Tomato plants are widely grown in temperate climates worldwide and are mostly cultivated as annuals. The objective of this study was to understand the interrelation between fruit quality of tomato, some soil biological parameters, and the addition of increasing biochar (BC) soil amendment doses. BC is an industrial product, made from organic waste by pyrolysis. Its use in the soil is known to improve fertility and several soil functions. Among organic, ecological conditions, a field experiment was performed in a type of slightly humous arenosol soil. Effect of increasing doses of biochar (BC) (0.5-, 1.0-, 2.5-, 5.0, 10 m/m% and control) was studied. Nutrient content and Total Soluble Solid (TSS) of the fruits, the ripeness, and the marketable/non-marketable ratio of yield were assessed. The presence of some cultivable microbial physiological groups (fungi, bacteria) and the soil-dehydrogenase activity (DHA) was estimated. Results represented that the changes of fruit TSS content was not linear with the increasing doses of BC. The increased yield (+53%) had an inverse correlation with the TSS content of the berry's pulps, and the content was lowest at the highest BC dose. Optimum doses of BC were considered, like 1–2.5 m/m%, supported by the nutritive element content (+55% N, +76% P, +83% K) and enhanced microbial activities (+45% DHA). Grouping the parameters by Pearson Correlation Coefficient, the biochar amendment was a driving factor for tomato growth, with certain dose limits in the studied organic agricultural practice.

Biologically active compounds, sugars, acids and antioxidants are key-important ingredients of healthy tomato food. Objective of the study is to investigate if specific industrial bioeffector products are improving the taste and some quality parameters of tomato fruits and how those results are appearing in pots and among organic field conditions? Spore-forming industrial Bacillus amyloliquefaciens FZB42 (RhizoVital) as single inoculums and combinations either with other Bacillus strains (Biorex-1), or with N2-fixing-siderophore-producing strains (Biorex-2) were applied on Solanum lycopersicon Mill. var. Mobil test plant. Soil microbial counts, phosphorus availability and fruit quality, such as total soluble solids (TSS), content of some essential organic acids (citric-, maleic) and sugars (glucose, fructose) were assessed. The results found, that single industrial inoculums of FZB42 product had positive effect on Pavailability and fruit quality in the pots, combinations of other biofertilizers, however, did not give additional results. Fruit quality parameters, TSS content, soluble sugars and organic acids were significantly improved (p<0.05) at both experimental conditions. Such better fruit taste was correlated significantly by the most probable number (MPN) total microbial counts with greater positive values in pot experiment, compared with more variable environmental field condition. The recommendation of using such bioeffector products is supported by the positive interrelation among measured soil characteristics and inside healthy quality parameters of organically grown tomato fruits. Meningitis caused by Listeria monocytogenes is a rare disease in immunocompetent individuals, however, in the presence of certain risk factors with clinical signs indicating infection of the central nervous system it should not be ignored. In this case report, we present the medical history of a 72-year-old man, suffering from hypertension and liver cirrhosis, who was diagnosed with meningitis caused by L. monocytogenes. The patient was admitted to our Department with the symptoms of weakness, dizziness, high fever and urinary incontinence. Laboratory tests showed elevated inflammatory and liver enzyme values as well as low white blood cell and platelet counts were confirmed. Imaging tests did not prove any abnormalities. Due to septic parameters, after microbiological samples were collected, empiric ceftriaxon and metronidazol treatment was started. Despite our therapeutic efforts, the condition of the patient had not improved significantly. The patient still suffered from high fever; increasing agitation and tremor, coordination disorder appeared, raising the possibility of a bacterial infection of the central nervous system. Examination of the cerebrospinal fluid confirmed the diagnosis of bacterial meningitis. In the meantime, findings of microbiological cultures verified the infection of L. monocytogenes, however, cerebrospinal fluid culture did not detect any pathogen. Following that, the therapy continued with targeted ampicillin which resulted in rapid improvement of the patient's condition; fevers and neurological symptoms have ceased to exist. We considered the case worthy of presentation because of the pitfalls of the diagnosis, the emerging differential diagnostic difficulties and the favorable outcome due to the effectiveness of targeted antibiotic treatment. 36. Characterisation of Luvisols Based on Wide-Scale Biological Properties in a Long-Term Organic Matter Experiment.

Soil organic matter is a biological system that functions as an integrated whole. These assemblies have different properties, functions, and decomposition times. SOM is one of the main determinants of soil productivity. Our studies were carried out in a temperate deciduous oak forest on Luvisols soil. In the DIRT Project (Detritus Input and Removal Treatments), the following treatments were applied: Double Litter, Double Wood, Control, No Litter, No Root and No Input. Our objective was to compare the effect of withdrawal or doubling of organic matter on the protein pattern of the soil and the biological activity and changes in labile C (permanganate-oxidizable carbon) content in a long-term organic matter manipulation experiment. Patterns of thermostable proteins, soil dehydrogenase enzyme activity, CO
2 emission, and POXC content were measured at the most biologically active soil depth of 0-5 cm after 23 years of treatment. Our results show that the enzyme activities of the litter removal treatments were significantly reduced compared to the doubling treatments, as were the values of soil respiration. The same significant difference was also detected in the C content of the soils of the treatments. Based on cluster analysis of the protein profile of the soil samples, the No Litter and No Input treatments were significantly different from the other treatments. This shows that specific organic matter is needed to enhance soil biological activity and the associated POXC content.- Published

37. Photoacoustic Flow Cytometry Using Functionalized Microspheres for Selective Detection of Bacteria.

- Abstract
Photoacoustic flow cytometry is a method to detect rare analytes in fluids. We developed photoacoustic flow cytometry to detect pathological cells in body fluids, such as circulating tumor cells or bacteria in blood. In order to induce specific optical absorption in bacteria, we use modified bacteriophage that precisely target bacterial species or subspecies for rapid identification. In order to reduce detection variability and to halt the lytic lifescycle that results in lysis of the bacteria, we attached dyed latex microspheres to the tail fibers of bacteriophage that retained the bacterial recognition binding sites. We tested these microsphere complexes using Salmonella enterica ( Salmonella ) and Escherichia coli ( E. coli ) bacteria and found robust and specific detection of targeted bacteria. In our work we used LT2, a strain of Salmonella , against K12, a strain of E. coli . Using Det7, a bacteriophage that binds to LT2 and not to K12, we detected an average of 109.3±9.0 of LT2 versus 2.0±1.7 of K12 using red microspheres and 86.7±13.2 of LT2 versus 0.3±0.6 of K12 using blue microspheres. These results confirmed our ability to selectively detect bacterial species using photoacoustic flow cytometry.

Introduction: A wealth of physiological, pathophysiological and clinical evidence of the beneficial effects of childhood fever exists already. Nevertheless, the public perception of fever has become persistently negative. Sociological research attributes this to a number of factors: unjustified fear, help-seeking behaviour, complex behavioural patterns of symptom avoidance and comfort-seeking. One of the keys to this change in attitudes, in the light of recent research, is linked to changes in the awareness and understanding of health among health professionals and lay people. The role of the young generation using media is crucial., Objective: To establish a long-term research project to reduce the use of medication (antipyretics and antibiotics) and the number of medical consultations and to improve attitudes towards fever, using media-based e-health tools., Method: An observational, adaptive, prospective cohort study was conducted. The intervention under study is a publicly available application and linked knowledge base. We collect self-reported data from caregivers. The application takes these into account and provides a decision-supporting condition classification based on a differential diagnosis algorithm., Results: 1) The parameters, primary and secondary criteria to be captured in the application as well as the data collection and data processing methodology for the assessment were defined by 100% consensus of the expert partners in a Delphi process. 2) Based on the available national and international guidelines, the above parameters were used to create the condition assessment, decision aid algorithm, which can be a starting point for machine learning in the long term. 3) We evaluated baseline data on demographics, febrile events and antipyretic use from 01/11/2020 to 15/06/2022., Conclusion: The FeverFriendTM project can contribute to reduce the burden of medicalisation and care burden on the existing healthcare system through evidence-based modern fever management in the care of children and adults with fever. 39. Long-Term Changes in Organic Matter Content and Soil Moisture Determine the Degree of Root and Soil Respiration.

Carbon in soil is one of the most important indicators of soil fertility. Part of the carbon stored in them is returned to the atmosphere during soil respiration. Climate change and inappropriate land use can accelerate these processes. Our work aimed to determine how soil CO
2 emissions change over ten years as a result of litter manipulation treatments. Plots at the Síkfőkút DIRT (Detritus Input and Removal Treatments) experimental site include doubling either leaf litter or wood, and removing all aboveground litter, all root inputs, or removing all litter inputs. With the help of this, we were able to examine not only the effects of the different organic matter intake but also the effects of the different microclimates that occur as a result of the treatments. Total soil respiration (root and microbial respiration) is a result of a persistent lack or excess of soil organic matter relative to soil moisture. Based on our studies, the increase in the intensity of root respiration on wetter soils was only half of the increase in respiration associated with decomposition activity. The sustained growth of leaf litter significantly increases soil respiration, which can be partly explained by the more favorable supply of nutrients to the decomposing organisms, and partly by the more favorable microclimatic conditions, however, these effects were only valid in the case of wetter soils. In the dry summer environment, we experienced higher CO2 emissions during litter removal treatments. In the first period between 2002 and 2004, even wetter root removal treatments showed a significantly higher CO2 emission, while in the period 2010-2012, surface litter removal treatments. The permanent removal of surface litter in the drier summer period resulted in the formation of a dense crack network, which increased the CO2 emission of these soils, which increases the soil organic carbon loss of the soil. Our study proves the advantages of mulching in terms of a more favorable microclimate of the soil surface and a balanced carbon balance of the soil-plant system.- Published

The EU Water Framework Directive requires the monitoring and evaluation of surface water sediment quality based on the assessment of risk posed by contamination on the biotic receptors. Floodplain sediments are important receptors of potentially toxic element (PTE) contamination from the upstream catchment areas, and floodplains host climate-sensitive riverine ecosystems and fertile agricultural areas at the same time. This study investigates the effect of PTE contamination on microbial communities in floodplain sediments and soils using the fast, inexpensive and reliable fluorescein diacetate (FDA) method in order to estimate its applicability for sediment quality monitoring and preliminary toxicity-based risk assessment. Sediment and soil samples were collected from the actively flooded alluvial plain and the river terrace areas along a 130-km stretch of the large Drava River floodplain known to be widely contaminated by historical mining, smelting and the associated industry in the upstream Alpine region. Results of detailed data analysis show that the total microbial activity represented by the measured FDA values is related to PTE (As, Cu, Zn, Cd, Pb) concentrations, but this relationship shows significant heterogeneity and depends on the spatial location and on the soil properties such as organic matter content, dissolved salt and nutrient content, and it is specific to the toxic elements. Results show that some microbe species appear to be able to adapt to the elevated PTE concentrations in toxic soil micro-environments, over time. Despite the observed heterogeneity of microbial activity, the results revealed a breakpoint in the FDA dataset around the FDA = 3 FC (fluorescein concentration) value suggesting that microbial activity is controlled by thresholds., (© 2022. 41. Food loss in the Hungarian food industry - Examining quantification problems and attitudes of managers and their impact on policy.

The food industry is estimated to be the second biggest generator of food losses after households. The novelty of this paper is that it focuses on this less analysed sector, conducting exploratory research based on a detailed primary survey among Hungarian food processors with a uniquely large sample of 175 respondents. A SPSS program was used to analyse the data, generating descriptive statistics, statistical tests for exploring relationships between variables and cluster analysis for classifying respondents based on their attitudes towards the problem. In-depth interviews (19) and a workshop were also applied, refining the results. Only half of the respondents maintain exact records of food loss. The leaders of the companies usually do not consider the generation of losses as a serious problem, but instead tend to see it as a natural phenomenon related to technological processes. Based on the cluster analysis carried out, the companies could be classified into two distinguishable groups. Larger companies perceived the loss to be a significantly more serious problem, they recorded it more accurately than the other group and they had more important motivations such as the desire to achieve cost reduction and better allocation of resources. Food processors face several barriers during food loss management: regulatory issues, the lack of a developed market for by-products, insufficient stakeholder collaboration along the food chain and currently, insufficient incentive to reduce losses. Reduction and better utilisation of food loss can only be achieved if there is a fundamental change in food loss regulation: incentives should be provided to drive companies to avoid losses and to make the best use of the by-products., (Copyright © 2021 Elsevier Ltd. Rhizobacteria-based technologies may constitute a viable option for biological fertilization and crop protection. The effects of two microbial inoculants (1) PPS: Pseudomonas protegens , P. jessenii and Stenotrophomonas maltophilia biocontrol bacterium strains and (2) TPB: Trichoderma atroviride , Pseudomonas putida , and Bacillus subtilis fungi, bacteria biocontrol, and biofertilizer combinations were examined on potato ( Solanum tuberosum L. var. Demon) in three consecutive years in irrigated organic conditions. The number of tubers showing symptoms of Streptomyces sp. and Rhizoctonia sp. was recorded. The severity of symptoms was evaluated based on the damaged tuber surface. There was a large annual variability in both the symptoms caused by soil-borne pathogens, and the effect of bio-inoculants. In the first and second year, with a stronger Rhizoctonia and Streptomyces spp. incidence, the bacterial and fungal combination of TPB inoculums with both the potential plant nutrition and biocontrol ability of the strains seemed to have a better efficiency to control the diseases. This tendency was not supported in the third year, and this may be attributed to the relatively high natural precipitation. Further studies are required to investigate the agronomic benefits of these inoculants and to tailor their application to the soil microbial characteristics and weather conditions.

- Abstract
Probiotics have been reported to have a positive impact on the metabolic control of patients with type 2 diabetes. We aimed to systematically evaluate the effects of probiotics on cardiometabolic parameters in type 2 diabetes based on randomized controlled studies. MEDLINE, Embase, and CENTRAL databases were reviewed to search for randomized controlled trials that examined the effects of probiotic supplementation on cardiometabolic parameters in patients with type 2 diabetes. 32 trials provided results suitable to be included in the analysis. The effects of probiotics were calculated for the following parameters: BMI, total cholesterol levels, LDL, triglycerides, HDL, CRP, HbA1c levels, fasting plasma glucose, fasting insulin levels, systolic and diastolic blood pressure values. Data analysis showed a significant effect of probiotics on reducing total cholesterol, triglyceride levels, CRP, HbA1c, fasting plasma glucose, fasting insulin levels, and both systolic and diastolic blood pressure values. Supplementation with probiotics increased HDL levels however did not have a significant effect on BMI or LDL levels. Our data clearly suggest that probiotics could be a supplementary therapeutic approach in type 2 diabetes mellitus patients to improve dyslipidemia and to promote better metabolic control. According to our analysis, probiotic supplementation is beneficial in type 2 diabetes mellitus.

- Abstract
The aim of the present study is to extend the applicability of MRI measurements similar to those used in human diagnostics to the examination of water barriers in living plants, thus broadening their use in natural sciences. The cucumber, Cucumis sativus, and Phillyrea angustifolia, or false olive, were chosen as test plants. The MRI measurements were carried out on three samples of each plant in the same position vis-a-vis the MRI apparatus using a Siemens Avanto MRI scanner. Two different relaxation times were employed, T
1 , capable of histological mapping, and T2 , used for the examination of water content. In the course of the analysis, it was found that certain histological formations and branching cause modifications to the intensity detected with relaxation time T2 . Furthermore, these positions can also be found in T1 measurements. A monotonic correlation (cucumber: ρ = 0.829; false olive: ρ = -0.84) was observed between the T1 and T2 measurements. In the course of the statistical analysis of the signal intensities of the xylems it was concluded that they cannot be regarded as independent in a statistical sense; these changes rather depend on the anatomic structure of the plant, as the intensity profile is modified by nodes, leaves and branches. This serves as a demonstration of the applicability of MRI to the measurement of well know plant physiological processes. The special parametrization required for this equipment, which is usually used in human diagnostics, is also documented in the present study.- Published

Myostatin, a TGF-β superfamily member, is a negative regulator of muscle growth. Here we describe how myostatin activity is regulated by syndecan-4, a ubiquitous transmembrane heparan sulfate proteoglycan. During muscle regeneration the levels of both syndecan-4 and promyostatin decline gradually after a sharp increase, concurrently with the release of mature myostatin. Promyostatin and syndecan-4 co-immunoprecipitate, and the interaction is heparinase-sensitive. ShRNA-mediated silencing of syndecan-4 reduces C2C12 myoblast proliferation via blocking the progression from G1- to S-phase of the cell cycle, which is accompanied by elevated levels of myostatin and p21(Waf1/Cip1), and decreases in cyclin E and cyclin D1 expression. Our results suggest that syndecan-4 functions as a reservoir for promyostatin regulating the local bioavailability of mature myostatin., (© 2018 The Authors. Biochar is a solid material obtained from reductive, oxygen-free processes, i.e. the thermo-chemical conversion of biomass in oxygen-limited environment. The obtained products have high carbon sequestration potential and strong nutrient-water absorption capacities because of the enlarged carbon surfaces. It is not yet clear how carbon stimulates agrochemical parameters in soil and how those characteristics are developing as time goes on a long-term basis. Samples of ancient (25, 35, 80 years old) plant coal-affected soils were collected in a temperate deciduous forest site located in the south part of the Bükk Mountains (in North Eastern Hungary). Physical-chemical soil characteristics, such as soil pH, cation exchange capacity (CEC), the organic and inorganic nitrogen (NH
4 + , NH3 - ) and the available nutrients (P2 O5 and K2 O), were estimated beside organic matter (SOM) content, measured by two different methods. Levels of polycyclic aromatic hydrocarbon (PAH) compounds in soil and in various biochar samples were assessed in relation with permissible limit values and potential toxicity. Positive correlation was found between the amount of available nutrients, total organic nitrogen content, cation exchange capacity and the age of plant coal-affected soils. The sample soils were exposed to continuous plant coal biochar effect for 25 years, during which macronutrients absorbed and accumulated in the plant coal surfaces. After this period, the degradation of carbon developed simultaneously with the reduction of the amount of available nutrients, till the end of the studied 80-year-affecting period. Measured CEC level indicated positive correlation with nutrient availability and the age of biochar-affected soils. Our results support the hypothesis that biochar in soil can improve its general agrochemical characteristics in relation with its persistence in a specific soil-plant system. Potential PAH content and toxicity of biochar products are key issues of developing proper application rates in sustainable agricultural practices.- Published

- Abstract
The TGFβ family member myostatin (growth/differentiation factor-8) is a negative regulator of skeletal muscle growth. The hypermuscular Compact mice carry the 12-bp Mstn(Cmpt-dl1Abc) deletion in the sequence encoding the propeptide region of the precursor promyostatin, and additional modifier genes of the Compact genetic background contribute to determine the full expression of the phenotype. In this study, by using mice strains carrying mutant or wild-type myostatin alleles with the Compact genetic background and nonmutant myostatin with the wild-type background, we studied separately the effect of the Mstn(Cmpt-dl1Abc) mutation or the Compact genetic background on morphology, metabolism, and signaling. We show that both the Compact myostatin mutation and Compact genetic background account for determination of skeletal muscle size. Despite the increased musculature of Compact s, the absolute size of heart and kidney is not influenced by myostatin mutation; however, the Compact genetic background increases them. Both Compact myostatin and genetic background exhibit systemic metabolic effects. The Compact mutation decreases adiposity and improves whole body glucose uptake, insulin sensitivity, and
18 FDG uptake of skeletal muscle and white adipose tissue, whereas the Compact genetic background has the opposite effect. Importantly, the mutation does not prevent the formation of mature myostatin; however, a decrease in myostatin level was observed, leading to altered activation of Smad2, Smad1/5/8, and Akt, and an increased level of p-AS160, a Rab-GTPase-activating protein responsible for GLUT4 translocation. - Abstract
Myostatin is an important negative regulator of skeletal muscle growth. The hypermuscular Compact (Cmpt) mice carry a 12-bp natural mutation in the myostatin propeptide, with additional modifier genes being responsible for the phenotype. Muscle cellularity of the fast-type tibialis anterior (TA) and extensor digitorum longus (EDL) as well as the mixed-type soleus (SOL) muscles of Cmpt and wild-type mice was examined by immunohistochemical staining of the myosin heavy chain (MHC) proteins. In addition, transcript levels of MHC isoforms were quantified by qPCR. Based on our results, all investigated muscles of Cmpt mice were significantly larger compared with that of wild-type mice, as characterized by fiber hyperplasia of different grades. Fiber hypertrophy was not present in TA; however, EDL muscles showed specific IIB fiber hypertrophy while the (I and IIA) fibers of SOL muscles were generally hypertrophied. Both the fast TA and EDL muscles of Cmpt mice contained significantly more glycolytic IIB fibers accompanied by a decreased number of IIX and IIA fibers; however, this was not the case for SOL muscles. In summary, despite the variances found in muscle cellularity between the different myostatin mutant mice, similar glycolytic shifts were observed in Cmpt fast muscles as in muscles from myostatin knockout mice.

- Abstract
We report on an in-situ observation of the colloidal silver nanoparticle self-assembly into a close-packed monolayer at the air/water interface followed by a 2D to 3D transition. Using the fast tracking GISAXS technique, we were able to observe the immediate response to the compression of the self-assembled nanoparticle layer at the air/water interface and to identify all relevant intermediate stages including those far from the equilibrium. In particular, a new nonequilibrium phase before the monolayer collapse via the 2D to 3D transition was found that is inaccessible by the competing direct space imaging techniques such as the scanning and transmission electron microscopies due to the high water vapor pressure and surface tension.

- Abstract
A 44-year-old female patient was admitted to our department with diagnosis of malignant lymphoma. The abdominal USG and CT showed multiple liver lesions with partial portal vein thrombosis, moderately increased alfa-fetoprotein (AFP), ASAT, ALAT (2x normal value), serology was negative for HBV and HCV. Liver transplantation was suggested but refused because of portal vein thrombosis. ATRA (45 mg/m2/day orally) was given on the basis of the assumption that HCC and acute promyelocytic leukaemia share similar oncogenic pathway (alter the RAR alpha and beta receptors). She was gained 15 kg-s and has resumed her work as a teacher for the last 20 months. Abdominal CT showed a complete regression of the intrahepatic tumour.
