Das duktale Pankreasadenokarzinom (PDAC) wird meist im fortgeschrittenen Stadium diagnostiziert, häufig mit Lebermetastasen. Krebsstammzellen (CSC) wurden bereits im PDAC nachgewiesen und spielen eine Rolle bei Entstehung des Primärtumors und Therapieresistenz. Ihr Beitrag zur Metastasierung ist bislang wenig verstanden. Ziel dieser Studie war es zu untersuchen, welchen Einfluss die hepatische Mikroumgebung, abhängig vom inflammatorischen Zustand, auf Pankreasgangepithelzellen (PDEZ) hinsichtlich CSC-Eigenschaften hat. Dafür wurden prämaligne und maligne PDEZ in einem indirekten Kokultursystem mit Hepatozyten alleine, einer Kombination aus Hepatozyten und 5 % hepatischen Sternzellen (HSC), repräsentativ für eine physiologische Leberumgebung, oder Hepatozyten und 5 % hepatischen Myofibroblasten, als Modell für ein entzündliches Lebermilieu, kultiviert. Insgesamt konnte ein größeres Koloniebildungspotential der malignen im Vergleich zu prämalignen PDEZ festgestellt werden. Die größte Anzahl an Kolonien bildete sich in beiden Zelllinien in Gegenwart von Hepatozyten und 5 % HSC, was mit einer erhöhten Expression der CSC-Marker Nestin und Nanog einherging. Weiterhin konnten verschiedene Kolonieformen detektiert werden. Um Unterschiede des CSC-Potentials dieser Kolonieformen zu untersuchen, wurden PDEZ aus Holo-, Mero- bzw. Paraklonen expandiert und charakterisiert. In vivo führte die Inokulation von Paraklon-Zellen in 2/10 SCID-beige Mäusen zu Tumorwachstum, die von Holoklon-Zellen in 7/10 Tieren. Die Tumore waren durch eine hohe Expression von Nestin sowie von mesenchymalen Markern gekennzeichnet. In vitro waren Holoklon- verglichen mit Paraklon-Zellen durch erhöhte CSC-Eigenschaften charakterisiert, was zusätzlich mit einem stärker mesenchymalen Phänotyp einherging. Diese Daten deuten darauf hin, dass die hepatische Mikroumgebung ein wichtiger Modulator von CSC-Eigenschaften disseminierter PDEZ ist und so zu der Entwicklung von Lebermetastasen des PDAC beiträgt. Pancreatic ductal adenocarcinoma (PDAC) is mostly diagnosed in an advanced state, often accompanied with liver metastasis. Cancer stem cells (CSCs) have been detected in PDAC and play a role in development of the primary tumour and therapy resistance. Their contribution towards the process of metastasis formation, however, is still poorly understood. This study aimed at understanding the impact of the hepatic microenvironment, depending on its inflammatory conditions, on pancreatic ductal epithelial cells (PDEC) regarding their CSC-properties. For these analyses, premalignant and malignant PDEC were cultured in an indirect coculture system together with either hepatocytes alone, a combination of hepatocytes and 5 % hepatic stellate cells (HSC), representing a physiologic liver milieu, or hepatocytes and 5 % hepatic myofibroblasts that serve as a model for an inflammatory liver microenvironment. Malignant PDEC displayed a higher colony formation potential than premalignant cells. In both cell lines, the highest number of colonies was detected after coculture with hepatocytes and 5 % HSC and was associated with an enhanced expression of CSC-markers Nestin and Nanog. Furthermore, different colony forms could be detected. To analyse differences between those colony forms regarding their CSC-potential, PDEC were expanded from holo-, mero- and paraclones and were characterised. In vivo, the inoculation of paraclone cells led to tumour formation in 2/10 SCID-beige mice, while holoclone cells evoked tumour formation in 7/10 animals. These tumours displayed high expression levels of Nestin as well as mesenchymal markers. In vitro, holoclone cells were characterised by enhanced CSC-characteristics together with a stronger mesenchymal phenotype compared to paraclone cells. These data point towards the hepatic microenvironment as an important modulator of the CSC-properties of disseminated PDEC and thereby a contributor to the formation of liver metastasis of PDAC.

International audience; The somatotropic axis plays an important role for embryonic growth and development. However, less is known about the effect of early pregnancy (embryo signaling) on the maternal GH-IGF-system. The aim was to investigate endocrine endpoints of the somatotropic axis in pregnant vs. non-pregnant dairy heifers until d18 of pregnancy. Blood samples of Holstein Friesian heifers (n=30) were taken on d0 (day of ovulation, defined by ultrasonography) and days 7, 14, 16 and 18 in repeated consecutive pregnant (p) and non-pregnant (np) periods. Additionally, liver biopsies were taken on d18. Heifers received artificial insemination or embryo transfer and pregnancy was verified either on d18 (P4 >2.0 and successful recovery of a conceptus [n=12]) or retrospectively on d42 (detection of embryonic heart beat [n=21]). The cows were defined as non-pregnant on d18 if P4 was
