Abstract

Cells need to couple intracellular actin flows with the substrate to generate forward movement. This has traditionally been studied in the context of specific transmembrane receptors, particularly integrin adhesion receptors, which link extracellular adhesive molecules to the actin cytoskeleton. However, leukocytes and other cells can also migrate using integrin-independent strategies both in vivo and in vitro, though the cellular and environmental requirements for this mode are not fully understood. In seminal recent work, Reversat et al.1 develop a range of innovative 2D and 3D engineered microdevices and probe the biophysical mechanisms underlying T lymphocytes and dendritic cells in conditions of limited substrate adhesion. They identify a physical principle of mechano-coupling between retrograde actin flow and irregular extracellular confinement, which allows the cell to generate mechanical resistance and move in the absence of receptor-mediated adhesion. Through the combined use of experiments and theoretical modeling, this work resolves a long-standing question in cell biology and establishes mechanical interaction with an irregular-shaped 3D environment which may be relevant to cell migration in a range of tissue contexts.

Abstract

The rapid increase of transportation infrastructure during the recent decades has caused a number of effects on bird species, including collision mortality, habitat loss, fragmentation and noise. This paper investigates the effects of traffic noise and road-induced fragmentation on breeding bird richness in Cyprus. Cyprus, situated along one of the main migratory routes for birds, has a rich and diverse avifauna threatened by an ever-expanding road network and a road density among the highest in Europe. In this first island-wide study we used data from 102 breeding birds recorded in 10 km x 10 km grid cells. Within every cell we calculated road traffic noise and eight road-related properties. Most of the grid cells are subject to intense fragmentation and traffic noise with combined impact hotspots located even within protected areas (such as Cape Greco, and the Troodos Massif). Results from variance partitioning indicated that road-related properties (total road extent and road length) accounted for a combined 59% of variation in species richness, followed by fragmentation-related properties and noise properties. The study posits the need for further in-depth research on the effects of road networks on birds, and road construction, particularly in protected areas within Mediterranean islands.

Abstract

To investigate the transition from non-cancerous to metastatic from a physical sciences perspective, the Physical Sciences-Oncology Centers (PS-OC) Network performed molecular and biophysical comparative studies of the non-tumorigenic MCF-10A and metastatic MDA-MB-231 breast epithelial cell lines, commonly used as models of cancer metastasis. Experiments were performed in 20 laboratories from 12 PS-OCs. Each laboratory was supplied with identical aliquots and common reagents and culture protocols. Analyses of these measurements revealed dramatic differences in their mechanics, migration, adhesion, oxygen response, and proteomic profiles. Model-based multi-omics approaches identified key differences between these cells' regulatory networks involved in morphology and survival. These results provide a multifaceted description of cellular parameters of two widely used cell lines and demonstrate the value of the PS-OC Network approach for integration of diverse experimental observations to elucidate the phenotypes associated with cancer metastasis.

32. Dysarthria in early Parkinson's disease

Abstract

The aim of the present study was threefold. First, to examine the incidence of dysarthria in patients in the beginning of Parkinson's disease by using a standardised test (Frenchay Dysarthria Assessment/FDA) and an intelligibility assessment tool. Second, to identify differences in speech and in measures of phonation between the Parkinsonian group and a matched control geriatric group using the FDA and electrolaryngography. Finally, to identify the effect of medication on speech and phonation in the dysarthric Parkinsonian group. The results showed that 8 out of 12 (66%) Parkinsonian subjects exhibited lower scores in the FDA compared to controls. Qualitative differences between the two groups were found in the isolated movements of the articulators but not in running speech and speech intelligibility. An improvement in the FDA scoring was found 3-3.5 months after medication. This improvement focused on the areas of tongue and lips and was accompanied with significant increases in intelligibility. No differences in measures of phonation were found either between the two groups or in the same group after medication. The above results suggest that in the beginning of Parkinson's disease, dysarthria is expressed as slowness and may be related to the primary diagnostic symptom of bradykinesia. Due to the small sample and the lack of dosage control, the significance of these findings appears to be inconclusive and warrants further investigation. Future research should employ instrumental quantitative measures on isolated movements of the articulators that may correlate with running speech and will aim to find clinical markers of speech in the diagnosis of Parkinson's disease.
