Since the renewable sources, which have gained great attention due to the low-carbon policies, are inherently intermittent, the conventional power generation systems will be in use to meet the power demand. These systems, however, must be capable of operating along with renewables, which will lead to a need for more operational flexibility with frequent system ramps. Therefore, understanding and control of thermal stresses and clearances are essential for improving flexibility of conventional power plants. Computational fluid dynamics tools are of great importance in predicting the turbomachinery flows design since the direct measurements of detailed and spatial flow and temperature distribution are often not trivial in the real engines. During shut-down regimes of steam turbines, natural convection takes place along with relatively weak forced convection which is not strong enough to prevent a rising thermal plume leading to a non-uniform cooling in the turbine cavities. Although natural and forced convection have been studied separately in the literature, mixed type of flows in turbine cavities have not been investigated extensively. This paper provides unique experimental data set for validation and development of the predictive tools, which is generated from the detailed flow field measurements in a test facility designed for mixed type of flows in the turbine casing cavities with engine representative conditions. Additionally, large eddy simulations have been performed and validated against the generated experimental data, to gain deeper insight into the flow field. Thus, this paper offers a great insight in these complex flow interactions and unique experimental data for enabling the flexible operations and the development of advanced turbulence modelling.

Antimicrobial resistance (AMR) of bacterial pathogens, including enterococci, is a global concern, and plasmids are crucial for spreading and maintaining AMR genes. Plasmids with linear topology were recently identified in clinical multidrug-resistant enterococci. The enterococcal linear-form plasmids, such as pELF1, confer resistance to clinically important antimicrobials, including vancomycin; however, little information exists about their epidemiological and physiological effects. In this study, we identified several lineages of enterococcal linear plasmids that are structurally conserved and occur globally. pELF1-like linear plasmids show plasticity in acquiring and maintaining AMR genes, often via transposition with the mobile genetic element IS1216E. This linear plasmid family has several characteristics enabling long-term persistence in the bacterial population, including high horizontal self-transmissibility, low-level transcription of plasmid-encoded genes, and a moderate effect on the Enterococcus faecium genome alleviating fitness cost and promoting vertical inheritance. Combined with its broad host range, the linear plasmid is an important factor in the spread and maintenance of AMR genes among enterococci.

Vancomycin-resistant Enterococcus faecium carrying the vanD1 gene on plasmid pEF-D was isolated from a fecal sample of a hospitalized patient in Japan. The strain JH5687 showed moderate resistance to vancomycin (MIC, 16 μg/mL) but remained susceptible to teicoplanin (MIC, 1 μg/mL).

We investigate the Reynolds analogy over riblets, namely the analogy between the fractional increase in Stanton number $C_h$ and the fractional increase in the skin-friction coefficient $C_f$ , relative to a smooth surface. We investigate the direct numerical simulation data of Endrikat et al. (Flow Turbul. Combust., vol. 107, 2021, pp. 1–29). The riblet groove shapes are isosceles triangles with tip angles $\alpha = {30}^{\circ }, {60}^{\circ }, {90}^{\circ }$ , a trapezoid, a rectangle and a right triangle. The viscous-scaled riblet spacing varies between $s^+ \approx 10$ to $60$ . The global Reynolds analogy is primarily influenced by Kelvin–Helmholtz rollers and secondary flows. Kelvin–Helmholtz rollers locally break the Reynolds analogy favourably, i.e. cause a locally larger fractional increase in $C_h$ than in $C_f$ . These rollers induce negative wall shear stress patches which have no analogue in wall heat fluxes. Secondary flows at the riblets’ crests are associated with local unfavourable breaking of the Reynolds analogy, i.e. locally larger fractional increase in $C_f$ than in $C_h$ . Only the triangular riblets with $\alpha = {30}^{\circ }$ trigger strong Kelvin–Helmholtz rollers without appreciable secondary flows. This riblet shape globally preserves the Reynolds analogy from $s^+ = 21$ to $33$ . However, the other riblet shapes have weak or non-existent Kelvin–Helmholtz rollers, yet persistent secondary flows. These riblet shapes behave similarly to rough surfaces. They unfavourably break the global Reynolds analogy, and do so to a greater extent as $s^+$ increases.

A two-phase ejector is a device to induce a suction flow without pump or electricity. The flow in the two-phase ejector consists of a drive flow and a suction flow. As the driving flow expands blowing out of a drive flow nozzle, the thermal energy potential is converted into momentum, and by giving it to the suction flow, it is possible to induce the flow without using external power. In a nuclear power plant, a two-phase ejector can be utilized as a device to drive coolant flow in the cases of power failure. Mixing of the drive flow and the suction flow accompanied with evaporation or condensation at the gas-liquid interface depends on thermal hydraulic parameters and flow rate, and it is necessary to control them to maintain the driving force, but it can easily come out of operation range with a slight change in balance. There is little knowledge about heat and mass transfer to find and design operating conditions and ejector configurations. In this study, a heat and mass transfer model of the gas-liquid interface in a critical two-phase flow was developed. To handle thermally non-equilibrium two-phase flow with phase changes occurring simultaneously at the interface, we implemented constitutive equations into CFD tool, such as a correlation for interfacial area concentrations, and we evaluated evaporation coefficient, which is an important parameter to determine the phase change rate, based on the physical property of the working fluid. The CFD simulation method was validated using the experimental data in the literature of a two-phase ejector. In the validation, the flow rates of the drive flow and the suction flow, and pressure distribution inside the ejector were compared. Then, the validity of the developed CFD simulation method have confirmed.

Thymidine-dependent small-colony variant (TD-SCV) of Escherichia coli was isolated from urine of a septuagenarian female patient on hemodialysis suffering from recurrent cystitis. The patient had been treated with frequent administrations of trimethoprim sulfamethoxazole (SXT), every time her cystitis symptoms developed. In the TD-SCV isolate, the deletion was detected in the thyA gene associated with thymidylate synthase. Interestingly, the isolate was found to produce extended-spectrum β-lactamase (ESBL), and the experiment on conjugational transfer of the resistance trait was successful. By means of genetic analysis, the isolate was found to carry blaCTX-M-1 group. To the best of our knowledge, this is the first report of urinary tract infection caused by the transmissible ESBL-producing TD-SCV of E. coli. MICs of the TD-SCV were obtained only on the Mueller Hinton agar media supplemented with appropriate concentrations of thymidine, which might lead to the difficulty for proper chemotherapy in daily medicine. Furthermore, transmission of the ESBL gene via plasmid should be of concern.

Background VanD is a rare type of vancomycin resistance worldwide. However, the host diversity of the vanD gene cluster and the structural similarity of their genomic islands are not well understood. Methods Three VanD-type Enterococcus faecium strains (AA620, AA622 and AA624) isolated from a Japanese patient who underwent vancomycin treatment in 2017 were analysed. This study utilized WGS analysis to characterize the three VanD-type E. faecium strains and describes the diversity of hosts possessing VanD-carrying genomic islands. Results The three isolates exhibited variable MICs of vancomycin. In the relatively vancomycin-resistant AA620, mutations were identified in vanSD and ddl. The strains AA622 and AA624 had intact ddl and harboured two vanD gene clusters. qRT-PCR results revealed the ddl mutation to be a factor affecting the high vancomycin resistance range of AA620. WGS data showed the 155 kb and 185 kb genomic islands harbouring the vanD gene cluster inserted in the coding region of the lysS gene, located in the chromosome in AA620 and AA622/624, respectively. Comparing the VanD-carrying genomic islands to available sequences of other enterococci and enteric anaerobes revealed how the genomic islands of these organisms isolated worldwide shared similar core genes and backbones. These anaerobes belonged to various genera within the order Eubacteriales. The phylogenetic cluster of the genomic island core genome alignment did not correlate with the host-species lineage, indicating horizontal gene transfer in the gut microbiota. Conclusions By horizontal gene transfer, various bacteria forming the gut microbiota maintain VanD-carrying genomic islands.

Five novel strains of Serratia fonticola that produce FONA, a minor extended spectrum beta-lactamase (ESBL), were isolated during the routine surveillance of ESBLproducing Enterobacteriaceae in imported chicken meat in Japan in 2017 and 2018. These strains exhibited a clear ESBL phenotype in susceptibility tests carried out in the presence of clavulanic acid; however, they were all negative in the multiplex polymerase chain reaction assay used to detect the TEM, SHV and CTX-M β-lactamase genes. After identification of the bacterial species as S. fonticola, full blaFONA genes were amplified and the DNA sequences were determined. The blaFONA genes from all five strains were different from those previously reported (blaFONA-1 to blaFONA-6); they clustered closely with one another but were distinct from previously reported blaFONA genes in a phylogenic analysis based on amino acid sequences.

This article shows the first parametric study on turbulent multi-jet impingement cooling flows using large-eddy simulations (LES). We focus on assessing the influence of the inter-jet distance and the cross-flow conditions on the heat transfer at the impingement wall. The LES setup is thoroughly validated with both experimental and direct numerical simulation data, showing an excellent agreement. The inter-jet distance effect on the heat transfer is studied comparing three different distances, where the full Nusselt number profile decreases in amplitude when the jet distance is increased. To evaluate the cross-flow effects, we prescribe both laminar and turbulent inflow conditions at different cross-flow magnitudes ranging between 20% and 40% of the impinging jet speed. Large cross-flow intensities cause a jet deflection which reduces the maxima in the Nusselt number distribution, and it increases the heat transfer in the areas of the wall less affected by the jet impingement. Adding realistic turbulent fluctuations to the inflow enhances the cross-flow effects on the heat transfer at the impingement wall.

Two-phase flows are observed in various industrial plants and piping systems. Understanding two-phase flow behaviors such as flow patterns and unsteady void fraction in horizontal and vertical pipes are crucial in improving plant safety. Notably, the flow patterns observed in a large diameter pipe (approx. 4–6 in or larger) are significantly different from those observed in a medium diameter pipe. In a vertical large diameter pipe, no slug flow is observed due to the instantaneous slug bubble breakup caused by the surface instability. Besides, in a horizontal pipe, flow regime transition from stratification of liquid and gas to slug (plug) flow that induces unsteady flow should be taken into account. From this viewpoint, it is necessary to predict the flow regime in horizontal and vertical large diameter pipes with some elbows and to evaluate the unsteady flow regime. In this study, the simulation method based on the two-fluid model is developed. The two-fluid model is considered the most accurate model because the governing equations for mass, momentum, and energy transfer are formulated for each phase. When using the two-fluid model, some constitutive equations should be given in computing the momentum transfer between gas and liquid phases. In this study, several state-of-art constitutive equations of the bubble diameter, the interfacial drag force and non-drag forces such as the lift force and the bubble-bubble collision force, are implemented in the platform of ANSYS FLUENT. The developed simulation method is validated with visualization results and force from an air-water flow at the elbow of the piping system.

The aim of this work is to improve numerical predictions of natural convection heat transfer inside of gas turbines. It is desirable for engineers to use numerical simulations be able to accurately predict the heat transfer inside of a gas turbine casing during engine shutdown. This will help them to understand and alleviate problems such as engine casing distortion. A natural convection test rig was constructed to provide simulation validation data. The rig represented a section of a large gas turbine casing and operated at the same Rayleigh number as a real engine. The casing rig was able to provide a unique dataset that could not be obtained through real engine measurements. Analysis of the casing rig was performed using a large eddy simulation and RANS simulations. It was found that the baseline RANS simulation did not predict the heat transfer accurately enough for engineering use. The simple gradient diffusion hypothesis (SGDH) turbulent heat flux model used in the baseline simulations was replaced by more advanced generalised gradient diffusion hypothesis (GGDH). A modification to the GGDH model, called GGDH+, was developed to account for buoyancy effects on the turbulent heat flux. The GGDH+ model was then able to give heat transfer predictions comparable to the LES results.

Due to an increase in the power generation from renewable sources, steam and gas turbines will be required to adapt for more flexible operations with frequent start-ups and shut-downs to provide load leveling capacity. During shut-down regimes, mixed convection takes place with natural convection dominance depending on the operating conditions in turbine cavities. Buoyant flows inside the turbine that are responsible for nonuniform cooling leading to thermal stresses and compromise clearances directly limits the operational flexibility. Computational fluid dynamics (CFD) tools are required to predict the flow field during these regimes since direct measurements are extremely difficult to conduct due to the harsh operating conditions. Natural convection with the presence of cross-flow -mixed convection has not been extensively studied to provide detailed measurements. Since the literature lacks of research on such flows with real engine representative operating conditions for CFD validation, the confidence in numerical predictions is rather inadequate. This paper presents a novel experimental facility that has been designed and commissioned to perform very accurate unsteady temperature and flow field measurements in a simplified turbine casing geometry. The facility is capable of reproducing a wide range of Richardson, Grashof, and Reynolds numbers which are representative of engine realistic operating conditions. In addition, high fidelity, wall resolved large eddy simulation (LES) with dynamic Smagorinsky subgrid scale model has been performed. The flow field as well as heat transfer characteristics have been accurately captured with LES. Finally, the inadequacy of Reynolds-averaged Navier–Stokes (RANS) for the mixed type of flows has been highlighted.

AST-120 (Kremezin) is used to treat progressive chronic kidney disease by adsorbing uremic toxin precursors produced by the gut microbiota, such as indole and phenols. Previously, we found that AST-120 decreased drug tolerance and virulence in Escherichia coli by adsorbing indole. Here, we show that AST-120 adsorbs phenazine compounds, such as pyocyanin, produced by Pseudomonas aeruginosa including multidrug-resistant P. aeruginosa strains, and suppresses pyocyanin-associated toxicity in A-549 (alveolar adenocarcinoma) and Caco-2 (colon adenocarcinoma) cells. Addition of fosfomycin, colistin and amikacin, which are often used to treat P. aeruginosa, inhibited the bacterial growth, regardless of the presence or absence of AST-120. These results suggest a further benefit of AST-120 that supports anti-Pseudomonas chemotherapy in addition to that of E. coli and propose a novel method to treat P. aeruginosa infection.

Convective heat transfer in rough wall-bounded turbulent flows is prevalent in many engineering applications, such as in gas turbines and heat exchangers. At present, engineers lack the design tools to accurately predict the convective heat transfer in the presence of non-smooth boundaries. Accordingly, a new turbulent boundary layer facility has been commissioned, where the temperature of an interchangeable test surface can be precisely controlled, and conductive heat losses are minimized. Using this facility, we can estimate the heat transfer coefficient (Stanton number, St), through measurement of the power supplied to the electrical heaters and also from measurements of the thermal and momentum boundary layers evolving over this surface. These methods have been initially investigated over a shorter smooth prototype heated surface and compared with existing St prediction models. Preliminary results suggest that we can accurately estimate St in this facility.

Sub-MICs of the 14-membered macrolides erythromycin (EM) and clarithromycin (CAM) decreased the growth of Pseudomonas aeruginosa PAO1 and increased its sensitivity to endogenous and exogenous nitrosative stress. However, a 16-membered macrolide, josamycin (JM), was not or less effective. In 9 of 13 non-multidrug-resistant P. aeruginosa (non-MDRP) and 9 of 27 MDRP ST235 strains, the sub-MIC of EM induced significant reductions in bacterial numbers following treatment with a nitric oxide donor.

Background VanB-type vancomycin (VAN) resistance gene clusters confer VAN resistances on Enterococcus spp. over a wide range of MIC levels (MIC = 4–1000 mg/L). However, the epidemiology and the molecular characteristics of the VAN susceptible VanB-type Enterococcus still remain unclear. Results We characterized 19 isolates of VanB-type Enterococcus faecium that might colonize in the gut and were not phenotypically resistant to VAN (MIC = 3 mg/L). They were obtained from two hospitals in Japan between 2009 and 2010. These isolates had the identical vanB gene cluster and showed same multilocus sequence typing (MLST) (ST78) and the highly related profiles in pulsed-field gel electrophoresis (PFGE). The vanB gene cluster was located on a plasmid, and was transferable to E. faecium and E. faecalis. Notably, from these VanB-type VREs, VAN resistant (MIC≥16 mg/L) mutants could appear at a frequency of 10− 6–10− 7/parent cell in vitro. Most of these revertants acquired mutations in the vanSB gene, while the remainder of the revertants might have other mutations outside of the vanB gene cluster. All of the revertants we tested showed increases in the VAN-dependent expression of the vanB gene cluster, suggesting that the mutations affected the transcriptional activity and increased the VAN resistance. Targeted mutagenesis revealed that three unique nucleotide substitutions in the vanB gene cluster of these strains attenuated VAN resistance. Conclusions In summary, this study indicated that stealthy VanB-type E. faecium strains that have the potential ability to become resistance to VAN could exist in clinical settings. The emergence of drug-resistant bacteria has led to the high demand for new antibiotics. In this report, we investigated membrane-active antimicrobial β-cyclodextrins. These contain seven amino-modified alkyl groups on a molecule, which act as functional moieties to permeabilize bacterial cell membranes. The polyfunctionalization of cyclodextrins was achieved through a click reaction assisted by microwave irradiation. A survey using derivatives with systematically varied functionalities clarified the unique correlation of the antimicrobial activity of these compounds with their molecular structure and hydrophobicity/hydrophilicity balances. The optimum hydrophobicity for the compounds being membrane-active was specific to bacterial strains and animal cells; this led to specific compounds having selective toxicity against bacteria including multidrug-resistant pathogens. The results demonstrate that cyclodextrin is a versatile molecular scaffold for rationally designed structures and can be used for the development of new antibiotics.

Emergence of drug-resistant bacterial pathogens and the concurrent demand for new antibiotics has led to membrane-active antimicrobial cyclodextrin (CD) development. CDs contain polyalkylamino groups; molecule polyfunctionalization was achieved via a click reaction. A survey using CDs with systematically varied functionalities clarified the unique correlation of their antimicrobial activity with the molecules’ hydrophobicity/hydrophilicity balance. The optimum hydrophobicity of the membrane-active molecule was specific to bacterial strains and animal cells, leading to selective toxicity against bacteria including multidrug-resistant bacteria such as methicillin-resistant Staphylococcus aureus. The results demonstrate that CDs have a good molecular scaffold to pursue rationally designed structures with a desired activity for new antibiotic development.

An investigation was conducted into the formation and development of streamwise thermal vortices, which were observed in a natural convection test rig. The formation mechanism of these vortices and the mechanism which determines the vortex wavelength is not well understood. A numerical investigation into these mechanisms using Large Eddy Simulations was conducted. A method for the formation of the streamwise vortices has been proposed. The mechanism involved the interaction of an incoming boundary layer vortex with buoyancy forces from a heated wall. This interaction causes the incoming boundary layer vortex to roll up into counter-rotating vortex pairs. Studies to understand factors affecting the wavelength of the vortices were conducted. One study looked at the effect of different wall temperatures on the wavelength. It was found the variation in wavelength with temperature agreed well with existing experimental data. Another study looked at the effect of fluid properties on the wavelength. This study involved simulations that doubled or halved the fluid viscosity and thermal conductivity. It was then shown that the variation in wavelength with temperature was not due to changes in fluid properties with temperature.

A STRUCTure-based (STRUCT) second generation (2G) URANS turbulence model is presented in this paper and assessed on the simulation of unsteady turbulent flow, with application to thermal striping evaluation in T-junctions. The closure methodology leverages the robustness of the Unsteady Reynolds-Averaged Navier-Stokes (URANS) formulation and enhances it with the addition of physics-based local resolution of flow structures. Numerical predictions of the velocity and temperature fields for the STRUCT model compare favorably with experimental data, where the main inlet Reynolds number ranges from 8790 to 16,300. Spectral analyses of the temperature signals are also performed in order to investigate the mechanism of high-cycle thermal fatigue in the flow mixing region. The computed spectra from the STRUCT solution agree with the reference LES calculations and experimental data, accurately reproducing both shape and magnitude of the power spectra density. Overall, the STRUCT model demonstrates significant potential to accurately and efficiently model the thermal striping phenomenon. Its application produces LES quality results, while the computational cost is comparable to the URANS solution.

The classical gradient-diffusion hypothesis has known deficiencies when applied to cooling applications. In this paper, the gene-expression programming (GEP) method, a machine learning approach, has been applied to develop scalar-flux models via symbolic regression. The scalar-flux, the unclosed term of the mean passive-scalar transport equation, is treated by considering the polynomial basis and scalar invariants available from computable Reynolds-averaged quantities. This method has been applied to develop and then assess a model for the test case of jet in crossflow. A high-fidelity database was first probed for insight into which of the candidate bases are the most suitable as modelling terms. The high dimensionality of the function space, spanned by the basis, was then reduced by basic statistical techniques. The resulting data-driven model is presented and tested for a range of different jet in crossflow cases. Compared with eddy-diffusivity models, the new model is shown to produce reliably more accurate results. This demonstrates that the current framework can be used for scalar-flux modelling in complex three-dimensional flows and has potential to provide generalized form closures with improved predictive accuracy for the same classes of flows they were trained on.

We here report a case of Dysgonomonas mossii bacteremia with cholangitis. An 85-year-old male patient with recurrent hepatitis B surface antigen-negative/anti-hepatitis C virus-negative hepatocellular carcinoma came to our hospital in poor physical condition. Two sets of blood cultures revealed a positive result for D. mossii. As matrix-assisted laser desorption/ionization time-of-flight mass spectrometry failed to identify D. mossii, analysis of 16S rRNA gene sequencing was performed; however, this gene is not specific enough to distinguish between D. mossii and D. oryzarvi. Finally, D. mossii infection was confirmed by gyrB and recA sequencing. To our knowledge, this is the first report of D. mossii causing human infection, which was identified in culture and confirmed using a combination of 16S rRNA, gyrB, and recA sequencing.

In nuclear plant piping, when the high-temperature water penetrates from main pipe, a thermal stratification interface may be formed in branch pipe with a closed end. The penetrating flow of high-temperature water is called a cavity flow. If a thermal stratification interface is formed in an elbow of vertical-horizontal branch pipes, thermal fatigue may occur by a periodic temperature fluctuation. Because high cycle thermal fatigue may have significant influence on structure integrity of pipe, it is important to evaluate the position which is called penetration depth of a cavity flow in which a thermal stratification interface is formed. In present evaluation guideline which was formulated by the Japan Society of Mechanical Engineers, applicable piping is limited only to 50 mm diameter of vertical-horizontal branch pipe. Therefore, it needs to expand the applicable diameter range of vertical-horizontal branch pipe. In this research, 200 mm diameter piping test is conducted to confirm characteristic phenomena, and to build evaluation method of penetration depth in large diameter piping. This paper presents about modeling of evaluation method for cavity flow penetration depth in large diameter piping. First, the mass transport model at the interface of layer in natural circulation region is arranged with Reynolds number and Richardson number based on visualization and the temperature measuring result. Second, the mass transport model at a thermal stratification interface between a cavity flow domain and a natural circulation domain is built based on time variation of lithium tracer concentration. The evaluation model of cavity flow penetration depth applicable to large diameter piping is proposed by combining new mass transportation models and existing evaluation guideline. In conclusion, predictive accuracy of cavity flow penetration depth in 200 mm diameter piping became the same order as the evaluation result of 50 mm diameter piping by using present evaluation guideline.

Purpose. The decline in methicillin-resistant Staphylococcus aureus (MRSA) isolation rates has become a general observation worldwide, including Japan. We hypothesized that some genetic shift in MRSA might cause this phenomenon, and therefore we investigated the genetic profiles among MRSA clinical isolates obtained from three different epidemic phases in Japan. Methodology. A total of 353 MRSA isolates were selected from 202 medical facilities in 1990 (pre-epidemic phase), 2004 (epidemic phase) and 2016 (post-epidemic phase). Molecular typing was performed by PCR detection of 22 genes using the polymerase chain reaction (PCR)-based ORF typing (POT) system, including an additional eight genes including small genomic islets and seven toxin genes. Results. Isolates with a POT1 of score 93, identified as presumed clonal complex (pCC)5-staphylococcal cassette chromosome mec (SCCmec) type II including ST5-SCCmec type II New York/Japan clone, represented the major epidemic MRSA lineage in 1990 and 2004. In 2016, however, a marked decrease in isolates with a POT1 score of 93, along with changes in the epidemiology of toxin genes carried, was noted, where the carriers of tst genes including the tst-sec combination were markedly reduced, and those possessing the seb gene alone were markedly increased. Rather, isolates with a POT1 score of 106, including pCC1 or pCC8 among the isolates with SCCmec type IV, which often links to community-associated MRSA, were predominant. Interestingly, the pCC1 and pCC8 lineages were related to sea and tst-sec carriage, respectively. Conclusions. Over time, a transition in MRSA genetic profiles from a POT1 score of 93 in 1990 and 2004 to 106 in 2014 was found in Japan.

Dry-out phenomena in packed beds or porous media may cause a significant digression of cooling/reaction performance in heat transfer/chemical reactor systems. One of the phenomena responsible for the dry-out in packed beds is known as the counter-current flow limitation (CCFL). In order to investigate the CCFL phenomena induced by gas–liquid two-phase flow in packed beds inside a pool, a natural circulation packed bed test facility was designed and constructed. A total of 27 experimental conditions covering various packing media sizes (sphere diameters: 3.0, 6.4 and 9.5 mm), packed bed heights (15, 35 and 50 cm) and water level heights (1.0, 1.5 and 2.0 m) were tested to examine the CCFL criteria with adiabatic air–water two-phase flow under natural circulation conditions. The emergence of drug-resistant bacteria has led to the high demand for new antibiotics. In this report, we investigated membrane-active antimicrobial β-cyclodextrins. These contain seven amino-modified alkyl groups on a molecule, which act as functional moieties to permeabilize bacterial cell membranes. The polyfunctionalization of cyclodextrins was achieved through a click reaction assisted by microwave irradiation. A survey using derivatives with systematically varied functionalities clarified the unique correlation of the antimicrobial activity of these compounds with their molecular structure and hydrophobicity/hydrophilicity balances. The optimum hydrophobicity for the compounds being membrane-active was specific to bacterial strains and animal cells; this led to specific compounds having selective toxicity against bacteria including multidrug-resistant pathogens. The results demonstrate that cyclodextrin is a versatile molecular scaffold for rationally designed structures and can be used for the development of new antibiotics.

New colony multiplex PCR assays for detection of seven types of vancomycin-resistance determinants and eight types of Enterococcus species were developed. For 135 enterococcal isolates examined in this study, these assays showed high sensitivity and specificity, and could provide the rapid and accurate detection of vancomycin-resistant determinants and Enterococcus spp.

Bacterial infections to anaerobic site are often hard to be treated because the activity of most of antimicrobials decreases under anaerobic conditions. However, fosfomycin rather provides a greater activity under anaerobic conditions than aerobic conditions. Previously, we found that expression of glpT and uhpT, fosfomycin symporters in enterohaemorrhagic Escherichia coli (EHEC) was upregulated by FNR, a global regulator during the anaerobiosis of the bacterium, which led to increased uptake and susceptibility to this drug. In this study, we showed that expression of glpT and uhpT is induced by CRP-cAMP, the regulator complex under both aerobic and anaerobic conditions. The activity of CRP-cAMP in EHEC was elevated under anaerobic conditions because levels of both CRP and cAMP were higher in the cells when grown anaerobically than those when grown aerobically. Results of expression study using mutants indicated that CRP-cAMP is indispensable for expression of glpT but not uhpT—whereas that of uhpT requires UhpA that is the response regulator composing of two-component system with the sensor kinase, UhpB. The CRP-cAMP protein bound to a region that overlaps RNA polymerase binding site for glpT and region upstream of UhpA binding site for uhpT. FNR bound to a region further upstream of CRP-cAMP binding site on region upstream of the glpT gene. These combined results suggested that increased antibacterial activity of fosfomycin to EHEC under anaerobic conditions is due to activation of FNR and increment of CRP-cAMP activity. Then, FNR enhances the expression of glpT activated by CRP-cAMP while CRP-cAMP and FNR cooperatively aids the action of UhpA to express uhpT to maximum level.
