Uneven road surfaces are the primary source of excitation in the dynamic interaction between a bridge and a vehicle and can lead to errors in bridge weigh-in-motion (B-WIM) systems. In order to correctly reproduce this interaction in a numerical model of a bridge, it is essential to know the magnitude and location of the various roadway irregularities. This paper presents a methodology for measuring the 3D road surface using static terrestrial laser scanning and a numerical model for simulating vehicle passage over a bridge with a measured road surface. This model allows the evaluation of strain responses in the time domain at any bridge location considering different parameters such as vehicle type, lateral position and speed, road surface unevenness, bridge type, etc. Since the time domain strains are crucial for B-WIM algorithms, the proposed approach facilitates the analysis of the different factors affecting the B-WIM results. The first validation of the proposed methodology was carried out on a real bridge, where extensive measurements were performed using different sensors, including measurements of the road surface, the response of the bridge when crossed by a test vehicle and the dynamic properties of the bridge and vehicle. The comparison between the simulated and measured bridge response marks a promising step towards investigating the influence of unevenness on the results of B-WIM.

Background: Computer users continue to report eye and upper body discomfort even as workstation flexibility has improved. Research shows a relationship between character size, viewing distance, and reading performance. Few reports exist regarding text height viewed under normal office work conditions and eye discomfort., Objective: This paper reports self-selected computer display placement, text characteristics, and subjective comfort for older and younger computer workers under real-world conditions., Methods: Computer workers were provided with monitors and adjustable display support(s). In Study 1, older workers wearing progressive-addition lenses (PALs) were observed. In study 2, older workers wearing multifocal lenses and younger workers were observed., Results: Workers wearing PALs experienced less eye and body discomfort with adjustable displays, and less eye and neck discomfort for text visual angles near or greater than ergonomic recommendations. Older workers wearing multifocal correction positioned displays much lower than younger workers. In general, computer users did not adjust character size to ensure that fovial images of text fell within the recommended range., Conclusions: Ergonomic display placement recommendations should be different for computer users wearing multifocal correction for presbyopia. Ergonomic training should emphasize adjusting text size for user comfort.

A new multifunctional α2,3-sialyltransferase has been discovered in Pasteurella dagmatis. The enzyme, in short PdST, was identified from the P. dagmatis genome by sequence similarity with sialyltransferases of glycosyltransferase family GT-80. In addition to its regioselective sialyltransferase activity (5.9 U/mg; pH 8.0), purified PdST is alternatively active at low pH as α2,3-sialidase (0.5 U/mg; pH 4.5) and α2,3-trans-sialidase (1.0 U/mg; pH 4.5). It also shows cytidine-5'-monophosphate N-acetyl-neuraminic (CMP-Neu5Ac) hydrolase activity (3.7 U/mg; pH 8.0) when no sialyl acceptor substrate is present in the reaction. After sialyltransferase PmST1 from P. multocida, PdST is the second member of family GT-80 to display this remarkable catalytic promiscuity. A unique feature of PdST, however, is a naturally occurring Ser-to-Thr substitution within a highly conserved Y(112)DDGS(116) sequence motif. In PmST1, the equivalent Ser(143) is involved in binding of the CMP-Neu5Ac donor substrate. Reversion of the natural mutation in a T116S-PdST variant resulted in a marked increase in α2,3-trans-sialidase side activity (4.0 U/mg; pH 4.5), whereas the major sialyltransferase activity was lowered (3.8 U/mg; pH 8.0). The Michaelis-Menten constant for CMP-Neu5Ac was decreased 4-fold in T116S mutant when compared with wild-type PdST (KM=1.1 mM), indicating that residue 116 of PdST contributes to a delicate balance between substrate binding and catalytic activity. D-Galactose and various β-D-galactosides function as sialyl acceptors from CMP-Neu5Ac, whereas other hexoses (e.g. D-glucose) are inactive. Structure comparison was used to rationalize the particular acceptor substrate specificity of PdST in relation to other GT-80 sialyltransferases that show strict α2,3-regioselectivity, but are flexible in using α/β-galactosides for sialylation.

The effect of neuroleptics on phospholipase A2 (PLA2) activity in rat brain plasma membranes was studied. Chlorpromazine (10 mg/kg), fluphenazine (5 mg/kg), thioridazine (5 mg/kg), trifluoperazine (5 mg/kg), haloperidol (2 mg/kg), and sulpiride (100 mg/kg) were administered to rats intraperitoneally as a single dose or long-term treatment (4 weeks). The PLA2 activity was determined 24, 48, and 72 h after the last injection of a drug. The enzyme activity was decreased after a single or 4-week administration of chlorpromazine, trifluoperazine, haloperidol, and sulpiride. Fluphenazine and thioridazine caused an increase of PLA2 activity in rat brain both after a single dose and long-term administration. For the first time it was shown that neuroleptics cannot only inhibit but also increase, PLA2 activity. Elucidation of this fact requires further studies.
