5. Evaluation of the In2Care Mosquito Station at low deployment density: a field study to manage Aedes aegyptiand Culex quinquefasciatus(Diptera: Culicidae) in North Central Florida

Abstract

In the last 2 decades, there has been an increase in the geographic range and frequency of vector-borne diseases. Management of mosquito populations has become challenging due to increasing rates of resistance to existing insecticidal products and formulations. Several alternative tools have emerged to suppress or replace mosquito populations. One of these tools is the In2Care Mosquito Station (In2Care station). This dual-action station contains the insect growth regulator pyriproxyfen which disrupts the development of immatures and the entomopathogenic fungus Beauveria bassiana (B. bassiana) strain GHAwhich kills exposed adult mosquitoes. The In2Care stations have previously been shown to effectively control Aedes aegyptiin field settings at a density of 6 stations/acre rather than the label-recommended 10 stations/acre. To further test the efficacy of low station density deployment, we deployed In2Care stations in the Pleasant Street Historic District of Gainesville, Florida, at a density of 3 stations/acre over a period of 2 years in the presence or absence of ground larvicidal applications. The deployment of stations resulted in no measurable impact on Ae. aegyptiand Culex quinquefasciatusadult or immature abundance suggesting that the low-density deployment of In2Care stations is insufficient to reduce Ae. aegyptiand Cu. quinquefasciatusabundance within treatment areas.Graphical Abstract

11. Species complex of mosquitos captured by mechanical traps and an equine host

Abstract

Field studies were conducted to evaluate the efficacy of commercially available mosquito traps to capture suspected West Nile Virus (WNV) vectors and compare the relative abundance, and species composition of mosquitoes captured by traps. Traps included the Mosquito Magnet (MM) Pro, MM-Liberty, MM-X, the CDC-1012, and the Home and Garden (HGM) prototype trap. In the first study, conducted at the University of Florida Horse Teaching Unit (HTU), Gainesville, FL, from May 17 to September 30, 2002, the CDC-1012 caught significantly more mosquitoes than the other traps. The HGM trap did not catch any mosquitoes and was replaced with a horse on July 17. Percentages of WNV vector species captured in traps and vacuumed from the horse, respectively, were Culex nigripalpus Theobald 85-91% and 27%, and Mansonia titillans (Walker) 2-5% and 40%. In the second study, performed for 5 days in April 2003, the MM-Pro, MM-X, and CDC-1012 traps caught significantly more mosquitoes than did the MM-Liberty trap. All traps captured primarily Cx. salinarius Coquillett, Cx. erraticus (Dyar and Knab), and Anopheles crucians Wiedemann. The horse attracted a greater percentage of Ma. titillans and less Cx. erraticus than did the mechanical traps. The mechanical traps may offer horses some degree of protection from suspected WNV species, however, a large number of Ma. titillans were recovered from the horse but the traps captured very few.

Abstract

Mosquitoes use chemical codes to locate and discriminate among vertebrate hosts to obtain a blood meal. Recent advances have allowed for the identification of the chemical codes used by mosquitoes to locate and discriminate humans from other vertebrate hosts. Humans are incidental “dead-end” hosts for the West Nile virus, which is maintained in an enzootic cycle, primarily through its transmission between infected birds by Culex mosquitoes. Host-seeking Culex mosquitoes are attracted to the odor of chicken, which are used in sentinel traps to monitor West Nile virus transmission. Using combined gas chromatography and electroantennography and mass spectrometry we identify a blend of volatile organic compounds present in chicken emanates, including mostly salient bioactive compounds previously identified in human emanates. When released at their identified ratios, this blend elicits behavioral responses of Culex pipiens molestus and Culex quinquefasciatus similar to that to the natural chicken odor. Tested under field conditions, this blend attract Culex spp. and other species of mosquitoes using birds among their hosts. This study provides evidence for conserved chemical codes for resource location by mosquitoes, and highlights the intricate role of CO2 for host-seeking mosquitoes. The identification of conserved chemical codes, which drive innate preference behaviors that are fundamental for survival and reproduction, provides important substrates for future control interventions targeting disease vector mosquitoes.

Abstract

Volatile compounds emanated from human skin were studied by gas chromatography/mass spectrometry (GC/ MS). The purpose of this study was to identify compounds that may be human-produced kairomones which are used for host location by the mosquito, Aedes aegypti (L.). The procedure used to collect volatiles was chosen because of prior knowledge that attractive substances can be transferred from skin to glass by handling. Laboratory bioassays have shown that the residuum on the glass remains attractive to mosquitoes until the compounds of importance evaporate. The sampling and analytical procedures modeled the above-cited process as closely as possible except that the evaporation of compounds from the glass surface was accomplished by thermal desorption from glass beads in a heated GC injection port. This made possible the solventless injection of volatiles onto the column. The compounds were cryofocused on the head of the column with liquid nitrogen prior to GC separation. A single stage of mass spectrometry on a triple quadrupole instrument was used for mass analysis. A combination of electron ionization and pulsed positive ion/negative ion chemical ionization modes on two different GC columns (one polar, one relatively nonpolar) was used to identify most of the 346 compound peaks detected by this technique.

36. Olfactometric Comparison of the Volatile Insecticide, Metofluthrin, Through Behavioral Responses of Aedes albopictus(Diptera: Culicidae)

Abstract

Testing behavioral response to insecticidal volatiles requires modifications to the existing designs of olfactometers. To create a testing apparatus in which there is no chemical memory to confound tests, we detail the technical aspects of a new tool with design influences from other olfactometry tools. In addition, this new tool was used to evaluate a novel formulation of metofluthrin for use as an outdoor residual treatment. After sourcing materials to prioritize glass and metal construction, a modular wind tunnel was developed that hybridizes wind tunnel and olfactometer specifications. Volatile contaminants were removed by strong ultraviolet light within the chamber before and between trials. Repellent trials were conducted with an experimental formulation of metofluthrin and a commercial formulation of esfenvalerate, prallethrin, and piperonyl butoxide (Onslaught Fast Cap) against Aedes albopictus(Skuse). Toxicant vapors were delivered with attractants from a lure with behavioral responses scored 20 min post-exposure. Upwind attraction to the attractant lure and the Onslaught Fast Cap plus lure resulted in 90 and 75% capture, respectively. In contrast, metofluthrin vapors resulted in less than 10% attraction, while also causing repellency, disorientation, knockdown, and mortality effects. Our findings demonstrated that an inert modular wind tunnel was functional for mitigating toxic secondary exposures of spatial repellents amidst complex behavioral analysis in mosquitoes. The resulting observations with formulated metofluthrin positively reinforce the merit of transitioning metofluthrin into expanded roles in mosquito management.
