoring is discussed for application in studying normal and abnormal metabolism, toxic and nontoxic drug effects, and human tissue biopsies.

30. Minimally invasive technique based on ultraslow ultrafiltration to collect and store time profiles of analytes

Abstract

A device is described to collect and store continuously time profiles of analytes over periods of 24 h suitable to sample freely moving individuals (humans and animals). The device consists of a hollow fiber ultrafiltration probe, a long capillary and a nonmechanical unit (a disposable medical syringe) driven by vacuum to withdraw fluid. The principle is that at low rates ([less than or equal to] 100 nL/min), sample fluid is collected through the ultrafiltration probe into the capillary. A time resolution of less than 5 min over a 24-h collection and storage period was achieved for lactate and glucose. To illustrate an in vivo application, devices were fixed under the wing of freely moving broiler chickens, with subcutaneous or intravenous probe placements. The device can be produced as a disposable, and it may become applied for ex vivo and in vitro monitoring.

Abstract

Classical microdialysis has some drawbacks. Two main issues in this respect are the time-consuming calculations (due to partial recovery of an analyte) and depletion near the sampling site. In this paper we describe a sampling method, called ultraslow microdialysis (usMD), and compare this with ultrafiltration (UF) at flow rates between 100 and 300 nL/min. As an example of an application of this method, we conducted a dual-enzyme analysis for simultaneous measurement of the glucose and lactate concentrations, demonstrating these sampling techniques in the subcutaneous tissue of the rat. For UF, the flow rates in vitro and in vivo were the same. For usMD, the recovery for glucose and lactate at the applied flow rates was 100%, in contrast to conventional microdialysis. Both sampling techniques have the potential of on-line in vivo monitoring, as well as the measurement of time profiles of analytes by continuous collection and off-line analysis afterward.
