Abstract

We report on the fabrication and characterization of single crystal field-effect transistors (FETs) based on diphenylbenzo diselenophene (DPh-BDSe). These organic field-effect transistors (OFETs) function as p-channel accumulation-mode devices. At room temperature, for the best devices, the threshold voltage is less than -7V and charge carrier mobility is nearly gate bias independent, ranging from 1cm2/Vs to 1.5 cm2/Vs depending on the source-drain bias. Mobility is increased slightly by cooling below room temperature and decreases below 280 K.

Abstract

We report the isolation of a gene encoding a novel member of the family of melanocortin receptors. The mouse melanocortin-5 receptor (mMC5R) responds to melanocortins with an increase in intracellular cyclic 3', 5'-adenosine monophosphate (cAMP) concentrations. Stimulation of the mMC5R by the melanocortins revealed a hierarchy of potency in which [alpha]-melanocyte stimulating hormone ([alpha]-MSH)> [beta]-melanocyte stimulating hormone ([beta]-MSH)> adrenocorticotropic hormone (ACTH)> [gamma]-melanocyte stimulating hormone ([gamma]-MSH). Further structure-activity studies indicated that amino- and carboxyl-terminal portions of [alpha]-MSH appear to be key determinants in the activation of mMC5R whereas the melanocortin core heptapeptide sequence is devoid of pharmacological activity. Northern blot analysis demonstrated the expression of mMC5R mRNA in mouse skeletal muscle, lung, spleen, and brain.

45. Effects of growth factors and gut hormones on proliferation of primary cultured gastric mucous cells of guinea pig.

Abstract

Almost completely homogenous gastric mucous epithelial cells of guinea pigs were grown to confluence in the presence of 10% fetal calf serum (FCS). FCS, epidermal growth factor (EGF), and insulin significantly increased 5-bromo-2'-deoxyuridine (BrdU) uptake by the cells and EGF together with insulin increased the cells' [3H] thymidine uptake. Basic fibroblast growth factor (bFGF) enhanced EGF-induced DNA synthesis by the cells, but vasoactive intestinal peptide (VIP), secretin, prostaglandin E2 (PGE2), and dibutyryl cyclic AMP (dbcAMP) neither induced DNA synthesis nor enhanced the effect of EGF on DNA synthesis by the cells. Gastrin, cholecystokinin-octapeptide (CCK8), and carbamylcholine chloride (CCh) also did not enhance the effect of EGF on DNA synthesis. 125I-EGF, 125I-bFGF, and 125I-gastrin binding to the gastric mucous cells revealed the presence of high-affinity receptors for EGF and bFGF, but not for gastrin. Northern blot analysis showed the expression of EGF receptor mRNA, but not gastrin receptor mRNA. These results suggest that EGF, insulin, and bFGF may cooperatively regulate gastric mucous cell growth, but that gastrin and other gastrointestinal hormones do not have a direct stimulatory effect on mucous cell growth in the guinea pig. 47. Analysis of Ballpoint Pen Inks by Field Desorption Mass Spectrometry

Abstract

Destructive identification of ballpoint pen ink was performed using field desorption mass spectrometry (FDMS) to determine the basic dyes in ballpoint pen ink. Seven different brands of black, blue, and red ballpoint pen inks (total: 21 samples) were examined in this study. A 1-mm section was cut from an ink line drawn on paper and used as the sample. Extraction was performed with methanol. Analysis of each extract by FDMS showed the molecular ion peak of each dye and the black, blue, and red inks were then classified into 6, 6, and 6 types, respectively, based on the ions detected. The results indicated that it was possible to distinguish between manufacturers of ballpoint pens. This analysis of ballpoint pen inks was found to be effective and the method was applied to the analysis of an actual forensic sample.

48. Structural determinants of the melanocortin peptides required for activation of melanocortin-3 and melanocortin-4 receptors.

Abstract

The melanocortins are peptide products of proopiomelanocortin post-translational processing that, among other functions, are thought to influence cognition. Recently, we isolated genes encoding two human melanocortin receptors, the melanocortin-3 receptor (hMC3R) and the melanocortin-4 receptor (hMC4R), which are expressed primarily in brain. We undertook the present studies to examine the structural features of melanocortins that determine activation of these two receptors. For our studies we expressed the coding regions of the hMC3R and hMC4R genes in Hepa cells using the eukaryotic expression vector CMVneo and examined the generation of intracellular cyclic 3',5'-adenosine monophosphate in response to stimulation with various melanocortins. Our findings indicate that the core heptapeptide sequence common to most of the melanocortins (amino acids 4-10 of adrenocorticotropic hormone [ACTH]) is the primary determinant for activation of hMC3R but, in addition, tyrosine2 is necessary for maximal response. Activity of hMC4R is heavily dependent on proline12, but full activity also requires a contribution by tyrosine2. These findings may provide insight into the development of targeted ligands for the brain melanocortin receptors.

50. Somatostatin receptors on rat cerebrocortical membranes. Structural characterization of somatostatin-14 and somatostatin-28 receptors and comparison with pancreatic type receptors.

Abstract

Somatostatin binding and cross-linking to its receptors on rat cerebrocortical membranes were characterized with [125I-Tyr1]somatostatin-14 and [125I-Leu8, D-Trp22, Tyr25]somatostatin-28. When [125I-Tyr1]somatostatin-14 was cross-linked to its receptors with the photoreactive cross-linker, N-(5-azido-2-nitrobenzoyloxy)succinimide, the hormone was specifically associated with a Mr = 72,000 protein band in the presence or absence of reducing agents. Affinity labeling of the Mr = 72,000 protein band was decreased with increasing concentrations of unlabeled somatostatin-14 and nonhydrolyzable guanine nucleotide analog, guanyl-5′-yl imidodiphosphate (Gpp(NH)p). Pretreatment of cerebrocortical membranes with islet-activating protein resulted in a decrease in subsequent labeled somatostatin-14 binding and affinity-labeling of the protein and abolished an inhibitory effect of somatostatin-14 on vasoactive intestinal peptide-stimulated increase in adenylate cyclase activity. When the affinity-labeled protein was solubilized with Zwittergent 3-12 and adsorbed to wheat germ agglutinin-agarose, it was eluted by N-acetylglucosamine. [125I-Leu8, D-Trp22, Tyr25]somatostatin-28 cross-linking to cerebrocortical and pancreatic membranes with the same photoreactive agent revealed specifically labeled protein bands of a Mr = 74,000 in cerebrocortical membranes and a Mr = 94,000 in pancreatic membranes, respectively. These results suggest that: 1) somatostatin receptor on cerebrocortical membranes is a monomeric glycoprotein with a Mr = 70,000 binding subunit, coupled to guanine nucleotide regulatory protein, and 2) the Mr = 70,000 protein may be a common receptor for somatostatin-28 and somatostatin-14 and is distinct from a common pancreatic type receptor.
