We present Zeeman-Doppler images of the active K2 star II Peg for the years 2004 and 2007. The surface magnetic field was reconstructed with our new ZDI code "iMap" which provides a full polarized radiative transfer driven inversion to simultaneously reconstruct the surface temperature and magnetic vector field distribution. II Peg shows a remarkable large scale magnetic field structure for both years. The magnetic field is predominantly located at high latitudes and is arranged in active longitudes. A dramatic evolution in the magnetic field structure is visible for the two years, where a dominant and largely unipolar field in 2004 has developed into two distinct and large scale bipolar structures in 2007., Comment: Proceedings of IAU Symposium 259, Cosmic Magnetic Fields: from Planets, to Stars and Galaxies Ed.: K. Zeeman-Doppler Imaging (ZDI) is a powerful inversion method to reconstruct stellar magnetic surface fields. The reconstruction process is usually solved by translating the inverse problem into a regularized least-square or optimization problem. In this contribution we will emphasize that ZDI is an inherent non-linear problem and the corresponding regularized optimization is, like many non-linear problems, potentially prone to local minima. We show how this problem will be exacerbated by using an inadequate forward model. To facilitate a more consistent full radiative transfer driven approach to ZDI we describe a two-stage strategy that consist of a principal component analysis (PCA) based line profile reconstruction and a fast approximate polarized radiative transfer method to synthesize local Stokes profiles. 6. A fast method for Stokes profile synthesis -- Radiative transfer modeling for ZDI and Stokes profile inversion

The major challenges for a fully polarized radiative transfer driven approach to Zeeman-Doppler imaging are still the enormous computational requirements. In every cycle of the iterative interplay between the forward process (spectral synthesis) and the inverse process (derivative based optimization) the Stokes profile synthesis requires several thousand evaluations of the polarized radiative transfer equation for a given stellar surface model. To cope with these computational demands and to allow for the incorporation of a full Stokes profile synthesis into Doppler- and Zeeman-Doppler imaging applications as well as into large scale solar Stokes profile inversions, we present a novel fast and accurate synthesis method for calculating local Stokes profiles. Our approach is based on artificial neural network models, which we use to approximate the complex non-linear mapping between the most important atmospheric parameters and the corresponding Stokes profiles. A number of specialized artificial neural networks, are used to model the functional relation between the model atmosphere, magnetic field strength, field inclination, and field azimuth, on one hand and the individual components (I,Q,U,V) of the Stokes profiles, on the other hand. We performed an extensive statistical evaluation and show that our new approach yields accurate local as well as disk-integrated Stokes profiles over a wide range of atmospheric conditions. The mean rms errors for the Stokes I and V profiles are well below 0.2% compared to the exact numerical solution. Errors for Stokes Q and U are in the range of 1%. Our approach does not only offer an accurate approximation to the LTE polarized radiative transfer it, moreover, accelerates the synthesis by a factor of more than 1000., Comment: A&A, in press

Our main objective is to develop a denoising strategy to increase the signal to noise ratio of individual spectral lines of stellar spectropolarimetric observations. We use a multivariate statistics technique called Principal Component Analysis. The cross-product matrix of the observations is diagonalized to obtain the eigenvectors in which the original observations can be developed. This basis is such that the first eigenvectors contain the greatest variance. Assuming that the noise is uncorrelated a denoising is possible by reconstructing the data with a truncated basis. We propose a method to identify the number of eigenvectors for an efficient noise filtering. Numerical simulations are used to demonstrate that an important increase of the signal to noise ratio per spectral line is possible using PCA denoising techniques. It can be also applied for detection of magnetic fields in stellar atmospheres. We analyze the relation between PCA and commonly used well-known techniques like line addition and least-squares deconvolution. 8. Zeeman-Tomography of the Solar Photosphere -- 3-Dimensional Surface Structures Retrieved from Hinode Observations

AIMS :The thermodynamic and magnetic field structure of the solar photosphere is analyzed by means of a novel 3-dimensional spectropolarimetric inversion and reconstruction technique. METHODS : On the basis of high-resolution, mixed-polarity magnetoconvection simulations, we used an artificial neural network (ANN) model to approximate the nonlinear inverse mapping between synthesized Stokes spectra and the underlying stratification of atmospheric parameters like temperature, line-of-sight (LOS) velocity and LOS magnetic field. This approach not only allows us to incorporate more reliable physics into the inversion process, it also enables the inversion on an absolute geometrical height scale, which allows the subsequent combination of individual line-of-sight stratifications to obtain a complete 3-dimensional reconstruction (tomography) of the observed area. RESULTS : The magnetoconvection simulation data, as well as the ANN inversion, have been properly processed to be applicable to spectropolarimetric observations from the Hinode satellite. For the first time, we show 3-dimensional tomographic reconstructions (temperature, LOS velocity, and LOS magnetic field) of a quiet sun region observed by Hinode. The reconstructed area covers a field of approximately 12000 by 12000 km and a height range of 510 km in the photosphere. An enormous variety of small and large scale structures can be identified in the 3-D reconstructions. The low-flux region (B_{mag} = 20G) we analyzed exhibits a number of "tube-like" magnetic structures with field strengths of several hundred Gauss. Most of these structures rapidly loose their strength with height and only a few larger structures can retain a higher field strength to the upper layers of the photosphere., Comment: accepted for A&A Letters

Late-type stars in general possess complicated magnetic surface fields which makes their detection and in particular their modeling and reconstruction challenging. In this work we present a new Zeeman-Doppler imaging code which is especially designed for the application to late-type stars. This code uses a new multi-line cross-correlation technique by means of a principal component analysis to extract and enhance the quality of individual polarized line profiles. It implements the full polarized radiative transfer equation and uses an inversion strategy that can incorporate prior knowledge based on solar analogies. Moreover, our code utilizes a new regularization scheme which is based on local maximum entropy to allow a more appropriate reproduction of complex surface fields as those expected for late-type stars. In a first application we present Zeeman-Doppler images of II Pegasi which reveal a surprisingly large scale surface structure with one predominant (unipolar) magnetic longitude which is mainly radially oriented., Comment: Astronomische Nachrichten / Astronomical Notes Vol. 328, Issue 10, p. 1043

10. The Meso-Structured Magnetic Atmosphere -- A Stochastic Polarized Radiative Transfer Approach

We present a general radiative transfer model which allows the Zeeman diagnostics of complex and unresolved solar magnetic fields. Present modeling techniques still rely to a large extent on a-priori assumptions about the geometry of the underlying magnetic field. In an effort to obtain a more flexible and unbiased approach we pursue a rigorous statistical description of the underlying atmosphere. Based on a Markov random field model the atmospheric structures are characterized in terms of probability densities and spatial correlations. This approach allows us to derive a stochastic transport equation for polarized light valid in a regime with an arbitrary fluctuating magnetic field on finite scales. One of the key ingredients of the derived stochastic transfer equation is the correlation length which provides an additional degree of freedom to the transport equation and can be used as a diagnostic parameter to estimate the characteristic length scale of the underlying magnetic field. It is shown that the stochastic transfer equation represents a natural extension of the (polarized) line formation under the micro- and macroturbulent assumption and contains both approaches as limiting cases. In particular, we show how in an inhomogeneous atmosphere asymmetric Stokes profiles develop and that the correlation length directly controls the degree of asymmetry and net circular polarization (NCP). In a number of simple numerical model calculations we demonstrate the importance of a finite correlation length for the polarized line formation and its impact on the resulting Stokes line profiles.

- Abstract
Cancer-associated fibroblasts (CAF) are key regulators of tumorigenesis. Further insights into the tumor-promoting mechanisms of action of CAFs could help improve cancer diagnosis and treatment. Here we show that the formin mDia2 regulates the positioning and function of mitochondria in dermal fibroblasts, thereby promoting a protumorigenic CAF phenotype. Mechanistically, mDia2 stabilized the mitochondrial trafficking protein MIRO1. Loss of mDia2 or MIRO1 in fibroblasts or CAFs reduced the presence of mitochondria and ATP levels near the plasma membrane and at CAF-tumor cell contact sites, caused metabolic alterations characteristic of mitochondrial dysfunction, and suppressed the secretion of protumorigenic proteins. In mouse models of squamous carcinogenesis, genetic or pharmacologic inhibition of mDia2, MIRO1, or their common upstream regulator activin A inhibited tumor formation. Consistently, co-upregulation of mDia2 and MIRO1 in the stroma of various human cancers negatively correlated with survival. This work unveils a key role of mitochondria in the protumorigenic CAF phenotype and identifies an activin A–mDia2–MIRO1 signaling axis in CAFs with diagnostic and therapeutic potential.

These guidelines are a consensus work of a considerable number of members of the immunology and flow cytometry community. They provide the theory and key practical aspects of flow cytometry enabling immunologists to avoid the common errors that often undermine immunological data. Notably, there are comprehensive sections of all major immune cell types with helpful Tables detailing phenotypes in murine and human cells. The latest flow cytometry techniques and applications are also described, featuring examples of the data that can be generated and, importantly, how the data can be analysed. Nahinfrarotspektroskopie ist eine etablierte Methode zur Qualitätsbestimmung von Obst und Gemüse. Neue Anwendungsgebiete, wie z. B. die mobile Lebensmittelanalyse mittels handgetragener und preisgünstiger Mikrospektrometer, verlangen nach neuen Ansätzen zur Multiprodukt-Kalibrierung. Zur produktspezifischen Kalibrierung existieren bereits geeignete Methoden wie partial least squares regression (PLSR). Der Versuch von Micklander et al. zeigt jedoch auf, dass die Multiprodukt-Kalibrierung noch eine ungelöste Herausforderung darstellt. Nichtlineare Ansätze wie neuronale Netze und lokale Regression erzielten hier bessere Ergebnisse als konventionelle Methoden wie PLSR. Vorläufige Untersuchungen zur Multiprodukt-Kalibrierung zur quantitativen Analyse von Lebensmitteln mittels NIR Spektroskopie lieferten vielversprechende Ergebnisse durch Memory-Based Learning (MBL) und Classification-Prediction-Hierarchy (CPH). In dieser Arbeit werden drei Ansätze zur Multiprodukt-Kalibrierung untersucht. Hierzu werden drei unterschiedliche Apfelsorten, Birnen und Tomaten mit bekanntem Zuckergehalt (in ○Brix) mittels bildgebender NIR Spektroskopie im Bereich von 900 nm bis 2400 nm analysiert. Die Genauigkeit eines linearen PLSR-Modells und zweier nichtlinearer Modelle (CPH und MBL) sowie unterschiedliche Vorverarbeitungsmethoden werden untersucht und evaluiert. Zur Bestimmung von Fehlermaßen dienen Leave-One-Out- und Leave-One-Product-Out-Kreuzvalidierungen.

- Abstract
Notch2 and B cell antigen receptor (BCR) signaling determine whether transitional B cells become marginal zone B (MZB) or follicular B (FoB) cells in the spleen, but it is unknown how these pathways are related. We generated Taok3 -/- mice, lacking the serine/threonine kinase Taok3, and found cell-intrinsic defects in the development of MZB but not FoB cells. Type 1 transitional (T1) B cells required Taok3 to rapidly respond to ligation by the Notch ligand Delta-like 1. BCR ligation by endogenous or exogenous ligands induced the surface expression of the metalloproteinase ADAM10 on T1 B cells in a Taok3-dependent manner. T1 B cells expressing surface ADAM10 were committed to becoming MZB cells in vivo, whereas T1 B cells lacking expression of ADAM10 were not. Thus, during positive selection in the spleen, BCR signaling causes immature T1 B cells to become receptive to Notch ligands via Taok3-mediated surface expression of ADAM10.

From the analysis of a data sample corresponding to an integrated luminosity of 4.63 pb-1 taken during the 1990 run of LEP at centre of mass energies between 88.2 GeV an 94.2 GeV, the tau decays {Mathematical expression} and their charge conjugates have been studied. The following branching ratios have been measured; {Mathematical expression}, {Mathematical expression}, Br(τ- → π- (K-)vτ)=11.9±0.7±0.7%, BR (τ- → ρ- vτ)= 22.4±0.8±1.3%, in good agreement with world averages. The measured electronic and muonic branching ratios lead to a measurement of the strong coupling constant, αs (mτ) = 0.26-0.12 +0.09. Extrapolating the αs value from mτ to mZ yields αs (mZ) = 0.109-0.028 +0.012. The average polarization Pτ of taus produced in Z → τs τs decays has also been measured using the above decay modes. The weighted mean of the polarizations obtained from the four decay modes is Pτ=-0.24±0.07. This value of Pτ gives, in the improved Born approximation, a ratio between the axial and vector coupling constants of the tau of υτ/aτ = 0.12 ± 0.04, and hence a value of the effective electroweak mixing parameter sin2 θW(mZ 2). © 1992 Springer-Verlag., 0, SCOPUS: ar.j, info:eu-repo/semantics/published
