In this study we determined the effect of transformation of a mature sessile oak forest stand into a coppiced forest, and of thinning and throughfall reduction in a coppice stand on soil water content (SWC) and soil CO efflux. The precipitation reduction was induced by installing parallel drainage channels in both unthinned and thinned coppice stands. The driving factor for temporal dynamics of soil CO efflux in all plots was soil temperature. The other factor was soil water content but only up to about 15%. Above this threshold, there was no more effect on CO efflux. We found no clear difference in SWC or soil CO efflux between the mature and coppiced stand eight years after harvesting. On the other hand, thinning of the coppice stand resulted in increase in SWC up to 22% in proportion, which we assume to be a result of increased gap fraction of the canopy. However, no effect on soil CO efflux was observed two years after the thinning. Installation of the drainage channels in two plots covering 30% of the ground area resulted in decrease in SWC up to a proportional 30.5% and thus contributed up to 50.7% reduction in soil CO efflux.222222

Group A Streptococcus (GAS) is a strict human pathogen possessing a unique pathogenic trait that utilizes the cooperative activity of NAD+-glycohydrolase (NADase) and Streptolysin O (SLO) to enhance its virulence. How NADase interacts with SLO to synergistically promote GAS cytotoxicity and intracellular survival is a long-standing question. Here, the structure and dynamic nature of the NADase/SLO complex are elucidated by X-ray crystallography and small-angle scattering, illustrating atomic details of the complex interface and functionally relevant conformations. Structure-guided studies reveal a salt-bridge interaction between NADase and SLO is important to cytotoxicity and resistance to phagocytic killing during GAS infection. Furthermore, the biological significance of the NADase/SLO complex in GAS virulence is demonstrated in a murine infection model. Overall, this work delivers the structure-functional relationship of the NADase/SLO complex and pinpoints the key interacting residues that are central to the coordinated actions of NADase and SLO in the pathogenesis of GAS infection.

Recent changes in water availability can be crucial for the development, growth and carbon budget of forests. Therefore, our aim was to determine the effect of reduced throughfall and severe summer drought on stem CO2 efflux as a function of temperature and stem increment. Stem CO2 efflux was measured using the chamber method on oak and hornbeam under four treatments: coppice, thinned coppice, and both coppice and thinned coppice with 30 %-reduced throughfall. The first year of the experiment had favourable soil water availability and the second year was characterized by a dry summer. While reduced throughfall had no effect on stem CO2 efflux, the summer drought decreased efflux by 43–81 % during July and August. The stem CO2 efflux was reduced less severely (by 13–40 %) in September when the drought persisted but the stem increment was already negligible. The stem increment was also strongly affected by the drought, which was reflected in its paired relationship with stem CO2 efflux over the two experimental years. The study showed that summer dry periods significantly and rapidly reduce stem CO2 efflux, whereas a constant 30 % rainfall reduction needs probably a longer time to affect stem properties, and indirectly stem CO2 efflux. Injection moulding of polymers is a well-established technique for the production of increasingly complex articles, and residual molecular orientation is a matter of considerable interest. A single matrix of commercial atactic polystyrene was systematically blended with deuterated polystyrene chains of five different weight fractions and the orientation investigated using small angle neutron scattering (SANS), optical birefringence and thermal shrinkage. The strengths and limitations of the experimental techniques used to measure residual orientation are explored and highlighted the unique contribution of SANS to observe ensemble averaged molecular orientation using deuterated polymers. The residual molecular orientation was found to vary with the molecular weight of the deuterated chains with an increase with increasing molecular weight. The observed correlations lead to a better understanding of the processing parameters that influence the degree of residual orientation on the molecular scale. Injection moulded polymer articles often have residual macromolecular or crystalline orientation which can have a significant impact on the optical and mechanical properties of the moulded article. Small angle neutron scattering (SANS) was used to measure the molecular shape and orientation of deuterated blends of injection moulded polystyrene. For ~1-mm-thick mouldings of uniform rectangular cross-section, the eccentricity in the SANS pattern gave a direct measure of the residual molecular orientation over the length scale ~100-1,500 [Angstrom]. The residual orientation was found to vary significantly with injection moulding conditions with comparative residual orientation decreasing with decreasing mould fill-time, and increasing with mould thickness and moulding temperatures. The orientation was found to be a minimum in the centre of the mould and highest near the surface and the average orientation at a particular position in the mould was found to be strongly correlated with the volume of material deposited as a solid skin layer during injection moulding. This study examines the soil environment of eight charcoal kiln platforms and the neighboring soil in Czech and Bohemian low-altitude forest stands. Both mixed and undisturbed soil samples were used to assess the hydrophysical soil properties, nutrient content, cation exchange capacity, enzyme activity, and soil active carbon content, while soil color, stoniness, root density, and horizon thickness were estimated in the field. Charcoal-rich horizons had high total organic carbon concentrations and total nitrogen content (about 150% and 40% higher than in the organomineral horizons of the control plot, respectively), with total carbon stocks being higher than those in neighboring forest soils. Fine root density was highest in the charcoal-rich horizons, encouraged by high soil porosity, aeration, and favorable chemical properties. Enzyme group activity differed between individual soil horizons and kiln and control plots, with depolymerization enzyme activity highest in charcoal-rich horizons and humification enzyme activity highest in mineral horizons. Phosphatase, chitinase, and phenoloxidase activity were highest in charcoal-rich horizons, while β-glucosidase activity remained similar across horizons. After long-term abandonment, kiln sites shift from inhospitable sites to localized hotspots for plant and microbial growth, having more favorable physical, enzymatic, and chemical soil properties than the surrounding areas. This study confirmed that kiln production platforms act as microhabitat hotspots, also providing information on a wide spectrum of soil properties linked with soil microorganisms and root growth.
