Background: The artificial intelligence (AI)-based platform ChatGPT (Chat Generative Pre-Trained Transformer, OpenAI LP, San Francisco, CA, USA) has gained impressive popularity in recent months. Its performance on case vignettes of general medical (non-ophthalmological) emergencies has been assessed - with very encouraging results. The purpose of this study was to assess the performance of ChatGPT on ophthalmological emergency case vignettes in terms of the main outcome measures triage accuracy, appropriateness of recommended prehospital measures, and overall potential to inflict harm to the user/patient., Methods: We wrote ten short, fictional case vignettes describing different acute ophthalmological symptoms. Each vignette was entered into ChatGPT five times with the same wording and following a standardized interaction pathway. The answers were analyzed following a systematic approach., Results: We observed a triage accuracy of 93.6%. Most answers contained only appropriate recommendations for prehospital measures. However, an overall potential to inflict harm to users/patients was present in 32% of answers., Conclusion: ChatGPT should presently not be used as a stand-alone primary source of information about acute ophthalmological symptoms. Purpose: To evaluate the rate of misdiagnosis of aneurysmatic pachychoroid type 1 choroidal neovascularization/polypoidal choroidal vasculopathy (PAT1/PCV) among cases diagnosed as non-aneurysmatic pachychoroid neovasculopathy (PNV) and to define optical coherence tomography (OCT) features facilitating their distinction., Methods: The database of the Department of Ophthalmology, Ludwig-Maximilians University Munich, was screened for patients diagnosed with PNV. Multimodal imaging was screened for the presence of choroidal neovascularization (CNV) and aneurysms/polyps. Imaging features facilitating the diagnosis of PAT1/PCV were analysed., Results: In total, 49 eyes of 44 patients with a clinical PNV diagnosis were included, of which 42 (85.7%) had PNV and 7 (14.3%) represented misdiagnosed PAT1/PCV. SFCT was comparable (PNV: 377 ± 92 vs. PAT1/PCV: 400 ± 83 µm; p = 0.39). Whereas no difference was detected in total pigment epithelium detachment (PED) diameter (p = 0.46), maximum PED height was significantly higher in the PAT1/PCV group (199 ± 31 vs. 82 ± 46, p < 0.00001). In a receiver operating characteristic (ROC) analysis, the optimum cutoff for defining "peaking PED" was 158 µm with an area under the curve of 0.969, a sensitivity of 1.0 (95% CI: 0.59-1.0), and a specificity of 0.95 (95% CI: 0.84-0.99). Sub-retinal hyperreflective material (SHRM; p = 0.04), sub-retinal ring-like structures (SRRLS; p < 0.00001), and sub-RPE fluid (p = 0.04) were significantly more frequent in eyes with PAT1/PCV., Conclusion: A relevant percentage of eyes diagnosed with PNV might instead suffer from PAT1/PCV. Many social animals display collective activity cycles based on synchronous behavioural oscillations across group members. A classic example is the colony cycle of army ants, where thousands of individuals undergo stereotypical biphasic behavioural cycles of about one month. Cycle phases coincide with brood developmental stages, but the regulation of this cycle is otherwise poorly understood. Here, we probe the regulation of cycle duration through interactions between brood and workers in an experimentally amenable army ant relative, the clonal raider ant. We first establish that cycle length varies across clonal lineages using long-term monitoring data. We then investigate the putative sources and impacts of this variation in a cross-fostering experiment with four lineages combining developmental, morphological and automated behavioural tracking analyses. We show that cycle length variation stems from variation in the duration of the larval developmental stage, and that this stage can be prolonged not only by the clonal lineage of brood (direct genetic effects), but also of the workers (indirect genetic effects). We find similar indirect effects of worker line on brood adult size and, conversely (but more surprisingly), indirect genetic effects of the brood on worker behaviour (walking speed and time spent in the nest).

Neural asymmetries of the bilateral parts of the nervous system are found throughout the animal kingdom. The relative low complexity and experimental accessibility of the insect nervous system makes it well suited for studying the functions of neural asymmetries and their underlying mechanisms. Recent findings in insects reveal hardwired asymmetries in their peripheral and central nervous systems, which affect sensory perception, motor behaviours and cognitive-related tasks. Non-alcoholic fatty liver disease (NAFLD) is the most common cause of liver disease worldwide. In adults with NAFLD, fibrosis can develop and progress to liver cirrhosis and liver failure. However, the underlying molecular mechanisms of fibrosis progression are not fully understood. Using total RNA-Seq, we investigated the molecular mechanisms of NAFLD and fibrosis. We sequenced liver tissue from 143 adults across the full spectrum of fibrosis stage including those with stage 4 fibrosis (cirrhosis). We identified gene expression clusters that strongly correlate with fibrosis stage including four genes that have been found consistently across previously published transcriptomic studies on NASH i.e. COL1A2, EFEMP2, FBLN5 and THBS2. Using cell type deconvolution, we estimated the loss of hepatocytes versus gain of hepatic stellate cells, macrophages and cholangiocytes with advancing fibrosis stage. Hepatocyte-specific functional analysis indicated increase of pro-apoptotic pathways and markers of bipotent hepatocyte/cholangiocyte precursors. Regression modelling was used to derive predictors of fibrosis stage. Unveiling the molecular mechanisms of tissue remodelling following injury is imperative to elucidate its regenerative capacity and aberrant repair in disease. Using different omics approaches, we identified enhancer of zester homolog 2 (EZH2) as a key regulator of fibrosis in injured lung epithelium. Epithelial injury drives an enrichment of nuclear transforming growth factor-β-activated kinase 1 (TAK1) that mediates EZH2 phosphorylation to facilitate its liberation from polycomb repressive complex 2 (PRC2). This process results in the establishment of a transcriptional complex of EZH2, RNA-polymerase II (POL2) and nuclear actin, which orchestrates aberrant epithelial repair programmes. The liberation of EZH2 from PRC2 is accompanied by an EZH2-EZH1 switch to preserve H3K27me3 deposition at non-target genes. Loss of epithelial TAK1, EZH2 or blocking nuclear actin influx attenuates the fibrotic cascade and restores respiratory homeostasis. Accordingly, EZH2 inhibition significantly improves outcomes in a pulmonary fibrosis mouse model. The collective motion of swarms depends on adaptations at the individual level. We explored these and their effects on swarm formation and maintenance in locusts. The walking kinematics of individual insects were monitored under laboratory settings, before, as well as during collective motion in a group, and again after separation from the group. It was found that taking part in collective motion induced in the individual unique behavioral kinematics, suggesting the existence of a distinct behavioral mode that we term a "collective-motion-state." This state, characterized by behavioral adaptation to the social context, is long lasting, not induced by crowding per se, but only by experiencing collective motion. Utilizing computational models, we show that this adaptability increases the robustness of the swarm. Insect locomotion represents a fundamental example of neuronal oscillating circuits generating different motor patterns or gaits by controlling their phase coordination. Walking gaits are assumed to represent stable states of the system, often modeled as coupled oscillators. This view is challenged, however, by recent experimental observations, in which in vitro locust preparations consistently converged to synchronous rhythms (all legs oscillating as one), a locomotive pattern never seen in vivo. To reconcile this inconsistency, we developed a modeling framework to capture the trade-off between the two competing mechanisms: the endogenous neuronal circuitry, expressed in vitro, and the feedback mechanisms from sensory and descending inputs, active only in vivo. We show that the ubiquitously observed double-tripod walking gait emerges precisely from this balance. The outcome is a short-lived meta-stable double-tripod gait, which transitions and alternates with stable idling, thus recovering the observed intermittent bouts of locomotion, typical of many insects' locomotion behavior., (Copyright © 2019 The Author(s). Higher motor centers and central pattern generators (CPGs) interact in the control of coordinated leg movements during locomotion throughout the animal kingdom. The subesophageal ganglion (SEG) is one of the insect head ganglia reported to have a role in the control of walking behavior. Here we explored the functional relations between the SEG and the thoracic leg CPGs in the desert locust. Backfill staining revealed about 300 SEG descending interneurons (DINs) altogether. Recordings from an in-vitro isolated chain of thoracic ganglia, with intact or severed connections to the SEG, during pharmacological activation were used to determine how the SEG affects the centrally generated motor output to the legs. The SEG was demonstrated to both activate leg CPGs and synchronize their bilateral activity. Purpose: To study the glistening formation in various hydrophobic-acrylic intraocular lens (IOL) models, and to evaluate the effect of glistenings on light scattering in these IOLs., Design: Laboratory investigation., Methods: The susceptibility of the hydrophobic-acrylic material to develop glistenings was evaluated in 6 IOL models. Accelerated lens aging was induced by immersing the IOLs in a solution at 45 C for 24 hours and cooled to 37 C for 2.5 hours. Light microscopy and image acquisition were performed. Glistening statistics-that is, microvacuoles' (MV) number and size-were derived from image analysis. Light scattering was measured using a clinical device featuring an adaptation for in vitro IOL assessment., Results: The number of glistenings differed among the studied IOLs and ranged from 0 to 3532 MV/mm 2 . In 1 model, glistenings were found only at the periphery, with diffuse light scattering observed centrally despite the absence of microvacuoles. The mean size of glistenings ranged from 5.2 to 10.2 μm. The mean straylight parameter of the IOLs increased from 0.6 to 5.0 deg 2 /sr after accelerated aging. Straylight elevation demonstrated a proportional relationship with the glistening number., Conclusions: We showed that hydrophobic-acrylic lenses differ in their resistance to glistenings, as one group proved to be glistening-free, but the other models revealed varying grades of glistenings. Moreover, we demonstrated that the presence of glistenings results in increased straylight, and that straylight proportionally depends on the glistenings number irrespective of the IOL model.
