Objectives Cancer therapy including chemotherapy causes gland atrophy, resulting in low salivary secretion in cancer patients. Since saliva plays an important role in oral health, the dysfunction may exacerbate oral ulcerative mucositis (OUM), which is another side effect. Here, we investigated the effect of hyposalivation on OUM using sialoadenectomized rats and examined the effects of anticancer drugs on the salivary glands. Design As models for hyposalivation, the bilateral submandibular and sublingual glands except (2EXT) or together with (3EXT) the parotid glands were extracted. At 16 days after the procedure, OUM was experimentally developed by topical acetic acid treatment on the labial fornix region of the inferior incisors, and the severity and bacterial loading level were evaluated. The salivary gland weights and histology were analyzed after administration of the representative anticancer drugs 5-fluorouracil or cisplatin. Results The severity of OUM was greater in both the 3EXT and 2EXT rats and delayed the healing process compared with that in sham rats without salivary gland extraction. The healing process in the 3EXT rats was longer than that in the 2EXT rats. The number of colony-forming units in the ulcerative region from the 3EXT rats was 10-fold greater than that in the sham rats. Both 5-fluorouracil and cisplatin reduced glands weights and damaged the salivary glands. Conclusions These results suggest that chemotherapy-induced hyposalivation exacerbates OUM and delays healing, most likely due to loss of salivary clearance and antimicrobial functions. This study illustrates the significance of oral health care for cancer patients undergoing chemotherapy.

Objective Recent studies have demonstrated that mouthwash made with the traditional Japanese medicine hangeshashinto exhibits anti-inflammatory action and alleviates oral mucositis scores, including pain complaints, in patients undergoing chemoradiotherapy. However, no study has demonstrated the mechanism underlying how hangeshashinto provides pain relief in oral ulcers. Design The analgesic effects on pain-related behaviors following the topical application of hangeshashinto were evaluated in an oral ulcer rat model treated with acetic acid using recently developed methods. Indomethacin, the representative anti-inflammatory agent, was intraperitoneally administered. The tissue permeability of the oral mucosa was histologically evaluated after applying the fluorescent substance FluoroGold. Results The topical application of hangeshashinto in ulcerative oral mucosa suppressed mechanical pain hypersensitivity over 60 min, without any effects on healthy mucosa. The same drug application also inhibited oral ulcer-induced spontaneous pain. Indomethacin administration failed to block the mechanical pain hypersensitivity, though it did largely block spontaneous pain. Topical anesthesia with lidocaine showed hyposensitivity to mechanical stimulation in healthy mucosa. In the ulcer regions in which the oral epithelial barrier was destroyed, deep parenchyma was stained with FluoroGold, in contrast to healthy oral mucosa, in which staining was limiting to the superficial site. Conclusions Hangeshashinto leads to long-lasting analgesic effects, specifically in the ulcer region by destroying the epithelial barrier. Hangeshashinto alleviates oral ulcer-induced pain in inflammation-dependent and/or independent manner.

In many patients with cancer, chemotherapy-induced severe oral ulcerative mucositis causes intractable pain, leading to delays and interruptions in therapy. However, the pain mechanism in oral ulcerative mucositis after chemotherapy has not been extensively studied. In this study, we investigated spontaneous pain and mechanical allodynia in a preclinical model of oral ulcerative mucositis after systemic administration of the chemotherapy drug 5-fluorouracil, using our proprietary pain assay system for conscious rats. 5-Fluorouracil caused leukopenia but did not induce pain-related behaviors. After 5-fluorouracil administration, oral ulcers were developed with topical acetic acid treatment. Compared with saline-treated rats, 5-fluorouracil-exposed rats showed more severe mucositis with excessive bacterial loading due to a lack of leukocyte infiltration, as well as enhancements of spontaneous pain and mechanical allodynia. Antibacterial drugs, the lipid A inhibitor polymyxin B and the TRPV1/TRPA1 channel pore-passing anesthetic QX-314, suppressed both the spontaneous pain and the mechanical allodynia. The cyclooxygenase inhibitor indomethacin and the TRPV1 antagonist SB-366791 inhibited the spontaneous pain, but not the mechanical allodynia. In contrast, the TRPA1 antagonist HC-030031 and the N-formylmethionine receptor FPR1 antagonist Boc MLF primarily suppressed the mechanical allodynia. These results suggest that 5-fluorouracil-associated leukopenia allows excessive oral bacterial infection in the oral ulcerative region, resulting in the enhancement of spontaneous pain through continuous TRPV1 activation and cyclooxygenase pathway, and mechanical allodynia through mechanical sensitization of TRPA1 caused by neuronal effects of bacterial toxins. These distinct pain mechanisms explain the difficulties encountered with general treatments for oral ulcerative mucositis-induced pain in patients with cancer and suggest more effective approaches.

Background Stomatitis induces severe and painful hypersensitivity to pungency and physical contact during meals. Many studies have used anesthetized animals to examine evoked nociception in the oral mucosa, but no reports have used traditional behavioral assays to evaluate nociception in conscious animals. New methods We developed two new methods of applying chemical or mechanical stimulation directly to the oral mucosa of the mandibular vestibule of conscious rats. Nociceptive evaluations were performed by measuring facial grooming time and the head withdrawal threshold to von Frey stimulations. (1) For the intraoral dropping method, rat mucosa was transiently exposed by hand, and a drop of a pungent solution was applied. (2) For the stable intraoral opening method, rat mucosa was long-term exposed following piercing surgery of the mental skin after habitual training for 2–3 weeks. Results In the intraoral dropping method, the application of 100 μM capsaicin or 100 mM allyl isothiocyanate prolonged mouth-rubbing time. Capsaicin-induced mouth-rubbing time was further enhanced following the development of an acetic acid-induced ulcer. The stable intraoral opening method enabled stable measurements of the mechanical withdrawal threshold in the oral mucosa of conscious rats. Ulcer development decreased the mechanical threshold, whereas topical lidocaine treatment increased the threshold. Comparison with existing methods These new methods enable the evaluations of motivational nocifensive behaviors in response to intraoral stimulations without any anesthetic effects. Conclusions The intraoral dropping and stable intraoral opening methods can be used in combination with traditional behavioral assays to evaluate nociception in the oral mucosa of conscious rats.

We propose a new mechanism of sensory modulation through cutaneous dopaminergic signalling. We hypothesize that dopaminergic signalling contributes to differential cutaneous sensitivity in darker versus lighter pigmented humans and mouse strains. We show that thermal and mechanical cutaneous sensitivity is pigmentation dependent. Meta-analyses in humans and mice, along with our own mouse behavioural studies, reveal higher thermal sensitivity in pigmented skin relative to less-pigmented or albino skin. We show that dopamine from melanocytes activates the D1-like dopamine receptor on primary sensory neurons. Dopaminergic activation increases expression of the heat-sensitive TRPV1 ion channel and reduces expression of the mechanically-sensitive Piezo2 channel; thermal threshold is lower and mechanical threshold is higher in pigmented skin.

Summary Substantial acute and chronic intakes of alcohol or ethanol (EtOH) severely influence oral sensations, such as thirst and oral dryness (dry mouth, xerostomia). Thirst sensation and oral dryness are primarily caused by the activation of neurons in brain regions, including the circumventricular organs and hypothalamus, which are referred to as the dipsogenic center, and by a decrease in salivary secretion, respectively. The sensation of thirst experienced after heavy-alcohol drinking is widely regarded as a consequence of EtOH-induced diuresis; however, EtOH in high doses induces anti-diuresis. Recently, it has been proposed that the ethanol metabolite acetaldehyde induces thirst via two distinct processes in the central nervous system from EtOH-induced diuresis, based on the results of animal experiments. The present review describes new insights regarding the induction mechanism of thirst sensation and oral dryness after drinking alcohol.

Recent evidence implicates endothelin in nociception, but it is unclear how endothelin activates trigeminal ganglion (TRG) neurons. In the present study, we investigated the expression of the endothelin receptors ETA and ETB and endothelin-induced responses in rat TRG neurons. Double-immunofluorescence studies demonstrated that ETA and ETB were expressed in TRG neurons and that 26% of ETA- or ETB-expressing neurons expressed both receptors. During whole-cell patch-clamp recording, endothelin-1 enhanced an induced current in response to capsaicin, a TRPV1 agonist, in approximately 20% of dissociated neurons. The enhancement was blocked by the PKC inhibitor chelerythrine and by the ETA antagonist BQ-123, but not by the ETB antagonist BQ-788. Ca2+-imaging showed that endothelin-1 increased the intracellular Ca2+ concentration in more than 20% of the dissociated neurons. Importantly, unlike the effect of endothelin-1 on capsaicin-induced current, the Ca2+ response was largely suppressed by BQ-788 but not by BQ-123. These results suggest that ETA-mediated TRPV1 hyperactivation via PKC activation and ETB-mediated Ca2+ mobilization occurs in different subsets of TRG neurons. These endothelin-induced responses may contribute to the induction of orofacial pain. The ETB-mediated function in TRG neurons is a special feature in the trigeminal system because of no ETB expression in dorsal root ganglion neurons.

This review article provides doctors and dentists with an introduction to the clinical applications and sig- nificance of magnetic resonance cisternography (MRC) for the evaluation of neurovascular compression of the trigeminal nerve in patients with trigeminal neu- ralgia (TN). Specifically, the method and characteris- tics of MRC are first explained using the relevant MR sequences. Next, clinical applications for the diagnosis of TN are discussed. Finally, the significance and util- ity of MRC for effective treatment of patients with TN, such as carbamazepine administration and/or local anesthetic blocks, are presented.

The traditional Japanese herbal medicine hangeshashinto (HST) has beneficial effects for the treatment of oral ulcerative mucositis (OUM) in cancer patients. However, the ingredient-based mechanism that underlies its pain-relieving activity remains unknown. In the present study, to clarify the analgesic mechanism of HST on OUM-induced pain, we investigated putative HST ingredients showing antagonistic effects on Na+ channels in vitro and in vivo. A screen of 21 major ingredients using automated patch-clamp recordings in channel-expressing cells showed that [6]-gingerol and [6]-shogaol, two components of a Processed Ginger extract, considerably inhibited voltage-activated Na+ currents. These two ingredients inhibited the stimulant-induced release of substance P and action potential generation in cultured rat sensory neurons. A submucosal injection of a mixture of [6]-gingerol and [6]-shogaol increased the mechanical withdrawal threshold in healthy rats. In a rat OUM model, OUM-induced mechanical pain was alleviated 30min after the swab application of HST despite the absence of anti-bacterial and anti-inflammatory actions in the OUM area. A swab application of a mixture of [6]-gingerol and [6]-shogaol induced sufficient analgesia of OUM-induced mechanical or spontaneous pain when co-applied with a Ginseng extract containing abundant saponin. The Ginseng extract demonstrated an acceleration of substance permeability into the oral ulcer tissue without an analgesic effect. These findings suggest that Na+ channel blockage by gingerol/shogaol plays an essential role in HST-associated analgesia of OUM-induced pain. This pharmacological mechanism provides scientific evidence supporting the use of this herbal medicine in patients suffering from OUM-induced pain.

The aims of this study were to investigate the involvement of dopamine (DA) in drinking behaviour related to body fluid balance. All experiments were performed in rats. Water intake induced by intracerebroventricular injection of angiotensin II (ANGII) was suppressed by co-injection of DA in a dose-dependent manner. RT-PCR revealed the presence of mRNAs for all known DA receptors, D1-D5, in the subfornical organ (SFO), a brain region that plays a key role in regulating drinking behaviour. Extracellular recordings and whole-cell patch-clamp recordings from SFO neurons showed that DA or the D4 selective agonist PD168077 inhibited spontaneous electrical activity. The D4 antagonist L745870 blocked DA-induced inhibition of spontaneous electrical activity in SFO neurons. Under conditions of synaptic blockade, the inhibitory effects of DA and PD168077 still remained, but the D2/D3 agonist quinpirole and the D1/D5 agonist SKF38393 had almost no effect on electrical activity. While DA induced excitation in a small number of neurons, these excitatory responses almost disappeared following synaptic blockade. All neurons with firing rates that were suppressed by DA were excited by ANGII. In voltage clamp mode, we found that DA and quinpirole, but not SKF38393, suppressed GABAergic miniature inhibitory post-synaptic currents. These results suggest that DA inhibits neuronal activity in ANGII-sensitive SFO neurons primarily through the postsynaptic D4 receptor subtype. This may be a cause of the suppression of ANGII-induced water intake by DA. In addition, the inhibitory DA responses in SFO neurons may be modulated by presynaptic suppression of GABAergic inhibitory inputs through D2/D3 receptor subtypes.

The subfornical organ is an essential central nucleus for angiotensin II-induced body fluid regulation. Similar to angiotensin II, centrally injected neurokinin B (NKB) may induce cardiovascular responses by the subfornical organ; however, it does not induce water intake. To clarify this inconsistency, we investigated the neuronal effects of NKB on subfornical organ neurons in slice preparations along with its behavioral effects in vivo. In electrophysiological extracellular recordings, NKB excited angiotensin II- insensitive and inhibited angiotensin II-sensitive neurons. Centrally injected NKB inhibited peripherally injected angiotensin II-induced water intake. These results suggest that NKB-mediated neuronal effects on the subfornical organ are likely to be involved in antidipsogenic responses in addition to the previously reported cardiovascular responses.

Although central injections of either neurokinin B (NKB) or angiotensin II (ANGII) induce a pressor response, they show different involvements in fluid intake behaviors. The aim of the present study was to elucidate the mechanisms by which these two peptides regulate body fluid balance in rats. We demonstrate that intracerebroventricular injections of NKB (1 nmol) and ANGII (0.1 nmol) both induce pressor responses. However, only ANGII induced significant water intake and increased sodium preference. Co-injection of NKB suppressed the ANGII-induced sodium preference but did not affect the ANGII-induced water intake. Immunohistochemistry for c-Fos, a marker of neuronal activation, revealed that both NKB and ANGII increased neuronal activation in the circumventricular organs and the hypothalamic paraventricular and supraoptic nuclei. In contrast, only ANGII significantly increased c-Fos immunoreactivity in the paraventricular thalamic nucleus, the central amygdala (CeA) and the ventrolateral bed nucleus of the stria terminalis (BSTvl). Co-injection of NKB suppressed the ANGII-induced c-Fos expression in the CeA and BSTvl. These results suggest that centrally injected NKB and ANGII lead to common cardiovascular responses by neuronal pathways through the circumventricular organs and hypothalamus but that they regulate fluid intake behaviors through different pathways. It is likely that the opposing effects of these two peptides on sodium preference can be explained by their differential actions in the CeA and BSTvl, both of which are inhibited by NKB and activated by ANGII.

In the present study, we subclassified acutely dissociated trigeminal ganglion (TRG) cells of rats using a current signature method in whole cell patch-clamp recordings. Using modified criteria for cell classification for the dorsal root ganglion (DRG), TRG cells were subclassified into nine cell types: 1–5, 7–9, and 13. Types 1, 3, and 7 were in the small cell groups (15–24 μm); types 4, 5, and 8–13 were in the medium cell groups (25–38 μm); and type 2 was a mixed group of both cell sizes. Types 1–3, 5, and 7 showed high-input resistance and types 1, 2, and 7 showed more depolarized resting membrane potentials. Types 1, 2, and 5–13 expressed long-duration action potentials (APs), but types 3 and 4 expressed short-duration APs. Sensitivities to capsaicin, protons, and adenosine 5′-triphosphate (ATP) in TRG cell types largely corresponded to DRG cell types. However, different from the matched DRG types, half of TRG type 1 cells were capsaicin insensitive, showing desensitizing proton-induced currents, and types 5, 7, and 9 exhibited slow-desensitizing ATP-induced currents. Types 4, 5, and 8–13 had nicotine sensitivity, but the other cell types were insensitive. These results indicate that the “current signatures” classification is a useful means to separate TRG cells into internally homogeneous subpopulations that were distinct from other cell types. Furthermore, the data suggest some specific differences in the chemical responsiveness of some cell types between the TRG and DRG.

We have recently classified dissociated trigeminal ganglion cells into nine types using electrophysiological current signatures. In the present study, we investigated the relationship between isolectin B4 (IB4) binding and the cell types in rat trigeminal ganglion cells. We found that IB4 was bound to all type 2 cells and more than 70% of cell types 1 and 13; however, it was bound to less than 20% of cell types 7 and 8 and did not bind at all to cell types 3–5 and 9. Thus, each trigeminal ganglion cell type showed high homogeneity in IB4 binding. These results correspond to reported IB4 binding profiles in the matched dorsal root ganglion cell types, except for types 5 and 7.

Saliva is secreted from the salivary glands, and its production is controlled by the autonomic nervous system. Acetylcholine released by the parasympathetic nervous system stimulates muscarinic receptors and triggers the secretion of serous saliva. We found that intraperitoneally injected pilocarpine, which is known as a cholinergic muscarinic receptor agonist and a sialogue, increases not only saliva secretion but also drinking behavior in rats. This indicates that while pilocarpine increases wetness in the oral cavity, it also evokes the sensation of thirst and elicits water intake. Therefore, the thirst sensation can be evoked independently of oral wetness. Further, we found that nicotine, which is a stimulant of cholinergic nicotinic receptor, evokes the thirst sensation. We demonstrated that thirst was induced both by nicotine acting on nicotinic receptors in the brain and by pilocarpine acting on muscarinic receptors in the brain using behavioral, electrophysiological and molecular biological experiments. In this review, we discuss the relationship between oral dryness and thirst, and the different mechanisms underlying these two phenomena. We propose that while the symptom of xerostomia includes both oral dryness and thirst, it is important to understand that oral dryness and the sensation of thirst are intrinsically different.

It is well known that patients with orofacial cancer suffer from cancer-induced pain which produces feeding difficulties. To understand the mechanisms of pain associated with orofacial cancer, we have recently created a model for rat orofacial cancer by inoculation with Walker carcinosarcoma 256B-cells into the vibrissal pads. The present study used both behavioral and immunohistochemical techniques to investigate changes in pain-related and ingestive behavior, along with c-Fos expression in the medullary dorsal horn which is a site for processing orofacial pain. The tumor mass grew gradually and contacted the nerve trunks within days after the inoculation of tumor cells. Physical difficulties in ingestion were observed after day 10 post-inoculation and facial grooming periods were prolonged. Sensitivities of the inoculated vibrissal pads to mechanical and thermal stimuli were increased on days 4 and 7, suggesting the development of mechanical allodynia and thermal hyperalgesia. Although hyposensitivity to mechanical and thermal stimulation was observed in the inoculated region after day 10, hyperalgesia developed on the margin of the tumor, suggesting that the hypersensitive region spread with growth of tumor mass. In the medullary dorsal horn, the levels of c-Fos immunoreactivity of the ipsilateral side increased significantly on days 4, 7 and 10, supporting the behavioral observations. These results indicate that the rat model shows symptoms similar to those in patients with orofacial cancer, for example, induction of feeding disorder and neuropathic pain.

The effects of Ca2+-free perfusion medium on excitatory postsynaptic currents (EPSCs) and potentials (EPSPs) were studied by whole-cell recordings from neurons of the supraoptic nucleus (SON) in trimmed slice preparations of mouse hypothalamus. EPSCs evoked with either focal stimulation to the SON or perfusion of slices with high K+-medium, spontaneous EPSCs (sEPSCs) and miniature EPSCs (mEPSCs) recorded from neurons of the SON were blocked by the glutamate receptor antagonist kynurenic acid (1 mM). While EPSCs evoked by focal stimulation were abolished in the presence of Ca2+-free perfusion medium; sEPSCs and mEPSCs remained. Neither the frequency nor the amplitude of the sEPSCs and mEPSCs significantly changed during the application of Ca2+-free perfusion medium. Perfusion of slices with high K+-medium increased the mEPSC frequency compared with that recorded in normal Ca2+-containing perfusion medium. In contrast, mEPSC frequency did not change during perfusion with Ca2+-free high K+-medium. In current-clamp mode sEPSPs were observed during the perfusion with Ca2+-free medium. Some sEPSPs recorded in Ca2+-free medium were sufficiently large to evoke action potentials. These results imply that spontaneous glutamatergic synaptic inputs to the hypothalamic neurosecretory cells exist in Ca2+-free perfusion medium. Thus, the present study suggests that Ca2+-free medium does not always block the synaptic transmission in hypothalamic slice preparations.

The aim was to evaluate the utility of dynamic magnetic resonance (MR) sialographic images as a diagnostic tool for patients with xerostomia as chief complaint.Various parameters of dynamic MR sialographic images from 40 healthy volunteers were compared with those from 28 patients having dry mouth as chief complaint. Dynamic MR sialographic images were acquired using a 1.5-T full-body MR system and 2-dimensional fast asymmetric spin-echo (2D-FASE) sequences. Acquisition of the optimal section using 2D-FASE sequencing with single-section acquisition of thick sections was repeated every 30 s before and after a few drops of 5% citric acid was placed on the tongue. Student t test was used to examine differences between the healthy volunteer and patient groups regarding the maximum area of salivary gland ducts before and after citric acid stimulation.The maximum area of the detectable ducts in the patient group was significantly smaller (P0.001), whereas the change in detectable duct area was significantly lower (P0.001).This study suggests that dynamic MR sialographic images and data can be useful in the diagnosis of patients with dry mouth as chief complaint.

It is unclear which nicotinic acetylcholine receptor (nAChR) subtypes are involved in the nicotinic activation of cells in the subfornical organ (SFO). We investigated the nAChR subtype using molecular biological, electrophysiological, pharmacological and immunohistochemical techniques. The use of reverse transcription-polymerase chain reaction in rats demonstrated the presence of mRNAs for the alpha2, alpha3, alpha4, alpha6, alpha7, beta2 and beta4 subunits in the SFO. The characteristics and dose-dependency of nicotine-induced inward currents in many dissociated SFO neurons were similar to those induced by acetylcholine in the presence of atropine. The nicotine-induced currents were larger than those induced by cytisine in most responding cells, suggesting the predominance of the beta2- rather than the beta4-containing nAChR. NIC-induced currents were significantly inhibited by dihydro-beta-erythroidine (a relatively selective antagonist for alpha4-containing nAChRs, and a partial antagonist for alpha2 or alpha3) at 300 nM in all responding cells. Additionally, the currents were significantly inhibited by alpha-conotoxin MII at 10 nM (a selective antagonist for alpha3- and/or alpha6-containing nAChRs) in some but not all responding cells. Methyllycaconitine at 10 nM (a selective alpha7-nAChR antagonist) reduced the nicotine-induced current significantly, but to a lesser extent. Fluorescence-labeled alpha-bungarotoxin (a homomeric alpha7 subtype selective binding drug) binding and immunofluorescence for the alpha7 subunit showed that positive images almost overlapped with those immunopositive for an astrocyte marker. These results suggest that the alpha4beta2 subtype is the main functional receptor in SFO neurons while alpha2, alpha3, alpha6, and beta4 subunits have some effect, and homomeric the alpha7 subtype exists in SFO astrocytes.

Galanin inhibits electrical activation of angiotensin II-sensitive neurons in the subfornical organ, which is related to angiotensin II-induced drinking behavior and pressor responses. In this study, the authors investigated whether intracerebroventricular injection of galanin in conscious rats inhibits the responses. Water intake following intracerebroventricular injection of angiotensin II at 100 pmol was inhibited significantly by coinjection of galanin at over 25 pmol, although galanin alone did not affect water intake. Furthermore, angiotensin II-induced pressor responses were inhibited significantly by coinjection of galanin at 50 pmol while galanin alone elicited slight pressor responses. These results suggest that galanin inhibits angiotensin II-induced physiological responses.

Central injection of hypocretins/orexins in rats induces water intake. As the subfornical organ (SFO) plays an important role in drinking behavior, hypocretins may excite SFO neurons. In this study, effects of hypocretins on SFO neurons were investigated electrophysiologically in slice preparations. In extracellular recordings, hypocretin-1 excited SFO neurons, but hypocretin-2 did not or it was little. After the block of synaptic inputs, the excitatory responses to hypocretin-1 remained, but some disappeared. In whole-cell patch-clamp recordings, hypocretin-1 reduced the frequencies of miniature inhibitory presynaptic currents with inward currents occasionally in SFO neurons, but hypocretin-2 did not. These results suggest that hypocretin-1 excites SFO neurons via the activation of hcrtR1 on premembranes and postmembranes.

本学学習環境における学部学生の満足度への新本館の効果を知るため，学部学生を対象にして新本館利用直前の平成19年4月2・3日（平成18年度調査）と利用7から10か月の平成19年11月と平成20年2月（平成19年度調査）に同じ21項目のアンケート調査を行った．平成19年度は平成18年度に比べ，「図書館学習環境」，「図書館学術雑誌」，「図書館視聴覚資料」，「図書館電子ジャーナル」，「コンピュータ演習室」，「ネットワーク」，「パソコン」，「実習室」，「教室施設・設備」の満足度は有意に改善したが，同じ新本館の中に設置された「図書館図書」，「ホームページ」，「自習室」，「講義室」，「ロッカールーム」，「自主学習環境」の満足度は改善せず，大学全体の学習環境満足度にも変化がなかった．また，新本館と関連しないグラウンドの満足度は低下し，体育館満足度は低下傾向であった．学習環境満足度改善は女性で11項目もあったのに比し男性では4項目しか改善しておらず，今回の新本館の新施設設備は男性よりも女性の満足度改善に寄与していた．学習環境とは別に調査した教育関連満足度調査では有意に教育全体の満足度が低下しており，学習環境改善は必ずしも教育満足度改善とは結び付かないことが分かった．教育関連の満足度改善は今後検討すべき重要な問題点と考えられる．また，施設設備が新設したにもかかわらず学生満足度が改善されなかった「自習室」，「講義室」，「ロッカールーム」，「自主学習環境」についてはその原因を解明し満足度上昇のための方策を実践することが必要である．

It is unclear which factors in saliva contribute to its rheological properties such as viscosity and spinnbarkeit. In this study, we investigated relationships between the rheological properties (viscosity and spinnbarkeit) and two salivary mucin (MUC5B and MUC7) levels in saliva. Unstimulated (UWS) and chewing-stimulated (CWS) whole saliva were collected from healthy young adults, and the viscosities and spinnbarkeits were measured. The image densities of MUC5B and MUC7 bands in saliva were measured by Stains-All staining after electrophoresis. Both the viscosity and spinnbarkeit of UWS were significantly higher than those of CWS. The viscosities were correlated positively with the spinnbarkeit in UWS, but not in CWS. Image-density analysis of mucins demonstrated that the viscosities increased with the levels of MUC5B and the spinnbarkeit with the levels of MUC7. These results suggest that MUC5B contributes to viscosity and MUC7 to spinnbarkeit.

九州歯科大学では，大学院の教育・研究改革の一環として，大学院生の教育および研究に関する調査を行った．本調査では九州歯科大学大学院歯学研究科の大学院生2，3，4年生の全員（43名）を対象にマークシート方式のアンケート調査を行い，34名から回答を得ることができた．心身・生活実態に関して解析を行い，学部学生のそれと比較したので報告する．特筆すべき点は以下のとおりであった．1．健康に不安がある，あるいはどちらかといえば不安があると感じている大学院生は23%で，通院している大学院生は16%いた．健康管理室を利用した大学院生は23%で，学部学生に比べると少なかった．2．友人関係は良好と答えた大学院生が多く，不良あるいは非常に不良と答えた大学院生はいなかった．不良あるいは非常に不良と答えた学部学生が各学年に4～9%いたことと比較すると総じて良好であった．3．精神的な悩みがある，あるいはどちらかいえばあると答えた大学院生は44%で，学部学生よりやや多かった．カウンセリングルームを利用した大学院生は6%で，学部学生に比べて少なかった．しかし，本学以外でカウンセリングを受けている大学院生が9%いた．4．自宅から通学している大学院生は27%で学部学生（9～20%）に比べ多かった．1時間以内の通学圏に住んでいる大学院生が94%を占めていた．5．サークル活動を現在していると答えた大学院生は12%で学部学生（43～77%）に比べると極端に少なかった．サークル活動をしていたとしても費やす時間は少なかった．6．アルバイトをしている大学院生が85%と学部学生（39～60%）に比べて多かった．84%が生活費や学費のために当てていた．小遣い稼ぎ感覚でアルバイトをしている大学院生は3%で，学部学生が40～53%いたのに比べると極端に少なかった．アルバイトの時間の長さは学部学生と同じ程度であったが，大学院生の50%は土日・祝祭日などに，40%が平日の昼にアルバイトを行っていた．

Objective Intraperitoneal injection of a sialogogue, pilocarpine, at high concentrations induces salivation via peripheral pathways and thirst sensation via central pathways. In this study, we report that the effects of another sialagogue, cevimeline, on salivation and water intake in conscious rats differ from those of pilocarpine. Design We investigated that effects of peripherally and centrally injected cevimeline on parotid saliva flow rate and water intake in conscious rats. The results were compared with those of pilocarpine. Results The intraperitoneal injection of cevimeline induced salivation from the parotid gland, but not water intake. In contrast, the intracerebroventricular injection of cevimeline induced water intake without salivation. The concentration of cevimeline needed to induce salivation by intraperitoneal injection was several 10 times that of pilocarpine, but that needed to induce water intake by intracerebroventricular injection was over a 1000 times greater. Conclusions The finding that intraperitoneally injected cevimeline induces salivation without inducing water intake, suggests that the effects on the thirst center in the brain are weaker than those of pilocarpine.

The sensation of thirst experienced after heavy alcohol drinking is widely regarded as a consequence of ethanol (EtOH)-induced diuresis, but EtOH in high doses actually induces anti-diuresis. The present study was designed to investigate the introduction mechanism of water and salt intake after heavy alcohol drinking, focusing on action of acetaldehyde, a metabolite of EtOH and a toxic substance, using rats. The aldehyde dehydrogenase (ALDH) inhibitor cyanamide was used to mimic the effect of prolonged acetaldehyde exposure because acetaldehyde is quickly degraded by ALDH. Systemic administration of a high-dose of EtOH at 2.5 g/kg induced water and salt intake with anti-diuresis. Cyanamide enhanced the fluid intake following EtOH and acetaldehyde administration. Systemic administration of acetaldehyde with cyanamide suppressed blood pressure and increased plasma renin activity. Blockade of central angiotensin receptor AT1R suppressed the acetaldehyde-induced fluid intake and c-Fos expression in the circumventricular organs (CVOs), which form part of dipsogenic mechanism in the brain. In addition, central administration of acetaldehyde together with cyanamide selectively induced water but not salt intake without changes in blood pressure. In electrophysiological recordings from slice preparations, acetaldehyde specifically excited angiotensin-sensitive neurons in the CVO. These results suggest that acetaldehyde evokes the thirst sensation following heavy alcohol drinking, by two distinct and previously unsuspected mechanisms, independent of diuresis. First acetaldehyde indirectly activates AT1R in the dipsogenic centers via the peripheral renin-angiotensin system following the depressor response and induces both water and salt intake. Secondly acetaldehyde directly activates neurons in the dipsogenic centers and induces only water intake.

Summary Objective A gender difference in the unstimulated whole saliva flow rate (UWSFR) may be due to a difference in the sizes of the salivary glands. In this study, we investigated the relationships among the UWSFR, gland sizes and body sizes of healthy young adult males and females. Design Unstimulated whole saliva was collected for 5 min by the spitting method in 50 healthy young adults, and the flow rate of the saliva was measured. Heights and weights were measured, and body mass indices (BMI) were calculated. The sizes of the salivary glands were measured by use of a magnetic resonance imaging technique. Results Parotid and submandibular gland sizes and flow rates in females were significantly smaller than those in males, as were also the weights, heights and BMI. In both males and females, there were significant positive correlations between gland sizes and the flow rates, weights and BMI. The variations of the flow rates were reduced by standardizing them with gland sizes, weights and BMI. Conclusions These results suggest that the lower UWSFR in females as compared with males is due to the smaller gland sizes due to the smaller body sizes.

The effects of noradrenaline (NA) and its analogs on subfornical organ (SFO) neurons in rat slice preparations were investigated by using whole cell patch-clamp recording. In the current-clamp mode, the application of NA at 10–100 μM produced membrane depolarization (63%, 17 responsive neurons/27 neurons tested) and hyperpolarization (22%, 6/27 neurons). In the voltage-clamp mode, NA application at 1–100 μM produced inward currents (69%, 42/61 neurons) and outward currents (23%, 14/61 neurons). These currents remained in the presence of TTX or both glutamate and GABA receptor antagonists. In most of the neurons (25/31 neurons) showing inward currents in the presence of NA, the membrane conductance was not changed by voltage ramps or hyperpolarizing pulse stimulation. Similar responses were obtained by the application of the α1-agonist phenylephrine. The phenylephrine-induced inward currents were inhibited by the α1-antagonist prazosin. The α2-agonist clonidine decreased the frequency of spontaneous GABAergic inhibitory postsynaptic currents (4/10 neurons). In addition, RT-PCR assay and immunohistochemical staining showed the existence of α1-adrenoceptors in the SFO. The results suggest that SFO neurons in rats are activated postsynaptically through α1-adrenoceptors and that the activation is enhanced by suppressing GABAergic inhibitory synaptic inputs through presynaptic α2-adrenoceptors.

Summary Objective The aim of this study was to examine the relationships between gland sizes and the flow rate and composition of the unstimulated whole saliva in humans. Design In 28 healthy young adults, the sizes of the three major salivary glands were estimated by use of a magnetic resonance (MR) imaging technique. Unstimulated whole saliva was collected for 5 min by the spitting method, and the flow rate and the concentrations of total protein, Na + and K + and pH were measured. Results The estimated sizes of the parotid and submandibular glands showed a significant positive correlation with the flow rate and the secretion rate of total protein in the unstimulated whole saliva, but that of the sublingual glands did not. Concerning the concentrations of Na + and K + and pH, there were no correlations with the salivary gland sizes. Conclusions The results suggest that the larger the sizes of the parotid and submandibular glands, the faster the fluid flow and protein secretion rates in unstimulated whole saliva.

Aquaporin-2 (AQP2) is responsible for the concentration of urine in the kidney collecting tubule under the regulation of vasopressin. The mRNA level of this water channel in polydipsic STR/N mice was extremely reduced compared with that in normal ICR mice. In male mice, reduction of the AQP2 mRNA level was not evident at 3 wk of age, at which time water intake was not increased. At 10 wk of age, however, the AQP2 mRNA level was reduced to 10% of that in control mice, whereas water intake was increased by 36%. At 44 wk, the water intake became five times that of the control ICR mice, and the AQP2 mRNA level in these polydipsic mice was only ∼5% of control. Similar changes were observed in the AQP2 protein level, suggesting that the mRNA level of AQP2 reflects the protein level of AQP2. These inverse changes in the AQP2 mRNA level and water intake were also evident in female mice. The data imply that polydipsia in STR/N mice may have affected AQP2 mRNA transcription in the kidney, resulting in reduced AQP2 expression, which would contribute to a reduction in overretention of water.

Although angiotensin II inhibits transient outward K+ currents (IAs) in subfornical organ neurones, there is no evidence concerning which Kv channels are involved. We investigated IA-generating Kv channels in dissociated rat subfornical organ neurones, using molecular, electrophysiological and pharmacological techniques, and studied the effects of angiotensin II. Conventional RT-PCR showed the presence of mRNAs for channels of the Kv3.4, Kv1.4 and Kv4 families, which are capable of generating IAs. Tetraethylammonium at 1 mm, which blocks Kv3 channel-derived currents, and blood-depressing substance-I, a Kv3.4-specific blocker, at 2 μm suppressed the IA-like component of whole-cell outward currents in some neurones. 4-Aminopyridine at 5 mm inhibited IAs in the presence of tetraethylammonium at 1 mm. Cd2+ at 300 μm shifted the activation and inactivation curves of the 4-aminopyridine-sensitive and tetraethylammonium-resistant IAs positively. The tetraethylammonium-resistant IAs showed fast and slow components during the process of recovery from inactivation, but the slow component was not seen in all neurones. The time constant of the fast recovery component was less than 200 ms, while that of the slow recovery component was around 1 s. Using single-cell RT-PCR, mRNAs for Kv4.2 and Kv4.3L were detected frequently, but those for Kv1.4 and Kv3.4 were seen only rarely. Angiotensin II at 30 nm inhibited the fast recovery component of tetraethylammonium-resistant IAs in many neurones. These results suggest that the fast recovery component of the tetraethylammonium-resistant IA in subfornical organ neurones depends upon Kv4, and that it can be modulated by angiotensin II.

Opioid effects on synaptic transmission in the mouse supraoptic nucleus (SON) were investigated using whole-cell, patch-clamp techniques. The mu-opioid receptor agonist, [D-Ala(2), N-Me-Phe(4), Gly(5)-ol]-enkephalin (DAMGO) decreased the amplitude of both evoked excitatory postsynaptic currents (eEPSCs) and inhibitory postsynaptic currents (eIPSCs), and also decreased the frequency of both miniature EPSCs and IPSCs without effect on the amplitude. The selective mu-opioid receptor antagonist, D-Phe-Cys-Tyr-D-Trp-Orn-Thr-Pen-Thr-NH(2), and the nonselective antagonist naloxone, antagonized these inhibitory effects. The application of DAMGO suppressed the amplitude of both the first and second evoked postsynaptic currents with a paired-pulse stimulus protocol, but increased the paired-pulse ratios (second ePSC/first ePSC). DAMGO induced neither inward nor outward currents, and had no significant changes in either glutamate- or GABA-induced currents. When compared with the relatively selective kappa- and delta-opioid receptor agonists dynorphin and [D-Pen(2), D-Pen(5)]-enkephalin, DAMGO showed the most potent inhibitory effects on evoked and miniature postsynaptic currents. Taken together, these results imply that DAMGO strongly suppresses the release of glutamate and GABA via mu-opioid receptors in the mouse SON, and support the involvement of presynaptic regulation by opioids in the control of magnocellular neurosecretory neurones.

The sensitivity of subfornical organ (SFO) neurons in rat slice preparations to nicotine was studied using whole-cell patch-clamp recordings and immunohistochemistry. In the current-clamp mode, nicotine at 10 microM depolarized the membrane and increased the firing rate. In the voltage-clamp mode, nicotine elicited inward currents in a dose-dependent manner. The net current-voltage relationships of the nicotine-induced currents displayed inward rectification above -40 mV. The relatively alpha4beta2-selective nicotinic acetylcholine (ACh) receptor antagonist, dihydro-beta-erythroidine, reduced the peak amplitudes of the nicotine-induced inward currents. Immunohistochemical experiments with alpha4 antibody showed the existence of immunopositive cells in the subfornical organ. These results suggest that neurons in the subfornical organ are excited via alpha4beta2 nicotinic acetylcholine receptors.

Cholinergic muscarinic inputs to subfornical organ (SFO) neurones in rats were studied using histochemical, molecular-biological and electrophysiological techniques. Neurones in the medial septum and the diagonal band (MS-DBB) were retrogradely labelled by a tracer wheat germ agglutinin-conjugated horseradish peroxidase-colloidal gold complex injected into the SFO. Some in the MS-DBB were double-labelled by choline acetyltransferase (ChAT) antibody. Many ChAT-immunoreactive fibres were observed in the SFO. M3 muscarinic receptor subtype-like immunoreactivity, detected using a polyclonal antiserum, was observed in the SFO. In slice preparations, muscarine induced inward currents in a dose-related manner. The inward currents were suppressed by the relatively M3 muscarinic receptor selective antagonist 4-diphenylacetoxy-N-methylpiredine methiodide. In the whole-cell current mode, muscarine depolarized the membrane with increased frequency of action potentials. Reverse transcriptase-polymerase chain reaction showed the presence of M2-M5 receptor mRNA in the SFO tissues. These results suggest that the SFO receives cholinergic muscarinic synaptic inputs from the MS-DBB. Acetylcholine postsynaptically activates and depolarizes neurones in the SFO partly through specific muscarinic receptors, including M3 receptor subtypes.

Actions of atropine on GABAergic inhibitory postsynaptic currents to neurons in the subfornical organ, which is a circumventricular organ, were studied by using rat slice preparations with whole-cell clamp recordings. Atropine at 0.01-1 microM antagonized the decreased frequency of Inhibitory postsynaptic currents by carbachol (Xu et al., Am. J. Physiol. Integr. Comp. Physiol. 280, R1657-R1664, 2001). It acted as a muscarinic antagonist at relatively low concentrations. Although the low concentrations of atropine did not change frequency and amplitude of the inhibitory postsynaptic currents, atropine at 10 microM to 1 mM did decrease them in a dose-dependent manner. Glutamatergic excitatory postsynaptic currents were not influenced by atropine at 100 microM. Atropine at 100 microM suppressed GABA- and muscimol-induced outward currents, but not kainic acid-induced inward currents. In addition, decrease of membrane conductance and induction of inward currents by 100 microM of atropine at a holding membrane potential, -51 mV, were found in subfornical organ neurons. From voltage-current curves, a mean reversal potential was estimated to be -65.9 +/- 3.7 mV, near to an equilibrium potential of chloride channels. These imply that atropine at 100 microM suppresses openings of chloride channels. Taken together, it is suggested that, while atropine at low concentrations has an antagonistic action on muscarinic responses, atropine at high concentrations suppresses GABAergic synaptic transmission in subfornical organ neurons. These findings may be of considerable value in understanding the central mechanisms of extraordinary drinking behavior in atropine intoxication.

During dental treatments, intraoral appliances frequently induce traumatic ulcers in the oral mucosa. Such mucosal injury-induced mucositis leads to severe pain, resulting in poor quality of life and decreased cooperation in the therapy. To elucidate mucosal pain mechanisms, we developed a new rat model of intraoral wire-induced mucositis and investigated pain mechanisms using our proprietary assay system for conscious rats. A thick metal wire was installed in the rats between the inferior incisors for one day. In the mucosa of the mandibular labial fornix region, which was touched with a free end of the wire, traumatic ulcer and submucosal abscess were induced on day 1. The ulcer was quickly cured until next day and abscess formation was gradually disappeared until five days. Spontaneous nociceptive behavior was induced on day 1 only, and mechanical allodynia persisted over day 3. Antibiotic pretreatment did not affect pain induction. Spontaneous nociceptive behavior was sensitive to indomethacin (cyclooxygenase inhibitor), ONO-8711 (prostanoid receptor EP1 antagonist), SB-366791, and HC-030031 (TRPV1 and TRPA1 antagonists, respectively). Prostaglandin E2 and 15-deoxyΔ12,14-prostaglandin J2 were upregulated only on day 1. In contrast, mechanical allodynia was sensitive to FSLLRY-NH2 (protease-activated receptor PAR2 antagonist) and RN-1734 (TRPV4 antagonist). Neutrophil elastase, which is known as a biased agonist for PAR2, was upregulated on days 1 to 2. These results suggest that prostanoids and PAR2 activation elicit TRPV1- and TRPA1-mediated spontaneous pain and TRPV4-mediated mechanical allodynia, respectively, independently of bacterial infection, following oral mucosal trauma. The pathophysiological pain mechanism suggests effective analgesic approaches for dental patients suffering from mucosal trauma-induced pain.

The action of angiotensin II on subfornical organ (SFO) neurones was studied using whole-cell current and voltage-clamp recordings in rat slice preparations. In the current-clamp mode, membrane depolarization in response to angiotensin II was accompanied by an increased frequency of action potentials and an increased membrane conductance. In the voltage-clamp mode, angiotensin II elicited inward currents in a dose-dependent manner. The net angiotensin II-induced inward currents were voltage-independent, with a mean reversal potential of -29.8 +/- 6.2 mV. Amplitudes of the angiotensin II-induced inward currents were decreased during perfusion with a low sodium medium. The angiotensin II-induced inward currents were blocked by the AT1 antagonist losartan, and were partially blocked by the AT2 antagonist PD-123319. Neurones which were sensitive to angiotensin II were found in the peripheral region of the SFO, whereas neurones in the central region were less sensitive to angiotensin II. These results suggest that angiotensin II induces inward currents, with opening of nonselective cation channels through mainly AT1 receptors in a subpopulation of SFO neurones of rats.

Inhibitory postsynaptic currents (IPSCs) were recorded from subfornical organ (SFO) neurons in slice preparations of rats, using whole-cell voltage clamp techniques. Some SFO neurons showed bimodal distributions in amplitude with the large and small IPSCs. The large IPSCs vanished in the tetrodotoxin perfusion medium, but the small did not. Both sizes of the IPSCs were completely abolished by application of bicuculline and picrotoxin. Further subpopulation of SFO neurons with the bimodal distributions showed intermittent bursts of the large IPSCs. Immunohistochemical approach revealed existence of gamma-aminobutyric acid (GABA)-immunoreactive neurons and axons in the SFO. These suggest that spontaneously-active and intermittently-burst-firing GABA interneurons affect other SFO neurons in slice preparations of rats.

Cholinergic actions on subfornical organ (SFO) neurons in rat slice preparations were studied by using whole cell voltage- and current-clamp recordings. In the voltage-clamp recordings, carbachol and muscarine decreased the frequency of GABAergic inhibitory postsynaptic currents (IPSCs) in a dose-dependent manner, with no effect on the amplitudes or the time constants of miniature IPSCs. Meanwhile, carbachol did not influence the amplitude of the outward currents induced by GABA. Furthermore, carbachol and muscarine also elicited inward currents in a TTX-containing solution. From the current-voltage relationship, the reversal potential was estimated to be −7.1 mV. These carbachol-induced responses were antagonized by atropine. In the current-clamp recordings, carbachol depolarized the membrane with increased frequency of action potentials. These observations suggest that acetylcholine suppresses GABA release through muscarinic receptors located on the presynaptic terminals. Acetylcholine also directly affects the postsynaptic membrane through muscarinic receptors, by opening nonselective cation channels. A combination of these presynaptic and postsynaptic actions may enhance activation of SFO neurons by acetylcholine.

Changes in salivary secretion with different consistency of diet and dehydration were studied in male Wistar rats under unrestricted conditions. To measure the salivary secretion, a stop-flow method was used. There was little unstimulated salivary secretion from the parotid and submandibular glands, but eating solid, powdered, and liquid diets induced parotid and submandibular saliva. There was no significant change in the volume and flow rate of saliva in bilateral parotid glands during the eating of solid diets. The solid and powdered diets induced significantly more salivary secretion from the parotid gland than did the liquid. The salivary flow rate with solid diets was significantly greater from the parotid gland than from the submandibular. On the other hand, the salivary flow rate with the liquid diet was significantly smaller from the parotid gland than from the submandibular. Appreciable amounts of submandibular saliva, but little parotid saliva were secreted during grooming. Clearly, parotid and submandibular saliva have different roles in the rat. When injected intraperitoneally with 1.5 M NaCl solution or water-deprived for 24 h, rats took longer to eat the solid diets, and had increased salivary volume and decreased flow rate from the parotid gland. These results indicate that the moisture content of the diet and the dryness of the mouth alters the volume of parotid saliva secreted in rats and show that parotid saliva plays an important part in mastication and swallowing.

The subfornical organ (SFO), which is related to drinking and cardiovascular regulation, is activated by central application of nicotine (NIC) and angiotensin II (ANG). However, NIC-induced drinking is much smaller than ANG-induced one although approximately 60% of SFO neurons are affected by both NIC and ANG. Therefore, some specific subpopulations of SFO neurons for NIC or ANG may be related to such different drinking responses. To clarify subpopulations of NIC-sensitive neurons, electrophysiological properties of SFO neurons with the application of NIC was investigated at whole-cell patch-clamp recordings. Based on our developed electrophysiological criteria of the recovery kinetics of tetraethylammonium-resistant transient outward K + currents, two sub-types (F- and S-type neurons) were distinguished. Twenty-nine dissociated SFO neurons were examined to determine whether they showed NIC-induced inward currents. Most F-type neurons ( n = 19/21) showed NIC sensitivity, but most S-type neurons ( n = 7/8) did not. Our previous study had demonstrated that half of the F-type and all of the S-type units showed ANG sensitivity. These suggests that almost all of the NIC-sensitive SFO neurons were electrophysiologically classified as the F-type, but not S-type, and this differs in part from angiotensin sensitivity. The different subpopulations for chemical sensitivities in the SFO may be related to different drinking responses.

Central administration of C-type natriuretic peptide (CNP) affects various neuroendocrine systems. In the present study, we examined whether CNP acts directly on arginine-vasopressin (AVP) secretion from rat supraoptic nucleus (SON) neurons, using acute dissociated cell preparations. CNP inhibited the basal secretion of AVP in a dose-dependent manner (10(-11)-10(-6) M). A- type natriuretic peptide (ANP) and B-type natriuretic peptide (BNP) also suppressed the basal secretion of AVP, however, the effects were two-orders of magnitude less potent than CNP. CNP also suppressed All-induced AVP secretion, however, the inhibitory effect of CNP was less than that of ANP or BNP. These findings suggest that CNP inhibits the basal secretion of AVP through natriuretic peptide receptor (NPR)-B and has a role in the body water and electrolyte homeostasis in the central nervous system.
