In modern society, obesity has become a severe worldwide health problem and related diseases like metabolic syndrome and type 2 diabetes are intractable challenges for public health. Adipose tissues are one of the most important metabolic organs and the main energy storage tissues. In my dissertation I studied the functions of two important proteins; Kindlin-2 and PAT2 in adipose tissues. Kindlin-2, as an integrin activator, plays a crucial role in ECM signal transduction, but its function in adipose tissue is less known. In my study, Kindlin-2 knockout in adipose tissue causes lipoatrophy. Furthermore, angiogenesis in adipose tissue and whole body glucose homeostasis are also affected by Kindlin2 knockout. PAT2 is a surface marker for brown adipocytes, but its function in brown adipocytes is not clear. Several functions of PAT2 are revealed in my study. First, PAT2 is found to affect acidification during amino acid depletion induced autophagy through modulation of V-ATPase activity in brown adipocytes. Second, its intracellular localization is mTORC1 dependent. Third, PAT2 regulates mitochondrial function and cellular amino acid sensitivity in the brown adipocyte. My study demonstrates the importance of Kindlin-2 in adipose tissues and the crucial role of PAT2 in brown adipocyte to nutrient homeostasis and development. These two proteins can be the targets for the therapy of obesity related diseases. Research of Kindlin-2 and PAT2 function are helpful to understand the physiological functions of adipose tissues in more detail., In der heutigen Gesellschaft sind Adipositas, sowie die damit assoziierten Erkrankungen, wie das metabolische Syndrom und Typ 2 Diabetes zu einem globalen Problem und einer Herausforderung für die öffentliche Gesundheit geworden. Fettgewebe sind eine der wichtigsten metabolische Gewebe und der Hauptspeicherort für überschüssige Energie. In meiner Arbeit habe ich mich mit der Funktion von zwei wichtigen Proteinen, Kindlin-2 und PAT2, im Fettgewebe beschäftigt. Kindlin-2, als ein zentrales Element der Integrinaktivierung, spielt eine zentrale Rolle in der Signaltransduktion der Extrazellulären Matrix. Die Funktion von Kindlin-2 im Fettgewebe ist bislang jedoch nur lückenhaft untersucht worden. In meinen Studien konnte ich zeigen, dass der Knockout von Kindlin-2 im Fettgewebe zu einer Lipodystrophie führt. Des Weiteren führt der Verlust von Kindlin-2 im Fettgewebe zu Störungen der Vaskularisierung und der Glukosehomöostase. PAT2 wurde in vorhergehenden Studien als spezifischer Oberflächenmarker für braune Adipozyten identifiziert. Bislang ist jedoch sehr wenig über die Funktion von PAT2 im braunen Fettgewebe bekannt. In meinen Studien konnte ich mehrere Funktionen und Regulationen von PAT2 aufzeigen. Zuerst konnte ich zeigen, dass PAT2 die lysosomale Azidifizierung während der durch Aminosäuredepletion induzierten Autophagie, durch die Modulation der vATPase Aktivität, in braunen Adipozyten beeinflusst. Zweitens, ist die intrazelluläre Verteilung von PAT2 von der mTORC1 Aktivität abhängig. Drittens reguliert PAT2 mitochondriale Funktion und zelluläre Amionsäuresensitivität in braunen Adipozyten. Meine Studien zeigen die Bedeutung von Kindlin-2 im Fettgewebe und die Rolle von PAT2 in braunen Adipozyten in Bezug auf Nährstoffhomäostase und Entwicklung. Diese zwei Proteine stellen zwei potentielle Angriffspunkte für die Therapie von Adipositas bezogenen Erkrankungen dar. Forschung an Kindlin-2 und PAT2 hilft die physiologische Funktion des Fettgewebes besser zu verstehen.

This thesis investigates possibility to improve reliability and performance of low-cost aeronautical integrated navigation systems by making use of robust autonomous measurements, namely of inertial measurement unit, air data system, and 3-axis magnetometer. The central applied result of the research consists in proposing low size, weight, power, and cost design of a robust platform-autonomous fault-tolerant navigation system with the best in-IMU-class performance, which, in the events of failures, can gracefully degrade to attitude and heading reference system., Diese Arbeit untersucht die Möglichkeit die Zuverlässigkeit und Leistung kostengünstiger, integrierter Navigationssysteme für die Luftfahrt zu verbessern. Hierzu werden robuste, autonome Messungen wie etwa aus der Inertialen Messeinheit, Luftdaten sowie 3-Achsen Magnetometer verwendet. Als zentrales, anwendbares Ergebnis dieser Forschung der Entwurf eines kleinen, leichten und kostengünstigen plattform-autonomen fehlertoleranten Navigationssystems vorgeschlagen.

Flight guidance & control systems for safety critical applications have to handle sensor failures with high reliability. Fundamental prerequisite is a statistically consistent flight state estimate of the integrated navigation system. By applying error identification techniques for sensor, model and observation errors, heuristic navigation filter tuning methods are superseded by a physically motivated approach. Further emphasis is on filter initialization, fusion of out-of-sequence observations and Kalman filter stability., Flugführungs- und Regelungssysteme für sicherheitskritische Anwendungen müssen auf Sensorfehler mit hoher Zuverlässigkeit reagieren. Wichtige Voraussetzung ist eine statistisch konsistente Flugzustandsschätzung durch das integrierte Navigationssystem. Mit der Identifikation von Sensor-, Modellierungs- und Beobachtungsfehlern wird die heuristische Herangehensweise beim Filter-Tuning durch einen physikalisch motivierten Ansatz abgelöst. Weitere Themen sind Filterinitialisierung, Fusion aus der Reihe ankommender Messungen und Kalman-Filter Stabilität.

Controlling a ship in a berthing operation is carried out mainly by the change of state, such as velocity and yaw rate (turn rate), although the value of the change of state is very small at berthing. Very high precision is, therefore, required to determine the velocity and angular velocity. A sensor that has an accuracy of ±0.02°/s (1 s) is sought for determination of turn rate in a berthing system. Three-dimensional angular velocity can directly be determined, with 2 independent baselines of 3 GPS antennas, using instantaneous Doppler measurements or phase rate (temporal difference of phase) observations. This paper discusses the mathematical model for direct determination of angular velocity using GPS, and the comparison of the results of the angular velocity determination using the Doppler and phase rate. The precision of angular velocity determination is estimated using temporal difference of the attitude sensors (TSS and gyrocompass) on board a hydrographic sounding ship. The RMS values of the difference of yaw rate determination between the two systems were: ±0.16°/s using phase rate and ±0.31°/s using Doppler measurements with the separation of onboard antennas of ca. 1·34 m. 10 m baselines could satisfy the sensor requirements for angular velocity determination during berthing maneuvers.

This report considers the feasibility of using the Global Positioning System (GPS) to obtain three-dimensional positions, at decimeter (or better) accuracies, in real time for hydrographic survey vessels, dredges, and offshore tide gage buoys. The various technical constraints associated with using GPS to obtain high-accuracy positions of a moving platform are discussed. Real-time, decimeter-level kinematic differential GPS positioning can only be obtained if GPS carrier beat phase ambiguities can be resolved on the fly. Resolution of carrier beat phase ambiguities on the fly can only be achieved (if at all) with improved P-code pseudoranging. Dual-frequency receivers will be required. Codeless techniques for tracking the P-code may be almost as useful as code- correlation techniques in providing pseudorange measurements for carrier beat phase ambiguity resolution. Problems in four areas remain to be solved if decimeter-level systems are to be feasible: reducing pseudorange noise, avoiding multipath, overcoming possible adverse effects of selective availability, and overcoming the effects of vessel dynamics. This report documents the findings of a study to design a kinematic positioning system to support U.S. Army Corps of Engineers dredging and hydrographic survey operations. Survey quality GPS receivers are considered, due to the accuracy constraints (10 cm vertical positioning). Use of auxiliary sensors (i.e., INS and high quality clocks) is considered to improve navigation performance when GPS is unavailable. Predicted navigation performance indicates that the accuracy requirements can be met, except at the extremes of the expected separation distances from the reference station. The integer ambiguities associated with the carrier phase measurements can generally be resolved within 7 minutes by processing all available satellites. Measurement gaps of roughly 60 seconds can be tolerated without losing the integers when using a high quality INS....
