Treatment of these infections remains a major challenge due to the low intracellular efficacy of many antibiotics. Therefore, local delivery systems are urgently required to improve the therapeutic efficacy of antibiotics by enabling their intracellular delivery. Purpose: To assess the potential of gelatin nanospheres as carriers for local delivery of vancomycin into macrophages of zebrafish larvae in vivo and into THP-1-derived macrophages in vitro using fluorescence microscopy. Materials and methods: Fluorescently labeled gelatin nanospheres were prepared and injected into transgenic zebrafish larvae with fluorescent macrophages. Both the biodistribution of gelatin nanospheres in zebrafish larvae and the co-localization of vancomycin-loaded gelatin nanospheres with zebrafish macrophages in vivo and uptake by THP-1-derived macrophages in vitro were studied. In addition, the effect of treatment with vancomycin-loaded gelatin nanospheres on survival of S. aureus-infected zebrafish larvae was investigated. Results: Internalization of vancomycin-loaded gelatin nanospheres by macrophages was observed qualitatively both in vivo and in vitro. Systemically delivered vancomycin, on the other hand, was hardly internalized by macrophages without the use of gelatin nanospheres. Treatment with a single dose of vancomycin-loaded gelatin nanospheres delayed the mortality of S. aureus-infected zebrafish larvae, indicating the improved therapeutic efficacy of vancomycin against (intracellular) S. aureus infection in vivo. Conclusion: The present study demonstrates that gelatin nanospheres can be used to facilitate local and intracellular delivery of vancomycin. Despite the immense need for effective treatment of spinal cord injury (SCI), no successful repair strategy has yet been clinically implemented. Multifunctional biomaterials, based on porcine adipose tissue-derived extracellular matrix (adECM) and reduced graphene oxide (rGO), were recently shown to stimulate in vitro neural stem cell growth and differentiation. Nevertheless, their functional performance in clinically more relevant in vivo conditions remains largely unknown. Before clinical application of these adECM-rGO nanocomposites can be considered, a rigorous assessment of the cytotoxicity and biocompatibility of these biomaterials is required. For instance, xenogeneic adECM scaffolds could still harbour potential immunogenicity following decellularization. In addition, the toxicity of rGO has been studied before, yet often in experimental settings that do not bear relevance to regenerative medicine. Therefore, the present study aimed to assess both the in vitro as well as in vivo safety of adECM and adECM-rGO scaffolds. First, pulmonary, renal and hepato-cytotoxicity as well as macrophage polarization studies showed that scaffolds were benign in vitro. Then, a laminectomy was performed at the 10th thoracic vertebra, and scaffolds were implanted directly contacting the spinal cord. For a total duration of 6 weeks, animal welfare was not negatively affected. Histological analysis demonstrated the degradation of adECM scaffolds and subsequent tissue remodeling. Graphene-based scaffolds showed a very limited fibrous encapsulation, while rGO sheets were engulfed by foreign body giant cells. Furthermore, all scaffolds were infiltrated by macrophages, which were largely polarized towards a pro-regenerative phenotype. Lastly, organ-specific histopathology and biochemical analysis of blood did not reveal any adverse effects. In summary, both adECM and adECM-rGO implants were biocompatible upon laminectomy while establishing a pro-regenerative microenvironment, which justifies further research on their therapeutic potential for treatment of SCI.

High-carbon steels are normally used as tool materials. The use of such steels for construction is limited due to their increased brittleness and poor weldability. However, it appears that high-carbon steels possess certain hidden reserves for enhanced plasticity and strength if properly heat-treated. An unconventional heat treatment was applied to carbon eutectoid steel (0.8 wt.% C) in order to increase its strength and impact toughness simultaneously. Samples for tensile and impact testing were held at 800 °C for different time ranges from 3 min to 9 min with subsequent cooling in oil. It was established that for each type of sample, an optimal holding time exists that is responsible for increased strength and high impact toughness. The hardness and microhardness levels of the surface and under-surface regions of the samples reached 390 HV after optimal heat treatment. An X-ray revealed a shift of the (211)α-peak to the lower 2-theta angles after heat treatment with the optimal holding time; this indicates an increase in carbon content in alpha solid solutions of approximately 0.12 wt.%. Thus, a nanostructured mixture of low-carbon martensite and thin cementite plates is formed in the under-surface region of carbon eutectoid steel after heat treatment, with a controlled holding time at the austenitizing temperature.

This work presents the results of the study of structural, substructural and optical properties of Zn2SnO4 nanoparticles (NPs) and films. NPs synthesized by the hydrothermal method during 28 h were chosen to form a nanoink from which films were obtained by spraying the solution it on glass substrates. Obtained Zn2SnO4 films were annealed in Argon atmosphere at Ta = (250–500) °C for 30 min and 60 min to remove organic impurities and optimize the film's physical properties. Since the choice of mineralizer determines a number of important characteristics of nanoparticles, a comprehensive study of the obtained samples grown using KOH as a mineralizer was conducted. The samples were characterized by X-ray diffraction analysis, Energy Dispersive X-ray (EDX) analysis, high-resolution transmission (HRTEM) and scanning electron (SEM) microscopy, Raman and optical spectroscopy. Analysis of morphology showed that synthesized films have a [220] growth texture with inverse spinel structure and consist of NPs, the size of which increases with the annealing temperature (Ta). It was identified that the size of the coherent scattering domain (CSD) in the sample increases with increasing Ta, while the level of microdeformations decreases. Raman spectra showed F2g(2), F2g(3) and A1g modes corresponding to the structure of inverse spinel Zn2SnO4. It is found, that the optical bandgap Eg of NPs decreases from 4.02 eV to 3.85 eV with an increase in growth time. On another hand, the optical bandgap Eg of films decreases with an increase in annealing temperature, Ta, from 4.04 eV to 3.63 eV.

Metal-oxides hold promise as superior plasmonic materials in the mid-infrared compared to metals, although their integration over established material technologies still remains challenging. We demonstrate localized surface plasmons in self-assembled, hemispherical CdZnO metal-oxide nanoparticles on GaAs, as a route to enhance the absorption in mid-infrared photodetectors. In this system, two localized surface plasmon modes are identified at 5.3 and 2.7 μm, which yield an enhancement of the light intensity in the underlying GaAs. In the case of the long-wavelength mode the enhancement is as large as 100 near the interface, and persists at depths down to 50 nm. We show numerically that both modes can be coupled to infrared intersubband transitions in GaAs-based multiple quantum wells, yielding an absorbed power gain as high as 5.5, and allowing light absorption at normal incidence. Experimentally, we demonstrate this coupling in a nanoparticle/multiple quantum well structure, where under p-polarization the intersubband absorption is enhanced by a factor of 2.5 and is still observed under s-polarization, forbidden by the usual absorption selection rules. Thus, the integration of CdZnO on GaAs can help improve the figures of merit of quantum well infrared photodetectors, concept that can be extended to other midinfrared detector technologies.

Examined in this paper have been optical properties of polycrystalline films Zn2SnO4 deposited using the spray pyrolysis method within the range of substrate temperatures 250 °C to 450 °C in increments of 50 °C. The spectral dependences have been found for the following physical quantities: k(), n(), ε1(), ε2() and defined as they change under the influence of substrate temperature Тs. Moreover, using the model by Wemple–DiDomenico it was calculated the dispersion energy Ео and Ed for this oxide. Two independent methods defined band gaps Zn2SnO4, which decreases from 4.21…4.22 eV down to 4.04…4.05 eV with increasing Тs from 250 °C up to 450 °C.
