International Council for the Exploration of the Sea (ICES) Annual Science Conference 2019, 9-12 September 2019 Gothenburg, Sweden.-- 1 page, The International Council for the Exploration of the Sea (ICES) - Intergovernmental Oceanographic Commission of UNESCO (IOC) Working Group on Harmful Algal Bloom Dynamics (WGHABD) has entered data into the Harmful Algal Event (HAEDAT) database for the last 20 years. These entries report information about harmful algal events that result in management actions such as closures of shellfish harvesting areas or negative environmental impacts e.g. mortalities of marine mammals. These data, collected mainly from routine monitoring programmes, provide a wealth of information that is not routinely accessible for scientific assessment. WGHABD is producing a HAB status report based on the incidence of HAB events in the ICES area which will contribute to a Global HAB Status Report. These HAEDAT data reveal a regional distribution in harmful algal events in the North Atlantic area and changes have been observed over time. On the east coast of the USA and Canada, the majority of issues have been caused by paralytic and amnesic shellfish toxins. In contrast, diarrhetic shellfish toxins have been the dominant cause of problems in Europe while cyanobacteria events were restricted to the Baltic. Incidents of farmed/wild fish mortalities are more sporadically recorded. HAEDAT is an essential source of information to support investigation of environmental drivers such as climate change on the incidence of harmful algal events on a regional scale

These data, collected mainly from routine monitoring programmes, provide a wealth of information that is not routinely accessible for scientific assessment. WGHABD is producing a status report based on the incidence of HAB events in the ICES area which will contribute to a Global HAB Status Report. These HAEDAT data reveal a regional distribution in harmful algal events in the North Atlantic area. On the east coast of the USA and Canada, the majority of issues have been caused by paralytic and amnesic shellfish toxins (PST, AST). In contrast, diarrhetic shellfish toxins (DST) has been the dominant cause of problems in Europe while cyanobacteria events were largely restricted to the Baltic. HAEDAT also provides information about changes in the incidence of harmful algal events over time, e.g. the recent increased recording of DST events in the USA, a reduction in the incidence of PST closures in Scottish waters at the beginning of this century. Abstract

Under ongoing climate change and increasing anthropogenic activity, which continuously challenge ecosystem resilience, an in-depth understanding of ecological processes is urgently needed. Lakes, as providers of numerous ecosystem services, face multiple stressors that threaten their functioning. Harmful cyanobacterial blooms are a persistent problem resulting from nutrient pollution and climate-change induced stressors, like poor transparency, increased water temperature and enhanced stratification. Consistency in data collection and analysis methods is necessary to achieve fully comparable datasets and for statistical validity, avoiding issues linked to disparate data sources. The European Multi Lake Survey (EMLS) in summer 2015 was an initiative among scientists from 27 countries to collect and analyse lake physical, chemical and biological variables in a fully standardized manner. This database includes in-situ lake variables along with nutrient, pigment and cyanotoxin data of 369 lakes in Europe, which were centrally analysed in dedicated laboratories. Publishing the EMLS methods and dataset might inspire similar initiatives to study across large geographic areas that will contribute to better understanding lake responses in a changing environment.

10. EXPERIMENTAL AUTOIMMUNE MYOCARDITIS IN RATS AND THERAPEUTIC HISTAMINE H1 – H4 RECEPTOR INHIBITION.

Abstract

Myocarditis, a life threatening disease, is still not adequately treated. Histamine plays an important role in physiology and pathophysiology of cardiovascular system. All four histamine receptors (H1R – H4R), are present in the heart. Experimental autoimmune myocarditis (EAM) was used to investigate which histamine receptor had a greater impact on the disease’s progression. EAM was evoked in Lewis rats by porcine myosin immunization. Mepyramine, ranitidine and ciproxifan were used to inhibit H1R, H2R and H3R receptors, respectively, and 2,4-diaminopyrimidines: ST994, ST1012, ST1006 were ligands of H4R. Quinapril, an ACE inhibitor, served as a reference drug. Drugs were administered daily, either from 0 – 2 weeks or from 2 to 4 weeks post EAM induction. Cardiac dysfunction developed with significant decreases in left ventricular ejection fraction and fractional shortening due to dilatation and wall thickening. EAM rats treated with mepyramine and ST994 in weeks 0 – 2 had the lowest decreases. These treated with ST994, ST1012 or quinapril performed much better the following 2 weeks without therapy than did the other groups. On autopsy their hearts were smaller, less fibrotic, histopathological changes in them of a lower grade. When the treatment started with 2 weeks’ delay, the ST994-treated EAM rats showed the highest median survival. H4 receptor antagonism inhibits heart remodelling, preserves heart contractility, improves survival and may be of potent therapeutic relevance in human clinics. Abstract

In clinical practice there are no sufficient histopathological and immunohistochemical parameters to predicting progression of laryngeal cancer. We applied the new quantitative Anertoha, Blatsakisa and Luny cancer classification and expression of PCNA (proliferating cell nuclear antigen) to clinical findings to appreciate prognosis in laryngeal cancer.

Abstract

Systemic lupus erythematosus (SLE) is a chronic autoimmune disease that can be exacerbated by exposure to ultraviolet radiation (UVR). The number and phenotype of antigen presenting cells in the skin play a role in cutaneous immune response generation. Although antimalarials are widely used in SLE treatment, their mode of action is not completely elucidated. The aim of our study was to determine the effect of chloroquine treatment on HLA-DR+ and CD1a+ cell number in locally irradiated (three minimal erythema doses of UVB) and normal appearing skin in SLE patients and healthy subjects. A significantly higher number of HLA-DR+ and CD1a+ cells were found in both locations in SLE patients compared with controls. Following three months of daily chloroquine treatment (250 mg), the HLA-DR+ and CD1a+ cell counts were significantly reduced in both irradiated and unirradiated sites of SLE patients, although still higher than in controls. Chloroquine treatment reduces the number of antigen presenting cells in the skin of SLE patients, and this effect may explain the antimalarials beneficial immunoregulatory and anti-inflammatory properties. Abstract

Insight into how environmental change determines the production and distribution of cyanobacterial toxins is necessary for risk assessment. Management guidelines currently focus on hepatotoxins (microcystins). Increasing attention is given to other classes, such as neurotoxins (e.g., anatoxin-a) and cytotoxins (e.g., cylindrospermopsin) due to their potency. Most studies examine the relationship between individual toxin variants and environmental factors, such as nutrients, temperature and light. In summer 2015, we collected samples across Europe to investigate the effect of nutrient and temperature gradients on the variability of toxin production at a continental scale. Direct and indirect effects of temperature were the main drivers of the spatial distribution in the toxins produced by the cyanobacterial community, the toxin concentrations and toxin quota. Generalized linear models showed that a Toxin Diversity Index (TDI) increased with latitude, while it decreased with water stability. Increases in TDI were explained through a significant increase in toxin variants such as MC-YR, anatoxin and cylindrospermopsin, accompanied by a decreasing presence of MC-LR. While global warming continues, the direct and indirect effects of increased lake temperatures will drive changes in the distribution of cyanobacterial toxins in Europe, potentially promoting selection of a few highly toxic species or strains. © 2018 by the authors. Abstract

Under ongoing climate change and increasing anthropogenic activity, which continuously challenge ecosystem resilience, an in-depth understanding of ecological processes is urgently needed. Lakes, as providers of numerous ecosystem services, face multiple stressors that threaten their functioning. Harmful cyanobacterial blooms are a persistent problem resulting from nutrient pollution and climate-change induced stressors, like poor transparency, increased water temperature and enhanced stratification. Consistency in data collection and analysis methods is necessary to achieve fully comparable datasets and for statistical validity, avoiding issues linked to disparate data sources. The European Multi Lake Survey (EMLS) in summer 2015 was an initiative among scientists from 27 countries to collect and analyse lake physical, chemical and biological variables in a fully standardized manner. This database includes in-situ lake variables along with nutrient, pigment and cyanotoxin data of 369 lakes in Europe, which were centrally analysed in dedicated laboratories. Publishing the EMLS methods and dataset might inspire similar initiatives to study across large geographic areas that will contribute to better understanding lake responses in a changing environment.

Abstract

Until recently, the data on the diversity of the entire microbial community from the Baltic Sea were relatively rare and very scarce. However, modern molecular methods have provided new insights into this field with interesting results. They can be summarized as follows. (i) Although low salinity causes a reduction in the biodiversity of multicellular species relative to the populations of the North-East Atlantic, no such reduction occurs in bacterial diversity. (ii) Among cyanobacteria, the picocyanobacterial group dominates when considering gene abundance, while filamentous cyanobacteria dominate in means of biomass. (iii) The diversity of diatoms and dinoflagellates is significantly larger than described a few decades ago; however, molecular studies on these groups are still scarce. (iv) Knowledge gaps in other protistan communities are evident. (v) Salinity is the main limiting parameter of pelagic fungal community composition, while the benthic fungal diversity is shaped by water depth, salinity, and sediment C and N availability. (vi) Bacteriophages are the predominant group of viruses, while among viruses infecting eukaryotic hosts, Phycodnaviridae are the most abundant; the Baltic Sea virome is contaminated with viruses originating from urban and/or industrial habitats. Abstract

The cyanobacterial response to pharmaceuticals is less frequently investigated compared to green algae. Pharmaceuticals can influence not only the growth rate of cyanobacteria culture, but can also cause changes at the cellular level. The effect of diclofenac (DCF) as one of the for cyanobacteria has been rarely tested, and DCF has never been applied with cellular biomarkers. The aim of this work was to test the response of two unicellular cyanobacteria (Synechocystis salina and Microcystis aeruginosa) toward DCF (100 mg L -1 ) under photoautotrophic growth conditions. Such endpoints were analyzed as cells number, DCF uptake, the change in concentrations of photosynthetic pigments, the production of toxins, and chlorophyll a in vivo fluorescence. It was noted that during a 96 h exposure, cell proliferation was not impacted. Nevertheless, a biochemical response was observed. The increased production of microcystin was noted for M. aeruginosa. Due to the negligible absorption of DCF into cells, it is possible that the biochemical changes are induced by an external signal. The application of non-standard biomarkers demonstrates the effect of DCF on microorganism metabolism without a corresponding effect on biomass. Abstract

Selenium (Se) is an essential trace element and displays a narrow range of concentration between essentiality and toxicity. Se plays an important role in ameliorating mercury toxicity in organisms. Despite this there are only a few reports concerning Se concentration in plankton, the first link in the trophic chain that determines the uptake and transfer of Se to subsequent trophic levels. This paper aimed to determine Se concentration in water, phytoplankton, and zooplankton in the Baltic Sea, and factors affecting Se absorption from the environment and its transfer to higher trophic levels. Sea water and plankton samples were collected from the Gulf of Gdańsk during 5 cruises (4 seasons: 2019-2022) at 4 research stations. An additional cruise was undertaken in July 2020 in the open waters of the southern Baltic Sea. The median Se concentrations in the Gulf of Gdańsk was 0.25 μg·dm -3 . While the median of Se concentration in phytoplankton was 1.11 μg·g -1 and in zooplankton was 1.25 μg·g -1 . The biomass of organisms in the phytoplankton and zooplankton in the Gulf of Gdańsk showed an important role in shaping Se concentration. Seasonal trends in Se concentration in zooplankton could be the result of taxa composition changes, changes to dietary intake of Se, changes in growth dilution, or potentially some combination of factors. The highest biomagnification rate occurred in the summer. In contrast, in autumn and winter, when plankton biomass was dominated by the ciliate species Mesodinium rubrum, the highest Se concentration in plankton was measured.
