Abstract Aldehydes represent a versatile and favored class of flavoring substances. A biocatalytic access to odor-active aldehydes was developed by conversion of fatty acids with two enzymes of the α-dioxygenase pathway. The recombinant enzymes α-dioxygenase (α-DOX) originating from Crocosphaera subtropica and fatty aldehyde dehydrogenase (FALDH) from Vibrio harveyi were heterologously expressed in E. coli, purified, and applied in a coupled (tandem) repetitive reaction. The concept was optimized in terms of number of reaction cycles and production yields. Up to five cycles and aldehyde yields of up to 26% were achieved. Afterward, the approach was applied to sea buckthorn pulp oil as raw material for the enzyme catalyzed production of flavoring/fragrance ingredients based on complex aldehyde mixtures. The most abundant fatty acids in sea buckthorn pulp oil, namely palmitic, palmitoleic, oleic, and linoleic acid, were used as substrates for further biotransformation experiments. Various aldehydes were identified, semi-quantified, and sensorially characterized by means of headspace–solid phase microextraction–gas chromatography–mass spectrometry–olfactometry (HS–SPME–GC–MS–O). Structural validation of unsaturated aldehydes in terms of double-bond positions was performed by multidimensional high-resolution mass spectrometry experiments of their Paternò–Büchi (PB) photoproducts. Retention indices and odor impressions of inter alia (Z,Z)-5,8-tetradecadienal (Z,Z)-6,9-pentadecadienal, (Z)-8-pentadecenal, (Z)-4-tridecenal, (Z)-6-pentadecenal, and (Z)-8-heptadecenal were determined for the first time. Es sind Abrechnungen mit einer inhumanen Vergangenheit, sie atmen das Credo der Bilanz, einer Rechenschaftslegung, einer harten Selbstbefragung. Erst mit diesen – teils authentischen, teils fiktiven – Bekenntnisbuchern haben die Autoren ein sie qualendes Kapitel ihrer Lebensgeschichte, das zugleich ein unbewaltigtes Kapitel der Geschichte der Deutschen ist, niedergeschrieben und sich von einem ihr Leben belastenden Alpdruck befreit. Der Beitrag analysiert diese drei Werke und charakterisiert ihre jugendlichen Protagonisten. In welcher Beziehung steht der Autor zu seinem 16- bzw. 17jahrigen Alter Ego in dem jeweiligen Werk? Es wird herausgearbeitet, wie Muller sein Thema – den Verlust der Heimat – mit seines Protagonisten schmerzvoller Verabschiedung der Vergangenheit und dessen zunehmendem Begreifen von realer geschichtlicher Gegenwart verbindet. Fur Grass ist dagegen zu konstatieren, dass der geheime Motor fur sein Schreiben wohl immer die subjektiv empfundene Schuld – seine einstige Verstricktheit im Nationalsozialismus – gewesen war. This paper analyzes transversal thermoacoustic oscillations in an experimental gas turbine combustor utilizing dynamical system theory. Limit-cycle acoustic motions related to the first linearly unstable transversal mode of a given 3D combustor configuration are modeled and reconstructed by means of a low-order dynamical system simulation. The source of nonlinearity is solely allocated to flame dynamics, saturating the growth of acoustic amplitudes, while the oscillation amplitudes are assumed to always remain within the linearity limit. First, a reduced order model (ROM) which reproduces the combustor's modal distribution and damping of acoustic oscillations is derived. The ROM is a low-order state-space system, which results from a projection of the linearized Euler equations (LEE) into their truncated eigenspace. Second, flame dynamics are modeled as a function of acoustic perturbations by means of a nonlinear transfer function. This function has a linear and a nonlinear contribution. The linear part is modeled analytically from first principles, while the nonlinear part is mathematically cast into a cubic saturation functional form. Additionally, the impact of stochastic forcing due to broadband combustion noise is included by additive white noise sources. Then, the acoustic and the flame system is interconnected, where thermoacoustic noncompactness due to the transversal modes' high frequency (HF) is accounted for by a distributed source term framework. The resulting nonlinear thermoacoustic system is solved in frequency and time domain. Linear growth rates predict linear stability, while envelope plots and probability density diagrams of the resulting pressure traces characterize the thermoacoustic performance of the combustor from a dynamical systems theory perspective. Comparisons against experimental data are conducted, which allow the rating of the flame modes in terms of their capability to reproduce the observed combustor dynamics. Ultimately, insight into the physics of high-frequency, transversal thermoacoustic systems is created.

Nerve cells are continuously generated from stem cells in the adult mammalian subventricular zone (SVZ) and hippocampal dentate gyrus. We have previously noted that stem/progenitor cells in the SVZ and the subgranular layer (SGL) of the dentate gyrus express high levels of plasma membrane-bound nucleoside triphosphate diphosphohydrolase 2 (NTPDase2), an ectoenzyme that hydrolyzes extracellular nucleoside diphosphates and triphosphates. We inferred that deletion of NTPDase2 would increase local extracellular nucleoside triphosphate concentrations perturbing purinergic signaling and boosting progenitor cell proliferation and neurogenesis. Using newly generated mice globally null for Entpd2, we demonstrate that NTPDase2 is the major ectonucleotidase in these progenitor cell-rich areas. Using BrdU-labeling protocols, we have measured stem cell proliferation and determined long-term survival of cell progeny under basal conditions. Brains of Entpd2 null mice revealed increased progenitor cell proliferation in both the SVZ and the SGL. However, this occurred without noteworthy alterations in long-term progeny survival. The hippocampal stem cell pool and the pool of the intermediate progenitor type-2 cells clearly expanded. However, substantive proportions of these proliferating cells were lost during expansion at around type-3 stage. Cell loss was paralleled by decreases in cAMP response element-binding protein phosphorylation in the doublecortin-positive progenitor cell population and by an increase in labeling for activated caspase-3 levels. We propose that NTPDase2 has functionality in scavenging mitogenic extracellular nucleoside triphosphates in neurogenic niches of the adult brain, thereby acting as a homeostatic regulator of nucleotide-mediated neural progenitor cell proliferation and expansion. Cellular signaling via extracellular nucleotides appears to play a major role in the functioning of the peripheral nervous system. Information regarding the functional characterization of nucleotide P2 receptors or their expression pattern has been accumulating rapidly; however, very little is known regarding the distribution of ecto-nucleotidases in the periphery. The extracellular level of nucleotides is controlled by ecto-nucleotidases, whereby the three membrane-bound members of the ecto-nucleoside triphosphate diphosphohydrolase (E-NTPDase) family are of special functional importance. Using enzyme histochemistry and immunostaining, we demonstrate that NTPDase2 is associated with nonmyelinating Schwann cells of the rat sciatic nerve, whereas NTPDase1 is restricted to blood vessel walls. NTPDase2 immunoreactivity was detected from embryonic day E18 onward, suggesting that immature Schwann cells express the enzyme. With the onset of myelination, NTPDase2 immunoreactivity remained associated solely with nonmyelinating Schwann cells. NTPDase2 was absent from perisynaptic Schwann cells but was associated with fibroblasts covering the endplate at some distance. In addition, NTPDase2 immunoreactivity was associated with the satellite glial cells in dorsal root ganglia and sympathetic ganglia, and with the enteric glia surrounding the cell bodies of ganglionic neurons of the myenteric and the submucous plexus. In contrast to NTPDase1, NTPDase2 preferentially hydrolyzes nucleoside triphosphates over nucleoside diphosphates and thus can act either in inactivating or in producing P2 receptor ligands. Our results suggest that NTPDase2 plays an important role in the control of nucleotide-mediated activation of peripheral neurons or glia and in the dialogue between these two cell types.

In the adult nervous system, multipotential stem cells of the subventricular zone of the lateral ventricles generate neuron precursors (type-A cells) that migrate via the rostral migratory stream to the olfactory bulb where they differentiate into neurons. The migrating neuroblasts are surrounded by a sheath of astrocytes (type-B cells). Using immunostaining, in situ hybridization and enzyme histochemistry, we demonstrate that the ecto-ATPase nucleoside triphosphate diphosphohydrolase 2 (NTPDase2) is expressed in the subventricular zone and the rostral migratory stream of the adult rat brain. This enzyme hydrolyses extracellular nucleoside triphosphates to the respective nucleoside diphosphates and is thought to directly modulate ATP receptor-mediated cell communication. Double labelling for the astrocyte intermediate filament protein GFAP and the glial glutamate transporter GLAST identifies the NTPDase2-positive cells as type-B cells. During development the enzyme protein is first detected at E18, long before expression of the astrocyte marker GFAP. It gradually becomes expressed along the ventricular and subventricular zone of the brain, followed by complete retraction to the adult expression pattern at P21. NTPDase2 is transiently expressed in the outer molecular layer of the dentate gyrus and within the cerebellar white matter and is associated with select microvessels, tanycytes of the third ventricle, and subpial astrocytes of the adult brain. Our results suggest that NTPDase2 can serve as a novel marker for specifying subsets of cells during in vivo and in vitro studies of neural development and raise the possibility that ATP-mediated signalling pathways play a role in neural development and differentiation.

These receptors initiate a wide variety of signalling pathways that appear important for functional associations between neurons and glial cells and for the regulation of blood flow, haemostatic and inflammatory reactions in the brain. Ectonucleotidases are extracellular nucleotide-metabolizing enzymes that modulate P2 receptor-mediated signalling by the regulated hydrolysis of these agonists. A considerable number of ectoenzyme species with partially overlapping substrate and tissue distributions have been described. Major candidates for expression in the brain are members of the ecto-nucleoside triphosphate diphosphohydrolase (E-NTPDase or CD39) family. The production of cd39-/- mice and specific reagents have enabled us to analyse the specific cellular distribution of NTPDase1 (CD39), the prototype member of the enzyme family, in the mouse brain. Using monospecific antibodies and enzyme histochemical staining, we have identified NTPDase1 as a major ectonucleotidase associated with both microglia and the endothelial and smooth muscle cells of the vasculature. NTPDase1 is not expressed by neurons and astrocytes. Additional unidentified ectonucleotidase functional activity is observed at lower levels throughout the brain parenchyma. NTPDase1 may regulate P2 receptor-mediated functions of microglia as well as influence nucleotide signalling between neurons or astrocytes that are associated with multiple microglial ramifications. The expression of NTPDase1 by cerebrovascular endothelial and smooth muscle cells also suggests involvement in the regulation of blood flow and thrombogenesis.

Objective: we examined the diagnostic accuracy of different methods of body temperature measurement to diagnose infection in geriatric patients presenting to the emergency department (ED). Methods: this observational study was done in consecutive patients ≥75 years old presenting to the ED. Body temperature was determined by tympanal thermometry, temporal artery thermometry and rectal temperature measurement. Adjudicated final diagnosis of infection was done by two experts including patient history, clinical and laboratory findings as well as radiographic studies. Results: a total of 427 patients were included in the data analysis (age: 82.7 ± 5.1 years). Infection was present in 105 patients (24.6%). Respiratory rate, heart rate and body temperature were significantly higher in patients with infection, blood pressure was lower (P< 0.01). Body temperature measured by tympanal and temporal artery thermometry was correlated with rectal thermometry. Body temperature was significantly higher in patients with infection compared with those without infection independent of the method of body temperature measurement (P< 0.001). The diagnostic accuracy for infection quantified by the area under curve (AUC) was comparable among rectal [AUC: 0.72 (95% CI: 0.65–0.80)] and tympanal thermometry [AUC: 0.73 (95% CI: 0.66–0.81)], but significantly lower in temporal artery thermometry [AUC: 0.65 (95% CI: 0.57–0.73; P< 0.001)]. Compared with rectal measurement tympanal thermometry showed a higher bias than temporal artery thermometry (0.54 versus 0.03°C), while its limits of agreement were more narrow (−0.14 to 1.21°C versus −0.94–1.01°C). Conclusion: diagnostic accuracy for the identification of infection was comparable among tympanal and rectal thermometry and lower for temporal artery thermometry. Different cut-off points should be used to identify infection using tympanal (37.3° C) or rectal (37.9°C) thermometry. ATP-mediated synaptic transmission represents the only transmitter-gated Ca2+-entry pathway in neurons of the rodent medial habenula. In addition to direct purinergic receptor-mediated synaptic inputs, the medial habenula contains purinergic systems that modulate synaptic transmission. Purinergic signaling is modulated or terminated by ectonucleotidase, nucleotide-hydrolyzing enzymes of the cell surface. Here we identify the major ectonucleotidase responsible for the hydrolysis of extracellular ATP in the mouse medial habenula as ectonucleoside triphosphate diphosphohydrolase 2 (NTPDase2), using immunostaining and enzyme histochemistry. Double labeling experiments reveal that the enzyme is expressed by astrocytes enwrapping the densely packed neurons and also the myelinated fiber bundles of the stria medullaris. NTPDase2 immunoreactivity is absent from the lateral habenula. The analysis of mice expressing enhanced green fluorescent protein under the promoter of glial fibrillary acidic protein revealed that the medial habenula harbors a highly polar type of astrocytes with very long laminar cellular processes, untypical for grey matter astrocytes. Its morphology strongly differs from that of the stellate astrocytes in the adjacent lateral habenula. Our results suggest that the mouse medial habenula contains a specific perineuronal nonstellate subtype of astrocyte that expresses the ectonucleotidase NTPDase2 and is in a strategic position to modulate purinergic transmission in this subnucleus. © 2012 Wiley Periodicals, Inc.

Nucleotides comprise a major class of signaling molecules in the nervous system. They can be released from nerve cells, glial cells, and vascular cells where they exert their function via ionotropic (P2X) or metabotropic (P2Y) receptors. Signaling via extracellular nucleotides and also adenosine is controlled and modulated by cell-surface-located enzymes (ectonucleotidases) that hydrolyze the nucleotide to the respective nucleoside. Extracellular hydrolysis of nucleotide ligands involves a considerable number of enzymes with differing catalytic properties differentially affecting the nucleotide signaling pathway. It is therefore important to investigate which type of ectonucleotidase(s) contributes to the control of nucleotide signaling in distinct cellular and physiological settings. By using a classical enzyme histochemical approach and employing various substrates, inhibitors, and knockout animals, we provide, for the first time, a comparative analysis of the overall distribution of catalytic activities reflecting four ectonucleotidase families: ecto-5′-nucleotidase, alkaline phosphatases, ectonucleoside triphosphate diphosphohydrolases (E-NTPDases), and ectonucleotide pyrophyphatases/phosphodiesterases (E-NPPs). We place into perspective the earlier literature and provide novel evidence for a parenchymal localization of tissue non-specific alkaline phosphatase, E-NPPs, and E-NTPDases in the mouse brain. In addition, we specify the location of ectonucleotidases within the brain vasculature. Most notably, brain vessels do not express ecto-5′-nucleotidase. The preponderance of individual enzymes differs considerably between brain locations. The contribution of all types of ectonucleotidases thus needs to be considered in physiological and pharmacological studies of purinergic signaling in the brain.

Massachusetts An astrocyte-like cell population corresponding to residual radial glia represents the neuronal progenitors of the adult mammalian hippocampus. We show that radial glia-like cells of the dentate gyrus express surfacelocated ATP-hydrolyzing activity and are immunopositive for NTPDase2. This enzyme hydrolyzes extracellular nucleoside triphosphates such as ATP and UTP to their nucleoside diphosphates and is thus involved in the control of signaling via P2 receptors. NTPDase2 is expressed from embryonic day 17 onward. In the hippocampus, the embryonic pattern of NTPDase2 expression mirrors that of the dentate migration of neuroblasts. Double-immunolabeling revealed that NTPDase2 is associated with subpopulations of glial fibrillary acidic protein-, nestin- and doublecortin-positive radial cells. It is absent from mature granule cells or S100-positive astrocytes. NTPDase2-positive cells proliferate. Furthermore, after mitosis, progenitor cells preferentially reveal an NTPDase2-positive phenotype. Patch-clamp analysis demonstrates functional nucleotide receptors in progenitor cells expressing nestin promotor-driven green fluorescent protein. Our results identify the ecto-nucleotidase NTPDase2 and functional P2X receptors at hippocampal progenitor cells. Using antisera directed against octopamine (OA), we identified and mapped octopamine-immunoreactive (OA-ir) neurons and their projections in the fused, central ganglion complex of wandering spiders, Cupiennius salei. Labeled cell bodies are concentrated in the subesophageal ganglion complex (SEG) where they are arranged serially in ventral, midline clusters. OA-ir processes from these cells project dorsally. Some neurites end close to segmental septa; others merge into longitudinal tracts connecting the neuromeres. Labeled collaterals leaving these tracts project into peripheral neuropil. In the brain, OA-ir somata were found only in the two cheliceral hemiganglia, where a cluster of 4-5 relatively large cells (soma diameter 25 microns) lies next to a group of small somata (diameter10 microns). Neurites originating from the large somata descend into the SEG and merge into longitudinal tracts. The central body of the brain contains profuse ascending projections. Except for fine varicosities that are confined to the roots of nerves, we found no OA-ir fibers leaving the central nervous system (CNS). Within the CNS, however, OA-ir varicosities are concentrated in neuropil and near hemolymph spaces. This distribution suggests that OA acts as a neurotransmitter and/or local neuromodulator at central synapses, while it is also released into the hemolymph and presumably acts hormonally at peripheral sites. Using high-pressure liquid chromatography measurements, the hemolymph was in fact found to contain 12-40 nM of free octopamine.

Coxal hair-plate sensilla in the spider Cupiennius salei are described with respect to their innervation, central projection pattern, electrical response to mechanical stimulation, and putative behavioral function. 1. Hair plates, each comprising 25–70 hairs, are situated on the ventrolateral leg coxae close to the prosomal joint; during coxal movements they are deflected by the bulging joint membrane. Each plate hair is innervated by one sensory cell. 2. Threshold sensitivity lies at 0.5° to 1° of hair deflection. Only distalward deflection excites. During maintained deflections the spike rate declines slowly. These hairs differ from hair sensilla of insects in that we measure no ‘standing potential’, nor do we measure a ‘receptor potential’ accompanying a mechanical stimulus. 3. The central projection areas of both hair plates are limited to neuropil of the ipsilateral neuromere. 4. Natural stimulus situation and the spike response to maintained deflection suggest that these hairs are used in proprioception and graviception. Yet behavioral changes following selective hair-plate ablations are not very pronounced. Unilateral removal of hair-plates produced significant increases in average body height in 7 of 10 animals, while the angular orientation of the long body axis with respect to gravity remained unchanged after hair-plate removal.

Retrograde CoS-impregnation was used to trace and map the course of sensory nerves and the distribution and innervation of the various proprioceptor types in all leg segments of Cupiennius salei, a Ctenid spider. 1. Sensory nerve branches. In both the tibia and femur, axons of all proprioceptor types ascend in just two lateral nerves which do not merge with the main leg nerve until they reach the next proximal joint region. In the short segments — coxa, trochanter, patella, and tarsus — axons of the internal joint receptors often run separately from those of the other sensilla. Axons of the large lyriform slit sense organ at the dorsal metatarsus and of the trichobothria join with only a few hair axons and form their own nerve branches (Figs. 1, 2, 3). 2. Proprioceptors. Each of the seven leg joints is supplied with at least one set of the well-known internal joint receptors, slit sensilla (single slits and lyriform organs), and long cuticular hairs. In addition, we found previously unnoticed “hair plates” on both sides of the coxa, near the prosoma/coxa joint; they are deflected by the articular membrane during joint movements (Fig. 4). 3. Sensory cells and innervation. CoS-impregnation shows that each slit of the slit sense organs — be it a single slit or several slits in a lyriform organ — is innervated by two bipolar sensory cells (Fig. 6). We also confirm previous reports of multiple innervation in the internal joint receptors and in the long joint hairs and cuticular spines. Most of the ascending nerve branches run just beneath the cuticle for at least a short distance (Fig. 5); hence they are convenient sites for electrophysiological recordings of sensory activity even in freely walking spiders.

Rows of long, smooth hair sensilla situated on both sides of the leg coxae were examined in the spider Cupiennius salei (Ctenidae). The hair shafts point into the space between adjacent legs and are deflected when the hairs of one coxa touch the cuticle of the neighboring coxa. 1. Unlike the serrated hair shafts of the ubiquitous tactile and chemosensitive setae of spiders, these hairs are entirely smooth. At their base they are articulated in a socket with an asymmetrical groove that determines the direction of hair deflection. Hair shafts are up to 1000 μm long. The exact grouping of smooth hairs in rows is typical of the coxae for each pair of legs. 2. Unlike the other, multiply innervated cuticular sensilla of spiders, smooth hairs are supplied by only a single mechanosensitive neuron. This is confirmed by electrophysiological recordings from single hairs. Threshold deflection to elicit a spike response lies near 1°. The response to maintained, step-like stimuli declines rapidly. 3. All central endings of these hair receptors in the fused segmental ganglia are confined to dorsal neuropil of the ipsilateral neuromere. The specific arborization pattern resembles an elongated, three-pronged fork with a long central prong. Topography, natural stimulus situation, and the “phasic” response characteristic of smooth hairs suggest that spiders use these sensilla to monitor the relative distance between leg coxae during locomotion.
