Contains fulltext : 245643.pdf (Publisher’s version ) (Open Access), BACKGROUND: Genetics play a significant role in coagulation phenotype and venous thromboembolism risk. Resistance to the anticoagulant activated protein C (APC) is an established risk for thrombosis. Herein, we explored the genetic determinants of thrombin generation (TG) and thrombomodulin (TM)-modulated TG using plasma from the Human Functional Genomics Project. METHODS: Calibrated TG was measured both in absence and presence of TM using tissue factor as trigger. Genetic determinants of TG parameters and protein C pathway function were assessed using genome-wide single-nucleotide polymorphism (SNP) genotyping. Plasma samples were supplemented with purified apolipoprotein A-IV, prekallikrein, or kallikrein to test their influence on the anticoagulant function of TM and APC in TG. RESULTS: Thrombin generation data from 392 individuals were analyzed. Genotyping showed that the KLKB1 gene (top SNP: rs4241819) on chromosome 4 was associated with the normalized sensitivity ratio of endogenous thrombin potential to TM at genome-wide level (nETP-TMsr, P = 4.27 × 10(-8) ). In vitro supplementation of kallikrein, but not prekallikrein or apolipoprotein A-IV, into plasma dose-dependently augmented the anticoagulant effect of TM and APC in TG. Variations of rs4241819 was not associated with the plasma concentration of prekallikrein. Association between rs4241819 and nETP-TMsr was absent when TG was measured in presence of a contact pathway inhibitor corn trypsin inhibitor. CONCLUSIONS: Our results suggest that kallikrein plays a role in the regulation of the anticoagulant protein C pathway in TG, which may provide a novel mechanism for the previously observed association between the KLKB1 gene and venous thrombosis.

We have observed, and assigned, the fluorescence excitation spectrum of the 000 band in the 1B1←1A1 electronic transition of pyrimidine, at a resolution of ∼10 MHz. The rotational constants of the 1B1 state, the lowest excited singlet state, are A’=6352±3, B’=5853±3, and C’=3042.0±0.5 MHz. The magnitudes of these constants are not very different from those of the ground (1A1) state. However, the in-plane a and b inertial axes in the 1B1 state are rotated by 90° with respect to those of the 1A1 state. The spectrum also exhibits numerous perturbations, evidenced by the presence of extra lines, anomalous intensities and lifetimes, and shifts of the main lines from their expected positions. The perturbations are strongly magnetic-field dependent, demonstrating that they arise from an intramolecular coupling of the 1B1 state with nearly isoenergetic rovibronic levels of a lower triplet (3B1) state. Models are proposed to account for this behavior based on a deconvolution of the experimental spectrum and simulations of the observed Zeeman effects. The most satisfactory interpretation of the data (in the language of the zero-order states) is obtained if it is assumed that a single rovibronic 1B1 level is spin–orbit coupled to one or a few 3B1 levels, which in turn are coupled via rotationally dependent Coriolis interactions to a dense manifold of background levels, probably those of the 1A1 state. Because the latter coupling is small, typically less than the linewidths in the spectra, it is manifested only in a K’+1 dependence of the lifetimes of selected molecular eigenstates and the reduced g values required to fit the magnetic-field dependence of their spectra. - Abstract
Poland was among the first Eastern European countries to apply for membership of the European Union. In view of Polands tragic history and the economic liberalisation that has taken place since 1989, we believe that the EU should adopt a generous approach toward its application. Unfortunately it looks as if the EU wants Poland to conform with all its laws before it can join. Our research suggests that the EU should pursue an open trade, labour and investment policy. Polands membership will not significantly threaten EU employment. EU companies will not transfer all their industrial activities to Poland. Polish workers will not leave their country en masse. The Polish membership will have a certain impact on the EU budget, particularly in the fields of agriculture, regional and environmental aid but the authors believe that the EU should not cut Polands entitlements. Moreover the EU should not use the long overdue reform of its institutions as a pretext for postponing the Polish membership sine die.

Item does not contain fulltext, Determination of the level of exposure during and after a chemical incident is crucial for the assessment of public health risks and for appropriate medical treatment, as well as for subsequent health studies that may be part of disaster management. Immediately after such an incident, there is usually no opportunity to collect reliable quantitative information on personal exposures and environmental concentrations may fall below detectable levels shortly after the incident has passed. However, many substances persist longer in biological tissues and thus biological monitoring strategies may have the potential to support exposure assessment, as part of health studies, even after the acute phase of a chemical incident is over. Reported successful applications involve very persistent chemical substances such as protein adducts and include those rare cases in which biological tissues were collected within a few hours after an incident. The persistence of a biomarker in biological tissues, the mechanism of toxicity, and the sensitivity of the analysis of a biomarker were identified as the key parameters to support a decision on the feasibility and usefulness of biological monitoring to be applied after an incident involving the release of hazardous chemicals. These input parameters could be retrieved from published methods on applications of biomarkers. Methods for rapid decision making on the usefulness and feasibility of using biological monitoring are needed. In this contribution, a stepwise procedure for taking such a decision is proposed. The persistence of a biomarker in biological tissues, the mechanism of toxicity, and the sensitivity of the analysis of a biomarker were identified as the key parameters to support such a decision. The procedure proposed for decision making is illustrated by case studies based on two documented chemical incidents in the Netherlands.

Smoking and consumer products are indoor sources of BTX, whereas traffic and industrial activities are primary sources contributing to outdoor levels of BTX. The aim of this study was to characterize exposure of children to BTX by personal air sampling using diffusive samplers and by analysis of end-exhaled air. For this study, 101 children of 10-11 years of age were recruited from four primary schools in Southern Turkey during the warm season (May 2008). Two schools were situated in a residential area near primary and secondary iron and steel works (Payas) and two schools were located in a non-industrialized city (Iskenderun). The children and their parents were visited at home for an interview and to identify possible sources of BTX in the residence. Median concentrations of benzene determined by diffusive samplers were higher in Payas (4.1 microg/m(3)) than in Iskenderun (2.7 microg/m(3), p<0.001). For toluene, no differences were observed, whereas for xylene isomers air concentrations tended to be lower for children living in Payas. The median end-exhaled air concentrations were 8.2, 29, 3.8, and 5.7 pmol/L for benzene, toluene, m/p-xylene and o-xylene, respectively (Payas), and 6.9, 25, 4.9, and 6.0 pmol/L, respectively (Iskenderun). Concentrations of toluene in end-exhaled air were 50% higher in children living with household members who smoked indoors (p<0.05) and benzene in end-exhaled air was more than 3-fold higher for those children who were exposed to tobacco smoke inside a vehicle (p<0.001). End-exhaled concentrations of benzene were also higher in children living in a residence with an attached garage (p<0.05). These exposure modifying factors were not identified when using the results obtained with diffusive samplers.

Efficiency wage theories have been put forward as attractive ways of explaining different aspects of the labour market. To find direct evidence of efficiency wage payments has proven to be quite difficult. We model various vertical spillovers from wage determination in an upstream labour market to market share performance in a downstream product market and vice versa. We do this within Sutton''s (1991) general oligopoly model under alternative theories of the labour market. Rent sharing due to efficiency wages is shown to create only a downstream vertical spillover. While rent sharing due to wage bargaining creates a two way vertical spillover. The spillovers due to efficiency wage payments are shown to be the only downstream spillover that drives a positive relationship between a firm''s wage growth and product market performance. Using U.K. firm level panel data we constrain the data with our theory to pinpoint the downstream spillover due to efficiency wage payments. The spillover turns out to be significant and we claim this to be the first direct and general empirical evidence for efficiency wage payments.

This paper uses data from firm level surveys to investigate firm performance for state-owned, privatized and de novo private firms in Romania, Bulgaria and Hungary. - Abstract
The concentration of lead in tibia (Pb-T) was measured in vivo by a 109Cd K-shell X-ray fluorescence technique in 123 workers from a primary lead smelter (age: mean, 45 years; range, 30-61; duration of employment: mean, 20 years; range, 7-45). Their cumulative blood lead index (CBLI) was also calculated on the basis of the blood lead (Pb-B) records available from the company's medical files. Geometric mean for Pb-T was 49 micrograms Pb/g bone mineral (range, 15-167). The company's health surveillance programs, implemented since 1945, resulted in Pb-B values which rarely exceeded 70 micrograms Pb/dl whole blood. Pb-B at the time of Pb-T measurement averaged 31 micrograms Pb/dl (range, 6-62) and the geometric mean for CBLI amounted to 803 micrograms Pb/dl x year (range, 220-2130). Despite various assumptions and uncertainties inherent in the assessment of the cumulative lead dose through Pb-T measurement or CBLI calculation, the relation between both variables in the present lead smelter populations is very strong (rpearson = 0.80, P < 0.0001; age explained at the most 9.5% of the variance). The slope of the regression equation of log Pb-T vs log CBLI showed that a doubling of CBLI also corresponds to doubling of Pb-T. It may be concluded that a sound calculation of CBLI represents a valid surrogate for estimating the life time integrated dose of lead as assessed by the measurement of cortical bone lead (e.g., in tibia).

- Abstract
Unlike inorganic arsenic, inorganic trivalent antimony (Sb) is not methylated in vivo. It is excreted in the bile after conjugation with glutathione and also in urine. A significant proportion of that excreted in bile undergoes an enterohepatic circulation. In workers exposed to pentavalent Sb, the urinary Sb excretion is related to the intensity of exposure. It has been estimated that after eight hours exposure to 500 micrograms Sb/m3, the increase of urinary Sb concentration at the end of the shift amounts on average to 35 micrograms/g creatinine.
